Purpose In preparation for a Phase I clinical trial utilizing a combined cytotoxic/immunotherapeutic strategy using adenoviruses expressing Flt3L (Ad-Flt3L) and thymidine kinase (Ad-TK) to treat glioblastoma (GBM), we tested the hypothesis that Ad-TK+GCV would be the optimal tumor killing agent in relation to efficacy and safety when compared to other pro-apoptotic approaches. Experimental Design and Results The efficacy and neurotoxicity of Ad-TK+GCV was compared with Ads encoding the pro-apoptotic cytokines (TNF-α, TRAIL, FasL), alone or in combination with Ad-Flt3L. In rats bearing small GBMs (day 4), only Ad-TK+GCV or Ad-FasL improved survival. In rats bearing large GBMs (day 9), the …

A Functional Polymorphism In The Hmgcr Promoter Affects Transcriptional Activity But Not The Risk For Alzheimer Disease In Swedish Populations., Lina Keller, Charlotte Murphy, Steve Meaney

Articles

Variations in genes associated with cholesterol homeostasis have been reported to modify the risk of developing Alzheimer disease (AD). To date there have been few investigations into variations in genes directly involved in cholesterol biosynthesis and AD. We investigated the influence of the -911C>A polymorphism (rs3761740) in the hydroxy-methyl-glutaryl CoA reductase (HMGCR) gene promoter on basal and regulated transcription, plasma cholesterol levels and the association with AD. Under in vitro conditions the A allele was found to be significantly more responsive to SREBP-2 mediated regulation than the C allele. In an age and sex matched case-control study, the genotype …

Rapid Multi-Class Multi-Residue Method For The Confirmation Of Chloramphenicol And Eleven Nitroimidazoles In Milk And Honey By Liquid Chromatography Tandem Mass Spectrometry, Mark Cronly, Patrice Behan, Barry Foley, Liam Regan, Sheila Martin, Michael Doyle, Edward Malone

Articles

A confirmatory method has been developed to allow for the analysis of eleven nitroimidazoles and also chloramphenicol in milk and honey samples. These compounds are classified as A6 compounds in annex IV of council regulation 2377/90 and therefore prohibited for the use in animal husbandry. Milk samples are extracted by acetonitrile with addition of NaCl, while honey samples are first dissolved in water before a similar extraction. Honey extracts undergo a hexane wash to remove impurities. Both milk and honey extracts are evaporated to dryness and reconstituted in initial mobile phase. These are then injected onto an LC-MS/MS system and …

A Full-Range, Multi-Variable, Cfd-Based Methodology To Identify Abnormal Near-Wall Hemodynamics In A Stented Coronary Artery, Jonathan Murphy, Fergal Boyle Jun 2010

A Full-Range, Multi-Variable, Cfd-Based Methodology To Identify Abnormal Near-Wall Hemodynamics In A Stented Coronary Artery, Jonathan Murphy, Fergal Boyle

Articles

The benefit of coronary stent implantation is reduced by excessive intimal hyperplasia which re-narrows the artery and the prevention of which is still a primary concern for clinicians. Abnormal hemodynamics create non-physiological viscous stress on the artery wall, one of the root causes of intimal hyperplasia following stent implantation. A methodology to comprehensively evaluate the viscous stress on the artery wall following stent implantation would be useful to evaluate a stent’s hemodynamic performance. The proposed methodology employs 3D computational fluid dynamics, the variables wall shear stress (WSS), WSS gradient (WSSG), WSS angle gradient (WSSAG) and a statistical analysis to evaluate …

Rainwater Harvesting And Greywater Treatment Systems For Domestic Application In Ireland, Zhe Li, Fergal Boyle, Anthony Reynolds

Articles

Water shortage has been recognised as one of the key issues facing many countries. Fortunately, there are relatively abundant water resources available in Ireland because of its plenty of rainfall. However, Ireland will inevitably encounter water shortage in the future, especially in urban areas. The water consumption per capita per day in Ireland is one of the highest in Europe. The water demand is still increasing due to population growth and higher standard of living. The use of domestic rainwater harvesting and greywater treatment systems has the potential to supply nearly 94% of domestic water in Irish households. The utilisation …

