Assessment Of A Predicted Diguanylate Cyclase In Treponema Denticola, Dhara T. Patel Jan 2018

Assessment Of A Predicted Diguanylate Cyclase In Treponema Denticola, Dhara T. Patel

Theses and Dissertations

Periodontal disease is a progressive inflammatory condition that is characterized by the reabsorption of alveolar bone, the destruction of connective tissue, and edentulism. It is caused by a dysbiosis in the oral microbiome as a result of a shift from a Gram-positive aerobic bacterial population, to one that becomes more Gram-negative and anaerobic. Treponema denticola is thought to drive this diseased state based on its role as a keystone periopathogen. A major component of T.denticola’s invasiveness is its motility, which allows the spirochete to penetrate and disseminate through tissues. This motility, which has been seen to be crucial to …

Zebrafish Model Of Mll-Rearranged Acute Myeloid Leukemia, Alex J. Belt Jan 2018

Zebrafish Model Of Mll-Rearranged Acute Myeloid Leukemia, Alex J. Belt

Theses and Dissertations

Acute myeloid leukemia (AML) is the second most common type of leukemia and accounts for 80% of adult acute leukemia cases and is characterized by the accumulation of poorly or undifferentiated myeloid blast cells. Standard treatment includes chemotherapy, which if unsuccessful, is followed by more rigorous chemotherapy as well as stem cell transplantation. Considering most patients are over the age of 45, these more rigorous therapies are not always possible, and as such, new therapies must be developed. Furthermore, AML patients harboring a chromosomal rearrangement involving Multiple Lineage Leukemia (MLL) that results in the expression of an MLL fusion protein …

Understanding The Role Of Sport For Development In Addressing Health Disparities In Low-Ses Communities, Tiesha R. Martin

Theses and Dissertations

The purpose of this study was to understand the role of Sport for Development (SFD) in addressing health disparities in low-SES communities. This was done using a multiple case study design, in which administrators, staff, and youth participants from five SFD programs were interviewed. A theoretical model, consisting of the theory of fundamental causes (Link & Phelan, 1995), the classification of SFD programs (Coalter, 2007), and the ecological model of health promotion (McLeroy, 1988), was developed to guide this study.

Interviews were transcribed and then analyzed using a deductive coding process (Gilgun, 2005). The findings revealed that the SFD programs …

Examination Of The Role Of Dehumanization As A Potential Mechanism Underlying The Racial Disparities In School Disciplinary Measures., Ebony A. Lambert Jan 2018

Examination Of The Role Of Dehumanization As A Potential Mechanism Underlying The Racial Disparities In School Disciplinary Measures., Ebony A. Lambert

Theses and Dissertations

Schools should be safe and supportive spaces for all students, yet Black students tend to face biased treatment in the education system, which often results in harsh disciplinary measures. This research examined the role of animalistic dehumanization (i.e., perceiving others as animal-like and uncultured and denying uniquely human characteristics), in predicting choice of harsher disciplinary measures for Black students as opposed to White students. It was hypothesized that individuals who dehumanize Black students to a greater degree would be more likely to believe that Black students need to be disciplined through harsher measures. Additionally, it was hypothesized that the link …

Red-Headed Woodpecker Full Annual Cycle Ecology At Fort A.P. Hill, Virginia, Benjamin Nickley Jan 2018

Red-Headed Woodpecker Full Annual Cycle Ecology At Fort A.P. Hill, Virginia, Benjamin Nickley

Theses and Dissertations

The red-headed woodpecker is a primary cavity excavator found throughout the Eastern Deciduous Forest and parts of the Great Plains. Although widespread, the red-headed woodpecker is generally considered uncommon, with patchily distributed populations. Over the past 50 years, this species has experienced precipitous, range-wide declines. The red-headed woodpecker uses a variety of cover types to meet resource needs across the annual cycle, ranging from oak savannas and wetlands, to mature beech forests and urban parks. Given their apparent adaptability to such a wide range of habitats, causes of declines are perplexing. To understand and stem declines, recent studies have focused …

