Electronic Theses and Dissertations
In this historical moment, the United States is amidst an opioid crisis killing the young and the old; at least seventy-eight people die every day from an opioid-related overdose (Enomoto in Murthy III). Changing mindsets of the doctors who prescribe opioids is just as important as asking the patients who are prescribed them to demand an alternative medication. The different parties involved in the crisis all have a different agenda and their rhetorical bias is explored throughout this project. The pharmaceutical companies have launched aggressive marketing campaigns expressing the benefits of opioids and encouraged physicians to prescribe, the CDC has …
An Ethical Framework For Organizational Resource Allocation For Patient Services In Health Care, Natalie Dick
An Ethical Framework For Organizational Resource Allocation For Patient Services In Health Care, Natalie Dick
Electronic Theses and Dissertations
Unanswered questions on how to allocate scarce resources in health care settings are often left to either bedside decision-making or, at best, the organizational policy level. Yet, there is little attention and no consensus on how organizational policy should address resource allocation in health care. An ethically sound framework is needed to guide policy development for resource allocation within health care organizations. In addition, a more comprehensive study of concepts tied to moral obligations of health care organizations is needed in terms of human rights, health disparities, and patient care quality. Until now, human rights literature has largely focused on …

The tongue is an important muscle used in speech, mastication, swallowing, and breastfeeding. It plays an important role in facial development, dentition, and periodontal status. Posterior ankyloglossia, also known as posterior tongue-tie (PTT), is characterized by a tether beneath the lingual mucosa. Upon visual inspection, the frenulum is non-prominent but with the use of a grooved director it may appear abnormally prominent, short, thick, or cord-like. Palpation of the frenulum may reveal tightness. There is a lack of empirical data about PTT and the impact on feeding beyond early infancy. PTT is not well recognized and can be missed resulting …
Malaria In The Prehistoric Caribbean : The Hunt For Hemozoin., Mallory D. Cox
Malaria In The Prehistoric Caribbean : The Hunt For Hemozoin., Mallory D. Cox
Electronic Theses and Dissertations
With the increase in resistance to anti-malarials and global warming trends expanding the habitation range of the mosquito vector, research highlighting the biogeographical contexts of infected populations is critical to understanding epidemiological patterns. A bioarchaeological approach to epidemiology can shed light on previous disease patterns and aid in the prediction of future outbreaks of diseases like malaria. Currently, there is no direct evidence of malaria in the Americas prior to European contact; however, skeletal, archaeological, paleoenvironmental, historic, and ethnohistorical evidence strongly suggest the presence of Plasmodium spp. malaria in indigenous Caribbean skeletal remains held in the Yale Peabody Museum of …

The Effects Of Polychlorinated Biphenyls Exposure On Non-Alcoholic Fatty Liver Disease : Role Of Aryl Hydrocarbon Receptor., Hongxue Shi
Electronic Theses and Dissertations
Polychlorinated biphenyls (PCBs) are detectable in serum of 100% of adults in US, and has been associated with fatty liver disease in epidemiological studies. PCBs are classified as either dioxin-like (DL) or non-dioxin-like (NDL) PCB based on their ability to activate the aryl hydrocarbon receptor (AhR). We used exposures that reflect human bioaccumulation patterns, which resembles Aroclor 1260 with a low level of the DL-PCB, PCB 126. Our aim was to determine if this exposure will activate the human and mouse AhR and examine if receptor activation influences these steatotic responses due to PCB exposures. DL-PCBs exposure-induced AhR activation in …

Inflammasome Independent Leukotriene-B4 Production Drives Crystalline Silica Induced Sterile Inflammation., Bindu Hegde
Electronic Theses and Dissertations
Silicosis is an irreversible lung inflammatory disease caused by chronic exposure to crystalline silica (CS) and is one of the most prevalent occupational diseases worldwide. Lipid chemoattractant Leukotriene B4 (LTB4) plays an important role in neutrophilic inflammation that drives silicosis and promotes lung cancer. Previous studies in our laboratory have demonstrated that CS-induced neutrophilic inflammation and lung tumor burden in K-rasLA1 mice is abrogated in the LTB4-receptor deficient mice. Another pathway whose importance is well studied in the progression of silicosis is the Nalp3 inflammasome pathway. Studies have shown inflammasome- dependent IL-1b to be …
Vinyl Chloride Enhances Diet-Induced Liver Injury Via Metabolic Dyshomeostasis : Critical Role Of Mitochondria., Anna L. Lang
Vinyl Chloride Enhances Diet-Induced Liver Injury Via Metabolic Dyshomeostasis : Critical Role Of Mitochondria., Anna L. Lang
Electronic Theses and Dissertations
Background. Vinyl chloride (VC) is an environmental toxicant and has been shown to be directly hepatotoxic at high exposures. However, recent studies suggest low-level toxicant exposure can cause subtle changes to the liver. Given the high prevalence of non-alcoholic fatty liver disease (NAFLD) in the United States, it is important to determine the impact of low-level toxicant exposure on the progression of underlying liver injury when combined with other factors. Therefore, the overarching goal of this dissertation was to develop a model of VC co-exposure with high-fat diet (HFD) and to determine the mechanisms by which VC contributes to the …
Nutritional Virulence Of Legionella Pneumophila., Ashley M. Best
Nutritional Virulence Of Legionella Pneumophila., Ashley M. Best
Electronic Theses and Dissertations
Legionella pneumophila is an environment organism that parasitizes a wide range of protozoa. Growth within the environmental host primes L. pneumophila for infection of human alveolar macrophages when contaminated aerosols are inhaled. Intracellular replication within either host requires the establishment a replicative niche, known as the Legionella-containing vacuole (LCV). Biogenesis of the LCV depends on the type IVb translocation system, the Dot/Icm, to translocation >320 effectors into the host cytosol. Effectors are responsible for preventing lysosome fusion to the LCV, recruitment of ER-derived vesicles to the LCV, and modulation of a plethora of host processes to promote the intracellular …

