Introduction: Following laparoscopic surgery, there is a need, in many cases, to close trocar sites to prevent hernias. Currently, devices that exist on the market are suture based, but the lack of standardization in the suturing techniques together with the time-consuming nature of the procedure leads to the need for improvement in trocar site closure products. Trocar closure sites do not need to be fully closed on the fascial layer; rather, sufficiently blocking the hole at the abdominal wall can significantly reduce post-operative herniation. Understanding interactions between nanoparticles and biological systems is fundamental for the development of emerging nano-biotechnology applications. In this thesis, I present an investigation of zinc oxide (ZnO) nanoparticles interactions with biomolecules in two separate studies. The first section of my thesis covers tracking and detection of ZnO nanoparticles using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). ZinkicideTM is a bactericidal ZnO nanoparticle which has been developed for agriculture. The characterization of Zinkicide in biological media and in solution has been difficult due to its high dispersibility and ultra-small size. Preovulatory estradiol has been reported to play a crucial role in pregnancy establishment and maintenance, but the mechanism by which estradiol exerts its effects has not been well characterized. Specifically, the interactions between the maternal uterine environment and the developing conceptus can greatly impact pregnancy success or loss. The objective of this dissertation is to determine the effects of preovulatory estradiol exposure on uterine and trophectoderm transcriptomes, and uterine luminal fluid (ULF) protein composition. Recent research has led to a much better understanding of the evolutionary processes that mold and structure variation within and among populations. How populations diverge at the genome-wide level and how polymorphism is maintained within a species, however, remains unclear. We address these questions with two freshwater color morphs, red and black, of the threespine stickleback fish (Gasterosteus aculeatus) from the northwest United States, in which a shift from red to black nuptial coloration occurred in several locations following glacial retreat. This project is a study of the development of early modern racial categories in England - focusing on religion and skin color as primary modes of demarcation interwoven with other prevalent categories of language, ancestry/blood, nationality, and gender - as illuminated in William Shakespeare's The Merchant of Venice and Othello. Religion and skin color, then, are the primary modes of racializing individuals in early modern England and characters in Shakespeare's works. Deficits in Reelin expression and signaling play a pathogenic role in Alzheimer’s disease (AD). Thus, strategies aimed at correcting Reelin deficits may provide a novel therapeutic approach to treating AD. The cysteine-rich, whey protein supplement, Immunocal®, has recently been shown to rescue Reelin expression in a mouse model of Schizophrenia. Brian Massumi's concept of affect offers a model of change that relies on sensory-affective modes of resistance to neoliberal power relations. Influenced by Bergson's concepts of time and space, Massumi develops an account of perception as the capacity to entrain with ontological, affective flows of becoming before they are captured and reduced to quantifiable forms. This requires a radical reconfiguration of the body as a zone of indetermination between the virtual field of unformed potentialities and the realm of determined existence. Atmospheric aerosols are ubiquitous throughout the Earth’s atmosphere and can be important with respect to environmental systems and human health. Pollen particles are a class of primary biological aerosol particles (PBAPs) that cost the United States billions of dollars a year in loss of productivity and healthcare costs due to allergy and respiratory effects. Traditional methods of pollen detection rely on collection and subsequent identification by visual microscopy, yet few measurement stations exist in the United States. This thesis reviews the major theoretical frameworks and their outlook on the government spending, its effectiveness, the implied size of the multiplier and how they differed in empirical studies. This is followed by an estimation of the government multiplier for the major U.S. fiscal policy, namely the American Recovery and Reinvestment Act (ARRA), after the Great Recession of 2007-2009. In the South Bohemian region of the Czech Republic, the landscape is distinguished by a network of long narrow fields bordered by hedgerows clustered in small groups. These unique clusters of hedgerows have been interacting with their environment, effectively mitigating erosion, since they were first established in the High Middle Ages. In this research project I used historical maps to characterize land-use and land-cover (LULC) change relating to hedgerow features in one cadastral territory in the Blanice Watershed. Foraging bees use social information (e.g. presence of absence of other bees) to assess the quality of flowers when choosing a flower to visit. My research tests how bees choose to visit a particular flower once they have been recruited to a flower patch. I tested if neighborhood effects, or the relative number of bees on neighboring flowers compared to a focal flower, affected to which flower a foraging honey bee visited. This thesis investigates the archaeological remnants of an early 14th century settlement at Ballintober, Roscommon County, Ireland. An innovative methodology