Shared Genetic And Environmental Influences On Fear, Anxiety, Posttraumatic Stress, And Brain Morphometry, Chelsea Sawyers

Theses and Dissertations

Anxiety disorders (ADs) and stress-related disorders are some of the most common psychiatric disorders in the United States. Like other c0mplex psychiatric illness, genetics and neuroimaging research has focused on understanding their underlying neurobiology. Areas within the fear-network play important roles in threat perception, fear conditioning/learning, cognitive processing, and modulation of fear responses including contextual modulation and extinction and have been implicated in ADs as well as stress disorders such as posttraumatic stress disorder (PTSD). The primary gap in the current search for underlying biological mechanisms is in whether biomarkers associated with disorders share genetic influences with the disorders they …

Mechanisms Of B-Myb Oncogenicity In Ovarian Cancer, Audra N. Iness Jan 2018

Mechanisms Of B-Myb Oncogenicity In Ovarian Cancer, Audra N. Iness

Theses and Dissertations

High expression of B-Myb (encoded by MYBL2), an oncogenic transcription factor, is associated with cell cycle deregulation and poor prognosis in several cancers, including ovarian cancer. However, the mechanism by which B-Myb alters the cell cycle is not fully understood. In proliferating cells, B-Myb interacts with the MuvB core complex including LIN9, LIN37, LIN52, RBBP4, and LIN54, forming the MMB (Myb-MuvB) complex, and promotes transcription of genes required for mitosis. Alternatively, the MuvB core interacts with Rb-like protein p130 and E2F4-DP1 to form the DREAM complex that mediates global repression of cell cycle genes in G0/G1, including a subset …

5-Ht2b Receptor-Mediated Cardiac Valvulopathy, Pallavi Nistala Jan 2018

5-Ht2b Receptor-Mediated Cardiac Valvulopathy, Pallavi Nistala

Theses and Dissertations

5-HT2B receptor agonism causes cardiac valvulopathy, a condition characterized by thickening of the heart valves and as a result, regurgitation of blood within the heart. The anti-obesity drug fenfluramine, which was originally prescribed as an anorectic, was withdrawn from the market due to causing cardiac valvulopathy. Fenfluramine, after metabolism by N-dealkylation, produces the metabolite norfenfluramine, which acts as a more potent valvulopathogen. The same was seen with MDMA (ecstasy), a popular drug of abuse, which is metabolized by N-dealkylation to produce MDA, a more potent valvulopathogen. Glennon and co-workers. studied a series of 2,5-dimethoxy-4- substituted phenylisopropylamines (DOX type) hallucinogens …

Discrimination, Mental Health, And Preparedness For Aging In Trans(Gender)/Gender-Nonconforming Adults, Richard S. Henry Jan 2018

Discrimination, Mental Health, And Preparedness For Aging In Trans(Gender)/Gender-Nonconforming Adults, Richard S. Henry

Theses and Dissertations

This cross-sectional study examined relationships among discrimination, mental health (i.e., depression and anxiety), preparation for aging (i.e., familiarity and planning), social support, death attitudes, and aging anxiety among TGNC adults (N = 154). Neither discrimination nor mental health predicted preparation for aging familiarity or planning. Discrimination did, however, predict both anxiety and depression, although only the non-affirmation subscale was a unique predictor of both. As discrimination and mental health were not a significant predictor of preparedness for aging in the previous regressions, the hypothesized mediation model and subsequent moderated mediation models were not conducted. Additional exploratory multiple regressions were …

Mechanistic Studies Of Proton-Coupled Electron Transfer Reactions Involving Antioxidants, Kejie Meng Jan 2018

Mechanistic Studies Of Proton-Coupled Electron Transfer Reactions Involving Antioxidants, Kejie Meng

