Triple-negative breast cancer (TNBC) patients have a poor prognosis and rely on chemotherapeutic treatment as standard of care. Often, they develop chemotherapy resistance, which leaves them without more therapeutic options, like targeted therapy. New models have been developed to test targeted inhibitors in human tumors, and they are known as patient-derived xenografts (PDX). These tumors are obtained from patients, then established and maintained in mice where they areused for tumor studies. In this work, we characterized 14 PDXs for their primary tumor growth rate and investigated metastatic propensity using spontaneous and experiment metastasis models. Despite the proven effectiveness of mental health interventions, services remain limited across the country. Social workers have repeatedly advocated for increased funding, but mental healthcare gaps persist. Disparities could be addressed through the policy process, but critical proposals often do not pass. One of the biggest barriers is the concept of stigma, which could extend into legislatures and influence mental health-related policy outcomes as a form of structural stigma. Acute Myocardial Infarction (AMI) is one of the leading causes of morbidity for cardiovascular diseases (CVDs) in the western world. While mortality rates for AMI has reduced in the last 20 years due to advancements in clinical interventions, patients who have an AMI are significantly more likely to develop heart failure (HF) in the following years. Connectivity and habitat area are important landscape characteristics that drive patterns of abundance and diversity across scales. However, responses to connectivity and patch size are dependent on species traits. Riverine landscapes are highly dynamic both spatially and temporally with hydrologic connectivity being a major driver of abundance and diversity. Here we modeled densities of the Virginia river snail and skimmer dragonfly nymphys, two taxa with differences in their dispersal abilities and life histories, as a function of flooding, patch area, and season in over 300 riverine rock pools. Drug-resistant epilepsy (DRE) is a common clinical sequela of developmental cortical malformations such as polymicrogyria. Unfortunately, much remains unknown about the aberrant GABA-mediated circuit alterations that underlie DRE's onset and persistence in this context. To address this knowledge gap, we utilized the transcranial freeze lesion model in optogenetic mice lines (Somatostatin (SST)-Cre or Parvalbumin (PV)-Cre x floxed channelrhodopsin-2) to dissect features of the SST, PV, and pyramidal neuron microcircuit that are potentially associated with DRE. Metabolomics is the rapidly-growing field which focuses on the endpoints of biological processes, and has the capability to provide a snapshot of an organisms’ interaction with their environment on a molecular scale. The objective of the experiment herein was to develop a standard metabolic extraction technique coupled with ultrahigh-precision analytical tools to build a global metabolic pathway of the keystone species Crassostrea virginica. Spermiogenesis is the process through which undifferentiated germ cells develop into mature spermatozoa. While spermiogenesis is a very well-regulated process, the protein-protein interactions regulating it remain poorly understood. A knockout (KO) mouse for the ciliary protein SPAG17 was generated by our lab. Loss of SPAG17 has been shown to disrupt the transport of proteins important for acrosome biogenesis and manchette functions. With this information, we hypothesized that SPAG17 plays an essential role in protein trafficking during mammalian spermiogenesis. β-hemoglobinopathies affect millions of people around the world. Research into treatments for these conditions has focused on methods to increase γ-globin expression, because increased levels of γ-globin ameliorate or reduce the severity of symptoms. As more and more studies have been done, a few proteins have emerged as having crucial roles in γ-globin repression and have been established as key genes to study. Studies reveal that 10-27% of older adults chronically use anticholinergic medications. Increased cumulative anticholinergic burden scores are associated with increased risks of dementia. The mechanisms by which anticholinergic drugs negatively impact cognition remain elusive. However, researchers speculate that the drug’s impairment of cholinergic neurons promotes neuroinflammation.

