Fidelity Of Dna Polymerase In Vitro, Yannacos C. Philopoulos

The fidelity of DNA Polymerase I, isolated from B. licheniformis and B. stearothermophilus, was tested in the presence of carcinogens and anticancer compounds. All of these compounds inhibited (62.4-99.2%) the synthesis of DNA using Poly(dA-dT) as a template. The compounds were inhibitory at all concentration levels tested, 0.01, 1.0 and 100 mM. The fidelity of the enzyme appeared to be enhanced by the presence of these compounds, as indicated by misincorporation (50-70%), that is, the incorporation of incorrect nucleotide. It was the purpose of this study to examine the relationship between women's perception of self as being competent to occupational role.

Based on a review of the literature and research it was hypothesized that a relationship between occupational role and competency self-perceptions did exist. Also, it was hypothesized that there is a relationship between occupational role and the difference between perception of self and view of ideal self in competency. Finally, it was hypothesized that there is a relationship between occupational role and the discrepancy between perception of self and perception of typical women in competency skills.

Problem. The purpose of this project was to develop an instructional product that would enrich the student's knowledge about dating and marriage, and teach specific relational skills.

Method. A systematic procedure was undertaken in the development and validation of the product. First, the content of a pre-marriage course was identified through an examination of a large sample of family-life texts, a survey of teacher criticisms and recommendations pertaining to the text Marriage (Christensen, 1980), and an analysis of a group of 317 youth using the Search Institute instrument, Youth Research Survey (Strommen, 1977; see appendix E). A Microbial Reductive Conversion Of The Steffimycin Chromophore: Chemical And Mechanistic Studies On A Soluble, Nadhdependent, Nonflavin-Containing Reductase From Streptomyces Steffisburgensis, John Rollin Hofstetter

An understanding of a microbial reduction of the anthracycline antibiotic, steffimycin (1) was approached from two directions. First, the NADH-dependent enzyme responsible for the reduction was purified from the Streptomyces steffisburgensis cytosol and characterized. Second, information was obtained on the reduction and oxidation of 1.

The enzyme was a single polypeptide chain, acidic, and stable in solution. It had a molecular weight of 80,000 and no prosthetic groups.

The 5-step purification process resulted in greater than 1,000-fold purification and a 9% yield of the homogenous enzyme from extracts of S. steffisburgensis cells.

The purpose of the study was to develop and validate a model to retain adolescent child-bearers in the Jamaican school system. More specifically, the study was concerned with retaining pregnant students and adolescent parents in Jamaican secondary schools.

The analytical bases of the model were: (1) A review of the literature in adolescent child-bearing in Jamaica and the United States. (2) A survey of programmes which currently deliver comprehensive services to pregnant students and adolescent parents in Michigan. Five programmes were surveyed. This dissertation covers the separation of proteins via recuperative mode pH parametric pumping. Continuous pH parametric pumping separations using the model system haemoglobin and albumin on a CM-Sepharose cation exchanger have been experimentally and theoretically investigated.

The pH parametric pumping system has a feed containing the protein mixture to be separated, introduced alternately to the top and bottom of the column. The top and bottom products are withdrawn from the apparatus during the bottom and top feed, respectively. Both equilibrium and nonequilibrium cases were considered, and theoretical results agreed with the experimental data.

The purpose of this research study was to determine the similarities and disparities among the expected learning outcomes of three sets of performance objectives for science education: (1) the teacher-written science objectives of the Goals-Objectives-Test (GOT) Program for the seventh, eight, and ninth grades in the Kalamazoo Public School System; (2) the objectives prepared by the Department of Research and Development of the Kalamazoo Public School System from test items on Test 8: Science of the Metropolitan Achievement Tests (MAT) - Advanced Forms; and (3) the science objectives of the Michigan Department of Education (MDOE) for grades 7-9.

The breeding bird populations of a 40.9 acre grassland-thicket, the Colony Farm Tract, Kalamazoo, Michigan, were studied over 18 years, from 1961-1979. The purpose of the study was to discover any variations in the avian populations that resulted from the construction of an adjacent residential development. Particular attention was directed toward establishing the kinds of relationships that existed between the breeding bird populations and the adjacent residential development.

The purpose of this study was to measure the concentration of lead, cadmium, zinc and copper in runoff entering an urban lake ecosystem and to measure the distribution of these metals in selected tropic levels of the lake.
