Prostate cancer is the most common male malignancy and the second leading cause of cancer related death in the United States. Despite recent advances in the detection, diagnosis, and treatment of prostate cancer, there is a need for more effective therapies. Unfortunately, most conventional therapeutic modalities, such as androgen ablation therapy, frequently result in androgen-independent cancers. These cancers are typically more aggressive, metastatic, and resistant to chemotherapeutic agents than androgen-dependent prostate cancer. Therefore, agents that are effective against both androgen-sensitive and androgen-independent, as well as genetically diverse cancers are critically needed. The objective of the dissertation research was to address …
Identification And Validation Of Biodosimetry Markers In Multiple Models Of Radiation Exposure, Ashley Nicole Filiano
Identification And Validation Of Biodosimetry Markers In Multiple Models Of Radiation Exposure, Ashley Nicole Filiano
All ETDs from UAB
Following a radiological or nuclear disaster, radiation dose assessment is imperative to minimize morbidity and mortality through rationally directed medical intervention. Current methods of retrospective dosimetry are not amenable to mass exposure scenarios and remain limited to monitoring of clinical symptoms (nausea/vomiting and lymphocyte depletion) and cytogenetic analysis. The goal of this study was to identify radiation biomarkers capable of qualitative (non-irradiated/irradiated) and quantitative (dose) assessment of radiation exposure. Initial analyses revealed 17 radiation-responsive cytokine/chemokine genes in blood samples from 6 pediatric cancer patients undergoing fractionated total body irradiation (TBI). These 17 genes were combined with 29 additional IR-inducible genes …

All ETDs from UAB
Exposure to air pollutants are known to induce airway inflammation, decrease lung function, and worsen existing pulmonary conditions such as asthma. Additionally, air pollutant exposure has also been shown to impact cardiovascular health. Although these pollutant-induced cardiovascular effects were considered to be mediated by the direct action of pollutants upon the cardiovascular tissue and/or the indirect release of pro-inflammatory factors into the circulation, the relative contribution of the two pathways remained equivocal. Recently, several epidemiology studies associated ozone exposure with cardiovascular mortalities. However, relatively few studies to date have examined ozone in this context. In order to address our hypothesis …

Sir Thomas Wyatt's Rondeaux: Early Modern Perception During And After The English Reformation, Anne Begin Myer
All ETDs from UAB
This paper examines the implications of Sir Thomas Wyatt's use of the rondeaux during the English Reformation and the reception and editorial emendations of that French poetic form in subsequent anti-Reformation publications. The first chapter, "Contemporary Publications and Controversy," explores the two texts by which Wyatt's works would have been disseminated to the public, namely, Tottel's Songes and Sonettes (or Miscellany) and The Court of Venus. In addition to these texts, I also consider the relationship between Wyatt and his deeply Protestant patrons, Sir Thomas Cromwell and Anne Boleyn, and how these relationships would have influenced the public perception of …
Cgmp Pde As A Novel Molecular Target For The Prevention And Treatment Of Breast Cancer, Heather Nicole Tinsley
Cgmp Pde As A Novel Molecular Target For The Prevention And Treatment Of Breast Cancer, Heather Nicole Tinsley
All ETDs from UAB
Breast cancer remains a major health concern, despite efforts to develop improved therapeutics. Chemoprevention is a promising strategy for reducing breast cancer-related morbidity and mortality. However, with the exception of the selective estrogen receptor modulators (SERMs), which have limited efficacy and severe toxicities, no drugs have been approved for breast cancer chemoprevention. Studies demonstrate that certain nonsteroidal anti-inflammatory drugs (NSAIDs) display promising chemopreventive efficacy. Unfortunately, the depletion of physiologically important prostaglandins due to inhibition of the cyclooxygenase (COX) enzymes results in potentially fatal toxicities, which exclude the use of NSAIDs and COX-2 selective inhibitors for chemoprevention. Ample data, however, suggest …

