Understanding mast cell development is central to allergic disease pathophysiology. Our laboratory has previously shown that cytokines such as IL-4 and IL-10 inhibit mast cell development from bone marrow progenitors. These studies encouraged our interest in other regulatory cytokines, including transforming growth factor β1 (TGF- β1). TGF- β1 has many cellular sources, one of which is CD4+CD25 regulatory T cells (Tregs). We wanted to determine the effects of Transforming Growth Factor (TGF) βl on mast cell development. We find that TGFβl decreased FcεRI, c-Kit, T1/ST2 and FcεR expression, and inhibited granule formation in developing mast cells. Accessory cells were not …

Poverty, Inequality & Terrorism Relationship in TurkeyUsing empirical evidence criminological studies have identified a relationship between poverty and crime and many studies have concluded that a high crime rate is associated with a higher poverty rate. Other studies indicate that inequalities are a better determinant of crime than absolute poverty. Social disorganization theory, anomie strain theory and Marxist theory have been used to explain the phenomenon. Actor training, like the theatre in Brazil, has historically been a middle and upper class pursuit that followed European models, namely Stanislavski's system. Yet within Brazil there is a wealth of diverse cultures that are inherently theatrical and well suited for application in actor training. In this study I explore one such culture, the Afro Brazilian religion Umbanda. First, I examine its formation to illuminate how the religion itself performed (or served as a site for cultural interaction) throughout history. Mammalian cells require a continuous and sufficient supply of oxygen to carry out their functions. The oxygen pathway has an overall direction taking O2 from the air to the mitochondria, which is a result of the mitochondrial O2 consumption (VO2) NO various effects on the mitochondria: at low concentrations for short periods NO specifically and irreversibly inhibits cytochrome c and reversibly inhibits cytochrome c oxidase, to decrease VO2. Alzheimer's disease (AD) is characterized by the accumulation of amyloid beta plaques, neurofibrillary tangles (NFT) and loss of cortical neurons that control memory and cognition. The cause of NFTs and Aβ plaques is not clear, though it is known that they are formed by enzymes which are preferentially sequestered to membrane domains called lipid rafts. Sulfatide (ST) is a glycosphingolipid that is essential for the proper structure and function of lipid rafts. Bile acids regulate their own synthesis through a feedback regulatory mechanism of mainly two enzymes in the classic pathway, the 7α-hydroxylase and the 12α-hydroxylase. In the early 1990's it was shown that the regulatory responses of 7α-hydroxylase are mediated at the transcriptional level and since then many positive and negative transcription factors that mediate regulatory response have been identified. This research illustrates the experience of living with hallucinations from the perspective of being, and sought to answer the questions: what is the meaning of hallucinations and how do hallucinations connect to one's sense of self?A phenomenological Heideggerian hermeneutic approach was used to guide data collection and analysis. In this study, 12 individuals with schizophrenia or schizoaffective disorders were asked to describe their experience of hallucinations within the context of being. An overarching constitutive pattern emerged with four themes. Glioblastomas are an incurable type of brain tumor with a mean survival time of 9-12 months following diagnosis. One of the reasons for this poor prognosis is the ability of tumor cells to invade the surrounding normal brain tissue. Enzymes responsible for this invasive nature include the matrix metalloproteinase family. MMP-1 is a member of this family which has been well studied in many types of invasive tumors, with gliomas being an exception. I use images from the Japanese comic Manga: Ranma1/2 to develop units of instruction with goals of engaging students in relating the study of art to their visual culture outside the classroom. Free radical-mediated DNA double strand breaks (DSBs) are induced either directly by ionizing radiation or by certain chemicals like bleomycin. These breaks are terminated by 3'-PG (PO4CH2COOˉ) or 3'-phosphate groups formed as a result of fragmentation of deoxyribose. To study the nature of repair of these 3'-blocked breaks, we constructed substrates mimicking free-radical induced DSBs. Human and yeast tyrosyl DNA-phosphodiesterase (Tdpl) efficiently processed substrates with 3'-PGs, in either the presence or absence of magnesium, to give a 3'-phosphate. Gel filtration chromatography and western blotting codmed that the putative enzyme in human extracts that efficiently processed PG was indeed tyrosyl DNA-phosphodiesterase. …

Serotonin (5-HT) plays an important role as both a neurotransmitter and animmune modulator. The serotonin reuptake transporter (SERT) clears the extracellular space of 5-HT, which decreases the effects of 5-HT on target cells. This study demonstrated that the RAW264.7 macrophage cell line expresses SERT function, measured by assays of 3H-5HT uptake. The 5-HT uptake in RAW264.7 macrophages was more than 10-fold that of peritoneal macrophages, indicating that these cells are an excellent model for studying regulation of the SERT. Activation of macrophages with lipopolysaccharide (LPS) increased SERT activity in a time- and concentration-dependent manner and Western blots indicate that the …

