All ETDs from UAB

Traumatic Brain Injury (TBI) and Posttraumatic Stress Disorder (PTSD) have been referred to as "signature injuries" among veterans of Operation Iraqi Freedom (OIF) and Operation Enduring Freedom (OEF). The aims of the current study were to identify demographic and injury related variables associated with the co-occurrence of mild Traumatic Brain Injury (mTBI) and PTSD and to identify neuropsychological assessment measures that differentiate individuals with mTBI from those with co-occurring mTBI and PTSD. A total of 81 OIF/OEF veterans participated in this study comprising the mTBI (n=21) and combined (n=60) groups. The results of logistic regression analyses indicated that Caucasians were …

Mechanism Of Nfi-C Functionin During Tooth Root Formation, Ejvis Lamani Jan 2012

Mechanism Of Nfi-C Functionin During Tooth Root Formation, Ejvis Lamani

All ETDs from UAB

Currently, very little is known regarding tissue-specific gene regulation during the later stages of tooth development, especially those associated with root formation. Re-cent studies have identified nuclear factor I-C (NFI-C) as a critical factor for root for-mation in mammals. NFI-C functions as a cellular transcription factor and adenovirus DNA replication factor. Its role in root formation first became evident when Nfi-c null mice were shown to lack molar roots. A similar phenotype of incomplete or absent root formation is observed in patients with the autosomal dominant disease Radicular Dentin Dysplasia (RDD; MIM125400) also known as Rootless Teeth or Dentin Dysplasia …

Development Of An Adenoviral Vector Platform For Selective Gene Delivery To Neurons, Travis Benjamin Lewis Jan 2012

Development Of An Adenoviral Vector Platform For Selective Gene Delivery To Neurons, Travis Benjamin Lewis

All ETDs from UAB

Parkinson disease (PD) is a devastating movement disorder affecting between 500,000 and 1,000,000 individuals in the United States alone, and for which there is currently no cure. Therapies such as dopamine replacement help initially, but with disease progression these lose efficacy and do not alter the underlying degenerative process. Gene therapy is the delivery of exogenous genetic material to correct an underlying disorder. A major advantage of this approach is the ability to distribute potent therapies to specific targets over an extended duration, properties necessary for the treatment of progressive degenerative diseases such as PD. A number of clinical trials …

The Role Of The Mirror Neuron System In Mental State Attribution In Autism: An Fmri Study, Lauren E. Libero Jan 2012

The Role Of The Mirror Neuron System In Mental State Attribution In Autism: An Fmri Study, Lauren E. Libero

All ETDs from UAB

The ability to interpret others' intentions based on their actions is a vital skill that helps us infer and understand their thoughts and plans. However, individuals with autism have been found to have difficulty in attributing intentions to others (Baron-Cohen et al., 1985; Baron-Cohen, 1995; Happe & Frith, 1996; Williams et al., 2001). The present study investigated the neural mechanisms of inferring intentions from actions in high-functioning adults with autism spectrum disorders (ASD) and typically developing control participants. fMRI data were acquired while participants made judgments about the means (how an action is performed) and intentions (why an action is …

Regulation Of P27kip1 By Trip6 And Its Implications In Cancer Progression, Victor T G Lin Jan 2012

Regulation Of P27kip1 By Trip6 And Its Implications In Cancer Progression, Victor T G Lin

All ETDs from UAB

TRIP6 is a focal adhesion molecule that functions as an adaptor protein to mediate diverse cellular functions, including motility and antiapoptotic signaling, through a wide variety of protein-protein interactions. We were able to characterize a novel interaction between TRIP6 and p27KIP1, a CDK inhibitor which can both suppress and promote tumorigenesis, depending on its regulation. TRIP6 expression specifically promotes the AKT-mediated phosphorylation of p27KIP1 at T157, which leads to its cytosolic retention, and accelerates the protein turnover rate of p27KIP1. We also found that phosphorylation of T157 by itself promotes cell motility and can be linked to the formation of …

Multiscale Modeling Of The Mouse Femur, Shannon Lisenbee Jan 2012

Multiscale Modeling Of The Mouse Femur, Shannon Lisenbee

All ETDs from UAB

This paper seeks to advance the knowledge base for bones and how they respond to mechanical stimuli through the use of multiscale modeling. First, a simple but effective approach for segmenting bone from computed tomography images using MATLAB is presented. Using edge detection and gradient operators to identify areas of bone within an image allows for much more accurate and consistent bone segmentation than simpler techniques like thresholding. This technique is applied to the femur of a female mouse for use in further modeling and testing. Next a representative volume element (RVE) for lamellar cortical bone is created based on …

