Mechanisms Of Condition-Specific Regulation Of Mrna Stability By Puf Proteins: From Yeast To Humans, Joseph Russo Dec 2014

Mechanisms Of Condition-Specific Regulation Of Mrna Stability By Puf Proteins: From Yeast To Humans, Joseph Russo

Dissertations

RNA binding proteins regulate mRNA decay and translation, two key steps in the control of gene expression in cells. Controlling mRNA metabolism allows cells to respond rapidly to altering conditions by utilizing already available mRNA, bypassing the wait for newly transcribed mRNA. The Puf family of RNA binding proteins bind specific mRNAs through interactions with sequences located in the 3’ untranslated region (UTR). Puf proteins are conserved throughout eukaryotes and have diverse roles including stem cell maintenance, neuronal development, stress response and organelle biogenesis. In Saccharomyces cerevisiae, Puf proteins are conditionally regulated in response to the cells metabolic. Specifically, in …

Women In Kazakhstan: A Multifaceted Approach To Female Political Representation, Ashley Gaile Benedict Dec 2014

Women In Kazakhstan: A Multifaceted Approach To Female Political Representation, Ashley Gaile Benedict

Dissertations

This investigation focuses on two competing theories (historical institutionalism and social constructivism) and their explanatory value in regards to female political representation in Kazakhstan. Historical Institutionalism maintains that current institutional dynamics are constrained by past institutional formations, even when these past institutions are no longer relevant. Social Constructivism challenges this theory by upholding that institutions are culturally situated and a reflection of shared ideas rather than material forces as argued by historical institutionalism. Based on Hanna Pitkin’s (1967) four dimensions of representation (formal, descriptive, substantive, and symbolic), I examine how Kazakhstan’s Soviet past and its creation of a Kazakh ethnic-national …

Who Decides? A Study Of The Effects On Decision Unit Dynamics In Crisis And Crisis Transition, Janet Leigh Drake Dec 2014

Who Decides? A Study Of The Effects On Decision Unit Dynamics In Crisis And Crisis Transition, Janet Leigh Drake

Dissertations

Foreign policy decision-making is often an obscured process, particularly when it involves threats to national security or national interests. Despite the lack of transparency, though sometimes necessary, foreign policy decisions can have far-reaching consequences. Policymakers establish and affect relationships with other governments, and can commit state resources for cooperation or for conflict. The purpose of this study is to determine what types of decision units make foreign policy decisions and what factors influence the dynamics of the unit. I employ the decision units (DU) framework developed by Margaret Hermann to decisions made by the United States and Israel during the …

Electrochemical Synthesis Of Nanostructured Noble Metal Films For Biosensing, Jay Kishan Bhattarai Dec 2014

Electrochemical Synthesis Of Nanostructured Noble Metal Films For Biosensing, Jay Kishan Bhattarai

Dissertations

Nanostructures of noble metals (Au and Ag) are of interest because of their important intrinsic properties. Noble metals by themselves are conductive, physically robust, chemically inert, and strongly bind to thiols. However, when the nanostructures are formed, they in addition possess high surface area and unique optical properties tunable by adjusting the shape and size. All of these properties make nanostructures of noble metals suitable candidates to be used as a transducer for biosensing. Individual nanostructures might be easier to prepare but difficult to handle to be used as a transducer. Therefore, we prepared and analyzed nanostructured films/coating of noble …

Characterization Of The Early Development And Quality Of Red Snapper (Lutjanus Campechanus) Eggs And Larvae In Aquaculture Conditions, Agnès Bardon Albaret

Dissertations

The quality of red snapper Lutjanus campechanus eggs is highly variable and unpredictable in aquaculture, leading to high mortality during early larval rearing. In this work, the viability of red snapper eggs was investigated from fertilization until larvae expired due to exhaustion of vitellin reserves. The studied spawns were obtained via strip spawning wild-caught (n=17) and captive (n=7) females following hormonal induction. The fertilization rate, the hatch rate, and the duration of survival of unfed larvae post hatch were weakly correlated to each other, revealing occurrence of distinct and independent components of egg quality.

Spawns from captive females were characterized …

The Effect Of Functional Fixation In Problem Solving Among Preschool, Second Grade, And Ninth Grade Children, Michael Kenneth Nehring Dec 2014

The Effect Of Functional Fixation In Problem Solving Among Preschool, Second Grade, And Ninth Grade Children, Michael Kenneth Nehring

Dissertations

Functional fixedness is a cognitive function whereby an individual becomes fixated on a given function of an object, which prevents the individual from using the object in an alternative fashion to solve a problem (Duncker, 1935/1945). The current study analyzed the effect of functional fixedness on 36 children from three different age groups, preschool, second grade, and ninth grade. The children were presented with a problem solving activity based on a problem used by German and Defeyter (2000), in which they concluded that young children are immune to the effects of functional fixedness. Research conducted by Chrysikou (2006) indicated using …

Comprehending Male And Female Levels Of Engagement In Subsets Of The National Survey Of Student Engagement: Explicating The Dynamics Of Gender Role Conflict As A Mediating Factor For Males, Jacob Glen Arndt

Dissertations

Male enrollment and graduate rates in higher education have paled in comparison to female achievement since the early 1980’s, and explanations as to the reasons behind why males are falling behind have not been fruitful in addressing these issues. One area that has received very little attention in the literature is the role that gender role conflict may play in male student performance.

