This study examines the impact of experiences of colorism on the crime trajectory of African Americans. It is qualitative in nature and utilizes semi-structured interviews with incarcerated African American people. As previous research has shown the painful impact of colorism on its victims, this study ultimately seeks to unearth if that pain translates into crime commission and trajectory. The primary purpose of this research is to contribute to the existing body of research that seeks to support “colorism healing” in the African American community. Conceptualizing, quantifying, and evaluating frailty in human skeletal remains is critical to understanding and interpreting physiological health and stress among past populations. While many researchers focus on frailty in bioarchaeological samples, developing models for mortality risk and individual- and population-specific indices of stress, no current methods directly address frailty in forensic contexts. This study considers the applicability of a modified index for assessing frailty in forensic anthropology by comparing distributions of 8 biomarkers of stress (linear enamel hypoplasia; periodontal disease; caries; osteoarthritis; intervertebral disc disease; rotator cuff disorder; antemortem fracture; and surgical procedure) using the original skeletal frailty index (SFI) …

Given the diversity and complexity within immunoglobulin (IG) loci, effective mouse models first require characterization of intra-strain differences and construction of high-quality reference assemblies for IG loci in several representative strains. To understand light chain germline diversity across biomedically significant mouse strains, we profiled the expressed IGK and IGL repertoires of 18 commonly used laboratory mouse strains using AIRR-seq. Across strains, we observed germline IGKV sequences shared by three different IGK haplotypes and a more conserved IGLV germline repertoire among common laboratory strains. This dissertation explores the relationship between mitochondrial physiology and development of therapeutics. Mitochondrial dysfunction is associated with both acute and chronic forms of pathophysiology. This work aims to address development efforts at the cell culture and drug-target levels with respect to mitochondria. At the cell culture level, I characterize an approach that has been shown to improve the physiological dependency on mitochondria in tumor-derived cells. I demonstrate that prolonged replacement of glucose with galactose in culture medium induces a global metabolic shift in hepatocellular carcinoma (HepG2) cells to closer reflect a primary hepatocyte phenotype (Chapter 2). Few studies address the lived experiences of transgender and gender nonconforming (TGNC) students in engineering. Grounded in critical trans politics (Spade, 2015), this dissertation contributes to the literature on TGNC students in engineering by examining their experiences negotiating their identities while navigating interrelated systems of oppression in a field dominated by White, heterosexual, cisgender men. Using a critical constructivism framework, I conducted a narrative inquiry to explore the lived experiences of five TGNC students in engineering programs. Participants experienced TGNC oppression at their universities, built LGBTQ+ and TGNC communities, and described more welcoming climates in non-engineering contexts compared to engineering. …

