Johannes Kepler was a mathematician that changed the way that science was conceived and practiced in the early seventeenth century. Kepler’s method of scientific inquiry consisted of the joining of the theoretical aspects of astronomy with the solid empirical proof provided by observations. Kepler’s method would lead him to the discovery of his three laws of planetary motion, as well as change the fundamental questions asked by the subject of astronomy. The Effects Of Residual Baylage Leachate On The Germination And Growth Of Orchardgrass (Dactylis Glomerata L.) And Red Clover (Trifolium Pratense L.)., Sheena R. Moore

In early spring, areas of soil directly under baylage, spread for winter consumption by steers, is void of vegetation even after reseeding. This study was conducted to determine if: 1) residual baylage leachate is allelopathic to orchardgrass and red clover, 2) soil microorganisms have an effect on the toxicity of the leachate, and 3) toxicity decreases over time. Petri dishes, as well as potted soil/sand mixtures, were used to test for allelopathy. Leachate reduced seed germination and seedling length as compared to distilled water. There is evidence that soil microorganisms did not affect leachate toxicity. Toxicity of leachate to both …

Recent research in the areas of substance use and juvenile delinquency has revealed that impulsivity is a multi-dimensional construct. Additionally, researchers have expressed the need for more objective, behavioral measures of impulsive behavior. This study was conducted to examine how a delay-discounting procedure, continuous performance test and questionnaire-based measures of impulsivity relate to substance use and delinquent behavior. The sample was comprised of 42 adolescents (35 male) residing in an inpatient treatment facility for alcohol and substance use. Fixed orthodontic appliances including standard brackets and titanium minscrews have the ability to introduce mechanical plaque (biofilm) traps. These orthodontic appliances impair plaque removal, proper oral hygiene, and gingival health11. The purpose of this study is to determine the bacterial accumulation and subsequent biofilm formation on miniscrews and compare it to that of metal orthodontic brackets, in vivo and in-vitro. Plaque samples were taken from the orthodontic brackets and miniscrews in-vivo. S. mutans, Lactobacillus casei, and P. gingivalis were cultured from these samples. In light of the impending ban on synthetic methionine in organic poultry diets, researchers have focused on finding alternative strategies to supply this amino acid. The objectives of this study were to assess performance and carcass characteristics of broilers fed diets devoid of synthetic methionine using 1) a slow-growing and fast-growing genotype, 2) choice-feeding management, and 3) pasture access and seasonal variation. Inclusion of fish meal and high percentages of soybean meal enabled the specific genotype methionine requirement to be met. All diets were certified organic. The purpose of this study was to examine West Virginia Agricultural Education Teachers perceptions on involving students with exceptionalities in agricultural classrooms and laboratories. This study examined whether teachers felt confident, well-prepared, and if other students interact well with students with exceptionalities. This study also sought to determine if agricultural teachers felt they had adequate training to work with students with exceptionalities or if additional training was needed. The majority of teachers agreed that including students with exceptionalities in the classroom fosters understanding for diverse populations. This thesis examines the changes in authenticity in rap lyrics between 1997 and 2004. Throughout its American history, Black music has expressed the lived experiences, the psychological realities, of Africans in diaspora and has always functioned to help create, negotiate and maintain cultural identity or “Blackness”. This dissertation locates a problem in poststructuralism's approach to the concept of believing. Since poststructuralism establishes itself as an intellectual inquiry that deconstructs not only particular beliefs but also the act of believing, many poststructuralists overlook the fact that their own philosophical premises must be initially believed before their critical interventions gain credence. These critics do not explain how belief, as such, emerges within people or why people adopt one belief system over another. Three response reduction procedures commonly employed in applied behavior analysis are extinction, differential-reinforcement-of-other-behavior (DRO) schedules, and time-based schedules of event delivery such as the fixed-time (FT) schedule. Each of these three procedures holds promise for the amelioration of canine behavior problems, yet none has been studied systematically, alone or in comparison with other procedures, with domestic dogs. In this research, the buoy-touching responses of eleven dogs were shaped and maintained using edible reinforcers. Responding then was exposed either to extinction, a fixed or an escalating DRO schedule, or an FT schedule. Extinction reduced responding more rapidly than the escalating DRO …

This study explores African initiatives in the small scale business sector within Kisii town and the larger region of Gusiiland in the period from 1930 to 1978. Despite the transition to a market economy with the onset of colonial rule at the end of 19th Century, African participation in trading activities within Kisii town and other market centers had remained minimal up to 1930. Fibroblast Growth Factor -19: A Novel Factor That Inhibits Hepatic Fatty Acid Synthesis., Sushant Bhatnagar

Previous studies have shown that administration of fibroblast growth factor-19 (FGF-19) reverses diabetes, hepatic steatosis, hyperlipidemia, and adipose accretion in animal models of obesity. To investigate the mechanism for this effect, we determined whether FGF-19 modulated hepatic fatty acid synthesis, a key process controlling glucose tolerance and triacylglycerol accumulation in liver, blood, and adipose tissue. Incubating primary hepatocyte cultures with recombinant FGF-19 suppressed the ability of insulin to stimulate fatty acid synthesis. This effect was associated with a reduction in the expression of lipogenic enzymes. FGF-19 also suppressed the insulin-induced expression of sterol regulatory element-binding protein-1c (SREBP-1c), a key transcriptional …