Divergent Evolution Of Di-Lysine Er Retention Vs. Farnesylation Motif-Mediated Anchoring Of The Ankb Virulence Effector., John David Perpich
Electronic Theses and Dissertations
Legionella pneumophila is an aquatic organism capable of intracellular replication within a wide range of protozoan hosts and within human macrophages where it causes Legionnaires’ Disease. L. pneumophila manipulates a variety of host cell processes by translocating, into the host cell cytosol, more than 300 effector proteins via the type IVB secretion system (T4SS). The AnkB effector from the AA100/130b strain (AnkB-AA100) contains a C-terminal CaaX motif that rapidly becomes farnesylated upon translocation into the host cell and anchored to the cytosolic face of the Legionella-containing vacuole (LCV) membrane, which is essential for intracellular replication. We show that a …
A New Surgical Approach For O-C2 Fusion, Solving The Problem Of Dysphagia., M. Kathryn Mcclure
A New Surgical Approach For O-C2 Fusion, Solving The Problem Of Dysphagia., M. Kathryn Mcclure
Electronic Theses and Dissertations
The fusion of the second cervical vertebrae to the occipital bone (O-C2 fusion) for head stabilization can result in postoperative dysphagia and dyspnea, negatively impacting the patient’s quality of life. Currently, the O-C2 angle is used for head placement, which may not place the head neutrally. We hypothesize that aligning the external auditory meatus with midline of the C2 will reduce oropharyngeal stenosis, reducing dysphagia. One male patient with poor swallow quality of life who required a revision of a previous O-C2 surgery was evaluated via videofluoroscopy and completed the standard swallow quality of life questionnaire (SWAL-QOL) before and after …

A Model To Study The Effects Of Whole Life Chronic Exposure To Arsenic Or Cadmium On The Development Of Adult Metabolic Syndrome : Initial Characterization Of Hepatic Changes., Jamie L. Young
Electronic Theses and Dissertations
Metabolic syndrome (MetS) is a group of diseases affecting < 30% of adults. Although obesity is a major risk for the development of MetS, it does not account for all cases, suggesting contribution of other risk factors. We hypothesized that early life exposure to arsenic (As) or cadmium (Cd) may represent such a risk. The purpose of this study was to characterize a model to discern the effects of early life exposures to Cd and As on high fat diet (HFD)-induced MetS. Adult C57BL/6J mice were exposed to control or metals containing drinking water. Pregnant dams and offspring were continuously exposed to the same toxicants as their parents. At weaning, offspring were fed LFD or HFD and sacrificed 10 or 24 weeks later. Metal exposure caused time- and sex-dependent alterations in HFD-induced variables of liver damage. The initial results suggest that these toxicants enhanced obesity-induced liver injury.
Comparative Analysis Of Survival And Decay Of Fecal Indicator Bacteria In Bovine Feces And Freshwater Microcosms, Reem Tariq

Agricultural runoff can carry FIB that can pollute surface waters through the soil matrix. This study was designed to inspect the impact of temperature and matrix on the survival of FIB. The FIB were routinely enumerated over an 18-day period from fecal samples and freshwater microcosms maintained at 4oC, 22oC, and 35oC. It was found that the FIB studied underwent a primary growth of up to 1-log10 to 3-log10, highlighting the weakness of conventional FIB as indicators of pathogen contamination. The concentrations of FIB in the water phase were found to be significantly greater than those observed in the fecal …
Escaping The Arrhenius Tyranny: Metabolic Compensation During Exposure To High Temperature In Daphnia, Bret L. Coggins
Escaping The Arrhenius Tyranny: Metabolic Compensation During Exposure To High Temperature In Daphnia, Bret L. Coggins
Electronic Theses and Dissertations
Poikilothermic organisms experience trade-offs by differential physiological demands generated by temperature extremes. Many such organisms exhibit acclimatory effects, adjusting their metabolism and physiology to recently experienced temperatures. One such acclimatory effect is metabolic compensation, the deceleration of biological rates below Arrhenius expectations. Daphnia magna is eurythermal, and if acclimated to mildly stressful temperatures first, survives longer in lethal temperatures. This study examined the effect of ambient temperature (5°C-37°C) and acclimation history (lifetime at 10°C or 25°C) on the oxygen consumption rates of 8 genotypes of Daphnia with high or low acute temperature tolerance. There were decelerations of respiratory rates across …

The Effect Of Ethanol On Skeletal Muscle Endocrine Function And The Novel Myokines Myonectin And Irisin, Kendra Hagood
Electronic Theses and Dissertations
Excessive alcohol consumption is a leading cause of death and disability globally and can lead to diseases such as alcoholic skeletal myopathy. Skeletal muscle is the largest organ in the human body and functions to regulate whole-body energy homeostasis. Additionally, skeletal muscle can function as an endocrine organ via secretion of myokines. Two myokines, myonectin and irisin, present a wide range of effects upon metabolism, inflammation, and tissue survival- signaling. We hypothesized that chronic alcohol consumption will result in reduced circulating myonectin and irisin levels. Mice were fed an ethanol-containing or control diet for 10 days or 6 weeks. Tissues …
