Brain-derived neurotrophic factor (BDNF) and neurotrophin-4 (NT-4) are two neurotrophins that play distinct roles in geniculate (taste) neuron survival, taste innervation and taste bud formation. These two neurotrophins activate the same receptors tyrosine kinase B (TrkB) and a pan-neurotrophin receptor (p75). While the roles of these neurotrophins have been well studied, it remains unclear how much BDNF and NT-4 function through TrkB and p75 to regulate taste development in vivo. In chapter 2, I compared taste development in TrkB-/- mice and Bdnf-/-/Ntf4-/- mice to determine if these deficits were similar. If so, this would indicate that the functions of both …

With increasingly successful clinical trials, the use of prosthetic devices to replace the function of photoreceptors has now become a viable option to treat degenerative diseases of the retina, including age-related macular degeneration (AMD) and retinitis pigmentosa (RP). Using prosthetics, patients have demonstrated the ability to read large text, recognize small objects, and navigate. While promising, this represents coarse visual function, with the highest visual acuity equivalent to 20/560, still below the legal definition of blindness, 20/200. A population of the rare Southern Appalachian endemic species Lilium grayi, (Gray’s lily) Roan Mountain, TN/NC was monitored for 2 years to determine the cause and impact of an early season collapse. High concentrations of the Lilium spp. host-specific fungal phytopathogen, Pseudocercosporella inconspicua (G. Winter) U. Braun were associated with 19/20 symptomatic and 0/30 asymptomatic plants. Strength of the association between pathogen and disease and the replication of disease symptoms in 4/4 healthy hosts showed that P. inconspicua was the causal agent of the disease referred to as lily leaf spot. Olanzapine (OLZ) is an effective first-line treatment for schizophrenia and bipolar disorder. The benefits of OLZ are countermanded by side effects such as weight gain, glucose intolerance, dyslipidemia, and liver injury. These effects impact not only antipsychotic medication compliance, but also increase the health risks to patients. Most studies to date have focused on potential effects of OLZ on the central nervous system (e.g., hypothalamic regulation of satiety); however, peripheral changes in key metabolic organs such as the liver may also play a critical role. The following dissertation was aimed at characterizing the metabolic disruptions that occur with selenite treatment in human lung cancer. Specifically, the pathways of glycolysis, TCA cycle, nucleotide metabolism, pentose phosphate pathway, hexosamine biosynthetic pathway, and glutaminolysis were examined thoroughly. A stable isotope-resolved metabolomic (SIRM) approach utilizing the tracers 13C6-glucose and 13C5,15N2-Gln was employed in order to trace the flow of atoms through the metabolic network. By utilizing this approach and a combination of NMR and MS analyses, the metabolic pathways perturbed by selenite were reconstructed based on measured isotopologues and isotopomers and known biochemical reaction mechanisms. Safety net clinics across the country struggle with a lack of resources to tackle the needs presented. Screening programs set up for children and elderly have proven to be effective in triaging need, prioritizing care, and maximizing resources. These programs do not currently exist for working uninsured adults. Research was initiated to answer the question: Does the screening process improve patient care for the community clinic? During a 6-week pilot study a licensed dental hygienist performed 30 screenings in the community clinic setting. Findings were recorded and coded according to patient’s level of need identified. The current study examined the mediating role of possible food preferences on the temperamentweight relationship among 18-month-old toddlers. Parents of 37 typically developing toddlers completed the Early Childhood Behavior Questionnaire (ECBQ). During a lab visit toddlers’ weight and recumbent length were measured and recorded. Toddlers also completed a sequential touching task to examine their ability to categorize a healthy group of foods and an unhealthy group of foods. The only temperament measure found to associate with both child weight status and food categorization was inhibitory control. Toddlers’ food categorization was not found to mediate the relationship between inhibitory control and …

The purpose of this modified replication study is to examine the relationship between the stress levels and the wellness practices of U.S. radiologic technology program directors and to determine if correlations exist between wellness practices and stress levels of the group. Additionally, relaxation activities and the feelings, characteristics, and symptoms associated with stress were considered. The design of the study was ex post facto correlational. The population of the study was U.S. radiologic technology program directors working in JRCERT accredited programs. A barrier cited by primary care administrators in integrating behavioral health is financial risk. Fee-for-service billing mechanisms remain complex and there is little empirical guidance on cost-effective models. This study was an economic evaluation