The Effect Of Acetylcholine Receptor Activation On The Response Properties Of Retinal Ganglion Cells, Ye Long Jan 2012

The Effect Of Acetylcholine Receptor Activation On The Response Properties Of Retinal Ganglion Cells, Ye Long

All ETDs from UAB

Acetylcholine (ACh) is the first identified neurotransmitter and acts in areas including the peripheral nervous system (PNS) and the central nervous system (CNS). The effects of ACh are mediated by nicotinic and muscarinic acetylcholine receptors (AChRs), both of which are extensively distributed in CNS, including the retina. The expression and distribution of nicotinic AChRs (nAChRs) and muscarinic AChRs (mAChRs) have been studied with RT-PCR, western blot, and immunohistochemical techniques in the retinas of different species (Kaneda et al., 1995; Keyser et al., 2000; Yamada et al., 2003; Strang et al., 2005; Dmitrieva et al., 2007; Liu et al., 2009; Strang …

The Effects Of Venlafaxine On Functional Recovery Following Spinal Cord Injury, Don E. Mccormick Jan 2012

The Effects Of Venlafaxine On Functional Recovery Following Spinal Cord Injury, Don E. Mccormick

All ETDs from UAB

Spinal cord injury (SCI) can significantly alter a person's physical and psychological health over their lifetime. Depression in persons with SCI is often treated with antidepressants which alter serotonergic and adrenergic signaling, both regulators of plasticity. However, the effects of antidepressants functional recovery after SCI have never been evaluated. Therefore, we hypothesize that antidepressant therapy would alter functional recovery and neuropathic pain behaviors in a SCI rodent model. The aim of this study was to evaluate the effects of venlafaxine (VEN) administration on functional recovery in the rehabilitation setting using a SCI rodent model. Adult male Sprague-Dawley rats received a …

Using Mouse Models To Investigate Ciliopathies, Raymond Carl Pasek Jan 2012

Using Mouse Models To Investigate Ciliopathies, Raymond Carl Pasek

All ETDs from UAB

Cilia and flagella are microtubule based organelles found on nearly every cell type in the mammalian body. Flagella have an essential role in sperm locomotion and reproduction, while motile cilia have a known role in fluid movement in the brain, lungs, and portions of the female reproductive tract. While the role of most primary cilia remained unknown for decades, they are now recognized as being essential to mammalian development, renal homeostasis, and satiety, just to name a few. Because of their widespread presence and importance, dysfunction of cilia and flagella can lead to a variety of human diseases, collectively referred …

Targeting Mitochondrial Function In Diabetic Nephropathy, Colin Reily

All ETDs from UAB

TARGETING MITOCHONDRIAL FUNCTION IN DIABETIC NEPHROPATHY The incidence of diabetes has increased to epidemic proportions over the last 50 years, due to a complex interaction of lifestyle and dietary factors. Changes in physical activity, caloric intake, and the composition of foods consumed over the last century are thought to interact with metabolic syndrome prone subpopulations leading ultimately to diabetes. It is widely accepted that chronic hyperglycemia in the blood characterizes the diabetic condition. Since the discovery of insulin therapy in the first half of the 20th century, diabetics have extended their lifespan considerably, but at the cost of increased morbitity …

Cystine/Glutamate Transporters As Prognostic & Therapeutic Markers Of Primary Brain Tumors, Stephanie Marie Robert

All ETDs from UAB

Glioblastoma multiforme (GBM) are the most prevalent and aggressive malignant brain tumors. Current treatment - a combination of radiation, chemotherapy and resection - has limited effectiveness and offers poor prognosis. In this study, we examined the roles of system xc- (SXC) and excitatory amino acid transporters (EAATs), which transport the amino acids cystine and glutamate, on tumor growth, neurotoxicity, and peritumoral seizure activity. Tissue micro-arrays from 45 patients were examined by immuno-histochemistry, comparing tumor-bearing tissue and adjacent normal brain. Using a novel flank tumor propagation technique, we chose 3 glioma samples with varying SXC and EAAT expression levels to study …

Connexin 43 Remodeling Contribute To Slowed Conduction Velocity In An Irreversible Arrhythmogenic Canine Model Of Non-Ischemic Heart Failure, Jiajie Yan Jan 2012

Connexin 43 Remodeling Contribute To Slowed Conduction Velocity In An Irreversible Arrhythmogenic Canine Model Of Non-Ischemic Heart Failure, Jiajie Yan

