This dissertation examines the evolving role of digital technologies in archaeological research, focusing on the management and interpretation of extensive digital datasets generated by modern excavations. Drawing on six years of comprehensive fieldwork at San Miceli, Sicily, this study critically evaluates and updates the established four-field methodology proposed by Levy et al. (2012), encompassing acquisition, analysis, dissemination, and curation. While traditional archaeological documentation methods rely heavily on analog techniques, these approaches increasingly fall short due to the rapid expansion and complexity of digital data.

This investigation intended to examine the relationship between the financial literacy components of full-time pastors and their financial behavior components. Many pastors struggle with their finances and often retire unprepared due to having inaccurate or incomplete knowledge of relevant financial information and planning strategies.

This study adopted a quantitative, descriptive, and correlational approach to investigate the financial and demographic characteristics of pastors. The aim of this study is to undertake a fresh look into the concept of Israel in the Old Testament and its divine purpose in the context of the overarching theology of the Hebrew Bible. The working method used in this research is a theological and exegetical analysis of relevant Hebrew text along with incorporating historical and socio-archaeological background studies.

To explore the relationship between two recently identified bacterial species (S. anginosus and S. urinae), three aims were designed. First, niche ranges were identified via two metagenome databases. Different niche ranges were identified between the two groups. S. urinae was more specific to the urogenital tract. S. anginosus appeared to be present across human anatomical niches. Secondly, specific genes to the S. urinae group were identified. This gives insight into the genomic and phenotypic differences previously identified. Legumes are an incredibly diverse and agriculturally important family of plants. They have a special relationship with symbiotic bacteria known as rhizobia which fix nitrogen into ammonia within nodules on their roots. Astragalus is the largest plant genus on Earth, and the species Astragalus lentiginosus is extraordinarily diverse, consisting of approximately 40 varieties. However, the rhizobia of A. lentiginosus have never been studied. In this study, root nodules were extracted from 17 varieties of A. lentiginosus to obtain rhizobial strains and metagenomes. A. lentiginosus primarily associates with rhizobia from the genera Mesorhizobium and Allomesorhizobium. There was little evidence of horizontal …

In 2019, there was a 140.19% increase in deaths due to urinary tract infections (UTIs) compared to 1990, worldwide. One of the common uropathogens known to associate with UTIs in females is Escherichia coli. The prevailing belief is that E. coli in the bladder originates in the rectum, passing its way from the perineal, to the vagina, and finally into the bladder. Known as the Fecal-Perineal-Urethral (FPU) hypothesis, this “origin story” has been supported by several studies that have relied on genotyping or serotyping isolates collected from the bladder and rectum. This thesis sets out to critically engage with existing research on Christian nationalism. It argues that current research suffers from a methodological focus that is too broad, and theoretical grounding that is too imprecise, both of which prevent researchers from grasping the full scope and intent of Christian nationalism, as imagined by Christian nationalists themselves. Coronaviruses infect a wide range of hosts and have caused recent human pandemics. While much has been learned, gaps remain in understanding viral assembly and egress, particularly the role of the envelope (E) protein in specific infectivity. To study this, recombinant MHV viruses were created with E gene deletions and HiBiT-tagged spike (S) proteins to monitor virion release. In mouse cells, MHV∆E S-HiBiT secreted less spike and fewer infectious particles, indicating reduced specific infectivity. Comparisons between human HeLa-CEACAM and mouse DBT cells showed species-specific support for virus production. TMED10, a host membrane protein, was tested as a potential E-interacting partner …