We hypothesize that drugs with anticholinergic properties will induce inflammation in the brain. MCP1 and IL 6 are chemokines that contribute to neuroinflammation. The SUMMIT-P project is a multi-institutional endeavor to leverage interdisciplinary collaboration in order to improve the teaching of undergraduate mathematics courses in the first two years of college. One goal of this work is to establish collaborative communities among the institutions involved. As part of the project, institutions visit one another on site visits that are structured according to a common protocol. The site visits have been valuable to the project. Translingual Public Pedagogy, Precarity And Inquiry: Learned Limits And Limitlessness Through Memoir, Melisa Cahnmann Taylor, Sharon Nuruddin, Tairan Qiu

In this paper we document the precarity of translingual pedagogies (Canagarajah, 2013), those occurring when languages are in contact and mutually influencing each other with emergent meanings and grammars. Sharing translingual art and literature during a public Lunar New Year Celebration, we turn to memoir methodologies for understanding “trans” practices: those that transgress singular codes, modalities, human/nonhuman cultures, and personal-scientific boundaries. Our findings (and lost things) identify the challenges and possibilities of working in the liminal space of fractured social and linguistic identities.

In this contribution to the special issue on Precarity, the author builds salvaged stories that touch ecological precarities related to place-based discourses within art education. Tsing’s (2015) attentiveness to precarity and Alaimo’s (2012; 2016) suspicion of sustainability cascade upon the author’s thinking/living/writing with Ecologies of Girlhood, an interdisciplinary, intersectional, intergenerational arts program considering living feminist lives co-creatively with place (Ahmed, 2017). New material feminism moves alongside an Indigenous ontology of land-based pedagogies creating “inter-theoretic conversations” (Rosiek, Snyder, & Pratt, 2019) about sustainability and place. Major depressive disorder (MDD) affects approximately 17.3 million adults in the United States each year. For more than 50 years, the serotonin hypothesis of MDD, which hypothesizes that a deficiency of monoaminergic neurotransmitters results in depression, has been the foundation for neuropsychological research. However, studies reveal that only an estimated 50% of MDD patients respond to traditional, biogenic-amine-based antidepressants (ADs), like selective serotonin reuptake inhibitors (SSRIs). Data suggest that individuals’ purported dietary patterns influence others’ perceptions of them. However, few studies have investigated how adherence to specific popular diets might influence these perceptions. This study investigated female undergraduates’ (n = 463) perceptions of vignette characters described as adhering to specific dietary practices perceptions of a vignette character in a sample of 463 female undergraduates. Characters described as adhering to a Clean Eating diet were viewed most positively, followed by individuals on the Ketogenic diet or nondieters. Characters following an Intermittent Fasting diet, and those with Orthorexia Nervosa were viewed most negatively. These findings support the …

Plants are particularly vulnerable to physical disturbance in low productivity areas, due to a high cost of replacing lost plant tissue. In the eastern United States, serpentine grasslands are fragmented ecosystems with high concentrations of rare endemic plant species, low concentrations of soil nutrients, and uncontrolled deer overpopulation. Test article CC-325 is a potent oral cereblon (CRBN) modulator that has shown potent G1 to S phase transition 1 (GSPT1) degradation and anti-tumor activity in pre-clinical models. One of the adverse effects associated with CC-325 was dose dependent hypocalcemia, which was determined to be an on-target toxicity.

To investigate the mechanism of hypocalcemia, we conducted a toxicity study in human cereblon (huCRBN) knock-in (KI) mice with CC-325. In immune-competent individuals, tumor growth occurs due to a failure of the immune system to recognize and destroy malignant cells. Immune surveillance can be curtailed by the presence of immunosuppressive cells such as myeloid-derived suppressor cells (MDSCs) and T-regulatory cells (Tregs) which both serve to dampen antitumor immunity. These studies utilize a metronomic low-dose therapeutic approach to examine the effects of anticancer drugs against murine breast cancer and the effects on the immune cell compartments. The increasing use of quantitative real-time polymerase chain reaction (qPCR) as a method for quantifying gene expression has led to an increased demand for standardization of data analysis methods to ensure accurate reporting and robust, reproducible results. The exponential nature of qPCR amplification results in the potential magnification of what are usually very small sources of error. Relative gene expression calculations circumvent this issue by normalizing target gene expression data to within-sample expression of a previously validated, stably expressed reference gene or genes.
