Using An In Vitro-In Vivo Correlation For The ‘Bioequivalence By Design’ Development Of An Immediate Release Carbamazepine Product, Douglas Steinbach

Electronic Theses and Dissertations

The quality of a drug product may be characterized by the consistency with which its indicated clinical effect, and safety profile, is experienced by the patient. The concept that such quality should be built into a product is at the core of the United States Food and Drug Administration’s (FDA) quality by design (QbD) initiative. This vision for pharmaceutical product development emphasizes the risk-based identification of critical quality attributes (CQA) which summarize a product’s performance, the efficient refinement of critical product/process parameters (CPP) that can affect such attributes, and the systematic development of CPP limits, which assure appropriate performance of …

The purpose of this study was to identify an optimal load for the knee extension exercise done with Impulse (Newnan, GA). Forty-one women and twenty-nine men made six laboratory visits, including two familiarization sessions of the knee extension exercise and four real workout sessions which entailed four 30-second exercise sets, separated by 120-second rests, against four different loads (0Kg, 3.41Kg, 5.68Kg, 7.95Kg). A Latin Squares design was used to counterbalance the sequence of the sets. Average force (AF), peak force (PF), total work (TW), Impulse value, and the number of knee extension repetitions were each analyzed with a 2 (gender) …

Exercise Preconditioning And Tfam Overexpression Diminish Skeletal Muscle Atrophy., Nicholas Todd Theilen May 2018

Exercise Preconditioning And Tfam Overexpression Diminish Skeletal Muscle Atrophy., Nicholas Todd Theilen

Electronic Theses and Dissertations

This dissertation is an analysis of skeletal muscle atrophy and a molecular assessment of potential preventative treatments. Chapter I begins with a background of skeletal muscle atrophy along with an analysis of associated molecular pathways. Here, we discuss how skeletal muscle atrophy is the consequence of protein degradation exceeding protein synthesis and can occur when a muscle is abnormally disused. The development of therapies prior to skeletal muscle atrophy settings to diminish protein degradation is scarce and could be useful to prevent negative clinical outcomes of patients who must unload and disuse musculature over extended periods. Mitochondrial dysfunction is associated …

Tumor-Targeted Double Emulsions For Ultrasound-Triggered Delivery Of Molecular Therapeutics., Connor S Centner

Electronic Theses and Dissertations

Cancer is the second leading cause of death in the United States, with 1.74 million new cancer cases diagnosed and 610,000 cancer deaths expected in 2018 alone. Current treatments often result in negative systemic effects and ineffective treatment of the tumor. Drug delivery vehicles have been developed for more effective local delivery methods, but many drug delivery vehicles lack spatial and temporal control. Targeted double emulsions are a new class of drug delivery vehicles which present a promising option for a high payload and controlled delivery. The purpose of our project was to develop and characterize an aptamer-chelated double emulsion …

Fact Vs. Fiction: Uncovering The Experiences Of Homeless/Street Youths' Involvement In Survival Sex, Amber R. Mcdonald

Electronic Theses and Dissertations

Youth involvement in trading/selling sex has become of concern for policy makers, scholars, and the general public in recent years (Cecchet & Thorburn, 2014; Hossain, et al, 2010; Pub L No. 106-386; Reid & Piquero, 2013; Sapiro, Johnson, Postmus, & Simmel, 2016; Shared Hope International, 2015). The behavior of trading/selling sex is defined dichotomously within the literature. One body of literature, employs the term "survival sex" when empirical work focuses on trading/selling sex among lesbian, gay, bisexual, transgender, or queer (LGBTQ) identified individuals or youth that are homeless (Greene, Ennett & Ringwalt,1999; Halcon & Lifson, 2004; Walls & Bell, 2011). …

The Investigation Of Lactoferrin Nitration: Quantification, Function, And Inhibition, Amani Yahya Alhalwani