Optical Limiting Study Of Double Wall Carbon Nanotube-Fullerene Hybrids, Kang-Shyang Liao, Jun Wang, Daniel Fruchtl, Nigel Alley, Enrico Andreoli, Eoghan Dillon, Andrew Barron, Hansoo Kim, Hugh Byrne, Werner Blau, Seamus Curran

Articles

In order to merge complementary temporal and spatial nonlinear optical characteristics of Fullerene and carbon nanotubes, synthesis of double wall carbon nanotube-Fullerene hybrid was performed by covalently linking DWNT and C60 by amination reaction with polyethylenimine. DWNT-Fullerene hybrids were characterized by thermogravimetric analysis, UV-vis spectroscopy and transmission electron microscopy. Optical limiting performance of DWNT-Fullerene hybrids is superior to those of Fullerenes and SWNTs at the same level (~ 80%) of transmission. Whereas nonlinear scattering is an evident mechanism, reverse saturable absorption from Fullerene moieties has significant contribution. Charge transfer between the DWNT and Fullerene moieties may play an important role …

An Assessment Of The Antioxidant And Antimicrobial Activity Of Six Species Of Edible Irish Seaweeds, Sabrina Cox, Nissreen Abu-Ghannam, Shilpi Gupta

Articles

Six species of edible Irish seaweeds; Laminaria digitata, Laminaria saccharina, Himanthalia elongata, Palmaria palmata, Chondrus crispus and Enteromorpha spirulina were screened for potential bioactivity. Extraction of secondary metabolites was carried out using different solvents to determine antioxidant and antimicrobial properties of the dried extracts. The total phenolic contents of dried methanolic extracts were significantly different (p < 0.05). H. elongata exhibited highest phenolic content at 151.3 mg GAE/g of seaweed extract and also had the highest DPPH scavenging activity (p < 0.05) with a 50% inhibition (EC50) level at 0.125μg/ml of extract. H. elongata also had the highest total tannin and total flavonoid contents (p < 0.05) of 38.34 mg CE/g and 42.5 mg QE/g, respectively. Antimicrobial activity was determined using a microtitre method which allowed detection of bacterial growth inhibition at low levels. All methanolic seaweed extracts inhibited the food spoilage and food pathogenic bacteria tested; Listeria monocytogenes, Salmonella abony, Enterococcus faecalis and Pseudomonas aeruginosa, except C. crispus extracts. It was found that dried methanolic extracts of red and green seaweeds had significantly lower antimicrobial activity than the brown species; H. elongata had the highest antimicrobial activity with up to 100% inhibition. In the studied work, the antimicrobial activity of red and green seaweed extracts significantly increased when ethanol and acetone were used as extraction solvents (p < 0.05). Inhibition of E. faecalis with C. crispus extracts increased from 39.28 to 100% when ethanol and acetone were applied as solvents.

Influence Of Prescribed Fire On Ecosystem Biomass, Carbon, And Nitrogen In A Pinyon Juniper Woodland, Benjamin M. Rau, Robin Tausch, Alicia Reiner, Dale W. Johnson, Jeanne C. Chambers, Robert R. Blank, Annmarrie Lucchesi Mar 2010

Influence Of Prescribed Fire On Ecosystem Biomass, Carbon, And Nitrogen In A Pinyon Juniper Woodland, Benjamin M. Rau, Robin Tausch, Alicia Reiner, Dale W. Johnson, Jeanne C. Chambers, Robert R. Blank, Annmarrie Lucchesi

