Personal ontology studies human constitution and human nature, an increasingly debated topic in Christian theology. Historically, the most prominent models of personal ontology in Christian theology have been substance dualist models. More recently, physicalist models have offered prominent alternatives.
This dissertation studies the conflict of interpretations between these two major model groupings. By applying a canonical theology, it then presents an Edenic model of personal ontology that can address the current conflict of interpretations. In Milwaukee, Black babies die before their first birthday nearly three times as often as White and Hispanic babies. Prematurity is the major cause of infant mortality, and social determinants of health play a large role. Commitments from within Christian bioethical traditions can critique ethical frameworks commonly in use in US bioethics by calling for the incorporation of analysis of social power dynamics that is necessary for addressing this issue. Original ethnographic fieldwork that listens closely to Black mothers and health professionals uncovers key themes related to women's and infant health at the intersection of race, class, and gender. Adoptive cell transfer of T cell receptor (TCR) gene-modified T cells targeting specific tumor antigens is currently in clinical trials for patients with advanced malignancies. Despite the clinical responses, there are still hurdles to be overcome in achieving an effective and safe therapy. One of the limitations in the success of this type of therapy is the potential for cross-reactivity and unanticipated off-target reactivity which could lead to autoimmunity. Life requires biological membranes. Membrane-enclosed compartments separate and unite through dynamic fission and fusion reactions. These are catalyzed processes that are central in organismal biogenesis. This dissertation focuses on extracellular membrane fusions, which are central to several processes. (1) Enveloped viruses enter cells through membrane fusions. (2) Extracellular vesicles (EVs) also deliver molecules into cells through membrane fusions. (3) Entire cells also fuse together, generating fertilized zygotes, skeletal muscles, and giant cell macrophages.Mechanisms of extracellular membrane fusion are poorly understood. T lymphocytes are a critical component of the adaptive immune system and their responses in pathogenic and steady state conditions require strict regulation. One mechanism involved in this regulation is Transforming Growth Factor-β (TGF-β) signaling. TGF-β can act on activated T cells to facilitate the differentiation of TGF-β dependent T helper subsets or the suppression of T cell activation and proliferation. This study explored the associations between immersive exhibit design, mother-child engagement, conversational interactions, and children's learning. Participants were 41 mothers and their 6-8-year-old children (Mage=7.15, SD=.79). A within-subjects, mixed methods approach was utilized, including naturalistic observations, interview data, and surveys. Mother-child dyads were video and audio-recorded as they visited four different exhibits: two low immersion exhibits and two high immersion exhibits. Interview data was collected from children immediately following the visit to assess learning. Survey data was collected from mothers to assess education, environmental predispositions, and science-related career. In high immersion exhibits, mothers and children spent more time and asked …