Exopolysaccharides (EPS) secreted by the commensal bacterium Bacillus subtilis exhibit potent anti-inflammatory properties, yet the underlying molecular mechanisms remain incompletely understood. We hypothesized that EPS reprograms antigen-presenting cells toward a tolerogenic state by modulating TLR4–MyD88 signaling. To test this, we began using a chemically defined, high-yield EPS production and purification protocol that largely eliminates protein and DNA contaminants, thus improving functional reproducibility across experiments using this new method. In vitro, EPS engaged TLR4 on murine dendritic cells (DC2.4) without inducing classical NF-κB activation, seen by LPS stimulation. Glioblastoma (GBM) is the most common and most aggressive form of adult cancer in the central nervous system, with GBM accounting for the majority of all malignant brain tumors. GBM is currently considered to be an incurable form of cancer, with over 14,000 patients diagnosed in the United States last year. Given the success of immunotherapy for treating other types of cancer, our group has investigated the possibility of therapeutically neutralizing immunosuppressive indoleamine 2,3 dioxygenase 1 (IDO1) in patients with GBM. Despite similar breast cancer incidence rates, African American (AA) women experience a 40% higher mortality rate compared to Caucasian American (CA) women. This disparity is multifactorial and particularly pronounced in the Chicago area. AA women are twice as likely to be diagnosed with triple-negative breast cancer (TNBC), often at a younger age and with more aggressive disease. Innate immune cells serve as the first line of defense by detecting pathogens through pathogen- and damage-associated molecular patterns (PAMPs and DAMPs). Recognition of PAMPs triggers the synthesis and secretion of pro-inflammatory proteins, with the inflammasome playing a key role in the secretion of interleukin-1β (IL-1β), a crucial part of the innate immune response. Upon inflammasome activation, the release of signal-peptide lacking cytoplasmic proteins such as IL-1β occurs through non-classical pathways known as unconventional protein secretion (UCPS), which vary depending on the cell type and stimulus which helps tailor the extent of pro-inflammatory protein secretion. Typically, the creation of CG content is led by expert practitioners who guide computational algorithms towards satisfactory results. Thus, creating CG content often requires manual iterations encompassing algorithm design, parameter tuning, and aesthetic feedback. The nonprofit sector faces a significant issue: the underrepresentation of the communities it aims to serve, including Alaska Native communities. This research investigates the likely causes of this underrepresentation and explores potential solutions. It includes nine qualitative interviews with a majority of Alaskan Native leaders and consultations with other non-Native nonprofit leaders who have extensive experience in Alaska's nonprofit organizations.

Contemporary education research seeks causal inferences in order to guide policy and practice. Often well-controlled quasi-experimental and experimental designs are used as they are very useful for supporting and contextualizing causal inferences. Nevertheless, individual studies, even high-quality, resource-intensive randomized controlled trials, are not able to establish generalized claims. Because primary studies vary over dozens of variables (e.g., grade, subject, duration) simply conducting additional primary studies cannot change this. Purpose: This qualitative phenomenological study examines how middle school teachers in Riverside County, California, support the social-emotional learning (SEL) needs of students as students returned to in-person instruction after the COVID-19 pandemic. Methodology: This qualitative study explores how Riverside County middle school teachers addressed students' social and emotional needs upon returning to in-person learning after COVID-19. Twelve teachers from local public middle schools (grades 6-8) participated in semi-structured interviews, answering ten questions. This study, a sequential mixed methods design, used quantitative techniques and a qualitative, phenomenological approach. The quantitative phase addressed the research question: Does the reimbursement of services for the delivery of pediatric palliative care differ between types of funding, and is funding related to directors' perceptions of obstacles to the delivery of care? The qualitative phase addressed the research question: What are the barriers that CHI PACC directors' experience in their roles and how do they overcome them?

This research study examined administrators’ perceptions of the effectiveness of trauma-informed practices in disciplinary decision-making. Ten elementary school assistant principals were interviewed about their experiences with trauma-informed practices following a student referral. The participants also submitted behavioral expectation matrices and discipline flowcharts for analysis. The participants recognized and valued the underlying elements of trauma-informed practices (relationships, context, support, and cultural factors). However, they lack a clear understanding of and processes for full implementation of trauma-informed practices into disciplinary decision-making.