combining ground-penetrating radar, magnetic gradiometry, and archaeological excavations is utilized to reconstruct the medieval built environment, which was comprised of a masonry castle, nucleated settlement and wider arable agricultural landscape. By integrating the archaeological and historical records, I pose hypotheses related to the differential statuses of people at the settlement, their domestic and agricultural practices, and a timeline of their occupation and abandonment of the site. Embodied storytelling in Denver's public broadcast media establishes how the intersectional identities of storytellers influence narrative practices in Denver's public sphere. Five approaches to communicating identity informed my theoretical background: embodiment, visual anthropology, the public sphere, practice theory, and phenomenology. Rocky Mountain PBS, a 60-year-old broadcast institution, served as my research site during the summer of 2018. In my thesis, I overviewed the history of RMPBS and observations of production activities performed by the creators of the show Colorado Memories. Insulin secretory granules (SGs) mediate the regulated secretion of insulin, which is essential for glucose homeostasis. The basic machinery responsible for this regulated exocytosis consists of specific membrane proteins present both at the plasma membrane and on insulin SGs. The protein composition of insulin SGs thus dictates their release properties, yet the mechanisms controlling insulin SG formation, which determines this molecular composition, remain poorly understood. VPS41, a component of the endolysosomal tethering HOPS complex, was recently identified as a cytosolic factor involved in the formation of neuroendocrine/neuronal granules. Explicitly-rated anti-fat attitudes are correlated with weight-based discrimination, which is rampant in society today as many countries grapple with soaring rates of obesity. Early perceptual processes, such as conscious awareness and visual attention, may be biased based on the weight of the perceived or the perceiver, or any number of individual perceiver characteristics regarding weight-biased attitudes and experiences. The three experiments presented used continuous-flash suppression (CFS) to mask body stimuli, thereby hoping to gain insight into attentional capture of unseen images and its relation to anti-fat attitudes. The visual system often integrates information that "goes together". Once information has been integrated, summary information (e.g., average emotion or average size) can be extracted; this occurs during ensemble coding. Integration thus allows for fast and efficient generalizations about sets to be made. In contrast, the visual system sometimes segments input that does not go together. For example, the perception of objects can be exaggerated away from natural category boundaries (e.g., a perfect circle is a category boundary; it is neither "flat" nor "tall"). Parental experience can alter the developmental and rearing environments of offspring, resulting in parental effects on offspring traits. I addressed the consequences of stress-induced maternal, paternal, and joint parental effects from both ultimate (ecological/evolutionary) and proximate (physiological/epigenetic) perspectives. I used a full-factorial design in which threespine stickleback (Gasterosteus aculeatus) mothers, fathers, both, or neither were exposed to a model predator at developmentally appropriate times to test for predator-induced maternal, paternal, and joint parental effects on daughters’ mating behavior and egg glucocorticoids (stress hormones) and on offspring gene expression. Multicultural competencies (MCCs) have laid the foundation for therapists and researchers to strongly consider culture as an important factor in psychotherapy. More recently, Owen (2013) developed the multicultural orientation framework (MCO) to better explicate the MCCs for psychotherapy. The purpose of this dissertation was to develop a Multicultural Orientation Performance Task (MCO-PT) and coding system with therapists-in-training. For the MCO performance task, participants responded to eight brief simulated therapy situations that centered on the intersection of various cultural identities. The purpose of this project was to determine the ways in which signs within social media posts on Instagram and Facebook frame the way that outdoor recreational tourists make decisions about Colorado state parks' locations, services and activities. Surveys were conducted at 6 Colorado state parks in the 2018 spring, summer, and fall months. These surveys asked respondents to answer multiple choice and open ended questions about their social media habits, their outdoor recreation habits, and their opinions about images displayed on Colorado Parks and Wildlife's social media pages.

This thesis examines social responsibility within a capitalist economy by investigating socially responsible investing and, tangentially, corporate social responsibility. These concepts have been at the heart of economic and legal debates for hundreds of years, with no clearly defined consensus regarding how to account for multi-stakeholder welfare inside the market system. Mycofactocin is a putative ribosomally synthesized and post-translationally modified peptide (RiPP)-derived redox cofactor. Its biosynthesis is accomplished through the dedicated actions of the products of six conserved genes, mftABCDEF. The mycofactocin pathway is one of the most widely distributed RiPP systems in bacteria however, this distribution is heavily skewed towards the Mycobacteria genus including human pathogenic variants such as M. tuberculosis and M. ulcerans.
