Pre-operative anxiety is a common and anticipated response to patients expecting to undergo anesthesia in preparation for surgery. Pre-operative parental anxiety can lead to increased anxiety in children, which is associated with physiologic alterations in the child. Pre-operative video education is an inexpensive method to increase knowledge and decrease pre-operative parental anxiety related to the perioperative process. The first goal of this project was to evaluate the effectiveness of a pre-operative video in increasing knowledge of the perioperative experience in parents of children anticipating surgery by viewing a pre-operative video. Proteomic and metabolomic analyses provide information about altered metabolic processes in plants and animals. This information can be used to assess the impact of toxicant exposure or diseases on biological systems. In this dissertation, development of an ion mobility spectrometry - mass spectrometry (IMS-MS) strategy has been evaluated in addition to utilizing traditional liquid chromatography tandem mass spectrometry (LC-MS/MS) techniques for metabolite and protein analysis. Aortic compliance is important for dampening pulsatile flow and delivery of continuous flow to the periphery. Aortic compliance is regulated by extracellular matrix composition and endothelial derived nitric oxide. Disruption of aortic endothelium or extracellular matrix can lead to the development of aortic stiffness an independent risk factor of cardiac events and mortality. Over the past decade perivascular adipose tissue (PVAT) surrounding the vasculature has come to light as an important regulator of artery function. Recent investigations focusing on interventions to improve self-controlled choice have centered upon timing processes. However, speculation still remains regarding the mechanism of these interventions. To investigate how these interventions have their effects, timing in mice was assessed using an 18-s peak procedure (18-s FI trials; 54-s peak trials). During an intervention phase, mice in three treatment groups experienced differential reinforcement of low rate (DRL) schedules of reinforcement. A control group received continued exposure to the peak procedure. After 36 DRL sessions, timing was reassessed in the peak procedure. In contrast to previous reports, the DRL intervention resulted in less precise …

This study focused on examining the relationship between the child-care directors' years of experience in relation to the Activities subscale on the Early Childhood Environment Rating Scale-R. The data used in the study was from the Quality Rating and Improvement for West Virginia Child-Care grant, which was funded by the West Virginia Department of Health and Human Resources. The participants included 175 4-year-old classrooms across West Virginia. The Early Childhood Environment Rating Scale contains 43 items across seven subscales. This scale was used to measure the activities score of the 4-year-old classrooms. There was no significant relationship found between the …

Metastasis from the primary tumor site is the major cause of death in breast cancer, and acquisition of migration capacity is a key element for successful metastasis. Recently, it was found that expression of NEDD9, a focal adhesion associated scaffolding protein, could be used to switch between two types of single cell movement known as mesenchymal and amoeboid. Control of this "switch" can potentially open up new avenues of treatment against cancer metastasis. Simultaneous inhibition of both types of single cell movement may prevent cancer migration, thus decreasing cancer spreading and increasing survival of cancer patients. This written thesis is the supporting documentation for High Touch, my Master of Fine Arts thesis exhibition. The exhibition seeks to define my personal expression of beauty through domestic pottery that seeks to both elevate and celebrate mundane life experiences.;The research underpinning my thesis explores the connections between object and user. The use of pottery, particularly in the home, has the ability to breed familiarity, consolation, comfort and security. Animal well-being, evolving feed manufacturing regulatory programs, and consumer perception drive the continued need to provide safe feed to animals. Considering the performance benefits associated with feeding pelleted feed, the concepts of hygienic feed production and production of highly durable pellets may interact to benefit the broiler chicken. In addition to hygienic and physical attributes, poultry nutrition may be enhanced by cereal grains expressing enzymatic activity. The incidence of obesity has increased at an alarming rate during the past thirty years. Current therapies for the treatment and prevention of obesity and metabolic syndrome are ineffective and/or produce undesirable side effects. Fibroblast growth factor 21 (FGF21) is a liver derived hormone that mediates adaptive changes in hepatic glucose and lipid metabolism by elevating energy expenditure and reducing lipid storage. Pharmacological administration of FGF21 analogs in both animal and human models induces striking reversals of obesity and type 2 diabetes. However, the activity of FGF21 analogs exhibit short half-lives in blood circulation and are unsuitable for current treatment. …

American Crocodiles (Crocodylus acutus) are large-bodied crocodylians broadly distributed throughout coastal and lowland wetlands in the Americas. Despite their being large, charismatic megafauna, the study of American Crocodile ecology is still lacking in many areas. As such we assessed the behavior and bioacoustics of American Crocodiles in Belize to address the paucity of data regarding these two facets of American Crocodile ecology. We conducted behavioral observations from three sites in the coastal zone of Belize. We categorized American Crocodile behavior and recorded activity duration for observed behaviors. In a globalized world, wood products are constantly being shipped from one location to another, along with tiny hitchhikers in the form of insects and microorganisms. Euwallacea validus is a fungus-farming ambrosia beetle native to East Asia that likely made its way to the United States in wood packaging materials in the latter half of the twentieth century. E. validus cultivates two fungal symbionts in the U.S., an unnamed Fusarium sp. (AF-4) and Raffaelea subfusca. Fusarium symbionts of Euwallacea ambrosia beetles as well as Raffaelea symbionts of closely related ambrosia beetles have incited widespread disease on more than one-hundred hosts …

