Alcohol-associated liver disease (ALD) is a broad spectrum of diseases ranging from steatosis to severe hepatic cirrhosis, none of which have any FDA approved therapies. Downregulation of hepatic Sirtuin 1 (Sirt1) activity by ethanol has been shown to lead to an upregulation of the pro-lipogenic gene targets under its jurisdiction, resulting in increased lipogenesis and triglyceride accumulation. It was hypothesized that ethanol acts on α4*-nicotinic acetylcholine receptors (α4*-nAChRs) in hepatocytes to decrease Sirt1 expression, which ultimately dysregulates lipid metabolic homeostasis. In recent years, research into air pollution has shown that exposure to certain components in air pollution, primarily PM2.5 can accelerate biological aging and thereby lead to increased susceptibility to multiple diseases. We hypothesize that prolonged exposure to air pollutants can result in premature aging leading to extensive tissue dysfunction and susceptibility to diseases. To examine this, we exposed mice to PM2.5 for 9, 15, and 21 days, then measured the telomere lengths, cellular senescence, and histone methylation patterns of multiple cell types. We reported pulmonary artery hypertension (PAH) patients had elevated levels of Cd in both blood and urine, therefore, this study tested whether Cd directly induces PAH or facilitates PAH pathogenesis in mouse models. C57/6J mice were initially exposed to drinking water with or without 5ppm Cd for 8 weeks. Then, half the mice in both control and Cd groups were given SU5416 and hypoxia (SuHx) for 4 weeks to induce PAH, resulting in 4 subgroups: Control, Cd, PAH, and Cd+PAH. Porphyromonas gingivalis is a keystone oral pathogen in the progression of periodontal disease and has been implicated in the etiology of many other systemic human diseases. P. gingivalis produces primary virulence factors termed the gingipains, a group of cysteine proteases. There is a high demand for high purity gingipains for downstream in vitro studies where P.g. is implicated, both oral and systemic. Zinc deficiency is associated with metabolic dysfunction-associated steatotic liver disease (MASLD). Previous studies show zinc supplementation improves steatosis, but its therapeutic effects in established MASLD remain unclear. We developed a model to characterize the effects of zinc supplementation on high-fat diet (HFD) induced MASLD and hypothesized established MASLD would be attenuated. Mice were fed control diet or HFD for 12 weeks and then grouped into normal or zinc-supplemented diets for 8 additional weeks. Steatotic liver disease is brought upon by differing etiologies and its global prevalence is expected to keep increasing over the next several decades. A protection zone model represents a patchy environment with positive growth over the protection zone and strong Allee effect growth outside the protection zone. Generally, these models are considered through the corresponding eigenvalue problem, but that has certain limitations. In this thesis, a general protection zone model is considered. This research focuses on crucial aspects of the development process, including fabrication techniques, miniaturization, and integration for a more efficient collaborative human-robot interface. The fabrication process of robot skin sensors, designed to mimic human skin, is explored both within and outside cleanroom environments. We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. Big Bone Lick (BBL) is a late Pleistocene and Holocene fossil locality in the Ohio River valley of Kentucky. This study utilized stable isotopes (δ13C, δ18O, and 87Sr/86Sr) in tooth enamel to reconstruct mobility and diet in Bison, as well as diet in Bootherium bombifrons. Isotopic results from seven Bison and two B. bombifrons are reported. Results suggest Pleistocene Bison and Bootherium occupied different dietary niches. Although both had C3-dominated diets, Bison occupied more open environments than B. bombifrons. Two bison from the late Holocene deposit had diets that consisted …

This study examines how professional storytellers negotiated a new storytelling stage—the videoconference platform—as they pivoted their careers during mandated shutdowns due to COVID-19. An examination of the literature reveals extremely limited research involving either professional storytellers or live virtual storytelling. After interviewing five professional storytellers, I analyzed their stories through narrative inquiry. Analysis revealed that the storytellers negotiated the limitations and affordances of Zoom and adapted their storytelling to successfully connect with their audiences. Many HIV-positive (HIV+) males on Highly Active Anti-Retroviral Therapy (HAART) experience metabolic abnormalities, including Non-Alcoholic Fatty Liver Disease (NAFLD) and lipodystrophy. The intestinal microbiota and short chain fatty acids (SCFA), participate in bidirectional communication with their host. Dysbiosis in HIV+ males on HAART demonstrate a Prevotella-rich enterotype shaped by multiple factors including, medications, adiposity, diet, intestinal permeability, and lifestyle; our objective was to investigate these factors. 