Electronic Theses and Dissertations

Lactoferrin (LF) is an iron-binding glycoprotein of molecular mass ca. 80 kDa that is predominantly found in mammalian body fluids. Lactoferrin is a multifunctional protein that has a wide range of properties such as antibacterial, antiviral, anti-inflammatory, and anti-allergic functions. Tyrosine residues in the protein play a part in many lactoferrin functions. Protein tyrosine nitration modification represents an oxidative and nitrosative stress process which can be caused by the exposure of proteins to oxidants from air pollution or disease. Understanding the way nitrated lactoferrin interacts with the biochemical environment of the body is thus important to the broader goal of …

Development And Validation Of A Scale To Measure Misconceptions About Educational Psychology Among Pre-Service Teachers, Morgan Mcafee

Electronic Theses and Dissertations

Misconceptions are widespread or commonly held beliefs explicitly contradicted by empirical evidence. When teachers harbor misconceptions or unjustified beliefs about teaching, learning, and human motivation, the potential pedagogical consequences are profound, and these inaccurate beliefs may be instilled into future students through ineffective teaching strategies and gross misinterpretations of learning science. While existing research has examined misconceptions about general psychology and neuroscience among various populations, no prior work has evaluated pre-service teachers' misconceptions about topics of educational psychology, comprising inaccurate beliefs about teaching, learning, and human motivation. The purpose of this research is to describe the development and validation of …

Development Of A Non-Human Primate Model For Staphylococcus Aureus Nasal Carriage, Amanda Lasseter Jan 2018

Development Of A Non-Human Primate Model For Staphylococcus Aureus Nasal Carriage, Amanda Lasseter

Electronic Theses and Dissertations

Staphylococcus aureus nasal carriage (SANC) is largely asymptomatic, but presents a risk of autoinfection and dissemination to new immunocompromised hosts. SA disease states range from mild cutaneous infections to life-threatening bacteremia. Historically utilized rodent models do not naturally carry SA in the nose, are insufficient in longitudinal SANC experimentation, and lack immune factors that are vital in human clearance of SA. The nasal passages of non-human primates are similar anatomically and histologically, and reproductive mucosal studies have shown similar immune responses to pathogens and human-relevant microbial profiles. Seventeen captive pigtailed macaques (Macaca nemestrina) were found to naturally carry SA in …

Psychophysiology Meets Computer Science: Predicting The Magnitude Of Participant Physiological Response With Machine Learning, Avonie Parchment Jan 2018

Psychophysiology Meets Computer Science: Predicting The Magnitude Of Participant Physiological Response With Machine Learning, Avonie Parchment

Electronic Theses and Dissertations

The present inquiry uses methods from psychophysiology and machine learning to reduce overall error in classification models. The field of psychophysiology, though rooted in decades of experimentation, has never reached the same level of precision as some aspects of medical inquiry. In fact, while some medical regression models, when determining some way to classify a patient's illness based on certain symptoms, can result in highly significant results with large effect sizes, equal levels are virtually unheard of in psychophysiology. The present investigation attempts to unravel some part of this mystery and determines some possible reasons for the difficulty in finding …

"Touching the Unreal" follows the structure set out by Scott McCloud in Understanding Comics to argue that understanding cartoons is serious business and requires that we define the art form, outline its basic tenets, and theorize how the mind understands it. The dissertation argues for a new definition of 3D computer generated cartoons, beginning with the most basic definition applicable to all forms of animation and taking into account new technological developments before arriving at the 3D cartoon narratives of today. The dissertation outlines the basic facets of 3D cartoon narratives in terms of narrative and aesthetics, arguing that, in …

Regulation Of Extra-Pituitary Prolactin In Monocytes And Macrophages, Richard Barrett Jan 2018

Regulation Of Extra-Pituitary Prolactin In Monocytes And Macrophages, Richard Barrett