This dissertation is an investigation into the effects of high-probability tasks on the task completion rates of three students diagnosed with an emotional and behavioral disorder. It begins with a close look into best practices verses common practices, examines students supports currently available in schools, and highlights gaps present in today’s educational system for students who exhibit significant challenging behaviors. Retinal fibrosis is detrimental to vision. Retinal pigment epithelial (RPE) cells contribute to several retinal fibrotic diseases. Upon exposure to TGF-β, a key fibrotic cytokine, RPE cells trans-differentiate to myofibroblasts marked by the integration of α-SMA fibers into F-actin stress fibers, which confer strong contractility. Myofibroblasts produce and contract the collagen-rich fibrotic scar and disrupt retinal architecture. In this study, we investigated the in vitro effects of the putative endocannabinoid compound N-oleoyl dopamine (OLDA) on TGF-β2 induced porcine RPE cell contraction and α-SMA expression. This MFA thesis defines Contemporary Blackface Theatre as exemplified in Dennis McIntyre’s Split Second, which was produced in Fall 2021 by the Department of Theatre Arts at the University of Louisville. Meridiolestids are a diverse and distinct clade of Cretaceous–Miocene South American dryolestoid mammals ranging in size from small shrew-sized insectivores to large dog- sized omnivores. Cronopio dentiacutus from the Cenomanian La Buitrera locality, Rio Negro Province, Argentina (Candeleros Fm.) is the oldest and best known of them, represented by skull material and associated jaws. A juvenile skull as attributed to Cronopio dentiacutus, based on tooth size and morphology, was collected from the type locality. The specimen includes the skull and both articulated lower jaws. Cell-based immunotherapies are a new generation of “living-drug” treatments for cancer and other diseases. Chimeric antigen receptor (CAR) T-cell cancer therapy has shown promising results in lymphoma and B-cell malignancies. Currently, there are six FDA-approved CAR-T drugs on the market, and all of them use viral transfection for reprogramming. While viral transfection is effective, there are safety concerns due to inconsistent transfection that limit the use of CAR-T therapy. Current non-viral transfection techniques generally have lower transfection efficiency than viral transfection. My goal is to develop chemical processes for transforming waste to solve environmental problems and enhance sustainability. Environmental problems such as pollution and massive amounts of waste are the main drivers that stimulate my research ideas. I focused on creating novel, efficient catalytic processes for converting polymeric waste "feedstocks" into high-value chemicals by integrating my expertise in catalysis, materials science, and synthetic chemistry to develop porous solid catalytic materials. Major depressive disorder (MDD) is a leading cause of disability worldwide, with a lifetime prevalence rate of approximately 20%. Inadequate pharmacological treatment methods for MDD are a significant debilitating factor. Patient estimates suggest that the treatment resistance rate for pharmacological interventions is over 30%. Postmortem analyses of human tissue of individuals diagnosed with MDD have shown an increase in Poly (ADP-ribose) polymerase 1 (PARP-1) mRNA gene expression in prefrontal cortical white matter when compared to psychiatrically normal brain tissue. Nearly all multicellular organisms show changes in redox balance with aging leading to oxidative damage of macromolecules. This study investigated the role of the [NADP+]/[NADPH] redox couple in aging. This redox couple plays an important role in maintaining tissue redox balance and becomes slightly more oxidized in aged tissues. NADPH is a major source of reducing equivalents for enzymes that detoxify hydrogen peroxide. However, catalase detoxifies hydrogen peroxide independently of NADPH. But catalase is absent from mitochondria, a major source of hydrogen peroxide, where instead glutathione plays the major role in hydrogen peroxide detoxification in an NADPH-dependent manner. …

This thesis examines the existence of posthumanism in the dramas of Ben Jonson, Christopher Marlowe, and William Shakespeare – the three most prominent playwrights of the early modern period. Posthumanist theory, which gives scholars the opportunity to look at past works in a new and unique way, attempts to re-locate the human in the diverse creatures and objects in the world we inhabit. Niche partitioning is a major component in understanding community ecology and how ecologically similar species coexist. Temporal and spatial partitioning and differences in foraging strategy, including sensitivity to risk (variance), likely contribute to partitioning as well. Here, we approach this partitioning with fine resolution to investigate differences in overall strategy between two species of diurnal, orb-weaving spiders, Verrucosa arenata and Micrathena gracilis (Araneae: Araneidae), that share similar spatial positioning, temporal foraging window, and prey. Through field observation, we found that V. arenata individuals appear to increase spatial and temporal sampling to compensate for an overall risk-prone strategy that depends on …

Hypocretin/Orexin (HCRT) neurotransmission facilitates drug-seeking behavior. HCRT neurotransmission at HCRT-receptors 1 and 2 (HCRT-R1 and -R2, respectively) is implicated in addiction. During the shift to alcohol-dependency, adaptations in neurotransmitter systems occur in reward- and stress-related brain regions. Specifically, neurotransmission systems in the central amygdala (CeA) are modulated by alcohol drinking/exposure. Therefore, this study investigated Hcrtr1 and Hcrtr2 mRNA expression in the CeA of alcohol-dependent rats and in non-dependent controls during acute alcohol withdrawal. No single volume of mythological or philological research exists for Hades. In the one moment Hades appears in archaic Greek literature, speaking for only ten lines, Hermes stands nearby. Thus, to understand and journey to Hades is to reckon with Hermes’ close presence. As I synthesize research by writers from several different disciplines, may some light be brought into the depths. May we analyze Hades’ brief appearance in archaic Greek literature, examining how what I define as the “Hermetic” emits from his breath in the one moment he physically appears and speaks.

