Background: In the last several decades, the use of coal has become more prevalent in turn increasing the amount of coal ash being produced. Coal ash, the by-product of coal combustion, is composed of small particles that contain essential elements, hazardous metals, polycyclic aromatic hydrocarbons, and radioactive material. While a small proportion of coal ash is reused, the majority gets discarded in open-air landfills and ash ponds. Fly ash, the major component of coal ash, can become emitted into the air and potentially contribute to the air pollution and metal exposure in the surrounding community. Few studies, particularly in the …

Function Of The Enterobactin Operon Of A. Actinomycetemcomitans In The Presence Of Catecholmines And Iron., Taylor Johnson May 2017

Function Of The Enterobactin Operon Of A. Actinomycetemcomitans In The Presence Of Catecholmines And Iron., Taylor Johnson

Electronic Theses and Dissertations

Background: Aggregatibacter actinomycetemcomitans (Aa) possesses a two-component system, QseBC, which is key in biofilm formation/virulence and is activated in the presence of iron-catecholamine complexes. Aa does not synthesize enterobactin, a catechol-based siderophore, yet possesses the machinery for recognition and uptake. We hypothesize that Aa is able to acquire iron by catecholamine mediation through the enterobactin receptor/transporter. Methods: By insertional mutation, we attempted to delete the enterobactin permease from the genome using suicide vector pJT1. A growth curve of truncated mutant ΔfepA was also conducted to observe growth in the presence of iron and catecholamines. Statistical significance was determined by ANOVA …

Inhibition Of Chemotherapeutic-Induced Apoptosis In Esophageal Squamous Cancer Cells By The Keystone Periodontal Pathogen, Porphyromonas Gingivalis., Atul Kumar Agrawal

Electronic Theses and Dissertations

Recent evidence has shown that P. gingivalis, a Gram-negative, anaerobic oral bacterium, is negatively associated with the presence and outcome of esophageal squamous cell carcinoma (ESCC). While potential underlying mechanisms are yet to be established, P. gingivalis infection is known to inhibit apoptosis in gingival epithelial cells. Therefore, we hypothesized that P. gingivalis may also inhibit of the induction of apoptosis in human ESCC cells exposed to the commonly employed anti-ESCC chemotherapeutic agent, cisplatin. The capacity of P. gingivalis, at variant multiplicities of infection, to suppress cisplatin-induced necrosis and apoptosis in EC9706 ESCC cells was established by lactate …

Combating Malignant Melanoma With The Multifaceted Soy-Dervied Peptide Lunasin., Christopher Paul Shidal

Electronic Theses and Dissertations

Lunasin is a 44 amino acid peptide that has been shown to have cancer chemopreventative and chemotherapeutic properties. This study investigated the potential utility of Lunasin as a chemotherapeutic in melanomas. Studies showed that Lunasin had little activity against established melanoma cell lines using adherent culture methods; however, Lunasin’s in vitro activity was significantly higher in non-adherent colony-forming assays and oncosphere formation. These results led to the investigation of whether or not Lunasin has selective effects on cancer initiating cells (CIC) that are known to be present in melanomas. It was revealed that Lunasin selectively inhibited the proliferation of high-ALDH …

Development Of Recyclable Gold Nanoparticles For Routine Benchtop Catalysis In Organic Transformations., Shengzong Liang

Electronic Theses and Dissertations

Homogeneous gold catalysis is a popular research area due to its high efficiency. Various gold catalysts have been utilized in organic synthesis and their reaction mechanisms are also well understood. However, most gold catalysis suffers from high catalyst loading or catalyst decay problems. In addition, homogeneous gold catalysts are not recyclable. These disadvantages have limited their application in large-scale synthesis and also caused a serious waste of this precious metal. A solution for these problems is to use a recyclable heterogeneous gold catalyst. So in this study, with the aim of making gold chemistry greener we successfully used heterogeneous gold …

The Interaction Of Homoserine Lactones And Paraoxonase 2 Modulates Cell Death Signaling And Cell Proliferation., Aaron Mackallan Neely May 2017

The Interaction Of Homoserine Lactones And Paraoxonase 2 Modulates Cell Death Signaling And Cell Proliferation., Aaron Mackallan Neely

Electronic Theses and Dissertations

Pseudomonas aeruginosa produces N-(3-oxododecanoyl)-homoserine lactone (C12) as a quorum-sensing molecule that functions to facilitate bacteria-bacteria communication. C12 has also been reported to affect many aspects of human host cell physiology, including evoking cell death in various types of cells. However, the signaling pathway(s) leading to C12-triggerred cell death remains unclear. To clarify cell death signaling induced by C12, we examined mouse embryonic fibroblasts (MEFs) deficient in one or more caspases. Our data indicate that, unlike most apoptotic inducers, C12 evokes a novel form of apoptosis in cells, probably through the direct induction of mitochondrial membrane permeabilization. Importantly, C12 induces apoptosis …

Attachment To God, Locus Of Control, And Outcomes In The Brief Alcohol Screening And Intervention For College Students, Misty Schoephoerster

Electronic Theses and Dissertations

Research shows that aspects of religion and spirituality can influence drinking behavior among university students. Some believe that an internalized commitment to God, or to another spiritual force, is an important component in explaining relationships between religion, spirituality, and drinking behavior. One way of understanding and measuring this internalized commitment to God is based on attachment theory. Another way of measuring and understanding internalized beliefs about the degree to which God shapes behavior is based on the social learning theory concept called locus of control. Conceptually, low God locus of control indicates that one believes God is passive and not …

A Mixed Methods Examination Of Pregnancy Attitudes And Hiv Risk Behaviors Among Homeless Youth: The Role Of Social Network Norms And Social Support, Stephanie J. Begun

Electronic Theses and Dissertations

Homeless young women become pregnant at exceptionally high rates, and such pregnancies often pose serious emotional, social, and physical health concerns. Perhaps surprisingly, many homeless youth intentionally seek to become pregnant or involved in pregnancy, as pregnancy and parenthood are viewed as conduits toward accessing social services and meaningful social connections to others that this group often lacks. However, most prevention efforts focus solely on young females' pregnancy attitudes and behaviors at the individual level. Such approaches fail to acknowledge contextual factors, such as desired pregnancy and pregnancy ambivalence, the influence of youths' social networks and perceived social norms regarding …

Points Of Leverage: Interrupting The Intergenerational Transmission Of Adversity, Lisa Schlueter Jan 2017

Points Of Leverage: Interrupting The Intergenerational Transmission Of Adversity, Lisa Schlueter

Electronic Theses and Dissertations

Early life stressors, such as abuse and neglect, have been associated with poor physical and mental health outcomes in adulthood. Moreover, animal models suggest that caregivers' early life stress can have intergenerational effects that then impact the health and well-being of their offspring. Although animal models are compelling, and inter-generationally transmitted and co-occurring risks are well-documented, proximal mechanistic explanations for how caregiver's history of childhood adversity can result in changes to their child's stress physiology and outcomes have not yet been systematically tested in humans. Thus, among a sample of low-income, predominantly Latino families participating in Early Head Start (EHS), …
