Studies Of The Middle Class, Scott S. Stanard Jan 2014

Studies Of The Middle Class, Scott S. Stanard

Theses and Dissertations

I create oil paintings and drypoints that bring attention to scenes of American middle class life. I present crowds of people engaged in community activities and festivals. This enables me to depict a cross section of the residents of a given region. I also create a counterpoint to these peopled gatherings by painting and drawing the exteriors of middle class homes. In my work, I attempt to imbue a sense of intrigue and pathos.

I work from candid photos that I take of people and their living spaces. In these photos, I modify and delete elements to optimize narrative and …

Regulation And Mechanistic Functions Of Caspase-9 Rna Splicing, Ngoc T. Vu Jan 2014

Regulation And Mechanistic Functions Of Caspase-9 Rna Splicing, Ngoc T. Vu

Theses and Dissertations

Caspase-9 has two splice variants, pro-apoptotic caspase-9a and anti-apoptotic caspase-9b, and dysregulation of caspase-9 splice variant ratio or expression of caspase-9b isoform has been linked to augmentation of the anchorage-independent growth and tumorigenic capacity of non-small cell lung cancer (NSCLC) cells. This study focuses on cell signaling pathway(s) regulating the alternative splicing of caspase-9 pre-mRNA and mechanistic roles of caspase-9b in a certain oncogenic/survival pathway. In regards to the former, we have identified hnRNP U as a novel splice-enhancer associated with exon 3 of caspase-9 (C9/E3). Moreover, hnRNP U binds specifically to C9/E3 at an RNA cis-element previously reported …

The Effect Of Fluvastatin On Mast Cell Function: Genotype Dependence, Elizabeth M. Kolawole Jan 2014

The Effect Of Fluvastatin On Mast Cell Function: Genotype Dependence, Elizabeth M. Kolawole

Theses and Dissertations

Fluvastatin, the HMG-CoA reductase inhibitor known for its role in the treatment of hypercholesterolemia and cardiovascular disease, has more recently been shown to play a role in the immune response. Given the critical role that mast cells play in allergy and inflammatory diseases such as asthma, which effects one third of America’s population, we assessed the effect of fluvastatin on mast cell and basophils function. We demonstrate that fluvastatin downregulated IgE-mediated cytokine production. Additionally, in vivo studies showed that fluvastatin suppressed IgE-mediated anaphylaxis. Interestingly, the effects of fluvastatin showed dependence on genetic background, as C57BL/6 mast cells were sensitive, while …

The Effect Of Lactic Acid On Mast Cell Function, Andrew J. Spence Jan 2014

The Effect Of Lactic Acid On Mast Cell Function, Andrew J. Spence

Theses and Dissertations

This study shows for the first time the effect that L-(+)-lactic acid has on mast cell activation. Lactic acid is a byproduct of anaerobic glycolysis and is associated with inflammatory environments such as wounds, tumors and, asthma. In this study, pre-treatment with lactic acid altered cytokine production by bone marrow-derived mast cells (BMMC). Specifically, lactic acid enhanced cytokine secretion following IgE cross-linking, but decreased IL-33 mediated cytokine production. These effects were altered by genetic background, since C57BL/6 mast cells demonstrated the aforementioned result, but lactic acid had no effect on IgE-mediated cytokine production in 129/SvJ mast cells. The affected cytokines …

Analysis And Modeling Of The Roles Of Actin-Myosin Interactions In Bladder Smooth Muscle Biomechanics, Seyed Omid Komariza Jan 2014

Analysis And Modeling Of The Roles Of Actin-Myosin Interactions In Bladder Smooth Muscle Biomechanics, Seyed Omid Komariza