Inhibitor of differentiation protein-2 (Id2) is a dominant negative helix-loop-helix (HLH) protein, and a positive regulator of proliferation, in various cells. The N-terminal region of Id2 contains a consensus cdk2 phosphorylation sequence SPVR, which may be involved with the induction of apoptosis, at least in myeloid 32d.3 cells. However, the role of Id2 phosphorylation at serine 5 in skeletal muscle cells is unknown. The purpose of this dissertation was to determine if the phosphorylation of Id2 at serine 5 alters its cellular localization and role in apoptosis in C2C12 myoblasts. The actin filament associated protein of 110 kDA (AFAP-110) is a well-characterized protein composed of amino-terminal binding motifs that function upstream of a carboxy-terminal actin binding domain. In vitro studies demonstrate that the amino-terminal pleckstrin homology 1 (PH1) domain interacts with activated PKCα, relaying signals that activate Src family kinases via SH3 and SH2 binding. Serving as an adaptor protein, AFAP-110 relays signals that result in the assembly of motility structures. These findings could be of physiological as well pathological significance as AFAP-110 colocalizes with SFKs in the developing brain as well as in the glial-derived tumor, glioblastoma multiforme. There is need for an anesthetic to sedate fishes that does not require a mandatory post-exposure withdrawal period. Such an anesthetic could be used to sedate handled fishes as they are moved from tank to tank on the farm, surveyed in the wild, released from state and federal hatcheries, or harvested for human consumption. A proprietary form of clove oil, AQUI-S® , was recently submitted to the U.S. Food and Drug Administration for approval as a 0-day withdrawal anesthetic for use on aquatic organisms. Two approaches to enhance the understanding of organic reaction mechanisms are described. First, a new method for teaching organic reaction mechanisms that can be used in a Computer-Assisted Instruction (CAI) environment is proposed and tested (Chapter 1). The method concentrates upon the important intermediate structures, which are assumed to be on the reaction coordinate, and which can be evaluated and graded by currently available computer techniques. Cytochromes P450 are a large family of heme-thiolate enzymes which metabolize a wide variety of both endogenous and xenobiotic compounds. Human P450 1A1 and 1A2 are two major members of the P450 1A subfamily. They share 72% sequence identity but display different specificities towards various substrates. In order to further understand the structure-function relationships of these enzymes, we have performed several molecular modeling-based studies that examined various functional domains of P450 1As. In the first canto of Book 1 of Spenser's The Faerie Queene, Archimago divides Redcrosse and Una "into double parts," creating the impression that they are not just divided from each other, but divided from themselves. While this is the first time the phrase "divided into double parts" is mentioned, the theme of division and doubling has already appeared in the text. Foresters in West Virginia follow BMP guidelines by reseeding retired log landings with inexpensive grasses that quickly provide erosion control. However, these grasses typically are not native nor do they provide high quality forage for wildlife. I developed 3 native seed mixtures for log landing reclamation that would maintain sediment control, as well as enhance wildlife habitat. These mixtures included an erosion control mixture, a wildlife mixture, and a wildflower mixture. Genes specifically expressed in oocytes are important for the development of oocytes and early embryos. By analyzing ESTs from a rainbow trout oocyte cDNA library, we identified a novel EST sequence that does not show homology to any sequences in the GenBank. Analysis of tissue distribution by RT-PCR revealed that this gene was only expressed in unfertilized oocytes. Sequencing of the EST clone identified a cDNA of 3163 bp. Northern blot analysis showed the novel gene has a single transcript of 3.4 kb. Additional 5' sequence was obtained by 5' RACE, extending the novel cDNA to 3333 bp. Analysis of …

The amount of sediment delivered to stream channels and the determination of the topographical attributes responsible for the origination and transfer of sediment were investigated in a central Appalachian mixed hardwood forest from 2002 through 2005. Two study watersheds were chosen within the Indian Run watershed in north central West Virginia. One remained undisturbed (control) and the other underwent road construction and tree harvesting operations (treatment). Sediment samples were collected from silt fences which were installed along both sides of the stream channels in both watersheds to ensure all sediment material delivered from adjacent hill-slopes was captured and collected. Our research compared the effect of six season extension methods on soil and ambient temperatures and yields of warm and cool season vegetables in organic production. Goldenseal (Hydrastis canadensis L.) is an important medicinal plant, native to Canada and Eastern United States, used as an antibiotic, anticonvulsant and in treatments for inflammations and dyspepsia. Over collection of wild population and native habitat destruction has lead the species to be listed as an endangered species in the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), Appendix II. An intersex condition, defined as the presence of oocytes in the testes of male gonochoristic fish, has been observed in smallmouth bass in the South Branch of the Potomac River, West Virginia, which indicates exposure to exogenous estrogens. Endocrine disrupting chemicals (EDC's) are generally hydrophobic and would tend to be found within the sediment of aquatic environments. Few studies have attempted to show the effects of exposure to EDC's on fish using sediment chemical extracts.
