Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as nonalcoholic fatty liver disease, is associated with obesity and type 2 diabetes, and as the prevalence of these conditions increases, the incidence of MASLD is expected to rise. However, the mechanisms that underlie the development of MASLD are complex and incompletely understood. Previous work from our group has indicated that the human leukocyte antigen-F adjacent transcript (Fat10) is overexpressed in the LEW.1WR1 rat model, which develops features of MASLD. Hurricanes remain among the most destructive natural hazards, producing widespread impacts through extreme rainfall, flooding, and damaging winds. Rapid urbanization in coastal regions further amplifies these risks by modifying land surface characteristics and increasing population, while accurately forecasting hurricane evolution remains a major challenge. This dissertation addresses these challenges through three interconnected research objectives focusing on urbanization-induced hurricane modification, hydrological response, and AI-based hurricane forecasting. The first objective investigates how incremental multi-decadal urban growth modifies hurricane rainfall distribution and rainband evolution during Hurricane Harvey (2017). Liquid-Core Nuclear Thermal Rockets (LNTR) were originally investigated in the 1960s. Exploratory efforts have thus far been relegated to preliminary models that illustrate proof-of- concept or identify theoretical limits. Collectively they would report ranges for 𝐼𝑠𝑝 between 1000 −2000 𝑠 and thrust-to-weights between 0.5 −6. This work expands a model for the Bubbler class of LNTRs into an engineering model suitable for design exploration and system model integration. Bubblers use centrifugal force to trap molten fissile fuel while bubbling propellant radially inward to cool structural members and achieve temperatures >4500 𝐾. This dissertation explores continuity and development in the eschatological understanding of the seventh-day Sabbath within selected Adventist theological writings from its Millerite origins to the contemporary scholarship of Jo Ann Davidson. The eschatological aspects of the Sabbath have played a central role in both pioneer and contemporary Adventist thought, particularly in its identification as the seal of God in Revelation and as a sign of covenant loyalty in the final crisis. The purpose of this project was to examine the effectiveness of an intentional sermon series, Everyday Evangelism, in cultivating an evangelistic culture at Mountain Chapel Methodist Church, a Global Methodist congregation. The study used a program evaluation model and addressed the question, How does preaching cultivate an evangelistic culture in the local church, along with questions concerning growth in prayerfulness for the lost, gospel conversations, acts of compassion, invitations to Christian community, follow-up with seekers, and whether leaders exhibited greater change than the congregation as a whole. The study surveyed employees in the service and helping professions (for example retail, healthcare related, hospitality), to examine the impact of transformational leadership on employee organizational citizenship behavior (OCB). Specifically, it tested job satisfaction and organizational trust as possible mediators to the transformational leader to employee OCB relationship and evidence of perspective-taking as a possible moderator to either of these mediated relationships. Semi-supervised learning has emerged as a powerful paradigm for analyzing single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) data, where full annotation is often costly or impractical. scRNA-seq technologies measure the expression of thousands of genes across tens of thousands of cells, whereas ST additionally captures the spatial coordinates of gene expression within intact tissue sections. Annotation is a key step in both scRNA-seq and ST analysis pipelines, aiming to identify cell types, spatial domains, and latent biological structures. Single-cell and multi-omic technologies have transformed the dissection of cellular heterogeneity and regulatory dynamics in health and disease. However, the high dimensionality, technical variability, and biological complexity of these datasets present significant challenges for integration, annotation, and interpretation. In this dissertation, a suite of computational approaches is introduced to address key problems in single-cell and multi-omic data analysis through model-based innovations and applied statistical frameworks.

This qualitative study investigated K-12 teachers’ views about gamification as an instructional tool to increase student motivation, self-efficacy, and learning mastery. Kelly’s Curriculum Theory and Bandura’s Self-Efficacy Theory were used to investigate how gamified learning environments affect teacher practice and self-efficacy. Seventeen teachers participated in a researcher-designed Qualtrics survey that included both Likert-style and open-ended items. The researcher used descriptive statistics and thematic coding of responses, based on participants’ self-identified levels of gamification experience. American Indian/Alaska Native (AI/AN) adults experience higher rates of type 2 diabetes and worse glycemic control than non-Hispanic White Americans. Despite the importance of medication adherence for T2DM management, there is only one study in the last 15 years examining the relationship between medication adherence and A1C among AI/ANs. Patients with autosomal polycystic kidney disease (ADPKD) have an increased risk and worse outcomes after acute myocardial infarction, but the mechanism for this is unknown. Polycystin 2 (PC2), one of the two main proteins mutated in ADPKD, is a calcium permeant channel that localizes to the ER/SR. In non-cardiac cells, PC2 regulates the unfolded protein response (UPR) pathway under ER stress. However, whether PC2 is cardioprotective against ER stress is unknown. We found that PC2 expression increased in cardiomyocytes from human ischemic hearts and infarcted murine hearts. Cardiomyocyte-specific PC2 KO (PC2 KO) mice had worsened cardiac function after myocardial infarction …