All ETDs from UAB

Heart failure is associated with a high mortality due to ventricular tachycardia leading to ventricular fibrillation that could arise from impaired conduction velocity. Myocardial conduction relies on intercellular coupling maintained primarily by the gap junction protein connexin43. The objective of this research is to assess the correlation between connexin43 remodeling and altered conduction in our novel irreversible and arrhythmogenic HF large animal model that mimics human heart failure. A novel heart failure canine model was established with combined strategy of creating aortic insufficiency and aortic constriction. We measured conduction velocity in heart failure and control left ventricular epicardium (n=5, 5) …

Transcriptional Regulation Of Gfap, Sujeong Yeo Jan 2012

Transcriptional Regulation Of Gfap, Sujeong Yeo

All ETDs from UAB

Glial fibrillary acidic protein (GFAP) is an astrocytic intermediate filament protein whose levels are increased in response to CNS injuries. Aim one of my thesis is to find the regulatory elements of the GFAP promoter responsible for its expression properties using a transgenic approach, since previous studies showed inconsistent results between in vitro and in vivo approaches. Two contiguous DNA segments previously implicated in transcriptional control of the GFAP gene, the B region (bp -1612 to -1489), and the C1.1 and C1.2 regions (bp -1488 to -1399), were analyzed. Block mutation of each of 4 contiguous sequences together spanning the …

The Role Of Histone Methylation In The Medial Temporal Lobe During Long-Term Memory Formation, Swati Gupta Jan 2012

The Role Of Histone Methylation In The Medial Temporal Lobe During Long-Term Memory Formation, Swati Gupta

All ETDs from UAB

Several studies have established the importance of epigenetic regulation of gene transcription in the brain during memory consolidation. Epigenetic mechanisms involve modifying the DNA or the associated histone proteins to determine the transcriptional outcome of a gene. The work encompassed in this dissertation serves as an initial investigation for the role of histone lysine methylation mechanisms in regulating gene activation and suppression in the medial temporal lobe that includes entorhinal cortex (EC), hippocampus, and amygdala during memory consolidation. To this end, we found that di-methylation of histone H3 at lysine 9 (H3K9me2) which promotes gene suppression was specific to associative …

Neural Networks Involved In Eye Movements In Humans, Demet Gurler

All ETDs from UAB

Saccades, smooth pursuit and vergence eye movements either shift or maintain gaze on objects of interest so that the image of the object is, respectively, either brought to, or maintained on the fovea for detailed analysis. To date, the neural substrates of these eye movements have been characterized individually using a variety of techniques including electrophysiology, positron emission tomography (PET) and functional magnetic resonance imaging (fMRI) both in non-human primates (NHPs) and humans. The non-invasive nature of fMRI allows us to use the same tasks and techniques in both humans and nonhuman primates. The purpose of the present study was …

The Role Of Vpr-1 Msp Signaling In C. Elegans, Sung Min Han Jan 2012

The Role Of Vpr-1 Msp Signaling In C. Elegans, Sung Min Han

All ETDs from UAB

In C. elegans, the major sperm protein (MSP) is the most abundant protein in sperm and functions as an intracellular cytoskeleton protein and a secreted extracellular signal that induces oocyte maturation and sheath contraction. Evolutionarily, MSP domain-containing proteins originate from the VAPs (VAMP/synaptobrevin-associated proteins), which comprise a highly conserved protein family with an MSP domain at the N-terminus and a transmembrane domain at the C-terminus. I have been studying the signaling role of VAP MSP domains using the C. elegans model system. In collaboration with Hugo Bellen's lab at Baylor College of Medicine, we have shown that C. elegans, Drosophila, …

All ETDs from UAB

Ultrasound is a noninvasive, inexpensive imaging modality used to characterize biological tissues. Targeted contrast-enhanced ultrasound imaging is an emerging preclinical strategy which uses contrast agents for characterization of biological processes on a molecular level. These microbubble agents are lipid shelled micron-sized particles that have the capability to be coated with ligands such as antibodies or small peptides in order to bind to their target receptors. Molecular ultrasound imaging, using these targeted contrast agents, allows for a quantitative assessment of molecular targets with a high sensitivity. Although this technique is in preclinical research, further advances in contrast agent design and potential …

Mitochondrial Morphology And Function In Neuronal Cells Under Stress, Lonnie Schneider Jan 2012

Mitochondrial Morphology And Function In Neuronal Cells Under Stress, Lonnie Schneider

All ETDs from UAB

Neurodegenerative disease encompasses a wide range of conditions and pathologies that can manifest at any age depending on the etiology. A major factor in both early onset and age-related neurodegeneration is mitochondrial dysfunction. To investigate how mitochondrial bioenergetics is affected by cellular stress, we used an in vitro culture system to examine mitochondrial function in response to oxidative stress. We also studied an in vivo model of neuronal ceroid lipofuscinosis to determine the impact of deficient autophagy-lysosomal activity on mitochondrial morphology, composition and function. In vitro we found that retinoic acid-induced differentiation of dopaminergic neuroblastoma SH-SY5Y cells exhibited increased mitochondrial …