Theses and Dissertations

The objective of the research was to investigate proton-coupled electron transfer (PCET) reactions involving antioxidants to gain insight into the detailed mechanisms of glutathione (GSH), Trolox, and α-tocopherol (α-TOH). PCET reactions are complex redox reactions that transfer electrons and protons sequentially or in concert. These reactions are ubiquitous in natural or artificial processes that produce electrochemical energy that is extractable as electricity or as chemical fuels of high energy content. Examples of processes based on PCET are photosynthesis, respiration, nitrogen fixation, carbon dioxide reduction, redox fuel cells, and artificial photosynthesis. Antioxidants were selected as a PCET model to understand the …

Examining Two Structural Equation Models To Determine The Effect Of Parental Practices And Familial Cultural Values On Mexican Adolescents’ Sexual Health, Efren A. Velazquez

Theses and Dissertations

Sexual behavior is a normative part of adolescence and it is important to understand how familial factors influence Latina/o adolescents’ sexual health. The current study examined two structural equation models (SEM) and the associations between familial cultural values (i.e., familismo and filial piety), maternal and paternal monitoring (i.e., disclosure and solicitation) and Latina/o adolescent’s sexual behavior (i.e., intentions to have sex, sexual behavior, sexual responsibility). A hypothesized SEM examined whether paternal/maternal monitoring mediated the relation between familial cultural values and adolescents’ sexual health, while an alternative SEM examined whether familial cultural values mediated the association between paternal/maternal monitoring and adolescents’ …

Gender Beyond Binary, Sidra Zubairi Jan 2018

Gender Beyond Binary, Sidra Zubairi

Theses and Dissertations

Transgender individuals challenge the binary definition of gender accepted in Pakistani society and as a result, Pakistan’s transgender community is highly marginalized. Pakistani society regards transgender individuals as “abnormal,” because their physical appearance and behavior fail to conform with conventional expectations. Based on contextual research and interviews conducted with transgender individuals in Pakistan, my thesis explores the obstacles transgender individuals face in the course of everyday survival. My research responds to the physical realities of being transgender in Pakistani society and analyzes cultural norms associated with gender, which trigger harassment. By designing transformable apparel for these individuals, informed by primary …

Anxious?: Re-Designing Women’S Jewelry To Help Manage Symptoms Of Anxiety Disorders, Nourbanu Feras Hijazi

Theses and Dissertations

Anxiety is a common and often stigmatized condition. Destigmatizing mental disorders can positively impact people’s interaction and communication with others and can prompt conversations in which people share their experiences with mental health, leading to improved societal understanding and perception.

My design solution tackles behavioral symptoms of anxiety, specifically two conditions that fall under obsessive-compulsive behavior: Dermatillomania and Trichotillomania. By redesigning women’s jewelry to specialized accessories, my intention is for these objects to help destigmatize these conditions and relieve symptoms of maladaptive behaviors and hurtful impulses. By redirecting these irresistible urges to an external artifact, the objects are designed to …

Impaired Cardiorespiratory Fitness Following Thoracic Radiotherapy, Justin M. Canada Jan 2018

Impaired Cardiorespiratory Fitness Following Thoracic Radiotherapy, Justin M. Canada

Theses and Dissertations

Cancer (CA) is the second leading cause of death in the United States preceded only by cardiovascular disease (CVD). Over the past 30 years, the 5-year survival rate for all cancers combined has increased by more than 20%. This improved survival rate is due to early diagnosis and advances in treatment involving a multimodality treatment approach that includes radiotherapy [RT] with about half of all CA patients receiving some type of RT sometime during the course of their treatment. Cardiotoxicity is one of the most important adverse reactions of RT and leads to a meaningful risk of CVD-related morbidity and …

Utilizing Voltage-Gated Calcium Channels To Assess The Activity Of Cathinone Derivatives At Human Monoamine Transporters, Brian A. Ruiz Jan 2018

Utilizing Voltage-Gated Calcium Channels To Assess The Activity Of Cathinone Derivatives At Human Monoamine Transporters, Brian A. Ruiz