The purpose of this research is to explore male and female engagement levels, while attempting to understand if levels of male gender role conflict are a mediating factor of engagement for men. This study utilized the three …

Ancestor Worship In The Middle Sicán Theocratic State, Go Matsumoto Dec 2014

Ancestor Worship In The Middle Sicán Theocratic State, Go Matsumoto

Dissertations

The major focus of this dissertation is the ancestor worship that is inferred to have been practiced in the multiethnic Middle Sicán theocratic state (AD 950-1100) that prospered on the northern North Coast of Peru. The major objective is twofold: (1) demonstrating by archaeological means that ancestors were indeed worshipped in the Middle Sicán society and (2) elucidating the nature and role of the inferred ancestor cult and associated rituals and ceremonies. Ancestor (and the veneration of it) is one of the themes that have the deepest roots in the anthropological thoughts; nevertheless, many archaeologists have uncritically invoked ancestor veneration …

Psychological And Genetic Contributions To The Development Of Social Cognition In Children, Jacqueline Marie Klaver Dec 2014

"I Don't Want To Hurt Anyone's Feelings": Using Race As A Writing Prompt In First Year Writing, Dianna Shank

Dissertations

First Year Composition (FYC) is one of the most important courses for any incoming college student. This course (often designated as English 101) provides students the rhetorical tools to fully engage in critical thinking and writing on the college level. One of the most common methods of organizing FYC is to use a topic as the center of all the reading and writing prompts. The use of outside subject matter to teach FYC is a common practice that is rarely interrogated for its effectiveness. However, the Hairston debate in the early 1990s opened up a public discussion of how FYC …

Aminoglycoside-Induced Otoneurotoxicity: Analysis Of Inner Hair Cell Synaptic Plasticity Following Drug Exposure, Matthew M. Abernathy

Dissertations

Aminoglycoside antibiotics are powerful drugs for combating bacterial infections, but are limited in use due to their ototoxicity. This class of drug targets the auditory hair cells of the cochlea, causing cell death, which leads to a decline in auditory function. In spite of much research aimed at revealing a mechanism of damage, there are no co-therapies available to diminish the ototoxic liability of aminoglycosides. Existing research does show that there may be a neurodegenerative process that contributes to the observed toxicity. In an effort towards clarifying present understanding of issue, this dissertation project was conducted to characterize the neurodegenerative …

Probing The Unfolding And The Stability Of The Last Four Metal Binding Domains Of Wilson Disease Protein Using Circular Dichroism And Novel Spectroscopic Techniques, Ibtesam Yaseen Alja’Afreh

Dissertations

Wilson disease protein is a copper-transporting P1B type ATPase. It has large N-terminal copper binding domain which is composed of six homologous sub-domains. Each of these six domains is ~72 residues and connected to one another by linking regions of various lengths. They all possess similar ferrodoxin fold, and metal-binding motif, MXCXXC.

The need of having six metal binding domains and the manner in which they are communicating with each other is not well understood. To better understand how the last four metal binding domains function, I pursued a detailed biophysical characterization of these domains. Using molecular biology I …

Ultrafast Interfacial Electron Transfer Across Molecule-Tio2 Nanocomposites: Towards Solar Cells And Two Photon Absorption, Edwin Mghanga

Dissertations

Interfacial charge transfer (ICT) across the molecule-TiO2 nanoparticle interface has gained enormous research attention for applications in dye sensitized solar cells (DSSC), photo-catalysis, water splitting and nonlinear optics. DSSCs are promising clean alternative energy sources. However, current DSSCs suffer from lower efficiencies and higher cost. Better understanding of the ICT processes in DSSCs can help solve these problems. We have used two strategies to understand ICT in the context of DSSCs. Firstly, we used a computationally validated anchor group, acetylacetonate (acac) to bind molecules to the semiconductor surface and facilitate charge separation. Secondly, we used natural dye sensitizers, …

Effective Instructional Strategies Utilized In Successful And High Performing Secondary Schools In The Southern Region Of Mississippi, Jeanifer Lynn Pearson

Dissertations

The schools in the United States have implemented measures intended to close the achievement gap for over fifty years (Solomon, 2009). The mandates handed down by the federal government cannot be altered. Therefore, schools are required to determine specific measures that increase student academic success. Prior research has demonstrated that schools can be successful if effective leadership at the district, school, and classroom level was present (Gregory, 2003; Johnson, Livingston, & Schwartz, 2000). At the classroom level, it was the leadership of the individual teacher to determine the creation of innovative techniques aimed at academic success of all students (Farr …

Dissertations

A number of molecular probes have been designed and synthesized for the detection of Zn2+ and Fe3+ ions. Two types of functional groups have been incorporated into the molecular scaffolds to utilize different fluorescent mechanisms. The first class of receptors contains a pyrene moiety. These molecular probes use the excimer mechanism for the detection of Zn2+ ion. The probes work well in an organic solvent with a detection limit of 20 nM (one ppb). Alternatives are made to make them water soluble, but this proved to be difficult. An interesting ion-induced self-assembly system will also be discussed. …
