Distinguishing Between Homeless And Unstably Housed Men On Risk Factors For Homelessness, Valerie Holton Apr 2011

Distinguishing Between Homeless And Unstably Housed Men On Risk Factors For Homelessness, Valerie Holton

Theses and Dissertations

This study explored the risk factors for homelessness in single men, the largest group of people experiencing homelessness and a group about which little is known regarding their risks for homelessness. A case control design was used to differentiate risk factors between men who were homeless and men who were unstably housed. Risk factors included cumulative risk, negative life events, and demographic factors. Two models were tested using discriminant function analysis (DFA). The Cumulative Risk Model did not significantly differentiate between the two groups. However, the Negative Life Events Model yielded one discriminant function that significantly differentiated between the groups …

Developmental Trajectories Of Physical And Relational Aggression And Their Relation To Delinquency And Substance Use In Adolescence, Denicia Titchner

Theses and Dissertations

Although researchers studying adolescent aggression have proposed a conceptual distinction between physical and relational aggression, there is contradictory evidence regarding the degree to which they differ in their trajectories and relations to other outcomes. This study explored the importance of differentiating between these two forms of aggression based on comparisons of their trajectories, relation with each other, impact on delinquency and substance use, and gender differences. Data were collected as part of the Multisite Violence Prevention Project, conducted at 19 middle schools from four sites with a predominantly low-income, minority sample of students (N = 2,822). Growth curves showed significant …

Assessment Of Diabetes Regimen Disease Care In Youth With Type 1 Diabetes Via The Diabetes Behavior Rating Scale And The 24-Hour Diabetes Interview, Kathryn Maher

Theses and Dissertations

The psychometric properties of two measures of diabetes disease care, the Diabetes Behavior Rating Scale (DBRS) and the 24-hr Diabetes Interview (24-hr) were evaluated. The 24-hr is a widely used, structured interview while the DBRS is a self-administered, fixed-choice questionnaire. Both measures were administered to 250 youth with Type 1 Diabetes (aged 11–14 years) and their parents. Overall, both measures demonstrate adequate psychometric properties. The DBRS and the 24-hr demonstrated good incremental validity and low convergent validity with each adding significant additive value. Both measures demonstrated good concurrent validity with HbA1c. As expected, scores on the 24-hr demonstrated less than …

Exploring The Functional Interaction Between Camk-Ii And P53, Raymond Lai Apr 2011

Exploring The Functional Interaction Between Camk-Ii And P53, Raymond Lai

Theses and Dissertations

Calcium (Ca2+)/calmodulin-dependent kinase 2 (CaMK-II) is a multifunctional member of a family of Ca2+/calmodulin-dependent serine/threonine protein kinases that respond to transient intracellular calcium signaling. CaMK-II has been reported to be involved with transcription regulation, cell motility, neuronal development, cell cycle regulation, and more recently early development of vertebrates (Easley et al., 2008; Rothschild et al., 2009; Francescatto et al., 2010). Through previous work in the lab using tandem mass spectrometry and “substrate-trapping mutants”, tumor suppressor protein 53 (p53) was identified as a novel CaMK-II binding partner in tissue culture. In this study, I sought to provide characterization of the functional …

The Role Of The Propeptide And Its Residues In Activation And Secretion Of Elastase, An M4 Metalloprotease Secreted By Pseudomonas Aeruginosa, Emily Boice Apr 2011

The Role Of The Propeptide And Its Residues In Activation And Secretion Of Elastase, An M4 Metalloprotease Secreted By Pseudomonas Aeruginosa, Emily Boice

Theses and Dissertations

Pseudomonas aeruginosa secretes several proteases associated with pathogenesis, but the most abundant and active is elastase (M4 metalloendopeptidase). Elastase (lasB), is first synthesized as a preproenzyme, with a signal peptide, an 18-kDa N-terminal propeptide, and a 33-kDa mature domain. The propeptide functions as an intramolecular chaperone that is required for the folding and secretion of elastase, but ultimately is proteolytically removed and degraded. Previous research has identified the conserved residues in the propeptide of elastase as compared to other M4 protease precursors and showed some among them to be important for the production of active elastase. In this project, the …

3d Functional Modeling Of Dbs Efficacy And Development Of Analytical Tools To Explore Functional Stn, Deepak Kumbhare