of an integrated care model in a pediatric private practice clinic. One of the numerous complications of obesity is infertility. Leptin has been shown to reverse infertility; however, exact mechanism is poorly understood. Recent evidence indicates Ecadherin/ β-catenin complex, which is a structural constituent of adherens junction, is expressed in the rat ovary during folliculogenesis. We hypothesized that systemic leptin modulates the expression of E-cadherin and β-catenin in dietary-induced obese infertile rats to reverse infertility. Female Sprague-Dawley rats were fed either regular chow diet (RCD) (n=6) or high fat diet (HFD) (n=14). It is primarily found in peanuts, berries, grape skin, and red wine. Resveratrol has been found to exhibit anti-cancer, anti-inflammatory, anti-aging, and anti-oxidant properties. Although no reliable concrete data exist on the extent of human trafficking internationally, it is estimated that thousands of women are trafficked into the European Union every year. According to research conducted by the European Roma Rights Centre in 2010, a disproportionately high percentage of these victims are of Roma ethnicity. Host use in herbivores is determined by a variety of ecological drivers, including bottom-up and top-down selective pressures such as host abundance, host plant quality, and parasitism pressure. If the relative importance and strength of interactions among these selective conditions change over an herbivore’s geographic range, local patterns of host use should change in response, evident in differing diet breadths. Hospitalization data on 128 HF patients will be analyzed looking at 14 day, 30 day, and 6 month time points, assessing key aspects of treatment and patient characteristics as potential risk factors in predicting re-hospitalization trends. Cardiovascular Disease (CVD) is the leading cause of death in women and the risk is increased threefold in diabetics. In postmenopausal diabetics, the ovary responds to hyperinsulinemia by increasing secretion of testosterone precursors which increases the risk of CVD. Data from the National Health and Nutrition Examination Survey (1988-2010) and the Study of Osteoporotic Fractures were used to test the hypothesis that a history of bilateral oophorectomy -surgical removal of both ovaries-would decrease the risk of CVD among postmenopausal diabetic women due to reduced androgen levels. P. gingivalis is an opportunistic, anaerobic, Gram-negative oral pathogen strongly implicated in the pathogenesis of periodontal diseases. Clinical isolates of P. gingivalis exhibit great genetic diversity. Tobacco smoking greatly increases the risk of P. gingivalis infection. Furthermore, cigarette smoke extract (CSE) is an environmental stressor for P. gingivalis, differentially regulating multiple genes including ones that encode proteins involved in DNA translocation, replication and repair. Therefore, we hypothesized that CSE exposure would promote genomic instability in this important periodontal and systemic pathogen, which may contribute to increased virulence. This study evaluates the impact of vitamin D plus calcium supplementation as a primary intervention for heart failure (HF) prevention and examines whether preexisting conditions that are precursors of HF modify this relationship in a large cohort of postmenopausal women. Analysis included 35,113 postmenopausal women (17,595 intervention, 17,518 control) aged 50 to 81 years enrolled in the randomized trial of vitamin D plus calcium (CaD) in the Women’s Health Initiative (WHI) study. In this study, we propose a new method for compound identification using penalized linear regression. Compound identification is often achieved by matching the experimental mass spectra to the mass spectra stored in a reference library based on mass spectral similarity. In the context of the linear regression, the response variable is an experimental mass spectrum (i.e., query) and all the compounds in the reference library are the independent variables. Procyonids are well known biologically but knowledge of their fossil record is comparatively sparse. This study seeks to describe a sample of late Blancan procyonid fossils composed mostly of postcranial and dental material. Fossils are analyzed using linear and 2D geometric morphometrics and compared with a large sample of modern specimens from a wide geographic area to understand the range of intraspecific variation. Nutrition education is an integral part of dental education as well as a variety of other healthcare professions1, but interprofessional education (IPE) between the fields of dietetics and dental hygiene is limited. The purpose of this pilot study was to define areas of opportunity to establish a foundation for the implementation of complimentary curriculum between the dietetics and dental hygiene programs at ETSU. Student mobility is an issue for high poverty schools in the shadow of increased rigor and accountability for student performance. Whereas mobility is not a sole cause for poor achievement, it is a contributing factor for students in poverty who are already considered to be at risk of low achievement. Student mobility creates a hardship for schools and districts and hampers attempts to properly monitor the progress of students.
