Evaluation Of Pilo Substrate Specificity Using Normally Non-Glycosylated Proteins In Pseudomonas Aeruginosa, Matthew A. Henkel

P. aeruginosa 1244 (PA1244) possesses an O-linked glycosylation system by which the glycosyltransferase, PilO, transfers a single preassembled O-antigen repeating unit to the C-terminal serine residue of pilin as a posttranslational modification. Previous work has shown that the group II pilin of PA103 can be engineered for glycosylation by extending a C-terminal serine away from the surface with a short stretch of alanine residues (Horzempa et al., 2006a). As an extension of that study, several normally non-glycosylated proteins [PA683 pilin, PA1244 PilA-E. coli (EC) alkaline phosphatase (PhoA) fusion, and EC PhoA] were engineered with C-terminal amino acid extensions and assessed …

The United States federal government, through the No Child Left Behind Act, is holding schools accountable for students' proficiency on academic standards. However, standards-based curriculum and instruction, along with school-based support and remediation programs, are not enough to help 100 percent of students to reach proficiency. It is imperative that schools work with parents and communities to maximize each child's potential. Children of involved parents tend to exhibit higher educational goals and perform at academically higher rates than those of parents who do not get involved in their education.

The Use Of Relational, Social, And Overt Aggression In Overtly-Aggressive And Non-Overtly-Aggressive Females, Leigh Ann Comstock

In this study, the researcher sought to examine whether differences existed in the use of relational, social, and overt aggression among previously-identified-overtly-aggressive and non-overtly-aggressive female adolescents. Specifically, explored were the relationships among types of aggression (overtly-aggressive and non-overtly-aggressive), age level (middle or high school), and cultural background/ethnicity (Caucasian or non-Caucasian), and the self-reported use of relational aggression, social aggression, and overt aggression. Results revealed that Caucasian, non-overtly-aggressive middle and high school girls use relational aggression more often than Caucasian, overtly-aggressive middle and high school girls, respectively. Purpose: To determine wheather ot not the nutritional supplement PreRace containing L-Taurine, Citrulline Malate, Quercetin, DiMethyl Amino Ethanol, Caffeine, Metabromine, Catechin, and Malic acid will improve performance in an indoor 40 Kilometer cycling time trial.
Methods: Seven male cyclists ages 21-58 participated in a placebo-controlled investigation which consisted of two trials in which they consumed either the supplement PreRace or aloe vera juice. In each of the trials the placebo or supplement was mixed with a sports beverage. Subjects performed a 40 kilometer time trial and measurements of oxygen consumption and lactate were taken every 15 minutes as well as …

Objectives: 1.) To develop and characterize an optimal formulation of oil-in-water nano-emulsions of trifluoperazine for parenteral delivery. 2.) To perform short term stability testing of the optimal formulation and monitor the potency using high performance chromatography (HPLC).

Materials and Methods: Emulsions containing soybean oil, water, trifluoperazine hcl as an amphiphilic drug, phospholipon 90 and Tween 80 as surfactant blend were prepared using the Thin-layer hydration method. Z-average, polydispersity index, zeta potential of emulsions were determined. A fully randomized 2X2X2X2X2 statistical design was developed using JMP software. Until recently, celiac disease was thought to be rare in the United States. However over the past ten years, the reported prevalence has increased from 1 in 4600 persons to 1 in 133 persons. The latest estimate makes the prevalence comparable to the prevalence in Europe, where the disease is considered to be common.

