Brassica carinata crop production offers an exciting alternative approach to the energy crisis when looking at novel ways to power our motor vehicles. Bacterial endophytes, while residing in host-plant tissues, offer a wide array of plant growth promotion benefits such as nitrogen fixation to impart nitrogen (N) nutrition, phosphate (P) solubilization to make soil-bound phosphate more mobile in soil for plant uptake, can suppress pathogenic microbes such as fungus and produce plant growth hormones to offset stress incurred by the plant. Hesperia dacotae (Skinner, 1911) (Insecta: Lepidoptera), also known as Dakota skipper, is a northern Great Plains species of butterfly associated with tall or mixed grass prairies. Its range once extended from southern Manitoba, southeastern Saskatchewan, North Dakota, South Dakota, Minnesota, to Iowa and Illinois. Now H. dacotae is only found in small isolated pockets in southern Manitoba, southeastern Saskatchewan, North Dakota, South Dakota, and Minnesota. The species is often observed on alkaline prairies with poor soil not desirable for cultivation. Thus, many of the historical H. dacotae are used for pasturing cattle or as hay prairie. Influenza remains a global health risk and challenge. Currently, NA inhibitors are extensively used to treat influenza, but their efficacy is compromised by the emergence of drug resistant variants. Antibodies targeting influenza A virus surface glycoproteins are critical components of influenza therapeutic agents and may provide alternative strategies to the existing countermeasures. This study characterized both polyclonal and monoclonal antibodies produced from the transchromosomic (Tc) cattle platform. A polyclonal antibody, designated SAB-100, was generated from Tc cattle after immunized with H1N1,H3N2, and influenza B virus. Auxin and cytokinin are major plant hormones that orchestrate plant development. These hormones are involved in cell division and differentiation and their spatio‐temporal balance controls organ initiation and maturation. Even though independent transcriptional responses to these hormones have been characterized, their concomitant outputs have not been described. We developed methodology to determine auxin-cytokinin relative output (ACRO) ratios at cellular resolution. Hormone-specific fluorescent markers that localize to the nucleus were used to report transcriptional outputs of auxin and cytokinin. Infectious respiratory disease has long been identified as the cause of bighorn sheep (Ovis canadensis) historical declines and extirpations, and Mycoplasma ovipneumoniae (Movi) is the primary pathogen inducing disease and mortality. Population-level effects of pneumonia events range from mild to extirpation. Variable individual response to pathogen exposure emerges from dynamic interactions between competing evolutionary processes within the host and pathogen. Understanding impacts of this evolutionary warfare is essential to assessing long-term impacts of pathogen invasion and developing appropriate countermeasures to protect population health. The phenomenon of hydrodynamic cavitation involves the formation, growth, and subsequent collapse of bubbles when a given liquid experienced a reduction of pressure below its vapor pressure. The presence of cavitation limits the performance and the safe operation of many machinery and pumps. Cellulose Acetate (CA) based biomaterials are being used as substrates for bone ingrowth applications due to their nontoxic and nonirritant nature coupled with optimum morphology and stiffness. Electrospinning with additives and/or post-treatment has emerged as a viable protocol to further improve mechanical properties of CA and expand its utility. Herein, we highlight the role of potassium chloride (KCl) in association with airdrying to enhance the elastic modulus and tensile strengths of CA fibers. Sulfur mustard is a highly toxic and dangerous vesicant that has been utilized as a chemical warfare agent (CWA) since World War I. Despite its extensive history, an effective antidote to sulfur mustard exposure still does not exist. With detectable levels of unreacted sulfur mustard lasting days after initial contact, a window of opportunity exists to administer a “scavenger” to affected individual immediately and in the days following exposure in hopes of reducing harm by neutralizing unreacted sulfur mustard. In eukaryotes, small RNAs (sRNAs) are key regulators of RNA silencing which is also known as RNA interference (RNAi). RNAi is an essential mechanism known in plants, animals and fungi that regulates various biological activities, including defense against foreign nucleic acids and viruses. Fungal pathogens, such as Sclerotinia sclerotiorum, severely limit crop production in all parts of the world. Macrophages maintain tissue homeostasis by identifying and eliminating threats within their tissue microenvironment. Pattern recognition receptors allow macrophages to recognize and internalize specific ligands while macropinocytosis allows the internalization of all extracellular solutes from their environment. Without pattern recognition receptors, pathogens may grow unchecked if they cannot be detected, and without macropinocytosis, macrophages struggle to detect and move around their environment. This thesis presents the results of CRISPR/Cas9 whole-genome screens used to identify the regulators of both endocytosis (Chapter 2) and macropinocytosis (Chapter 3). Termites in the genus Reticulitermes (Blattodea: Rhinotermitidae) are distributed across the eastern United States, including the southern Appalachian Mountains, a region incredibly rich in biodiversity. The eastern subterranean termite, Reticulitermes flavipes, has been uninentionally introduced to South America and Europe, and is predicted to further expand its geographic range. My goal was to determine how eco-evolutionary processes, operating at both long and short timescales, may have contributed to R. flavipes becoming an invasive species. Ground nesting bird species are on a considerable decline and research efforts are being made to increase these populations throughout the Great Plains. Ground nesting bird communities found in the Northern Great Plains are driven greatly by varying amounts of cover and area size. Past research implemented patch-burn grazing to increase structural heterogeneity and to increase grassland bird habitat in the tallgrass prairie. While bird populations were very