Novel Strategies To Overcome Carboplatin Resistance In Triple Negative Breast Cancer Using Patient Derived Xenografts, Mohammad A. Al Zubi Jan 2020

Novel Strategies To Overcome Carboplatin Resistance In Triple Negative Breast Cancer Using Patient Derived Xenografts, Mohammad A. Al Zubi

Theses and Dissertations

Triple-negative breast cancer (TNBC) patients have a poor prognosis and rely on chemotherapeutic treatment as standard of care. Often, they develop chemotherapy resistance, which leaves them without more therapeutic options, like targeted therapy. New models have been developed to test targeted inhibitors in human tumors, and they are known as patient-derived xenografts (PDX). These tumors are obtained from patients, then established and maintained in mice where they areused for tumor studies. In this work, we characterized 14 PDXs for their primary tumor growth rate and investigated metastatic propensity using spontaneous and experiment metastasis models. We utilized RNA-sequencing to characterize contributions …

Analysis Of Stigmatic Content In State Mental Health Legislative Proposals, David L. Conley Jan 2020

Analysis Of Stigmatic Content In State Mental Health Legislative Proposals, David L. Conley

Theses and Dissertations

Despite the proven effectiveness of mental health interventions, services remain limited across the country. Social workers have repeatedly advocated for increased funding, but mental healthcare gaps persist. Disparities could be addressed through the policy process, but critical proposals often do not pass. One of the biggest barriers is the concept of stigma, which could extend into legislatures and influence mental health-related policy outcomes as a form of structural stigma. Factors that influence legislator voting behavior are found in the literature, but studies have not explicitly focused on structural stigma or mental health-specific policy outcomes. Thus, the present study aimed to …

The Role Of Interleukin-18 In Myocardial Ischemia/Reperfusion, Yogesh Dhakal Jan 2020

The Role Of Interleukin-18 In Myocardial Ischemia/Reperfusion, Yogesh Dhakal

Theses and Dissertations

Acute Myocardial Infarction (AMI) is one of the leading causes of morbidity for cardiovascular diseases (CVDs) in the western world. While mortality rates for AMI has reduced in the last 20 years due to advancements in clinical interventions, patients who have an AMI are significantly more likely to develop heart failure (HF) in the following years. Reperfusion of the ischemic tissue is a necessary procedure to restore blood delivery to cardiomyocytes, minimize myocardial dysfunction, and reduce infarct size. Still, the step of reperfusion itself induces myocardial damage in a phenomenon known as reperfusion injury. Considering that the degree of myocardial …

Can School Be A Source Of Trauma? Assessing Academic Traumatic Stress As A Mechanism Underlying The Health Outcomes Of Black Undergraduate Students, Ebony A. Lambert Jan 2020

Can School Be A Source Of Trauma? Assessing Academic Traumatic Stress As A Mechanism Underlying The Health Outcomes Of Black Undergraduate Students, Ebony A. Lambert

Theses and Dissertations

Research examining Black students’ school experiences demonstrates that exposure to oppressive power dynamics in schools may lead to adverse physiological and psychological consequences. Recent conceptualizations in public discourse further posit that traumatic educational experiences, operationalized here as academic trauma or the cumulative toll of adverse and oppressive experiences in academic settings, may influence Black students’ wellbeing even after they have graduated. However, academic trauma has yet to be investigated empirically, and the health contributions of such educational harm remain unstudied. Moreover, little is known about how culturally-relevant personal characteristics (e.g., emotion regulation strategies) influence Black students’ reactivity to academic trauma. …

Impact Of Performance-Based Budgeting On Quality Outcomes In U.S. Military Healthcare Facilities, Kimberly L. Decker Virginia Commonwealth University Jan 2020

Impact Of Performance-Based Budgeting On Quality Outcomes In U.S. Military Healthcare Facilities, Kimberly L. Decker Virginia Commonwealth University

Theses and Dissertations

Performance-based budgeting (PBB) is a variation of pay for performance that is applicable to government programs. It works by increasing or decreasing funding based on pre-established performance thresholds, which incentivizes organizations to improve performance. In late 2006, the U.S. Army implemented a PBB program in all of its healthcare facilities due to rising concerns over costs and quality in its facilities. The U.S. Army’s PBB program tied hospital-level funding decisions to performance on key cost and quality-related metrics. This study examined the impact of this program and a subsequent PBB program on quality improvement in U.S. Army healthcare facilities. Drawing …