In this work, hydrophilic fluorene-based derivatives that contain ethylene oxide substituents, have been synthesized and characterized for potential use as new fluorophores for bioimaging applications and for fluorescence sensing of heavy metals. Symmetrical and unsymmetrical fluorene derivatives based on structural types of acceptor-pi-acceptor, acceptor-pi-donor, and donor-pi-donor were characterized by TGA, UV-vis absorption, fluorescence emission, lifetime, anisotropy, and two-photon absorption (2PA) cross section. They were found to possess high thermal stability, high photostability, high fluorescence quantum yields, and generally large two-photon absorption cross sections, making them quite suitable for new probes in single-photon absorption and two-photon absorption fluorescence microscopy imaging. Purpose: Prenatal care (PNC) is the healthcare service most often provided to women of childbearing age throughout the world. Poorly understood and defined, based on culture, and expensive for any healthcare system, PNC remains a target for change and improvement. Selenoproteins are unique proteins in which selenocysteine is inserted into the polypeptide chain by highly specialized translational machinery. They exist within all three kingdoms of life. The functions of these proteins in biology are still being defined. In particular, the importance of selenoproteins in pathogenic microorganisms has received little attention. We first established that a nosocomial pathogen, Clostridium difficile, utilizes a selenoenzyme dependent pathway for energy metabolism. Following this initial characterization, we demonstrate that this pathway is linked to production of toxins by this organism. The purpose of this study was to determine the relationship between second-order change leadership behaviors and the grade assigned to schools in large urban districts by the Florida Department of Education (FLDOE). A total of 101 Title I elementary school principals from large urban school districts with 60+% students on Free and Reduced School Lunch participated in the study. Specifically, this study analyzed 7 of the 21 second-order change factor responsibilities. They include (a) knowledge of curriculum, instruction, assessment, (b) optimizer, (c) intellectual stimulation, (d) change agent, (e) monitoring/evaluating, (f) flexibility, and (g) ideals/beliefs. It can be argued that the current Black-White achievement gap provides evidence for a long-standing history of racial inequity within American society, as well as an informative barometer of progress toward educational parity. By all accounts, the measurements registered by this barometer continue to be cause for alarm. The disturbing Black-White achievement gap has been shown to be present in both mathematics and reading at every grade studied, from grades one through twelve (Jacobson, Olsen, Rice, Sweetland, & Ralph, 2001).

Maternal-fetal conflict refers to a perceived incompatibility between the interests of a pregnant woman and her fetus. Maternal-fetal conflict occurs when a pregnant woman declines a medical treatment or procedure that her physician believes is necessary to benefit her fetus. In some situations, the pregnant woman's health is also at risk. The question explored in this dissertation is: what is the pregnant woman's moral obligation to her fetus when a cesarean section is recommended to save the life of or to prevent serious harm to her term viable fetus? Obesity is one of the major public health issues in the United States, often regarded as part of a global crisis. Companies invest billions of dollars each year towards television advertising campaigns aimed at convincing audiences how their ground-breaking discovery 'battles the bulge' or somehow offers an increased health benefit. This study examined how advertisers presented health-related claims, including health and nutrient-content claims, in U.S. adult-targeted television food commercials. Genetic modification of plastids in the model plant tobacco (Nicotiana tabacum) has demonstrated that numerous foreign gene products can accumulate to high levels in this setting. Plastid biotechnology is maturing to encompass the improvement of food and feed species and the production of biopharmaceutical proteins for oral delivery necessitating development of stable transplastomic edible plants. In the interest of establishing an edible platform we have investigated the use of native and foreign regulatory elements in relation to foreign gene expression in plastids. The purpose of this study is to test the effectiveness of the Dove Evolution film as a one-shot media literacy treatment to change sociocultural attitudes toward appearance. Four speech classes at the University of Central Florida were used in a Solomon four-group design. Group 1 received a posttest; Group 2 received a pretest and a posttest; Group 3 received the treatment and posttest; and Group 4 received the pretest, treatment and posttest. Body image, broadly defined as an individual's general experience of his or her physical appearance, is a multidimensional phenomenon that has been found to affect functioning throughout the lifetime. Although some degree of dissatisfaction has been found to be a common aspect of the female experience, research suggests that a disturbance in body image can result in a number of clinical complications, particularly the development of an eating disorder (ED).