The objectives of the first study were to assess performance and digestible amino acid concentration of diets fed to broilers that differed in diet formulation and degree of thermal processing. Basal diets were corn, soybean meal, and DDGS based and formulations were balanced to be similar in calculated energy and digestible amino acids. Treatments were arranged in a 3 x 3 factorial that varied in diet formulation (Basal, Basal + Meat and Bone Meal (MBM), and Basal + Bakery Byproduct (BBP)) and degree of thermal processing (Unprocessed Mash, Pellet, Double Pellet). This contribution highlights and compares some recent achievements in the use of k-space and real space imaging(scanning probe and wide-filed microscope techniques), when applied to a luminescent color center in diamond, known as nitrogen vacancy (NV) center. These techniques combined with the optically detected magnetic resonance of NV, provide a unique platform to achieve nanometric magnetic resonance imaging (MRI) resolution of nearby nuclear spins (known as nanoMRI), and nanometric NV real space localization. Atomic size optically detectable spin probe. High magnetic field sensitivity and nanometric resolution. Non-invasive mapping of functional activity in neuronal networks.

PURPOSE. Recent studies indicate that mitochondrial proteins may contribute to the pathogenesis of primary open-angle glaucoma (POAG). In this study, we examined the association between POAG and common variations in gene-encoding mitochondrial proteins. METHODS. We examined genetic data from 3430 POAG cases and 3108 controls derived from the combination of the GLAUGEN and NEIGHBOR studies. We constructed biological-system coherent mitochondrial nuclear-encoded protein gene-sets by intersecting the MitoCarta database with the Kyoto Encyclopedia of Genes and Genomes (KEGG) database. Tumor microenvironment has been recognized as a key determinant of tumor formation and metastasis, but how tumor microenvironment is affected by nanomaterials is essentially unknown. Here, we investigated whether carbon nanotubes (CNTs), a widely used nanomaterial with known carcinogenic potential, can affect cancer-associated fibroblasts (CAFs), which are a key component of tumor microenvironment that provides necessary support for tumor growth. We show for the first time that single-walled CNT and to a lesser extent multi-walled and its COOH-functionalized form induced CAF-like cells, which are non-tumorigenic in animals, but promote tumor growth of human lung carcinoma and CNT-transformed lung epithelial cells. …

Purpose: Photoreceptor cells are born in two distinct phases of vertebrate retinogenesis. In the mouse retina, cones are born primarily during embryogenesis, while rod formation occurs later in embryogenesis and early postnatal ages. Despite this dichotomy in photoreceptor birthdates, the visual pigments and phototransduction machinery are not reactive to visual stimulus in either type of photoreceptor cell until the second postnatal week. Several markers of early cone formation have been identified, including Otx2, Crx, Blimp1, NeuroD, Trβ2, Rorβ, and Rxrγ, and all are thought to be involved in cellular determination. Determining bacterial gene expression during infection is fundamental to understand pathogenesis. In this study, we used dual RNA-seq to simultaneously measure P. aeruginosa and the murine host’s gene expression and response to respiratory infection. Bacterial genes encoding products involved in metabolism and virulence were differentially expressed during infection and the type III and VI secretion systems were highly expressed in vivo. Strikingly, heme acquisition, ferric-enterobactin transport, and pyoverdine biosynthesis genes were found to be significantly up-regulated during infection. In the mouse, we profiled the acute immune response to P. aeruginosa and identified the pro-inflammatory cytokines involved in acute response …

Colorectal cancer is the second most common cause of cancer deaths for both men and women, and the third most common cause of cancer in the U.S. Toxicity of current chemotherapeutic agents for colorectal cancer, and emergence of drug resistance underscore the need to develop new, potentially less toxic alternatives. Our recent cross-sectional study in a large Appalachian population, showed a strong, inverse, dose–response association of serum perfluorooctane sulfonate (PFOS) levels to prevalent colorectal cancer, suggesting PFOS may have therapeutic potential in the prevention and/or treatment of colorectal cancer. Mitigating stream and river impairment is complex, particularly in mixed-land-use watersheds given the likelihood of integrated responses of stream restoration to coupled and ongoing terrestrial ecosystem disturbance and the need for periodic reassessment and maintenance. Traditional biological sampling (e.g., macroinvertebrate sampling or other biological indices) alone seldom identifies the cause of biological community impairment and large fiscal investments are often made with no apparent improvement to aquatic ecosystem health. A stream physical habitat assessment (PHA) can yield information that, when paired with land-use data may reveal causal patterns in aquatic physical habitat degradation and help to identify sites for rehabilitation …

Fillet yield (FY, %) is an economically-important trait in rainbow trout aquaculture that affects production efficiency. Despite that, FY has received little attention in breeding programs because it is difficult to measure on a large number of fish and cannot be directly measured on breeding candidates. The recent development of a high-density SNP array for rainbow trout has provided the needed tool for studying the underlying genetic architecture of this trait.