All ETDs from UAB
The ubiquitin proteasome system (UPS) is a coordinated process by which the cell can control protein distribution and abundance. Proteins are marked for turnover by the construction of a polyubiquitin chain on the protein substrate. Once engaged by the proteasome, the ubiquitin side-chain is disassembled by proteasomal deubiquitinating enzymes (DUBs), preventing entry of ubiquitin into the proteasome and recycling it for use in future reactions. One of the DUBs that resides on the proteasome is Ubiquitin Specific Protease 14 (Usp14), which is mutated in the ataxia (axJ) mice. This mutation results in pronounced physical deficits and, unexpectedly, a deficit in …

Small Molecule Inhibitors Of Acid Sensing Ion Channel-1, Yawar J. Qadri
All ETDs from UAB
Acid Sensing Ion Channel 1 is one of the many proteins in the Epithelial Sodium Channel/Degenerin family. The proteins in this family interact to form cation channels with unique biophysical properties and can all be inhibited by the small molecule amiloride. Their expression in many different cell types underlies their involvement in a large variety of physiological and pathophysiological processes. ASIC-1 containing channels, specifically, are an important therapeutic target for ischemic stroke, nociception, the invasiveness of glioblastoma cells, and many other processes including anxiety and memory formation. Of the members of the ENaC/Deg family, chicken ASIC-1 is the only protein …

Overexpression Of Garp2 Increases Phototransduction Gain., Shanta Sarfare
All ETDs from UAB
The cGMP-gated cation channel (CNG) in the rod photoreceptors is an essential component of phototransduction, and may have a role in maintaining structural integrity of the rod outer segment (ROS). The ß-subunit of the CNG channel (CNGB1) in the rod photoreceptors is comprised of an N-terminal glutamic acid-rich region (GARP') and a C-terminal channel-like region. Additionally, two soluble proteins GARP1 and GARP2 are alternatively spliced from the CNGB1 gene. Although GARPs are known to interact with other proteins in the rod photoreceptors, their role is currently unknown. To determine the role of GARP2 in the rod photoreceptors, we generated transgenic …

The Comparison Of Mitochondrial Functions Of 2 Drosophila Species: Drosophila Ananassae And Drosophila Subobscura, Hye J. Shin
All ETDs from UAB
Recently, it has been suggested that differences in mitochondrial function enabled Homo sapiens to migrate out of Africa by enabling humans to adapt to colder environments as they migrated to higher latitude areas. Through mitochondrial mutations that partially uncoupled oxidative phosphorylation, humans that moved to colder areas were able to generate more heat for better survival, while producing less energy. However, due to challenges in working with human populations, no specific studies have been able to show the existence of such differences in mitochondrial function within healthy humans. Because of numerous advantages of Drosophila as a developmental model, such as …

Association Between Birth Outcomes And Aflatoxin B1-Biomarker Levels Among Pregnant Women In Kumasi, Ghana, Faisal Mohammed Barbas Shuaib
All ETDs from UAB
Aflatoxins are fungal metabolites that contaminate staple food crops in many developing countries. Although studies have linked these toxins to adverse health outcomes, few studies have investigated the association between birth outcomes and aflatoxin B1-lysine adduct levels among pregnant women. The objective of this dissertation was to investigate the association between birth outcomes and aflatoxin B1- lysine adduct levels among pregnant women in Kumasi, Ghana. In a cross-sectional study of 755 pregnant women, Aflatoxin B1-lysine adduct (AF-ALB) levels were determined by High Performance Liquid Chromatography. AF-ALB associations were assessed as both continuous and categorical values. Analysis of variance (ANOVA) was …

