Molecular Dissection Of I(A) In Cortical Pyramidal Neurons Reveals Three Distinct Components Encoded By Kv4.2, Kv4.3, And Kv1.4 Alpha-Subunits, Aaron J. Norris, Jeanne M. Nerbonne

The rapidly activating and inactivating voltage-gated K(+) (Kv) current, I(A), is broadly expressed in neurons and is a key regulator of action potential repolarization, repetitive firing, backpropagation (into dendrites) of action potentials, and responses to synaptic inputs. Interestingly, results from previous studies on a number of neuronal cell types, including hippocampal, cortical, and spinal neurons, suggest that macroscopic I(A) is composed of multiple components and that each component is likely encoded by distinct Kv channel alpha-subunits. The Novel Endosomal Membrane Protein Ema Interacts With The Class C Vps-Hops Complex To Promote Endosomal Maturation, Sungsu Kim, Yogesh P. Wairkar, Richard W. Daniels, Aaron Diantonio

Endosomal maturation is critical for accurate and efficient cargo transport through endosomal compartments. Here we identify a mutation of the novel Drosophila gene, ema (endosomal maturation defective) in a screen for abnormal synaptic overgrowth and defective protein trafficking. Ema is an endosomal membrane protein required for trafficking of fluid-phase and receptor-mediated endocytic cargos. In the ema mutant, enlarged endosomal compartments accumulate as endosomal maturation fails, with early and late endosomes unable to progress into mature degradative late endosomes and lysosomes. Defective endosomal down-regulation of BMP signaling is responsible for the abnormal synaptic overgrowth. Ema binds to and genetically interacts with …

Experiments explored the minimal kanamycin dosing regimen that renders protection against noise induced hearing loss in young CBA/J mice. We also tested the age-dependence of protection in CBA/J as well as the dependence of protection on a particular genetic background in experiments using young C57BL/6J and CBA/CaJ mice.

This project attempts to identify anatomic features that predict the range of the ‘normal’ endocochlear potential in young inbred mice. Cochlear lateral wall histologic metrics were compared in recombinant inbred (RI) mouse strains formed from BALB/c and C57BL/6 mice.

Alpha-synuclein has been implicated in the cellular mechanisms that control auditory sensitivity. In other systems it can also confer protection against cellular injury. Auditory brainstem response thresholds and immunohistochemistry were used to assess the ability of alpha-synuclein to protect against oxidative damage to the cochlea.