As a prelude to strain selection for domestication and future marker assisted selection, genetic variation revealed by microsatellite DNA was evaluated in yellow perch, Perca flavescens, from four wild North American populations collected in 2003-2004 (Maine, ME; New York, NY; North Carolina, NC; and Pennsylvania, PA), and two captive populations (Michigan, MI; Ohio, OH). For the loci examined, levels of heterozygosity ranged from He = 0.04 to 0.88, genetic differentiation was highly significant among all population pairs, and effective migration ranged from low (Nem = 0.3) to high (Nem = 4.5). Degenerate Oligonucleotide Primed-PCR (DOP-PCR) can potentially enhance analysis of low copy number DNA samples. Theoretically, this procedure replicates fragments of the genome that can then be used for downstream multiplex STR analysis. The objective of this study is to optimize DOP-PCR by examining ramplelongation times and cycle numbers in the non-specific amplification portion of DOP-PCR, and by modifying the degenerate primer. Additionally, other methods such as Multiple Displacement Amplification (MDA) and Low Copy Number PCR (LCN PCR) were examined for their ability to create accurate DNA profiles from low DNA input amounts. PROBLEM STATEMENT: Growing evidence indicates that psychological stress contributes to cardiovascular diseases through complex neuroendocrine mechanisms. Psychological stress leads to several physiological responses including increased heart rate (HR) and blood pressure (BP) as well as decreased heart rate variability (HRV) through alterations in the autonomic nervous system (ANS), specifically increased sympathetic nervous system (SNS) activity and decreased parasympathetic nervous system (PNS) activity. MDM2 has been shown to induce G0-Gl/S phase arrest. To determine the cell cycle step targeted by MDM2, flow cytometry was employed to detect induction of events during the G1-S phase transition in MDM2-arrested cells. MDM2 overexpression does not prevent expression of cyclin D, cyclin D-CDK mediated phosphorylation of Rb or cyclin E in normal, immortal or tumor-derived cells. However, MDM2 down-regulates cyclin A expression specifically in normal cells, which is associated with G1 arrest. The genes of the human β-globin locus are located on chromosome 11 in the order of their expression during development: 5' ε, γ, β 3'. During development, silencing of the 5' gene occurs with activation of the immediate 3' gene. This process occurs twice and is termed hemoglobin switching. Naegleria fowleri, a free-living ameboflagellate, is the causative agent of primary amebic meningoencephalitis. Healthy humans sporadically become infected with N. fowleri and develop fatal PAM after recreational or work exposure to freshwater; accordingly, there is a need for monitoring the presence of pathogenic amebeflagellates in public freshwater. The present study was conducted to determine whether a nested PCR assay could be used for detection of N. fowleri in freshwater habitats. Excitotoxic calcium influx and activation of calcium-regulated systems is a common event in several types of neuronal injury. This mechanism has been the focus of intense research, with the hope that a more complete understanding of how neuronal injury affects calcium-regulated systems will provide effective treatment options. This study examines one such calcium-stimulated enzyme, calcineurin, in the context of two common neurological pathologies, status epilepticus and traumatic brain injury.Status epilepticus was induced by pilocarpine injection. The objective of this study was to investigate the relationship between stressful events in adolescents and negative outcomes of substance use, depressive symptoms, and anxiety symptoms. Parental support and unsupportive parental responses were examined as moderators of this relationship. The research design was cross-sectional, and self-report data were collected from 100 adolescents in the 8th and 9th grades. As hypothesized, significant positive associations were found between perceived stressors and each of the three negative outcomes, as well as between unsupportive parental responses and the outcomes. Cross-sectional research of youth with type I diabetes has demonstrated that rote and working memory predict blood glucose monitoring (BGM) and carbohydrate consumption, respectively; however, to date, no longitudinal follow-up studies exist. Rote and working memory subtests from well-standardized memory measures, along with a problem-solving and executive functioning measures were administered to 118 youth with type I diabetes, aged 9-16 in two waves of data collection (mean interval = 2.07 years). GIS (Geographic Information Systems) and remote sensing techniques were used to predict relationships between bald eagle nest presences and land type, distance to land type and impervious surface cover area. Data plots revealed bald eagle nest presence decreases in response to an increase in area of bareland; increases with an increase in area of forested land; decreases with an increase in distance (m) to shoreline, and decreases in response to an increase in area of impervious surfaces. The physical and biophysical properties of a biological membrane model, phosphatidylcholine bilayers, were investigated using novel spiropyran/merocyanine molecular probes. The femtosecond to second dynamics of this system's photochemistry enabled bilayer viscosity and free volume to be studied over a broad time scale. Spiropyrans/merocyanines with different polarity were synthesized by changing the substitution of the indole moiety enabling determination of the trans-membrane properties of the bilayer. Serpins are a unique breed of proteins due to their enzymatic mechanism. Two systems were closely monitored during fluorescent binding studies, the ACT-CHY along with the AT:TRY interaction. Four different conformational variants of each system were studied including the native, cleaved, latent and complex forms. Three different fluorescent dyes were used to identify the conformations including ANS, TNS, and bis-ANS. The majority of research on the endogenous opioid system has focused on various pain assays and the efficacy of different opioid agonists. However, minimal attention has been focused on the effects of gonadal hormones and their impact on the opioid peptide system. The present study was designed to determine the effects of modulation of gonadal hormones on the opioid receptor protein levels in Complete Freund's Adjuvant (CFA)-treated arthritic and non-arthritic male and female Lewis rats.