The early Pliocene Gray Fossil Site (GFS) is a biodiverse site with a unique faunal assemblage that represents one of few sites of its age in eastern North America. A community structure analysis of the mammals at GFS was done to characterize species and better understand the paleoenvironment. Data and was gathered from twenty modern communities and five late Neogene sites to compare with GFS. Species from these 26 sites were categorized by body size, locomotor mode, cheek tooth crown height, and diet to characterize niches occupied. The study of human face-processing has granted insight into key adaptions across various social and biological functions. However, there is an overall lack of consistency regarding digital alteration styles of human-face stimuli. In order to investigate this, two independent studies were conducted examining unique effects of image construction and presentation. In the first study, three primary forms of stimuli presentation styles (color, black and white, cutout) were used across iterations of non-thatcherized/thatcherized and non-inverted/inverted presentations. Karst springs are an essential source of private water supply for about 10% of households in Tennessee. However, these springs, which can be easily polluted, are unregulated. This study, therefore, assesses water quality spatial patterns and water quality rating of roadside springs in northeast Tennessee.
Karst spring water samples collected from 50 springs were assessed using EPA Standard methods for pathogens, nutrients, radon, and physicochemical parameters. Springs generally met federal and state standards for physicochemical parameters, 90% of samples contained E. coli, and all samples contained fecal coliform. Previous work found significant differences in growth rates of native mussels at locations downstream from the regulated Walter’s Dam and the out-of-service, free-flowing Davy Crockett Dam. The purpose of this study is to investigate differences within the macroinvertebrate communities related to factors driving the differences in mussel growth between rivers. Macroinvertebrate samples were collected following the Tennessee Department of Environment and Conservation protocol for SQKICK collection and analyzed using the Tennessee Macroinvertebrate Index (TMI). However, research regarding the effects of nature exposure on social health is less developed. Prior research found that watching a video of nature improved state social motivation. The purpose of this study was to investigate the effect of an intervention package that included constant time delay (CTD) and sentence frames on correct word selection for sentences constructed using technology-aided instruction (TAI) during and following a story-based lesson (SBL) for participants ages 6-8 who have intellectual disability, developmental delays, and autism (IDD). A multiple probe across participants design was used to evaluate the efficacy of the intervention. Research on factors that predict opioid cravings is lacking. Anhedonia may be a predictor of cravings and particularly relevant to cravings when people struggle to regulate emotions but has yet to be examined among justice-involved populations. This study aimed to examine the relationship between anhedonia, opioid cravings, and emotion dysregulation (ED) in this population. Participants completed several measures. The results showed that anhedonia and opioid cravings were significantly related at the bivariate level, but not in moderation models. Cromolyn sodium (CS) has clinically shown to be an effective topical remedy for atopic dermatitis, however its physiochemical properties prevent efficient passive drug delivery beyond the outermost skin layer. This project aimed to optimize CS gel formulations and applications to improve drug delivery to the dermis of skin by examining various topical enhancement strategies. Oleic acid, salcaprozate sodium, and microneedles were investigated as enhancers for their effect on skin permeation of CS. The thesis demonstrates that Ezek 40–48 functions as a unified narrative of restoration and that it is profitable to understand its fantastic realism and postcolonial rhetoric as early precursors to the modern magical realism genre. The tools of narrative criticism are utilized to exegete the literary features of Ezek 40–48 and uncover its poetic elements and rhetorical thrust. After this analysis, Ezek 40–48 is viewed as a precursor to modern magical realist texts in a comparative study with Gabriel García Márquez’s novel One Hundred Years of Solitude and Salman Rushdie’s novel Midnight’s Children, paying special attention to the thematic emphases …