Theses and Dissertations

Introduction: Exploring the brain for optimal locations for deep brain stimulation (DBS) therapy is a challenging task, which can be facilitated by analysis of DBS efficacy in a large number of patients with Parkinson’s disease (PD). The Unified Parkinson's Disease Rating Scale (UPDRS) scores indicate the DBS efficacy of the corresponding stimulation location in a particular patient. The spatial distribution of these clinical scores can be used to construct a functional model which closely models the expected efficacy of stimulation in the region. Designs and Methods: In this study, different interpolation techniques were investigated that can appropriately model the DBS …

Parasitic Interiors, Michael Kanasink Apr 2011

Parasitic Interiors, Michael Kanasink

Theses and Dissertations

This project is an experiment in the adaptive reuse of an Art Deco style manufacturing facility in Richmond, VA. The building has fallen into disrepair and has been inhabited by two different owners over the years, but the structure remains very much as it did when it was erected in 1946. I will propose adapting to the defunct space, a forward thinking school based on hands-on, creative learning. This school will challenge accepted pedagogies and serve as an exapmple of future learning. I have always looked as adaptive reuse interior design as a parasitic form of designing. The new program …

Spatial Parameters In Therapeutic Spaces: Design Of A Residential Treatment Facility For Eating Disorder Patients, Sarah Beth Basinger Apr 2011

Spatial Parameters In Therapeutic Spaces: Design Of A Residential Treatment Facility For Eating Disorder Patients, Sarah Beth Basinger

Theses and Dissertations

Can the built environment positively influence the treatment of an eating disorder? Does the built environment play a role in the quality of care, the rehabilitation process and the lasting affects of treatment in eating disorders and if so, what role does it play? What are the important parameters to consider in designing such a space? Currently there are few facilities designed specifically for the treatment of this disorder. Of those facilities, there is a lack of research regarding the effects of the built environment. Therefore, the process of answering these questions must begin by assessing parameters in architectural precedents …

Electrophysiological Characterization Of Enteric Neurons Isolated From The Immortomouse, Edward G. Hawkins Apr 2011

Electrophysiological Characterization Of Enteric Neurons Isolated From The Immortomouse, Edward G. Hawkins

Theses and Dissertations

The availability of murine genetic models is extremely advantageous to studying gastrointestinal function, but the benefits afforded by studying enteric neurons in mice has been hindered by their accessibility. Fetal (E13) and 2 day post-natal (P2) enteric neuron cell lines (IM-FEN and IM-PEN, respectively) were recently developed from the H-2Kb-tsA58 immortomouse. Our goal was to identify the electrophysiological properties of these cell lines and clarify their utility as a model of enteric neurons. IM-PEN cells stained positively for the neuron specific markers βIII-tubulin and PGP9.5 and were negative for the glial cell marker S100. Detection of mRNA for TRPA1, TRPV1, …

The potential for radical landscape is considered.

Involvement Of P53 In The S-Phase Checkpoint During Nucleotide Deficiencies, Cortney Heyer Apr 2011

Involvement Of P53 In The S-Phase Checkpoint During Nucleotide Deficiencies, Cortney Heyer

Theses and Dissertations

Several classes of antimetabolites have been developed for the treatment of cancer, including numerous inhibitors of nucleotide biosynthesis. N-(phosphonacetyl)-L-aspartate (PALA) and hydroxyurea (HU) are two antimetabolites that inhibit nucleotide biosynthesis; PALA inhibits de novo pyrimidine synthesis and HU inhibits the conversion of ribonucleotide diphosphates to deoxyribonucleotide diphosphates. Due to the similar mechanisms, it was thought that cancer cells would respond similarly to HU and PALA treatment. However, studies in this dissertation revealed strikingly different responses to either HU or PALA treatment in HCT116 cells. A cytoprotective S-phase arrest was activated upon HU treatment while PALA treatment failed to activate the …

Adam10 Exacerbation Of Allergic Disease Is Potentially Explained By Its Role In Cd23 Exosomal Sorting., Joel Mathews

Theses and Dissertations

CD23, the natural negative regulator of IgE, has been shown to be involved in asthma progression through its regulation of IgE. To investigate if its sheddase, ADAM10, is also involved in asthma progression, three mouse models were utilized; an IgE/mast cell dependent model, an IgE dependent, mast cell independent model and a mast cell and IgE independent model. Experimental asthma was then induced in mice which were selectively deficient for ADAM10 in B cells (ADAM10-/-) and compared to WT controls. The ADAM-/- mice had decreased signs of asthma, including eosinophilia, AHR and IgE synthesis in the IgE dependent model compared …

Radiation Therapy Treatment Plan Optimization Accounting For Random And Systematic Patient Setup Uncertainties, Joseph Moore Apr 2011