Articles

Increases in pinyon and juniper woodland cover associated with land-use history are suggested to provide offsets for carbon emissions in arid regions. However, the largest pools of carbon in arid landscapes are typically found in soils, and aboveground biomass cannot be considered long-term storage in fire-prone ecosystems. Also, the objectives of carbon storage may conflict with management for other ecosystem services and fuels reduction. Before appropriate decisions can be made it is necessary to understand the interactions between woodland expansion, management treatments, and carbon retention. We quantified effects of prescribed fire as a fuels reduction and ecosystem maintenance treatment on …

Platelet Alpha- And Beta- Secretase Activities Are Not Significantly Affected By Dementia Or Mild Cognitive Impairment In Swedish Patients., Paul Gorham, Natalie Bark, Ingemar Bjorkhem, Steve Meaney, Milita Crisby

Articles

The processing of the Amyloid Precursor Protein (APP) is a critical event in the formation of amyloid plaques which are composed of the 4kDa amyloid β-peptide (Aβ). Processing of APP occurs through a non-amyloidogenic pathway, mediated by initial α-secretase cleavage or through an amyloidogenic pathway via sequential cleavage by β- and γ- secretase enzymes, which produces Aβ peptides. Currently, the diagnosis of probable or possible Alzheimer's disease (AD) is primarily based on neuropsychological and neuroradiological assessment. Recent reports indicate that platelet β- secretase activity is moderately increased in patients with AD and mild cognitive impairment (MCI). To our knowledge platelet …

Dual-Action Hygienic Coatings: Benefits Of Hydrophobicity And Silver Ion Release And Surface Analysis, Niall Stobie, Brendan Duffy, John Colreavy, Patrick Mchale, Steven Hinder, Declan Mccormack Feb 2010

Dual-Action Hygienic Coatings: Benefits Of Hydrophobicity And Silver Ion Release And Surface Analysis, Niall Stobie, Brendan Duffy, John Colreavy, Patrick Mchale, Steven Hinder, Declan Mccormack

Articles

Coatings that demonstrate reduced attachment of crystalline precipitates and the medical device colonising Staphylococcus epidermidis were prepared by the immobilisation of silver doped perfluoropolyether–urethane siloxane thin films on glass substrates. The presence of stratified hydrophobic perfluoropolyether groups protects the coating surface from the attachment of crystalline hydrophilic species such as chlorides and phosphates, whilst silver ion release inhibited attachment of S. epidermidis and subsequent biofilm formation in vitro. The release of silver ions protects the perfluoro groups from the hydrophobic interactions of S. epidermidis cells, which can reduce the hydrophobicity of the protective coating. These coatings also exhibited significant antibacterial …

Predicting Neointimal Hyperplasia In Stented Arteries Using Time-Dependant Computational Fluid Dynamics: A Review, Jonathan Murphy, Fergal Boyle Feb 2010

Predicting Neointimal Hyperplasia In Stented Arteries Using Time-Dependant Computational Fluid Dynamics: A Review, Jonathan Murphy, Fergal Boyle

Articles

This paper reviews the recent literature regarding the time-dependant computational fluid dynamics (CFD) analyses of blood flow through implanted coronary stents. The in vivo processes which result in arterial restenosis are identified. The definition and range of the computationally predicted variables which are believed to stimulate the restenosis processes are evaluated. The reviewed literature is subdivided into effect-based in which the effects of altering the flow model are investigated and design-based in which different geometric stent configurations are compared. Finally, conclusions are made regarding the body of work reviewed and recommendations are provided for future work in this field.

Fourier Transform Infrared Microspectroscopy And Multivariate Methods For Radiobiological Dosimetry, Aidan Meade, Colin Clarke, Hugh Byrne, Fiona Lyng

Articles

The scientific literature contains an ever-growing number of reports of applications of vibrational spectroscopy as a multivariate non-invasive tool for analysis of biological effects at the molecular level. Recently, Fourier transform infrared microspectroscopy (FTIRM) has been demonstrated to be sensitive to molecular events occurring in cells and tissue after exposure to ionizing radiation. In this work the application of FTIRM in the examination of dose-dependent molecular effects occurring in skin cells after exposure to ionizing radiation with the use of partial least-squares regression (PLSR) and generalized regression neural networks (GRNN) was studied. The methodology is shown to be sensitive to …