Securing A Successful Future For Children With Developmental Disabilities In Ghana And Zambia: A Mixed-Methods Study Of Parental And Service Provider Expectations, Princess-Melissa T. Washington-Nortey

Theses and Dissertations

Studies suggest that parental expectations can influence the developmental trajectory of a growing child. However, the role of parental expectations in the lives of children with disabilities such as intellectual disabilities or autism, and children in Africa in general, to date, has received little attention. Using a cultural psychology framework, the current study examined parental and service provider expectations for children with intellectual disabilities or autism in two African countries: Ghana and Zambia. A mixed-method approach involving the use of concept mapping and quantitative strategies was used. A total of 20 parents and 16 service providers participated in four separate …

Focal Augmentation Of Somatostatin Interneuron Function And Subsequent Circuit Effects In Developmentally Malformed, Epileptogenic Cortex, Nicole Ekanem Jan 2020

Focal Augmentation Of Somatostatin Interneuron Function And Subsequent Circuit Effects In Developmentally Malformed, Epileptogenic Cortex, Nicole Ekanem

Theses and Dissertations

Drug-resistant epilepsy (DRE) is a common clinical sequela of developmental cortical malformations such as polymicrogyria. Unfortunately, much remains unknown about the aberrant GABA-mediated circuit alterations that underlie DRE's onset and persistence in this context. To address this knowledge gap, we utilized the transcranial freeze lesion model in optogenetic mice lines (Somatostatin (SST)-Cre or Parvalbumin (PV)-Cre x floxed channelrhodopsin-2) to dissect features of the SST, PV, and pyramidal neuron microcircuit that are potentially associated with DRE. Investigations took place within developmental microgyria’s known pathological substrate, the adjoined and epileptogenic paramicrogyral region (PMR). As well, microcircuit relationships within the previously unexplored range …

Hospital Investment In Urgent Care: Impacts On Emergent, Outpatient, And Ambulatory Surgery Volumes, Hillary A. Linhart Jan 2020

Hospital Investment In Urgent Care: Impacts On Emergent, Outpatient, And Ambulatory Surgery Volumes, Hillary A. Linhart

Theses and Dissertations

As the popularity of urgent care centers (UCCs) has grown, hospitals have increasingly chosen to invest in this care model. Hospital interest in UCCs is largely driven by two key factors. First, hospitals aim to use UCCs to generate referrals, which generate additional patient volume for downstream hospital services. Secondly, hospital UCC investment presents an opportunity to ease the demand for non-urgent care in the emergency department (ED) by diverting low-acuity ED patients to the more appropriate UCC setting. No study to date has evaluated the impact of hospital UCC investment on referrals or downstream service volumes. A few studies …

An Exploration In Accountable Care Organization Structure, Contingency And Performance, 2015-2017, Siriporn P. Satjapot

Theses and Dissertations

The Patient Protection and Affordable Care Act of 2010 enacted one of the most significant reforms seen in the United States healthcare landscape. The Center for Medicare and Medicaid (CMS) led transformation efforts in regulatory reform and coverage expansion across the U.S. population. Since 2010, care delivery systems have been shifting from episodic, decentralized and fee-for-service models to value-based population health models, like accountable care organizations (ACO). ACOs have been specifically primed for local response to improve the health of their communities. ACO research has traditionally focused on performance measures like mortality, readmissions, quality outcomes and savings. ACO organizational characteristics …

Solid Phase Extraction And High-Precision Mass Spectrometry Analysis Methods For Crassostrea Virginica Metabolome, Alyssa Darling Jan 2020

Solid Phase Extraction And High-Precision Mass Spectrometry Analysis Methods For Crassostrea Virginica Metabolome, Alyssa Darling

Theses and Dissertations

Metabolomics is the rapidly-growing field which focuses on the endpoints of biological processes, and has the capability to provide a snapshot of an organisms’ interaction with their environment on a molecular scale. The objective of the experiment herein was to develop a standard metabolic extraction technique coupled with ultrahigh-precision analytical tools to build a global metabolic pathway of the keystone species Crassostrea virginica. To achieve this, C. virginica whole-body samples were extracted using acetonitrile and methanol, then split into a dehydrated group and a diluted group. Following reversed-phase solid phase extraction (RP-SPE) of both groups, metabolites were identified using …

Spag17 Is Important For Protein Trafficking In Mammalian Spermiogenesis, Virali Bhagat Jan 2020

Spag17 Is Important For Protein Trafficking In Mammalian Spermiogenesis, Virali Bhagat