Electronic Theses and Dissertations

Recently it has been shown that leukocytes are capable of producing prolactin (PRL). Evidence of extra-pituitary PRL (ePRL) production is so far been limited to primates and is not shared across other mammal species such as mice and rats. While ePRL is characterized as an identical protein to traditional pituitary PRL, it is controlled under an alternative promoter and is thus regulated differently from pituitary PRL. Little is known about what regulates ePRL or its direct role in human physiology, but given that PRL has well over 300 described functions, it is likely that the autocrine and paracrine effects of …

Characterization Of Neural Cells Derived From Reelin-Deficient Schizophrenic Patient Ips Cells, Nicole Roberts Jan 2018

Characterization Of Neural Cells Derived From Reelin-Deficient Schizophrenic Patient Ips Cells, Nicole Roberts

Electronic Theses and Dissertations

Reelin is a large, extracellular glycoprotein that binds to several membrane receptors on neural stem cells (HNSCs), neural progenitor cells (NPCs), and neuroblasts of mammals to direct their migration. Previously, our lab established the presence of Reelin increased migration of wild-type fetal-derived HNSC's, both in vitro and in vivo. In addition, we demonstrated that Reelin protein treatment also increases the formation of radial glia via Notch-1 signaling, in vitro. Radial glia are precursors to NPCs, as well as a scaffold for neuroblast migration during cortical lamination. Reelin has long been associated with Schizophrenia (SZ). Because post-mortem brains are limited to …

Metabolic Regulation Of Myocardial Adaptation To Exercise., Andrew Alan Gibb Dec 2017

Metabolic Regulation Of Myocardial Adaptation To Exercise., Andrew Alan Gibb

Electronic Theses and Dissertations

While the benefits of exercise affect several organs, a significant adaptive response occurs within the heart. Exercise promotes cardiac growth, increases angiogenesis, and enhances cardiac function and these adaptations are associated with a cardioprotective phenotype. Additionally, extensive evidence shows that exercise dynamically regulates myocardial metabolism. This can be attributed to both changes in hormonal stimulation, increases in bioenergetic demand, and the bioavailability of circulating substrates. However, it is unclear whether these changes in metabolism contribute to physiologic cardiac growth. We reason that exercise-induced changes in metabolism are required to balance the catabolic and anabolic reactions needed for growth. Specifically, we …

Characterization Of Filifactor Alocis And Its Immune Evasion Strategies Employed Against Human Neutrophils., Cortney Linn Armstrong Aug 2017

Characterization Of Filifactor Alocis And Its Immune Evasion Strategies Employed Against Human Neutrophils., Cortney Linn Armstrong

Electronic Theses and Dissertations

Periodontal disease is among the most common of inflammatory conditions and is caused by bacterial and host derived factors. The presence of bacteria drives the recruitment of neutrophils, professional phagocytes, to migrate to specific oral sites where they produce potent antimicrobials to kill their target. However, this inflammation and production of antimicrobials must be strictly regulated to minimize collateral host tissue damage. Human neutrophils recognized the oral pathogen Filifactor alocis through Toll-like receptor (TLR) 2 and upon binding, activated both p38 MAPK and ERK signaling pathways, known to be involved in neutrophil cell migration and degranulation. F. alocis also stimulated …

How Can The Use Of Human Enhancement (He) Technologies In The Military Be Ethically Assessed?, Philip Andrew Taraska

Electronic Theses and Dissertations

War is a terrible price to pay for the prospect of peace. Yet every nation has a moral obligation to protect its citizenry from unjust aggression and threats to security. To be sure, war is always a failure. It is a failure of mankind to come together in mutual respect for the inherent dignity of the human person. The issue of the use of HE in the military is relevant today because the Global War on Terror (GWOT) and the rapid rise of emerging technologies have led to a never-before-seen type of asymmetrical warfare. The rise of these technologies can …

Distress, Circadian Rhythms, Immunity, And Survival In Presurgical Breast Cancer Patients., Whitney Nicole Rebholz Dec 2016