Adoptive cell transfer (ACT) immunotherapy using antigen (Ag)-specific T cells is partially effective treating several malignancies but numerous challenges remain in order to improve its therapeutic potential. The roles of host factors, such as sex hormone receptor signaling, that can affect the T cell anti-tumor function remain understudied. Staphylococcus aureus is known to cause severe systemic infection with high mortality rates. Antibiotics are the only approved therapy for patients, but the widespread prevalence of antibiotic resistant strains limits treatment efficacy, and many patients succumb to the disease. Many probiotic agents are commercially available, but the mechanisms by which they benefit are not known. Chronic low back pain (CLBP) is a prevalent condition, often involving an inflammatory process. Those with CLBP frequently experience behavioral symptoms, including depressed mood, fatigue, and sleep disturbance, which may exacerbate pain and reduce quality of life (QOL). The purpose of this study was to identify behavioral symptom clusters (depressive mood, fatigue, poor sleep) in individuals with CLBP, and to determine whether there are differences in pain, QOL and inflammation (plasma IL-6) based on cluster membership. Research reveals that zero-tolerance policies lead to school suspensions of a disproportionate number of African American students in urban areas (Center for Civil [CCRR], 2015). Increasing number of genes encoding PLP-dependent transcription regulators, MocR/GabR type regulators, have been identified in various bacterial genomes. However, only a handful of them, including MocR, PdxR and GabR have been studied experimentally. They control different aspects of the bacterial metabolism. Only GabR has reported crystallographic structures. MocR/GabR regulators possess a chimeric structure consisted of a WHTH DNA binding domain and an Aminotransferase-like regulation domain, which can bind PLP as an effector in transcription regulation. Background: In 2010, more than 51.4 million procedures were performed in hospitals in the United States. Almost half of all patients had a surgical procedure prior to their hospital discharge and over 80% of patients experienced acute pain after surgery. When acute pain persists for two or more months after surgery, chronic pain develops, costing the United States up to $635 billion annually. Hispanics, in particular, disproportionately experience disparities in pain treatment as they are less likely to receive analgesics. Cell migration in complex micro-environments, that are similar to tissue pores, is important for predicting locations of tissue nucleation and optimizing scaffold architectures. Firstly, how fibroblast cells - relevant to tissue engineering, affect each other’s directional decisions when encountered with a bifurcation of different channel lengths was investigated. It was found that cell sequence and cell mitosis influence the directional choices that the cells made while chemotaxing. Specifically, the fibroblasts chose to alternate between two possible paths - one longer and the other shorter - at a bifurcation. The vision of highly parallel, automated manufacturing systems that can build macroscopic products by heterogeneous assembly of many small devices will have a major impact in manufacturing. In this study, a novel milli-scale robotic assembly machine with parallel capabilities, assisted with programmable magnetic field, is developed. The machine prototype consists of a 16x16 array of electromagnets. The dimensions of the electromagnets are 5mm high with an inner diameter of 1.1mm and outer diameter of 2.5mm. All the electromagnets are driven by a 16x16 array of H-Bridges, and an Arduino microcontroller is used to control and program the arrays.

Among these compounds are high levels of polycyclic aromatic hydrocarbons (PAHs) which are not only toxic but can also be carcinogenic and/or mutagenic. The direct incorporation of carboxylated carbon nanotubes (f-CNTs) into hydrophobic drug particles during their formation via anti-solvent precipitation is presented. The approach is tested using two drugs namely antifungal agent Griseofulvin (GF) and antibiotic Sulfamethoxazole (SMZ) that have very different aqueous solubility. It is observed that the f-CNTs dispersed in the water serve as nucleating sites for crystallization and are readily incorporated into the drug particles without altering crystal structure or other properties. The results show that the hydrophilic f-CNTs dramatically enhance dissolution rate for both drugs. The increased degree of functionalization leads to higher hydrophilicity and therefore faster dissolution …

Current tissue engineering approaches have fallen short of promoting fully functional cardiovascular cells and the post-myocardial infarction microenvironment is still not well understood. Listeria monocytogenes is a Gram-positive, facultative intracellular food-borne pathogen that causes the disease listeriosis. In order to establish an infection, L. monocytogenes must survive multiple stressors encountered within the gastrointestinal tract, including alterations in pH, bile, salt, and oxygen availability. This dissertation focused on understanding the stress response of L. monocytogenes to bile. Bile acts as a bactericidal agent by disrupting the membrane integrity and causing instability to macromolecules like DNA. Thus, a bacterium must be able to maintain its membrane architecture, composition and integrity.

This dissertation aimed at answering several fundamental questions, specifically: 1) What is the effect of scribing parameters on visual corrosion assessments? 2) What role do molecular weight, residual solvent, and polymer composition have on morphological changes in organic thin films? 3) What alternatives to visual assessments could be implemented to better understand and detect corrosion before macroscopic failure? This research definitively proved that scribes can vary in type and dimensions, and yet are still comparable if the scribe dimensions before exposure are known. Forest disturbance occurs on a wide gradient of selectiveness and creates new growth opportunities for adapted species. Across the spectrum of disturbance, anthropogenic disturbance influences community assembly in the Midwest more than other mechanisms but is its role in shaping and maintaining ecosystems is inadequately considered in most discussions on the historic range of variability (HRV). Forest resiliency is threatened by unprecedented agents of ecosystem change such as invasive species and reduced regeneration potential of native species.