Early-In-Life Bladder Inflammation Alters Kappa-Opioid Modulation Of Inflammatory Bladder Pain, Amber Dianne Shaffer Jan 2012

Early-In-Life Bladder Inflammation Alters Kappa-Opioid Modulation Of Inflammatory Bladder Pain, Amber Dianne Shaffer

All ETDs from UAB

In adult female rats, zymosan-induced inflammation of the bladder leads to bladder hypersensitivity, as reflected in augmented visceromotor reflex (VMR) responses to urinary bladder distension (UBD). Prior early-in-life (EIL) experience with bladder inflammation further increases bladder hypersensitivity. Naloxone increases bladder hypersensitivity in animals inflamed only as adults, but not in animals inflamed EIL, indicating that bladder inflammation recruits an inhibitory opioid system which may be impaired by EIL inflammation. This thesis examined what role the dynorphin/κ-opioid system played in mediating bladder hypersensitivity through the impairment of opioid inhibition. In all experiments, animals were treated EIL (P14-P16) and as adults with …

Innate Immune Mechanisms Against The Atypical Fungal Pathogen Pneumocystis Murina, Michael Paul Nelson Jan 2012

Innate Immune Mechanisms Against The Atypical Fungal Pathogen Pneumocystis Murina, Michael Paul Nelson

All ETDs from UAB

Pneumonia caused by the fungal pathogen Pneumocystis continues to be the leading cause of morbidity and mortality in AIDS patients. In addition, there are a number of other non-HIV-associated diseases that render an individual susceptible to Pneumocystis infection, including chronic obstructive pulmonary disease. Innate immune mechanisms are important for controlling Pneumocystis lung infection. As alveolar macrophages serve the front lines of lung host defense, our goal was to elucidate their responses during lung Pneumocystis infection. Here, we established a role for Src family kinases in the lung immune response to Pneumocystis, which uncovered a role for alternatively activated alveolar macrophages. …

Electrophysiological Investigation Of Rabbit Retinal Ganglion Cells, Przemyslaw Nowak Jan 2012

Electrophysiological Investigation Of Rabbit Retinal Ganglion Cells, Przemyslaw Nowak

All ETDs from UAB

The retina is the first stage of the visual system, responsible for transducing light into the neural signal and for subsequent processing of this signal before sending it to the higher visual centers in the brain. The output to the higher visual centers is mediated by retinal ganglion cells, which not only relay the signal, but also substantially contribute to its processing. Depending on what computations they perform, they are subdivided into different types. These cells have been extensively studied in various spices, one prominent example being the rabbit. In rabbits, one characteristic type comprises On-Off directionally selective retinal ganglion …

Association Between Cc Chemokine Ligand 3-Like-1 (Ccl3l1) Gene Copy Number And Rheumatoid Arthritis In African Americans, Mawuli Kwame Nyaku

All ETDs from UAB

Gene copy number of the CC Chemokine ligand 3-Like-1 (CCL3L1) located on chromosome 17 position q12 varies between ethnicities. Previously, gene copy number of CCL3L1 greater than the ethnic median copy has been associated with an increased risk of Rheumatoid Arthritis. Three later studies found no association between CCL3L1 and RA. All studies were conducted in non-African Americans and an objective of this study was to determine this association in an African American cohort. CCL3L1 shares significant homology with three other genes within the same cluster; CC Chemokine ligand 3 (CCL3), CC Chemokine ligand 3-Like-2 (CCL3L2) and CC Chemokine ligand …

Post-Transcriptional Control Of Type I Interferon Genes By Ksrp In The Innate Immune Response Against Viral Infection, Wei-Jye Lin Jan 2011

Post-Transcriptional Control Of Type I Interferon Genes By Ksrp In The Innate Immune Response Against Viral Infection, Wei-Jye Lin

All ETDs from UAB

Several strategies are used in eukaryotic cells in order to determine the gene expression level, including the transcriptional and post-transcriptional mechanisms. One of the post-transcriptional mechanisms is the regulation of mRNA stability, which varies considerably from one mRNA species to another. Previous studies have shown that inherently unstable mRNAs encoding cytokines and proto-oncogenes contain the AU-rich elements (AREs) in the 3' untranslated regions (UTRs). Their decay requires decay-promoting RNA-binding proteins that recognize the AREs and recruit mRNA decay machineries to facilitate the rapid decay of targeting mRNA. KH-type splicing regulatory protein (KSRP) was previously identified as an ARE decay promoting …