Theses and Dissertations

Spermiogenesis is the process through which undifferentiated germ cells develop into mature spermatozoa. While spermiogenesis is a very well-regulated process, the protein-protein interactions regulating it remain poorly understood. A knockout (KO) mouse for the ciliary protein SPAG17 was generated by our lab. Loss of SPAG17 has been shown to disrupt the transport of proteins important for acrosome biogenesis and manchette functions. With this information, we hypothesized that SPAG17 plays an essential role in protein trafficking during mammalian spermiogenesis. To further investigate this, immunofluorescence (IF) studies were performed in germ cells collected from both WT and SPAG17 KO mice to visualize …

Prediction Of The Vaginal Microbiome Using Urine Samples, Christopher R. Diachok Jan 2020

Prediction Of The Vaginal Microbiome Using Urine Samples, Christopher R. Diachok

Theses and Dissertations

The vaginal microbiome is associated with women’s health, including but not limited to the pathogenesis of bacterial vaginosis, adverse pregnancy outcomes including preterm birth and sexually transmitted infections. However, there has been very limited research on the urinary microbiome and its relationship to the vaginal microbiome. We sought to predict the vaginal microbiome profile using clean catch urine samples from the same person. Here, paired vaginal and urine samples were collected and sequenced by 16S ribosomal RNA sequencing. Alpha and beta diversity analysis showed that there was no significant difference between the paired vaginal and urine microbiomes. In fact, a …

Anticholinergic Burden And Risk Of Cognitive Impairment In Older Adults, Syeda Hashimi Jan 2020

Anticholinergic Burden And Risk Of Cognitive Impairment In Older Adults, Syeda Hashimi

Theses and Dissertations

Studies reveal that 10-27% of older adults chronically use anticholinergic medications. Increased cumulative anticholinergic burden scores are associated with increased risks of dementia. The mechanisms by which anticholinergic drugs negatively impact cognition remain elusive. However, researchers speculate that the drug’s impairment of cholinergic neurons promotes neuroinflammation.

We hypothesize that drugs with anticholinergic properties will induce inflammation in the brain. MCP1 and IL 6 are chemokines that contribute to neuroinflammation. We investigated the influence of diphenhydramine (Benadryl) on the production of MCP1 and IL-6 in Normal Human Astrocytes and Paroxetine on the production of MCP1 in Normal Human Astrocytes and Normal …

An Intuitive Eating Intervention For College Women With Disordered Eating: Evaluating Two Accessible And Affordable Approaches, Carolyn B. Burnette Jan 2020

An Intuitive Eating Intervention For College Women With Disordered Eating: Evaluating Two Accessible And Affordable Approaches, Carolyn B. Burnette

Theses and Dissertations

Undergraduate women of all racial and ethnic groups engage in disordered eating behaviors (DEBs) at alarming rates. Most women do not receive treatment, with the largest disparities observed in women of color and those at higher weights. Prevailing interventions have limitations that could exacerbate these disparities, such as an emphasis on the thin ideal, which is often considered less relevant for some women of color. This is concerning, as disordered eating is often chronic and symptom crossover is common. Thus, a transdiagnostic, inclusive secondary prevention approach could enhance outcomes by addressing the spectrum of DEBs in diverse women. The purpose …

Precarity In Feminism And Feminist Art Education: Decentering Whiteness Through Reproductive Justice Activism, Michelle Bae-Dimitriadis, Olga Ivashkevich Jan 2020

Precarity In Feminism And Feminist Art Education: Decentering Whiteness Through Reproductive Justice Activism, Michelle Bae-Dimitriadis, Olga Ivashkevich

Journal of Social Theory in Art Education

The article addresses precarity in mainstream feminism and feminist art education as a systemic dismissal and exclusion of the critical concerns and voices by disenfranchised women of color from its narratives and agendas. It draws on a case of the reproductive justice feminist activism to illustrate how the mainstream pro-choice feminist movement neglected the urgent and often life threatening reproductive concerns by Black, Brown, Indigenous and immigrant women, which led to an establishment of the reproductive justice coalitions by activists of color. The reproductive justice movement is an important call to action to challenge and decenter Whiteness in mainstream feminism …

Don’T Call This World Adorable & Other Salvaged Stories, Brooke A. Hofsess Jan 2020