Radiation Therapy Treatment Plan Optimization Accounting For Random And Systematic Patient Setup Uncertainties, Joseph Moore

Theses and Dissertations

External-beam radiotherapy is one of the primary methods for treating cancer. Typically a radiotherapy treatment course consists of radiation delivered to the patient in multiple daily treatment fractions over 6-8 weeks. Each fraction requires the patient to be aligned with the image acquired before the treatment course used in treatment planning. Unfortunately, patient alignment is not perfect and results in residual errors in patient setup. The standard technique for dealing with errors in patient setup is to expand the volume of the target by some margin to ensure the target receives the planned dose in the presence of setup errors. …

Wilms’ Tumor-1 (Wt1) Protein Expression In Glioma Cells Actuates Cellular Invasiveness- Identifying Its Target Genes, Archana Chidambaram Apr 2011

Wilms’ Tumor-1 (Wt1) Protein Expression In Glioma Cells Actuates Cellular Invasiveness- Identifying Its Target Genes, Archana Chidambaram

Theses and Dissertations

Previous studies in our laboratory demonstrated the expression of WT1 in a significant number of glioma cells and established its role in promoting tumor cell proliferation. Here, we noted the effect(s) of manipulating WT1 levels on the expression levels of genes that were previously shown to be regulated by WT1. We found no correlation between the expression levels of WT1 and PDGF-A, Snai1 and E-cadherin and a consistent inverse correlation between WT1 and IGF-1R expression in U251-MG cells. To ascertain whether the increased IGF-1R levels resulting from WT1 silencing could account for decreased cellular proliferation, we utilized siRNA mediated knockdown …

Joint Attention Interventions For Young Children With Autism Spectrum Disorders: Caregiver And Child Actions And Transactions, Abigail Vo Apr 2011

Joint Attention Interventions For Young Children With Autism Spectrum Disorders: Caregiver And Child Actions And Transactions, Abigail Vo

Theses and Dissertations

Early intervention is a critical component of efforts to optimize outcomes for children with autism spectrum disorders (ASD) and their families. One promising target for early intervention is joint attention, an early developing social-cognitive competency that is foundational to many other social, communicative, and cognitive skills; and a core deficit in children with ASD. While joint attention interventions are gaining interest among researchers, many are limited by their failure to situate joint attention development within the caregiver-child relationship and to adequately examine child and caregiver outcomes and transactional processes. The purpose of this study was to examine changes in child …

Relations Between Violence Exposure, Threat Appraisal, And Coping Among Typologies Of Victimized Adolescents, Katherine Taylor Apr 2011

Relations Between Violence Exposure, Threat Appraisal, And Coping Among Typologies Of Victimized Adolescents, Katherine Taylor

Theses and Dissertations

According to the transactional theory of stress and coping, threat appraisals influence coping and adjustment. Previous research has shown that threat appraisals mediate relations between violence exposure and adjustment, but few studies have examined links between threat appraisals and coping. The current study examined relations between violence exposure, threat appraisals, and coping among typologies of victimized adolescents. The sample included 159 predominately African American adolescents (M = 12.1). Path analyses were used to test whether threats of negative evaluation by self and others mediated relations between violence exposure and avoidance and positive reframing coping, respectively. Results did not indicate mediation …

Drug Design, Biological Activity, And Metabolic Consequences Of Cytotoxic Platinum Compounds: Utilizing Fluorescent Tagging To Understand Drug Action And Metabolism, Brad Benedetti

Theses and Dissertations

Platinum drugs are among the most commonly used chemotherapeutics for the treatment of testicular, head and neck, ovarian, small cell lung, and colorectal carcinomas. Although the current set of platinum chemotherapeutics has proven somewhat successful, the overall success of platinum based drugs is limited due to acquired drug resistance and a limited range of tumor types that are treatable with the current regime. The development of novel cytotoxic platinum based compounds, both trans- and polynuclear, provides for the promising treatment of clinical platinum drug resistant tumors. While the cytotoxic activity of platinum drugs provides for a hopeful outlook, the ultimate …

Sensory Modulation Disorder And Schizophrenia: Linking Behavioral Measures, Linda Olson

Theses and Dissertations

Schizophrenia is a devastating disorder affecting millions of people in the United States. Studies leading to new understanding of and intervention for this disorder are essential, as current interventions are minimally effective. The end result is high rates of re-hospitalization, impaired occupational performance and decreased community reintegration. According to the Diagnostic and Statistical Manual of Mental Disorders, symptoms of schizophrenia are categorized as positive or negative, with both types of symptoms impacting successful occupational engagement. Based on behavioral observations occupational therapists have hypothesized that this population may also experience sensory modulation disorder (SMD). Although electrophysiological studies support the presence of …