Differential Genome-Wide Array-Based Methylation Profiles In Prognostic Subsets Of Chronic Lymphocytic Leukemia., Nicola Cahill, Fergus Ryan Jan 2010

Differential Genome-Wide Array-Based Methylation Profiles In Prognostic Subsets Of Chronic Lymphocytic Leukemia., Nicola Cahill, Fergus Ryan

Articles

Global hypomethylation and regional hypermethylation are well-known epigenetic features of cancer, however, in chronic lymphocytic leukemia (CLL), studies on genome-wide epigenetic modifications are limited. Here we analyzed the global methylation profiles in CLL, by applying high-resolution methylation micro-arrays (27,578 CpG sites) to 23 CLL samples, belonging to the immunoglobulin heavy-chain variable (IGHV) mutated (favorable) and IGHV unmutated/IGHV3-21 (poor-prognostic) subsets. Overall, results demonstrated significant differences in methylation patterns between these subgroups. Specifically, in IGHV unmutated CLL, we identified methylation of 7 known or candidate tumor suppressor genes (e.g. VHL, ABI3 and IGSF4) as well as 8 unmethylated genes involved in cell …

Articles

Soybean [Glycine max (L.) Merr.] cultivars varied in their resistance to different populations of the soybean cyst nematode (SCN), Heterodera glycines, called HG Types. The rhg1 locus on linkage group G was necessary for resistance to all HG types. However, the loci for resistance to H. glycines HG Type 1.3- (race 14) and HG Type 1.2.5- (race 2) of the soybean cyst nematode have varied in their reported locations. The aims were to compare the inheritance of resistance to three nematode HG Types in a population segregating for resistance to SCN and to identify the underlying quantitative trait loci (QTL). …

Seeing Is Believing: Body Motion Dominates In Multisensory Conversations, Cathy Ennis, Rachel Mcdonnell, Carol O'Sullivan

Articles

In many scenes with human characters, interacting groups are an important factor for maintaining a sense of realism. However, little is known about what makes these characters appear realistic. In this paper, we investigate human sensitivity to audio mismatches (i.e., when individuals’ voices are not matched to their gestures) and visual desynchronization (i.e., when the body motions of the individuals in a group are mis-aligned in time) in virtual human conversers. Using motion capture data from a range of both polite conversations and arguments, we conduct a series of perceptual experiments and determine some factors that contribute to the plausibility …

The objective of the Endangered Species Act is to "recover" imperiled species and thus to render the Act's conservation tools unnecessary To achieve this goal, the drafters of the Act crafted a linear process that begins with an assessment of the threats facing the species and moves through the elimination of those threats to the recovery and delisting of the species. It has become increasingly apparent over the past decade that few species fit this model- most species face threats-altered habitats and competition with invasive species-that cannot be eliminated. These species are "conservation reliant" because they will require ongoing conservation …

Conservation-Reliant Species And The Future Of Conservation, Dale Goble

Articles

Species threatened with extinction are the focus of mounting conservation concerns throughout the world. Thirty-seven years after passage of the U.S. Endangered Species Act in 1973, we conclude that the Act’s underlying assumption—that once the recovery goals for a species are met it will no longer require continuing management—is false. Even when management actions succeed in achieving biological recovery goals, maintenance of viable populations of many species will require continuing, species-specific intervention. Such species are “conservation reliant.” To assess the scope of this problem, we reviewed all recovery plans for species listed as endangered or threatened under the Act. Our …

Real-Time Pcr Method For The Quantification Of Burkholderia Cepacia Complex Attached To Lung Epithelial Cells And Inhibitionn Of That Attachment, Ciara Wight, Gillian Herbert, Ruth Pilkington, Máire Callaghan, Siobhan Mcclean