Don’T Call This World Adorable & Other Salvaged Stories, Brooke A. Hofsess

Journal of Social Theory in Art Education

In this contribution to the special issue on Precarity, the author builds salvaged stories that touch ecological precarities related to place-based discourses within art education. Tsing’s (2015) attentiveness to precarity and Alaimo’s (2012; 2016) suspicion of sustainability cascade upon the author’s thinking/living/writing with Ecologies of Girlhood, an interdisciplinary, intersectional, intergenerational arts program considering living feminist lives co-creatively with place (Ahmed, 2017). New material feminism moves alongside an Indigenous ontology of land-based pedagogies creating “inter-theoretic conversations” (Rosiek, Snyder, & Pratt, 2019) about sustainability and place. These accounts are simply told and cultivated from everyday practices that explore a craftsmanship of attention …

The Substantiality Of The Neuroplasticity Hypothesis Of Major Depressive Disorder: The Prospective Use Of Ketamine-Like Drugs As Antidepressants, Roma Kankaria Jan 2020

The Substantiality Of The Neuroplasticity Hypothesis Of Major Depressive Disorder: The Prospective Use Of Ketamine-Like Drugs As Antidepressants, Roma Kankaria

AUCTUS: The Journal of Undergraduate Research and Creative Scholarship

Abstract

Major depressive disorder (MDD) affects approximately 17.3 million adults in the United States each year. For more than 50 years, the serotonin hypothesis of MDD, which hypothesizes that a deficiency of monoaminergic neurotransmitters results in depression, has been the foundation for neuropsychological research. However, studies reveal that only an estimated 50% of MDD patients respond to traditional, biogenic-amine-based antidepressants (ADs), like selective serotonin reuptake inhibitors (SSRIs). Research has noted that the neuroplasticity hypothesis, which posits that weakened excitatory synaptic transmission results in depression, offers an alternative mechanism by which ketamine-like drugs lacking the abuse liability and psychoactive effects of …

Curricular Change In Institutional Context: A Profile Of The Summit-P Institutions, Mary D.R. Beisiegel, Suzanne I. Dorée Jan 2020

Curricular Change In Institutional Context: A Profile Of The Summit-P Institutions, Mary D.R. Beisiegel, Suzanne I. Dorée

Journal of Mathematics and Science: Collaborative Explorations

There is a national call to improve the mathematics curricula in the first two undergraduate years to improve student success and engagement. But curricular change happens in an institutional context: Who are the students, and what do they need to succeed? What is the climate for change? Does the department regularly revise its courses and curriculum? Is it common for different departments to collaborate on curricular change? What supports or obstacles does the department, college, or university have for changing the curriculum? Who are the institutional stakeholders, and what practices build their buy-in? In the SUMMIT-P project, nine different institutions …

Stigma, Confinement, And Silence : On The Precarious Life And Death Of John Derby, Kevin Tavin, Mira Kallio-Tavin

Journal of Social Theory in Art Education

In this commentary, we take seriously the question of what does it mean to be in a precarious position and a precarious subject within educational institutions. Structured around three concepts, Stigma, Confinement, andSilence we discuss the life and death of art education scholar and colleague, John Derby. We attempt to address how John’s scholarship helped other researchers in art education orientate themselves and take a critical stance based on disability studies.

Exploring Whether Contextual Factors Relate To The Hoped-For And Feared Possible Selves Of Latinx Youth, Stephanie M. Romo Jan 2020

Exploring Whether Contextual Factors Relate To The Hoped-For And Feared Possible Selves Of Latinx Youth, Stephanie M. Romo

Theses and Dissertations

Youth’s hopes and fears for their future (i.e., hoped-for and feared possible selves) are related to their academic and health behavior outcomes. Much of the literature on possible selves focuses on African-American and European-American youth. In a sample of 132 Latinx youth, the present cross-sectional study utilized widely-used measures (i.e., Possible Selves Questionnaire, Beliefs about Education, Youth Risk Surveillance Survey) to identify the content and characteristics (e.g., strategies, balance) of their possible selves; investigate the relationship between contextual factors (e.g., sex, immigrant status) and possible selves; and, explore whether possible selves predicted youth’s beliefs about education and sexual risk behaviors. …

Hidden Gem: Realizing The Value Of Community In Healthcare Environments, Nicole Lee Jan 2020

Hidden Gem: Realizing The Value Of Community In Healthcare Environments, Nicole Lee

Theses and Dissertations

Holistic approaches in healthcare address a problem from its root causes. The American healthcare model – both its treatment and distribution methods – is among the many issues that divides the nation. We have found ourselves with millions of uninsured people among a growing population where chronic illness is on the rise among adults and children, especially within low-income demographics (Goodman & Conway, 2016). Though this is a widespread epidemic, only recently has there been any effort to question and reform traditional healthcare.

The way that wellness is assessed in America has been detrimental to patient outcomes …
