This thesis examines the build-up to and the events of the First Barbary War (1801-1805) with the purpose of demonstrating that ideological factors significantly influenced the decision to go to war – specifically the republican ideology of Thomas Jefferson and his belief that this ideology represented the essence of the United States and its Revolutionary heritage. As such, Jefferson will be the main focus of this analysis. This focus is justified because it is Jefferson who broke with the policies set by his predecessors and directly engaged the Barbary States with military force. Bone Formation Around Implants In Adult Transgenic Mice With Selective Runx2-Ii Deficiency, Jaime Rodrigo Rivera

Titanium-aluminum-vanadium (Ti-6Al-4V) tacks were placed in the tibiae of 10 female wild type (Runx2 flox/+ or Runx2 flox/flox) and heterozygous (Runx2+/-, ColII Cre) mice littermates to examine healing and bone response. Animals were divided into control and test groups of 5 each. A novel generation of Runx2 knock-out mice where the gene is only deleted in chondrocytes was used. Ten days after implant surgery, specimens were recovered and prepared for histological, histomorphometric, and micro-CT morphometric analysis. Histological examination revealed that wild type animals presented larger regions of bone formation and endochondral ossification when compared to heterozygous mice. Micro-CT morphometric analysis …

A variety of reptiles have temperature-dependent sex determination (TSD), where incubation temperature determines gender. This form of sex determination has significant conservational implications because it has the potential of producing highly biased sex ratios which can affect the reproductive output of a population. TSD is of particular interest to conservation programs that use hatcheries to artificially incubate the eggs of endangered sea turtles because such programs need to select specific incubation temperatures in an effort to produce a desired sex ratio. Over the years, various roles for neurotrophins have been revealed, being initially described as survival signals for neurons making their initial synaptic contacts in the developing brain. Eventually, it was discovered that these molecules also play important modulatory roles in the adult brain. The work encompassed in this dissertation serves to characterize one particular neurotrophin, brain-derived neurotrophic factor (BDNF), which has been implicated in long-term potentiation (LTP), and its effects at CA3-CA1 synapses in the hippocampus following acute application in vitro. Sortases are transpeptidases that covalently link surface proteins to the stempeptide on the cell wall peptidoglycan of Gram-positive bacteria. This work endeavored to characterize the various sortase enzymes and their effect on the virulence of Streptococcus pneumoniae. The genome of S. pneumoniae may encode up to four sortase enzymes. We found that 38 of 54 strains of serotype 6B clonal complex 14 (CC14) carry the genes for the secondary sortases srtB, srtC, and srtD. The presence of these sortases did not seem to influence the ability of strains within CC14 to infect the nasopharynx, lungs, or blood of strains within …

The grassroots path to public policy is characterized by citizen pressure on policy makers to produce desired change and has been especially successful in the realm of natural resources. It has proven successful in the obtainment of various local and state level policy to control light pollution in the United States. During the fall of 2005 and spring of 2006, habitat assessments and rapid bioassessments were conducted at six sites on 100 m reaches on Five Mile Creek in Jefferson County, Alabama and two sites on neighboring rural streams that were used as references. Habitat scores and index of biotic integrity (IBI) scores for the fish communities were calculated for all sites. The IBI scores on Five Mile Creek did not compare well with one of the reference streams and suggest impairment. The habitat scores for the Five Mile Creek sites were similar to the scores of the reference streams. The …

Lymphocytes are highly mobile cells that can travel throughout the body in response to a multitude of stimuli. Identifying lymphocyte trafficking patterns in vivo is essential for a complete understanding of immune function. Cell-cell and cell-tissue interactions in immune development and in lymphocyte response to stimuli can be comprehended with these investigations. Although the location of cell populations in various tissues at any given point in time may be investigated by techniques such as flow cytometry and immunofluorescence, these methods are not easily used in the assessment of dynamic cell migration patterns in vivo. According to the Central Brain Tumor Registry of the United States, the most common primary brain tumors are gliomas, tumors composed of cells of glial origin, most commonly astrocytes and oligodendrocytes. The most aggressive of these tumors are characterized by hyperproliferation, marked cellular and nuclear atypia, extensive infiltration into surrounding normal brain tissue, and large areas of cell and tissue death. Ischemic tissue and organ dysfunction occur in a variety of cardiovascular diseases. The hypoxic environment created during ischemic injury activates cytoprotective genes, of which heme oxygenase-1 (HO-1) is of great interest. This microsomal inducible enzyme is involved in the homeostasis of prooxidant heme, which is released from heme proteins upon injury. HO-1 responds to a variety of oxidative stress associated stimuli. HO-1 expression has been implicated as a protective response in diverse disease states which has been attributed to the beneficial properties of the by products iron, carbon monoxide and biliverdin/bilirubin. O-glycosylation, a type of posttranslational modification, is abundant and dynamic in the cell, yet its function in cell signaling and destiny regulation is far from known. This study examines the effects of O-glycosylation on muscle differentiation and metabolism, and the potential mechanism behind the effects: proteasome blockade. Proteasome action is required for muscle differentiation. We previously have shown that O-GlcNAc modification of the 19S cap of the proteasome can inhibit the proteolytic function of this organelle. By changing O-GlcNAc modification levels and proteasome function independently in C2C12 skeletal myoblasts, we have found that myoblast differentiation is largely dependent on both. …

Gender Bending And Comic Books As Art: Issues Of Appropriation, Gender, And Sexuality In Japanese Art, Harley Blue Acres

In Japanese high art, popular art (such as comic books and animation), theater, and music, representation of gender is a blurred line that is commonly crossed, more so than in the West. I argue that this fluidity is the result of disparate artistic sources that have filtered through Japanese culture since the Edo Period (1603-1867) with the flourishing of Kabuki Theater (in which men played women on stage) and the emergence of ukiyo-e (particularly shunga/erotica prints). This artistic lineage impacted arts of the Showa Period (1926-1989) notably Takarazuka Theater (which feature an exclusively female cast that portrays men) and the …