Cells interpret biochemical and mechanical signals through dynamic regulation of the cytoskeleton and adhesion complexes, enabling key processes such as shape adaptation, force generation and migration. As these signals vary in both time and space, they can be challenging to resolve using traditional bulk assays. To overcome this limitation, we employ light-based optogenetic tools to reversibly modulate biochemical signals and mechanical equilibrium within cells. This qualitative case study involved a sample of Title I elementary schools, their principal, academic coach, and district Title I supervisor, who are involved in the school strategic planning process. The study aimed to explore the strategies and processes Title I schools use to develop, implement, and monitor their school strategic plans to grow student achievement. The research work finds a solution to precision agriculture of cotton cultivation using artificial intelligence (AI) models. Two sets of model performance based on the application are selected namely a low resource and a high resource setting. This is because using drone surveys to capture images identifying the classes of stressed and unstressed cotton plantation requires limited model architecture and CPU based computation. This qualitative study explores faculty perspectives on student accommodation in the community college setting. The one-on-one, semi-structured, in-depth interviews with faculty and perspectives of accommodation experts will result in outcomes that better prepare faculty, as a workforce, to support students with accommodations. The study utilized the social cognitive career theory (SCCT) to understand how increasing faculty awareness and engagement through professional development can enhance their ability to provide services to students with accommodation requests. African American spirituals and gospel music are two foundational folk traditions that have profoundly shaped the American musical landscape and continue to influence cultural identity and sacred expression. Emerging from the resilience and faith of enslaved African Americans, spirituals often carried coded messages of liberation. Rape myths are false beliefs that explain sexual violence, and uphold current beliefs on sexual violence (Payne, Lonsway, & Fitzgerald, 1999). These myths can be learned and reinforced through a variety of sources, including formal school-based sexual education programs, pornography consumption, and peers. Through two studies, sexual education and rape myth were examined. In the first study, an exploratory content analysis of state sexual education guidelines was completed to better understand what information and guidance is provided by states to those providing sexual education. This study examines the relationship between university athletic subsidies and outcomes for the broader undergraduate student population. The athletic subsidy, referred to as an athletic tax, is defined as institutional and student fees used to support athletics. Although athletics are often viewed as enhancing institutional prestige and recruitment, most universities depend heavily on subsidies to sustain their programs. In 2019, National Collegiate Athletic Association (NCAA) institutions spent over $18.8 billion on athletics, with $8.3 billion, or 44 percent, coming from institutional and student sources rather than athletic revenue (NCAA, 2019a). Using shallow learning strategies such as rote memorization is similarly unfavorable for success in academic settings compared to using deep learning strategies such as generating examples. This quantitative study examined the factors influencing Mississippi state-tested teachers’ engagement in data-based decision-making (DBDM). The purpose of the study was to explore how self-efficacy, anxiety, and school culture shape teachers’ experiences using data to guide instruction. A researcher-developed instrument measuring four constructs self-efficacy in data interpretation, anxiety related to DBDM, self-efficacy and engagement in DBDM, and school culture and DBDM was administered to Mississippi teachers of state-tested subjects. Descriptive statistics, a one-way ANOVA, correlation analysis, and multiple regression were used to analyze the data.

Long considered impractical, overly idealistic, and/or politically dangerous, utopianism has been largely marginalized in political science and American culture. However, this project finds that there are instructors teaching utopianism and they are using the concept to challenge students to think differently about governance, justice, and institutional design. This dissertation evaluated the efficacy of Three Funny Things, a humor-based positive psychology intervention (PPI) designed to enhance subjective well-being (SWB) and reduce depressive symptoms. Subjective well-being was operationalized through self-reported happiness, measured by the Authentic Happiness Inventory (AHI), and depressive symptomatology, measured by the Center for Epidemiologic Studies Depression Scale (CES-D). A repeated-measures design was employed across three time points: baseline (Time 1), post-intervention (Time 2), and one-month follow-up (Time 3).