Theses and Dissertations

Cathinones are psychostimulant compounds heavily implicated as drugs of abuse. They exert their physiological actions at the monoamine transporters, which are responsible for maintaining synaptic neurotransmitter homeostasis. Monoamine transporters produce currents during transport and have been shown to depolarize cell membranes and activate voltage-gated calcium channels in mammalian expression systems. This phenomenon is harnessed in an assay which measures these induced calcium transients, allowing for quantification of pharmacodynamic effects of compounds at monoamine transporters. It is unknown if this electrical coupling occurs in neurons, but the implications if it does are significant. In the current work, fluorescent resonance energy transfer …

Impact Of Phosphoinositides On Regulation Of K-Atp By Atp And Hydrogen Sulfide, Tyler Hendon Jan 2018

Impact Of Phosphoinositides On Regulation Of K-Atp By Atp And Hydrogen Sulfide, Tyler Hendon

Theses and Dissertations

Hydrogen sulfide (H2S) reduces ischemia reperfusion (IR) injury by stimulating adenosine triphosphate (ATP) sensitive potassium channels (KATP) [1-5]. Demonstrating H2S stimulation is unique to KATP, as other inwardly rectifying potassium (Kir) channels demonstrate inhibition or are unaffected [6]. We recently showed that H2S inhibits Kir2 and Kir3 by decreasing channel sensitivity to phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2 or PIP2) [6]. Here, we test the hypothesis that H2S regulation of Kir6.2, a pore-forming subunit of the KATP channel, is also dependent on PIP2. Using whole-cell patch-clamp we show that H2S increases the activity of Kir6.2 channels expressed in HEK-293 cells. To study the …

Oncogene Induced Dna Replication In Lung Injury Repair And Carcinogenesis, Shilpa Singh Jan 2018

Oncogene Induced Dna Replication In Lung Injury Repair And Carcinogenesis, Shilpa Singh

Theses and Dissertations

Deregulation of genome duplication is a common theme in cancer cells. Although multitude of pathways has been discovered and implicated in the ability of oncogenes or loss of function tumor suppressors to induce oncogenesis, most of the pathways ultimately converge in deregulated genome duplication or uncoupling of DNA replication and segregation in cancer cells. It is important to determine how known oncogenes deregulate genome duplication, and if prevention of deregulated genome duplication could be a feasible target to thwart uncontrolled proliferation of cancer cells. Conventional paradigm ascribes the cell proliferative function of the human oncoprotein mouse double minute2 (MDM2) primarily …

Climate Change And Mountaintop Removal Mining: A Maxent Assessment Of The Potential Dual Threat To West Virginia Fishes, Lindsey R F Hendrick Jan 2018

Climate Change And Mountaintop Removal Mining: A Maxent Assessment Of The Potential Dual Threat To West Virginia Fishes, Lindsey R F Hendrick

Theses and Dissertations

Accounts of species’ range shifts in response to climate change, most often as latitudinal shifts towards the poles or upslope shifts to higher elevations, are rapidly accumulating. These range shifts are often attributed to species ‘tracking’ their thermal niches as temperatures in their native ranges increase. Our objective was to estimate the degree to which climate change-driven shifts in water temperature may increase the exposure of West Virginia’s native freshwater fishes to mountaintop removal surface coal mining. Mid-century shifts in habitat suitability for nine non-game West Virginia fishes were projected via Maximum Entropy species distribution modeling, using a combination of …

Development Of Novel Models To Study Deep Brain Effects Of Cortical Transcranial Magnetic Stimulation, Farheen Syeda Jan 2018

Development Of Novel Models To Study Deep Brain Effects Of Cortical Transcranial Magnetic Stimulation, Farheen Syeda

Theses and Dissertations

Neurological disorders require varying types and degrees of treatments depending on the symptoms and underlying causes of the disease. Patients suffering from medication-refractory symptoms often undergo further treatment in the form of brain stimulation, e.g. electroconvulsive therapy (ECT), transcranial direct current stimulation (tDCS), deep brain stimulation (DBS), or transcranial magnetic stimulation (TMS). These treatments are popular and have been shown to relieve various symptoms for patients with neurological conditions. However, the underlying effects of the stimulation, and subsequently the causes of symptom-relief, are not very well understood. In particular, TMS is a non-invasive brain stimulation therapy which uses time-varying magnetic …