19 HIV+ and 21 HIV- males were enrolled. Theories of microaggressions have been used in the last decade as a framework for studying subtle forms of discrimination against racial minorities and other marginalized groups. However, there is a dearth of research on the scope and types of microaggressions nontheists face. This qualitative study examines microaggressions against nontheists by interviewing 16 religious Christians on their experiences and opinions of the discrimination Christians and nontheists face. The narratives were analyzed for thematic patterns between the rhetoric used and the type of microaggressions employed. The study revealed nine categories of microaggressions. Sepsis is a life-threatening condition characterized by overwhelming inflammation, resulting in organ system damage, leading to a high mortality rate. Care in the clinical setting is supportive, and there are no approved sepsis-specific treatments. In septic mice, activation of the cholinergic anti-inflammatory pathway decreases cytokine secretion by leukocytes and improves survival. The cholinergic anti-inflammatory pathway is a reflex of the parasympathetic nervous system, converging on the α7 nicotinic acetylcholine (α7nAChR) at the surface of macrophages. Plants synthesize and store oil, mostly as triacylglycerols (TAG), in seeds that is transcriptionally controlled by WRINKLED1 (WRI1), an APETALA2 (AP2) transcription factor. Among the four Arabidopsis WRI paralogs, WRI2 is nonfunctional, while the others are expressed in a tissue-specific manner. Additionally, two rate-limiting enzymes, diacylglycerol acyltransferase (DGAT) and phospholipid diacylglycerol acyltransferase (PDAT) catalyze the terminal step in TAG assembly and contribute to oil accumulation. Avocado (Persea americana) mesocarp, a non-seed tissue, accumulates significant amounts of TAG (~70% by dry weight) enriched with heart-healthy oleic acid. The chief argument of this work rests on the idea that culture-blindness—especially White ethnic-cultural blindness—contributed as much as issues of race to the heavily documented social rift between predominantly Chicanx, Eastside punk and the mostly-White, Westside punk rock communities of Los Angeles, 1976-1981. To date, historical blame for the divide has centered on racism, including racist intent. Preterm birth is the number one cause of death in neonates, accounting for 35% of neonatal mortality. The preterm birth rate in the U.S. in 2021 was 10.5%, disproportionally affected by race and ethnicity. Obese women have an increased risk of preterm pregnancy, and if delivered before 37 weeks of gestation, the offspring have higher rates of complications that extend from the neonatal period into life-long metabolic and immune adverse outcomes. Among growing concerns over K–12 students’ computational thinking deficits, the research evaluated the impact of Lego EV3 and VEX IQ platforms. Data was sourced from lesson plans, student work surveys, and teacher interviews and then subjected to thematic analysis using a qualitative approach. This thesis aims to understand how the Christian virtue of detachment, understood as the proper ordering of one’s desires toward God, contributes to epistemic transformation. Yosemite Valley is a transformative landscape that helps to shape climbers’ identities and fosters a unique sense of community, which continually reinforces its status as a renowned and evolving climbing destination. The historical influence of Yosemite Valley on rock climbing began in the 1950s and has since defined itself as a prominent destination for climbers worldwide. Encounter-Oriented Subjectivity In Levinas And Barth: Conditioned Freedom In Resistance To Ethnonationalism, Michael Laminack

This dissertation examines philosophical and political subjectivity, or how an individual knows the world and relates to society, through a comparative analysis of the phenomenologist Emmanuel Levinas and the Christian theologian Karl Barth. Levinas and Barth share a deep structure of “encounter-oriented subjectivity,” meaning subjectivity oriented by encounter with the Other that calls into question the subject’s freedom and, in this same moment, calls the subject to concrete response. Encounter-oriented subjectivity is critical of the “unconditioned freedom” that orients liberal subjectivity and offers an alternative sense of a “passively conditioned” freedom, or a freedom always already called into question by …
