Single-cell and multi-omic technologies have transformed the dissection of cellular heterogeneity and regulatory dynamics in health and disease. However, the high dimensionality, technical variability, and biological complexity of these datasets present significant challenges for integration, annotation, and interpretation. In this dissertation, a suite of computational approaches is introduced to address key problems in single-cell and multi-omic data analysis through model-based innovations and applied statistical frameworks.

Effectively designed controllers for lower-limb exoskeletons to assist sit-to-stand (STS) and walking are crucial for delivering efficient, safe, and comfortable assistance during daily activities. Traditionally, controller optimization involves biomechanical modeling and user-specific customization. Hall effect thrusters (HETs) are the most widely used electric propulsion (EP) device for in-space satellite propulsion. EP devices provide high efficiencies for extended mission lifetimes and low thrust for precision maneuvers. The hollow cathode is a critical HET component and undergoes extensive lifetime testing in addition to the lifetime testing of the full HET. Significant density gradients, non-Maxwellian electron behavior, and magnetic fields sufficiently strong enough to magnetize electrons in hollow cathode plumes create favorable conditions for the formation of plasma instabilities, which have an impact on cathode lifetime. Bioactive peptides are peptide fragments derived from the proteolytic cleavage or maturation of functional proteins that have shown promising therapeutic potential in addressing major health challenges, including type 2 diabetes, hypertension, and pain management. Diabetes is characterized by high blood glucose levels due to insufficient insulin production or utilization. There are several therapeutic targets for managing diabetes, and one key target is dipeptidyl peptidase IV (DPP IV) due to its role in glucose metabolism. Angiotensin-converting enzyme 2 (ACE2) and the neutral amino acid transporter B0AT1, encoded by SLC6A19, are membrane proteins with pivotal roles in human physiology. ACE2 is involved in regulating blood pressure and serves as the cellular entry receptor for SARS-CoV-2, while B0AT1 facilitates amino acid absorption in the intestine. The interplay between ACE2 and B0AT1, particularly their physical interaction and co-expression in the intestine, underscores their relevance in both normal physiology and disease. The relapse and chemotherapy resistance are the major challenges of acute myeloid leukemia (AML) treatment, driven by a rare population of leukemia stem cells (LSCs). LSCs are unique types of cells which evade standard chemotherapies and can regenerate AML post-treatment. Background: Female student-athletes navigate unique psychosocial challenges balancing academics and athletics, yet limited research has explored the impact of university engagement on their social and emotional well-being. This study examines the impact of university engagement on sense of social fit (SSF), nonspecific psychological distress (NPD), and loneliness among Division I female student-athletes.

Video games have been examined for their effects on cognition, learning, health, and physiological arousal, yet research on social dynamics within video gaming is limited. Studies have documented the presence of derogation, racism, and discrimination in this anonymous medium. However, gamers‟ firsthand experiences are typically examined qualitatively. Thus, this study aimed to establish a quantitative baseline for the frequency of derogatory, racist, and discriminatory speech (DRDS) in gaming. Vascular smooth muscle cells (SMCs) surround many blood vessels throughout the body,where they dynamically alter vessel diameter to regulate blood pressure, provide structural integrity, and absorb shock on a beat-to-beat timescale. As smooth muscle function fails, profound vascular disease ensues, often with tragic results- even death. Smooth muscle myosin 2 (SM2) is the dominant motor protein that actuates contractility and allows SMCs to perform these vital functions. The current measures of competency for nurse anesthetists for recertification are continuing education units for each biennial recertification cycle, and records of current practice and state licensure. Recently, the National Board of Certification and Recertification of Nurse Anesthetists (NBCRNA) adopted new standards that include a written examination every 8 years, an increase of 40 continuing education units per year, and completion of four core competency modules each four-year recertification cycle. The primary focus of this research was to determine the effect of berberine (BBR), a clinically relevant plant-derived alkaloid, on follicular T helper (Tfh) cell activation and differentiation, with implications for the generation of T cell-dependent humoral responses. This research revealed a novel function of BBR as a suppressor of Tfh cell proliferation in secondary lymphoid organs of BBR-treated mice that underwent immunization for arthritis induction in a collagen-induced arthritis (CIA) mouse model. This project aimed to examine the effectiveness of a preoperative preparation program (PPP) in decreasing parental anxiety. When children needed anesthesia to get outpatient procedures completed, the accompanying parents received verbal education about the process on the day of the procedure and were assisted by a support person during and after the child's induction anesthesia. Parental anxiety was measured using the State-Trait Anxiety Inventory (STAI) tool before preoperative preparation and after the induction of anesthesia to assess whether the PPP improved parents' anxiety levels. The urinary tract contains a community of bacteria called the urinary microbiota (urobiota) thatmay be relevant to health; the genomic component of the urobiota is the urinary microbiome (urobiome). Urinary bacteria have been associated with both asymptomatic states and disease conditions, such as urinary tract infection (UTI), overactive bladder (OAB), and urge urinary incontinence (UUI). Some bacteria, such as E. coli, are considered urinary pathogens (uropathogens) but also can be commensals. Finally, various ways to incorporate self-care into already established graduate-level training programs at universities and practicum training sites were discussed to enhance the training experience as well as mitigate burnout in the novel clinician.

Open-heart surgery patients in ICU report anxiety and pain with chair rest despite administration of opioid analgesics. In addition, chair rest increases myocardial oxygen demand (MVO2). Non-pharmacological, complementary methods (such as sedative music and scheduled rest) used to reduce anxiety, pain, and MVO2 during chair rest warranted further investigation. The conceptual framework was based on the transactional model of stress and coping. A 3-group pretest-posttest repeated measures experimental design examined the effect of music and rest on anxiety, pain sensation, and pain distress and MVO2 during chair rest in adult postoperative open-heart surgery patients.