Distress, Circadian Rhythms, Immunity, And Survival In Presurgical Breast Cancer Patients., Whitney Nicole Rebholz

Electronic Theses and Dissertations

Women with breast cancer experience significant stress related to diagnosis and treatment, which may negatively affect physiology (e.g., circadian, endocrine, and immune) and contribute to cancer progression. Demographic (e.g., ethnic minority) and neighborhood factors can contribute to cancer-related stress. African American women with breast cancer experience more adverse disease pathology and earlier mortality; however, factors that explain these disparities rarely have been explored. It is hypothesized that psychosocial stressors (i.e., ethnicity, neighborhood stress, and cancerrelated stress) will be related to one another. Associations of psychosocial stress with physiological disruption are posited with regard to circadian, endocrine, and immune function. It …

Patterns In Dental Health: Investigating Dental Disease At The Archaeological Site Of Kuelap In Chachapoyas, Peru, Vu Tran

Electronic Theses and Dissertations

Social organization influences individual well-being and overall community health, which may lead to health disparities that manifest in teeth. The research in this thesis explores social variability by analyzing patterns in dental disease at the archaeological site of Kuelap in Chachapoyas, Peru. The ancient Chachapoya (AD 900-1535) were a complex society but the nature of that complexity is not well understood based on traditional archaeological correlates. Since burials at Kuelap lack grave goods that are often used to discriminate variation in social status or identity, bioarchaeological dental proxies were tested. The dental remains of 106 individuals (7 subadults, 54 adult …

Developing Methods For The Estimation Of Stature And Their Use As A Proxy For Health Among The Ancient Chachapoya Of Peru, Armando Anzellini

Electronic Theses and Dissertations

Population mean stature and patterns of health are often linked in anthropological studies, yet few studies control for the multifactorial nature of achieving adult standing height. This thesis explores the intersection of health and stature by analyzing the skeletal remains of 161 adult individuals from the archaeological site of Kuelap, in the eastern slopes of the northern Peruvian Andes, and also tests current biometric methods for estimating stature from skeletal remains. This Chachapoya site dates to the Late Intermediate Period (AD 900 – 1470) and Late Horizon (AD 1470 – 1536) and resides in the high altitude sub-tropical forests of …

Cognitive Flexibility: Using Mental Simulation To Improve Script Adaptation, Javier Rivera Jan 2016

Cognitive Flexibility: Using Mental Simulation To Improve Script Adaptation, Javier Rivera

Electronic Theses and Dissertations

Human behavior and decision-making depend largely on past experiences that generate specific action patterns (i.e., scripts, Gioia & Manz, 1985) for specific situations. In an ideal world, in which changes in the environment do not conflict with these action patterns, humans would be able to operate consistently, efficiently, and automatically. However, real-world environments are dynamic and fluid, thus altering behavior and forcing changes in scripts. Research suggests that to implement alternate solutions to changing situations, humans select from a "library" of learned scripts. Since humans tend to implement scripts to the degree that these are successful over a period of …

Differential Expression Of Proteins Involved In Vldl Trafficking Causes Reduced Vldl Secretion In Male Ames Dwarf Mice, Faisal Ahmed Moinuddin

Electronic Theses and Dissertations

Cardiovascular diseases (CVDs) have been recorded as the number one cause of death worldwide, accounting for 32% of total deaths annually. More than two-thirds of all CVD cases are associated with atherosclerosis, which is the accumulation of fats and other substances causing plaque formation in the interior walls of major arteries. This leads to narrowing of the lumen and hardening of the arteries, ultimately resulting in angina, heart attack and/or stroke. Studies have shown that the pathogenesis of atherosclerosis and associated CVDs is strongly linked to elevated secretion of liver-specific lipoproteins called very-low-density-lipoprotein (VLDL). VLDLs are crucial lipoproteins responsible for …