A Comparative Study Of Rotary Instrumentation Of The Maxillary First Premolar Buccal Root Utilizing Cone Beam Computed Tomography, Stephan Zigo Apr 2011

A Comparative Study Of Rotary Instrumentation Of The Maxillary First Premolar Buccal Root Utilizing Cone Beam Computed Tomography, Stephan Zigo

Theses and Dissertations

The study objective was to determine cementum-dentin wall thickness along the furcation groove in maxillary bifurcated first premolars after preparation with three successively larger, 0.04 tapered, nickel titanium rotary files. Pre-instrumentation and post-instrumentation imaging was accomplished utilizing Cone Beam Computed Tomography. All data was analyzed using an ANOVA. Instrumentation resulted in a significant reduction in dentin-cementum wall thickness (p < .001). At mid-groove, predicted dentin-cementum wall thickness (95% CI) was equal to or less than the proposed standard (0.50 mm) for apical file sizes 30, 35, and 40 respectively. Instrumentation of the mid-groove in maxillary first premolars reduces dentin-cementum wall thickness to levels that may be insufficient to ensure tooth integrity.

Accelerating Finite State Projection Through General Purpose Graphics Processing, Thomas Trimeloni Apr 2011

Accelerating Finite State Projection Through General Purpose Graphics Processing, Thomas Trimeloni

Theses and Dissertations

The finite state projection algorithm provides modelers a new way of directly solving the chemical master equation. The algorithm utilizes the matrix exponential function, and so the algorithm’s performance suffers when it is applied to large problems. Other work has been done to reduce the size of the exponentiation through mathematical simplifications, but efficiently exponentiating a large matrix has not been explored. This work explores implementing the finite state projection algorithm on several different high-performance computing platforms as a means of efficiently calculating the matrix exponential function for large systems. This work finds that general purpose graphics processing can accelerate …

Characterization And Genetic Analysis Of The Cellulolytic Microorganism Thermobifida Fusca, Yu Deng Mar 2011

Characterization And Genetic Analysis Of The Cellulolytic Microorganism Thermobifida Fusca, Yu Deng

Theses and Dissertations

Currently, one of the hurdles hindering efficient production of cellulosic biofuel is the recalcitrant nature of cellulose to hydrolysis. A wide variety of cellulase enzymes are found natively in microorganisms that can potentially be used to effectively hydrolyze cellulose to fermentable sugars. Thermobifida fusca is a high G-C content, thermophilic, gram-positive soil actinobacterium with high cellulolytic activity. The phenomenological and mechanistic parameters affecting cellulase activity were studied in T. fusca and two mechanisms have been found: 1) transcriptions of cellulase-related genes were not closely associated with measured differences in cellulase activity and 2) cellular energetics (intracellular ATP) correlated more closely …

The Relationship Of Physical Activity, Eating Behaviors, And Hunger Control To Weight Loss And Quality Of Life In Gastric Banding Patients, Nancy Baugh Mar 2011

The Relationship Of Physical Activity, Eating Behaviors, And Hunger Control To Weight Loss And Quality Of Life In Gastric Banding Patients, Nancy Baugh

Theses and Dissertations

Gastric banding as a weight loss surgery has increased in popularity in the United States since its approval by the Food and Drug Administration in 2001. Successful weight loss after weight loss surgery is most frequently defined as greater than 50 percent of excess weight loss (EWL). Systematic reviews show that the band is widely effective in achieving successful weight loss in most patients, however individual studies show more inconsistent outcomes. Although previous research supports gastric bypass surgery as having a positive effect on quality of life after gastric bypass, there is very little data to support this relationship in …

Relations Among Media, Eating Pathology And Body Dissatisfaction In College Women, Carrie Bair Mar 2011

Relations Among Media, Eating Pathology And Body Dissatisfaction In College Women, Carrie Bair

Theses and Dissertations

Research has identified a relation between exposure to thin-ideal magazine and television media images and eating disorder pathology. However, few studies have examined the potential influence of Internet media on eating disorder behaviors and attitudes. This study investigated the associations among appearance-orientated media exposure, body dissatisfaction, eating pathology and thin-ideal internalization in a sample of 421 female undergraduate students. Results indicate that undergraduate women spend significantly more time viewing appearance-oriented sources online, rather than reading appearance-orientated magazines. Appearance-oriented Internet consumption was also more strongly associated with eating disorder pathology than was use of other media (television and magazines). Relations between …