responsive to this management in the Northern Great Plains fire management is viewed negatively, especially for Midwest cattle ranchers. Bacterial endophytes have the capability to enhance plant growth by producing plant growth hormones, solubilizing phosphates, suppressing pathogenic fungi, and reducing plant stress hormones. These capabilities make them desirable limiting the amount of nutrients and pesticides that are applied to crops. Through these assays and isolations, it is possible to identify novel bacterial species. In-vitro testing had shown 9 of the 20 isolates possess the ability to produce indole-3-acetic acid (IAA) with Pantoea agglomerans BC09 producing a concentration of 30.2 ng/μl over 4 days. Fusarium head blight (FHB) is a severe disease of wheat (Triticum aestivum L.). It not only reduces the quantity of the harvested grains but also decreases the grain quality due to mycotoxins contamination, especially Deoxynivalenol (DON). Qfhb1 (or simply called Fhb1) is the most important quantitative trait locus (QTL) for FHB resistance. Our lab has previously identified wheat gene WFhb1-1 (aka. Wfhb1_c1) as a candidate for FHB resistance gene. Here we report that WFhb1-1 has been cloned. The high fiber content of corn dried distillers’ grains with solubles (DDGS) can limit its utilization in swine diets. Pretreatment with heat and enzymatic predigestion of whole stillage (WS; slurry material that is dried into DDGS) can alleviate negative effects of dietary fiber and can improve digestive and fermentation characteristics of the feedstuff. Anabaena sp. PCC 7120 is a filamentous, nitrogen-fixing cyanobacterium that uses spatial separation to perform photosynthesis and nitrogen fixation simultaneously. Under fixed nitrogen limiting conditions, Anabaena 7120 forms specialized cells, called heterocysts, to fix nitrogen. In this research, I sought to add to the knowledge surrounding the process of heterocystous nitrogen fixation in Anabaena 7120. Several decades of above average precipitation in South Dakota has increased the area of saline and sodic soils, which reduce crop yields and inhibit sensitive plant growth. Saline and sodic soils are difficult to remediate using traditional agricultural crops. Establishing salt tolerant perennial species may restore productivity to salt affected areas. The goal of the present study was to develop a nanoemulsion (NE) system for efficient and prolonged delivery of Fingolimod Hydrochloride (FTY720; FT), to the posterior segment of the eye through the topical route. FT is a sphingosine-1-phosphate receptor agonist, an immunosuppressive agent. This research is based on field observations in Ridgeville, a Southern city, at an LGBTQ bar, that I call Rainbow Bar and on in-depth interviews with Rainbow Bar drag performers. Through my study at Rainbow, I set out to answer: 1) What do drag kings consider to be “good drag” for themselves and for other kings?; and 2) Who is considered to have the best drag? The overarching goal of this research is to explore the biological mechanisms of aging and formulate novel therapeutic approaches to mitigate age-related impairments. The presence and pervasiveness of ~24 hour (circadian) rhythms in molecular and behavioral dynamics suggests that the mechanisms regulating these rhythms are integral to biological aging and possibly amenable to therapeutic interventions. Based on evidence presented in this dissertation, we conclude that the endocannabinoid system is a mediator of both central and peripheral circadian physiology. Additionally, the hormetic pharmacological properties of the synthetic cannabinoid CP55940 suggest that bidirectional modulation of circadian rhythms via cannabinoids is possible. Chapter …

Natamycin (NT) is a front-line drug in the management of ocular fungal infections (OFI). An ophthalmic marketed NT suspension (Natacyn®) is currently the only FDA approved medication prescribed in the pharmacotherapy of OFI. Current NT pharmacotherapy requires frequent topical administration (every hour or 2-hours over 6-8 times in a day) due to it being instilled as eye-drops. This leads to higher precorneal losses and subsequent poor permeation and bioavailability. Therefore, in this research study, alternative ocular formulations of NT were investigated with an intent to improve precorneal retention and corneal permeation in comparison to Natacyn®. Climatic conditions are particularly important to breeding birds, especially as recent global change has caused a shift in the timing and length of avian breeding seasons. Nest microclimate has been shown to influence avian development and parental care, however, little work has been done to examine whether increased heat poses a cost to altricial nestlings at different stages of their development. We manipulated the temperature of eastern bluebird (Sialia sialis) nest boxes to examine whether or not nestlings exhibit a heat shock response, and a difference in growth or altered parasite loads at both their early and late developmental stages. …

The founding leadership of the Mississippi Private School Association (MPSA) used individual experience and extensive networking via the Citizens’ Council along with the community’s belief and desire to maintain white-only schools to create a coalition of quickly formed but well-resourced private schools. This political and social clout afforded them the ability to create a large, powerful organization almost “instantly” during a pivotal moment of southern educational history. Scholar Kenneth T. The reproductive success of many aquatic insects is highly dependent on where they deposit their eggs. Not all habitats are created equal. Some are more favorable than others for larval development. Therefore it would be evolutionarily advantageous for an ovipositing female to differentiate between them and choose the most suitable for her offspring’s survival. The purpose of this phenomenological study is to describe the transition experience for student veterans enrolled in the three largest universities in Mississippi: Mississippi State University, The University of Mississippi, and The University of Southern Mississippi. Schlossberg’s Transition Theory was used as a lens for viewing the results of this study. This phenological approach will take a social constructivist worldview in order to describe the veteran transition experience. Data was collected using semi structured interviews, transcribed, and analyzed.