Mining Canonical Views From Internet Image Collections, Lin Yang Jan 2011

Mining Canonical Views From Internet Image Collections, Lin Yang

All ETDs from UAB

Recent years have seen an explosion of internet image collections. The explosion was brought about by the popularity of photo sharing sites such as Flickr and Picasa Web Albums, where millions of internet users upload their personal photos online and share these photos with the public. The wealth of images provides an excellent resource for perceiving the world through the eyes of others. However, the gigantic volume of images also poses a challenge to the consumption of this unorganized visual information. In this dissertation, we present research on canonical view mining. Given an image collection, we leverage a combination of …

Sox2/Parylated Parp1 Complexes Regulate Pluripotency, Yi-Shin Lai Jan 2011

Sox2/Parylated Parp1 Complexes Regulate Pluripotency, Yi-Shin Lai

All ETDs from UAB

Embryonic stem cells (ESCs) were first derived from the inner cell mass (ICM) of blastocyst-stage embryos. ESCs are distinguished from other cell types by their pluripotency and unlimited self-replication. ESCs are able to differentiate into multiple cell types that represent all three primary germ layers (ectoderm, mesoderm and endoderm) and can form an entire adult organism. Under defined conditions, ESCs are self-renewable and have an extended life-span without compromising pluripotency. The plasticity of ESCs provides opportunities for regenerative medicine. The recent production of induced pluripotent stem cells (iPSC) offers an ideal "personalized" substitute for ESCs. Overexpression of four transcription factors, …

The diamondback terrapin is the only obligate estuarine turtle in North America and is considered an integral member of the salt marsh ecosystem. Unfortunately, many populations throughout the terrapin's range have experienced size declines due to past overexploitation and have been unable to rebound due to current threats including crab trap mortality, habitat degradation, nest predation, and road mortality. The current study was the first comprehensive study examining various population and conservation parameters of Mississippi diamondback terrapins in Alabama. Through various field survey methods conducted in numerous salt marshes along the Gulf Coast of Alabama, it was concluded that Cedar …

Understanding Molecular Mechanisms Of Breast Cancer Metastasis Using Genetically-Engineered Mice, Leah M. Cook Jan 2011

Understanding Molecular Mechanisms Of Breast Cancer Metastasis Using Genetically-Engineered Mice, Leah M. Cook

All ETDs from UAB

Morbidity and mortality of breast cancer patients are drastically increased when primary tumor cells metastasize to distant organ sites. Effective treatment of metastatic disease has been limited; therefore, an increased molecular understanding to identify biomarkers and therapeutic targets is needed. Breast cancer metastasis suppressor 1 (BRMS1) suppresses development of metastases when expressed in a variety of cancer types, including metastatic mammary carcinoma. Little is known of Brms1 function throughout the initiation and progression of mammary carcinoma. Thus Brms1 transgenic mice (derived on C57BL/6 background) were generated by utilizing MMTV promoter expression (for mammary-selective overexpression) or the beta-actin promoter (for ubiquitous …

All ETDs from UAB

The development of the orofacial structure in mammals is a highly ordered process that is coordinated activity of epithelial and mesenchymal cells. Chondrocytes and osteoblasts work in close association for development of many cartilaginous skeletal structures and cartilaginous orofacial structures. Two major orofacial structures are the palate and the temporomandibular joint (TMJ). The transcription factor, Runx2, is obligatory for differentiation of mesenchymal cells into chondrocytes and osteoblasts. However, specific contributions of chondrocyte in cartilaginous and mineralized structures of the craniofacial skeleton are not known. Thus, the objective of this study was to discover the regulatory role of Runx2 in chondrocyte …

Limited Transplantation Of Antigen-Expressing Hematopoietic Stem Cells Induces Long-Lasting Cytotoxic T Cell Responses And Effect Of Altered Suppressive Myeloid Population On Hiv-Disease Progression, Warren L. Denning

All ETDs from UAB

While each type of cancer and chronic viral infection has its own specific pathology, they share two common mechanisms of immune evasion. The first mechanism is the exhaustion or deletion of antigen-specific T cells. The second mechanism is the formation of an immunosuppressive environment responsible for the block of T cell function. Elicitation of antigen-specific T cells be accomplished by immunotherapy in place of conventional treatments such as HAART and chemotherapy. In addition, immunotherapy can alleviate the side-effects associated with long-term use of conventional therapies while reducing the total cost. The results presented here provide an alternative to conventional methods …