Articles

To develop a rapid method to quantify the attachment of the cystic fibrosis pathogen, Burkholderia multivorans, to lung epithelial cells (16HBE14o(-)) using real-time PCR with a view to monitoring potential inhibition of lung cell attachment. Mammalian and bacterial DNA were purified from bacteria attached to lung epithelial cells. The relative amount of bacteria attached was determined by amplification of the recA gene relative to the human GAPDH gene, in the presence of SYBR Green. The method was thoroughly validated and shown to correlate well with traditional plating techniques. Inhibition of bacterial attachment with simple sugars was then evaluated by real-time …

Macrophage Responses To Cf Pathogens: Jnkmap Kinase Signaling By Burkholderia Cepacia Complex Lipopolysaccharide, Suzanne Mckeon, Siobhan Mcclean, Máire Callaghan

Articles

Chronic bacterial colonization of the airways with opportunistic pathogens is the primary cause of morbidity and mortality in cystic fibrosis (CF) patients. Burkholderia cepacia complex (Bcc) organisms pose a particular challenge in CF lung disease, due in part to their ability to trigger a fulminant pneumonia. This study compares the U937 macrophage response to two Bcc species, B. cenocepacia and Burkholderia multivorans, against Pseudomonas aeruginosa and Staphylococcus aureus. The two Bcc strains demonstrated higher levels of U937 macrophage internalization compared with both P. aeruginosa and S. aureus. Both the Bcc strains also stimulated significantly greater levels of tumor necrosis factor-a …

The Planned And The Unplanned: A Roundtable Discussion On The Legacies Of Michel Foucault And Norbert Elias, Sam Binkley, Paddy Dolan, Stefanie Ernst, Cas Wouters

Articles

When one considers the proximity of their concerns, it is perhaps surprising that the works of Norbert Elias and Michel Foucault have not been more systematically compared and discussed. However, the differentiation of disciplinary knowledge (particularly the boundary that separates philosophy from social theory), com-pounded by parochialisms fostered by the cult of the intellectual, have delayed this process far past its due. This conversation, which began in 2008 at a conference on the works of Elias and Foucault at the University of Hamburg, is, in this regard, an effort to make up for lost time. Fashioned from hours of discussion …

Reactive Oxygen Species Induced Cytokine Production And Cytotoxicity Of Pamam Dendrimers In J774a.1 Cells, Pratap Naha, Maria Davoren, Fiona Lyng, Hugh Byrne

Articles

The immunotoxicity of three generations of polyamidoamine (PAMAM) dendrimers (G-4, G-5 and G-6) was evaluated in mouse macrophage cells in vitro. Using the Alamar blue and MTT assays, a generation dependent cytotoxicity of the PAMAM dendrimers was found whereby G-6 > G-5 > G-4. The toxic response of the PAMAM dendrimers correlated well with the number of surface primary amino groups, with increasing number resulting in an increase in toxic response. An assessment of intracellular ROS generation by the PAMAM dendrimers was performed by measuring the increased fluorescence as a result of intracellular oxidation of Carboxy H2DCFDA to DCF both quantitatively using …

Ablation Of Chronic Total Occlusions Using Kilohertz-Frequency Mechanical Vibrations In Minimally Invasive Angioplasty Procedures., Garrett Mcguinness, Mark Wylie, Graham Gavin Jan 2010

Ablation Of Chronic Total Occlusions Using Kilohertz-Frequency Mechanical Vibrations In Minimally Invasive Angioplasty Procedures., Garrett Mcguinness, Mark Wylie, Graham Gavin

Articles

Certain minimally invasive cardiology procedures, such as balloon angioplasty and stent implantation, critically require that the site of an arterial blockage be crossed by an intraluminal guidewire. Plaques resulting in near or totally occluded arteries are known as chronic total occlusions (CTOs), and crossing them with conventional guidewires is a significant challenge. Among the most promising proposed solutions is the delivery of high power, low frequency ultrasonic vibrations to the occlusion site via an intraluminal wire waveguide. The vibrating distal-tip of the ultrasound wire waveguide is used to transmit energy to the surrounding plaques, tissues and fluids in order to …