Investigation Of Residues Of The Vesicular Stomatitis Virus Nucleocapsid Protein That Affect Transcription And Rna Replication, Djamila Harouaka
All ETDs from UAB
The template for transcription and RNA replication for vesicular stomatitis virus (VSV) and other negative-strand RNA viruses is a ribonucleoprotein (RNP) complex consisting of the viral RNA genome and associated nucleocapsid (N) protein. The structure of the RNP-like complex of VSV showed that the RNA is sequestered between two lobes of the N protein, and adjacent N monomers are linked by an extensive network of interactions. Long-range interactions mediated by the N-terminal arm and C-terminal loop of the N protein stabilize the side-to-side contacts between adjacent N monomers, and are required for RNA encapsidation. It is unclear how the polymerase …
H.D.'S Incantations: Reading Trilogy As An Occultist's Creed, Sarah Grace Harrell
H.D.'S Incantations: Reading Trilogy As An Occultist's Creed, Sarah Grace Harrell
All ETDs from UAB
The focus of this thesis is H.D.'s epic poem, Trilogy, composed during 1942-1944, while she was living in World War II London. This project's primary aim is to investigate how the poet's participation in occult spiritual practices impacts the work's language. This reading examines terms, concepts, and ritualistic language from a range of esoteric traditions. While H.D. was involved in a wide range of occult practices, the specific traditions with which this investigation is mainly concerned are: astrology, spiritualism, Kabbalism, and gnostic Christianity. Explicating Trilogy exclusively through this lens places significantly more importance on the poem's metaphysical, historical, and archeological …

O-Glcnac Transferase Modulates Jnk1 And Foxo4 Transcription Factor To Resist Acute Oxidative Stress, Shiuh-Rong Ho
All ETDs from UAB
O-GlcNAcylation is an abundant and dynamic post-translational modification on serine and threonine residues of nuclear and cytoplasmic proteins. O-GlcNAc Transferase (OGT) and Nuclear Cytoplasmic O-GlcNAcase and Acetyltransferase (NCOAT) are the only two enzymes and major regulators in this process. In the past two decades, many studies have demonstrated its important modulation roles in nutrient sensing, protein turnover, cell cycle progression, transcription, translation as well as stress resistance. Among these biological functions, the understanding of O-GlcNAcylation dependent oxidative stress is relatively unknown. This study aims to examine the effects of O-GlcNAcylation on JNK1 and FOXO4 transcription factors under acute oxidative stress …
Role Of Epithelial Sodium Channel And Acid Sensing Ion Channel In Glioblastoma Multiforme, Niren Kapoor
Role Of Epithelial Sodium Channel And Acid Sensing Ion Channel In Glioblastoma Multiforme, Niren Kapoor
All ETDs from UAB
Glioblastoma Multifrome is the most common and aggressive of the primary brain tumors. These tumors express multiple members of the Epithelial Sodium Channel (ENaC)/Degenerin (Deg) family, associated with a basally active amiloride sensitive cation current. We hypothesize that this glioma current is mediated by a hybrid channel composed of a mixture of Epithelial Sodium Channel (ENaC) and Acid Sensing Ion Channel (ASIC) subunits. To test this hypothesis we made dominant negative cDNAs for ASIC1, αENaC, and γENaC. D54-MG glioma cells transfected with the dominant negative constructs for ASIC1, αENaC, or γENaC showed reduced protein expression for each of the specific …

Young Children's Ways Of Playing "Concentration" (A Card Game), Tsuguhiko Kato
All ETDs from UAB
To explain why preschool children play a card game, Concentration illogically, 108 three-to-six-year-olds from a child care center and an elementary school in a Southern state were interviewed individually. At the beginning of the game, children were asked to match seven pairs of cards. The pairs were then separated into two rows each consisting of seven different cards. After the cards were turned face down and mixed within each row, three-year-olds often played Concentration by selecting two cards from the same row rather than one card from each row. On the basis of Piaget's theory, I hypothesized that children's low …

Characteristics Of Fundraising For Nonprofit Hospital Organizations, Cathleen Owens Erwin
All ETDs from UAB
This study addresses a gap that has been noted in the charitable fundraising literature, which is a scarcity of theoretically-based studies of fundraising by nonprofit organizations and the need to study fundraising within sub-sectors of organizations rather than the full nonprofit sector. The study uses institutional theory and strategic management constructs to examine the characteristics of the fundraising operations of nonprofit organizations. A categorization scheme is created based on nonprofit organizational effectiveness and performance characteristics to analyze fundraising by nonprofit hospital organizations as an organizational field. The categorization scheme identifies distinct clusters of organizations that are then investigated to determine …
