Identification Of Protein Interaction Between The Drosophila Runx1 Transcription Factor Lozenge And Ets-1 Factor Pointed Using Site Directed Mutagenesis And Yeast Two-Hybrid Analysis, Shalini Singh

Electronic Theses and Dissertations

Many classes of transcriptional regulatory proteins are known to function in both cell proliferation and differentiation. Runx1 proteins one such family of transcription factors, plays critical roles in hematopoiesis, osteogenesis and leukemogenesis and act as promoter organizers that cooperate with other transcription factors such as Ets-1 in the regulation of gene activation or repression. Genes that are regulated by the Runx1-Ets1 complex, frequently have multiple, adjacent consensus binding sites in their promoters. I have investigated a similar interaction in developing fly eye. Lozenge (DmRunx1) and Pointed P2 (DmEts-1) cooperate to upregulate expression of prospero, which has multiple Lz and Ets …

The Regulation Of Git1 Transcription In Response To Nutritional Factors, Wei Cheng Jan 2004

The Regulation Of Git1 Transcription In Response To Nutritional Factors, Wei Cheng

Electronic Theses and Dissertations

Glycerophosphoinositol is produced through deacylation of the essential phospholipid phosphatidylinositol. In Saccharomyces cerevisiae, when inositol is limiting in the extracellular environment, the glycerophosphoinositol can be transported into the cell through the permease encoded by GIT1 . In this study, Northern blotting and GIT1 promoter activity were analyzed to assess the nutritional factors responsible for the regulation of GIT1 transcription. Here, I report that GIT1 transcript levels are greater in cells starved for phosphate, with or without inositol limitation, than in cells only limited for inositol. Ino2p and Ino4p are required for full GIT1 expression when the medium lacks both inositol …

Development Of A Biochemical Probe For Arsenate Respiring Bacteria Using Bacillus Selenitireducens Strain Mls10, Mirunalni Thangavelu

Electronic Theses and Dissertations

To date, eighteen phylogenetically diverse prokaryotes have been found to utilize arsenic as the terminal electron acceptor in anaerobic respiration, converting arsenate into the more toxic and mobile arsenite. This process can lead to ground water contamination, making it imperative to detect in situ, active arsenate-respiring prokaryotes.

A biochemical probe that targets the catalytic subunit (ArrA) of the respiratory arsenate reductase (Arr) from Bacillus selenitireducens strain MLS10 was developed. Polyclonal antibodies were raised against a fifteen amino acids long highly conserved sequence at the C-terminus of ArrA. Highly specific antibodies obtained by affinity purification using the synthesized ArrA polypeptide, reacted …

Social Underdevelopment In Sub-Saharan Africa, Michelle L. Wingo Jan 2004

Social Underdevelopment In Sub-Saharan Africa, Michelle L. Wingo

Electronic Theses and Dissertations

For the past thirty years Africa has produced a more noticeably inferior reserve of human capital than other developing regions. This is puzzling because at the inception of independence, the future of Africa looked promising. However, during the 1970s both the political and economic situation in Africa began to deteriorate, and since 1980, the aggregate per capita GDP in sub-Saharan Africa has declined at almost one percent per fiscal year. Thirty-two countries are poorer now than they were twenty years ago, and sub-Saharan Africa is now the lowest-income region in the world despite the fact that during the last two …

The Osteogenic Effects Of 12 Weeks Of Oral Supplementation Of Androstenedione In Middle-Aged Men., Troy Matthew Wills Dec 2003

The Osteogenic Effects Of 12 Weeks Of Oral Supplementation Of Androstenedione In Middle-Aged Men., Troy Matthew Wills

Electronic Theses and Dissertations

Recent evidence suggests that declining bone mineral density (BMD) in males is related to declining circulating estrogens. The majority of endogenous plasma estrogens in males result from peripheral aromatization of plasma androgens. Thus, it was hypothesized that dietary supplementation with an aromatizable androgen (androstenedione) may stimulate increases in BMD.

BMD (measured by dual energy x-ray absorptiometry) and biochemical markers of bone turnover (1, 25 OH2 Vitamin D, calcitonin, deoxypyrodinoline, and parathyroid hormone) were assessed before and after 12 weeks of dietary androstenedione supplementation (200 mg/d). Twenty-four volunteers were randomized into either an androstenedione supplementation or placebo groups. Study volunteers also …

A Markov Model Of Secondary Prevention Of Osteoportic Hip Fractures., Stephanie Tapp Dec 2003

A Markov Model Of Secondary Prevention Of Osteoportic Hip Fractures., Stephanie Tapp

Electronic Theses and Dissertations

Low-trauma fractures of older women are a major public health burden. Fractures of the hip and spine are some of the primary causes of chronic pain, functional impairment, and disability among the elderly [1]. The underlying cause of these fractures is osteoporosis. Although the disease is common, only a small proportion (15%-20%) of women at risk currently receive preventative treatment [2]. We have focused our attention to white postmenopausal women who have sustained a fracture of the distal radius. We developed a Markov model using probabilities obtained from recent literature in order to evaluated the effects of three hypothetical interventions …