Theses and Dissertations

Muscle mechanical behavior potentially plays an important role in some of the most common bladder disorders. These include overactive bladder, which can involve involuntary contractions during bladder filling, and impaired contractility or underactive bladder, which may involve weak or incomplete contractions during voiding. Actin-myosin cross-bridges in detrusor smooth muscle (DSM) are responsible for contracting and emptying the bladder. The total tension produced by muscle is the sum of its preload and active tensions. Studies suggest that actin-myosin cross-links are involved in adjustable preload stiffness (APS), which is characterized by a preload tension curve that can be shifted along the length …

Modeling Acute Changes In Bladder Wall Tension, Shape And Compliance During Filling, Firdaweke Habteyes Jan 2014

Modeling Acute Changes In Bladder Wall Tension, Shape And Compliance During Filling, Firdaweke Habteyes

Theses and Dissertations

The bladder wall consists primarily of detrusor smooth muscle. Tension-sensitive nerves in the bladder wall are responsible for providing bladder fullness information that is interpreted as urgency. Bladder wall tension, and therefore nerve output, is a function of bladder volume, shape and material properties. Studies have shown that the bladder wall exhibits acutely regulated detrusor compliance. In addition, bladder shape throughout filling depends on intra-abdominal forces and material properties of the bladder wall, such as regulated detrusor compliance. This thesis focused on modeling the potential influence of acute changes in bladder compliance, shape and bladder wall tension during filling. Laplace’s …

The Creatures We “Assessinate”: Mental Testing As Science Fiction In Chicago Public High Schools In 1909, Clayton Funk Jan 2014

The Creatures We “Assessinate”: Mental Testing As Science Fiction In Chicago Public High Schools In 1909, Clayton Funk

Journal of Social Theory in Art Education

This article tracks the development of what educators and psychologists, in 1909, termed “mental testing” in relation to art education in Chicago Public Schools (CPS). According to CPS Superintendent Edwin G. Cooley (1857-1923) American civilization was in trouble due to the influx of Southern and Eastern European immigrants in Chicago. He and other educators sought to ward off the social collapse they feared with the efficiency of science. As part of what Sol Cohen termed the “medicalization of education,” Chicago’s Department of Child Study tested students for mental capacity and those considered less intelligence were placed in technical classes, while …

The Group Iva Cytosolic Phospholipase A2/C1p Interaction And Its Role In Eicosanoid Synthesis And Inflammation, Jennifer A. Mietla Jan 2014

The Group Iva Cytosolic Phospholipase A2/C1p Interaction And Its Role In Eicosanoid Synthesis And Inflammation, Jennifer A. Mietla

Theses and Dissertations

In the presented study, we demonstrate that the interaction of group IVA cytosolic phospholipase A2 and ceramide-1-phosphate is crucial for production of eicosanoid synthesis in inflammation. Inflammation is a critical component of many disease states including anaphylaxis, cancer, cardiovascular disease, rheumatoid arthritis, diabetes and asthma. Eicosanoids are well established mediators of inflammation, and the initial rate limiting step in the production of eicosanoids is the liberation of arachidonic acid (AA) from membrane phospholipids by a phospholipase A2 (PLA2). The major phospholipase involved in this liberation of AA during the inflammatory response is group IVA cytosolic phospholipase A2 (cPLA2α). Previous studies …

Examining The Effects Of Human-Animal Interaction On Individuals With Developmental Disabilities, Jennie Feinstein Jan 2014

Examining The Effects Of Human-Animal Interaction On Individuals With Developmental Disabilities, Jennie Feinstein

Theses and Dissertations

Companion animals play a pivotal role in typical human development. It remains unknown how animals affect individuals with developmental disabilities. Based on the knowledge that companion animals help typically developing individuals, this research examined the effects of human-animal interactions on individuals with developmental disabilities.

Human-animal interactions are based on the Biophilia hypothesis, an assertion that an emotional and beneficial relationship exists between humans and nature, in which there is an “innate tendency to focus on life and lifelike processes.” These are the shared, dynamic associations between people and animals, and the effects of those relationships on health and well-being. Sparse …

Speech Is My Hammer, It's Time To Build: Hip Hop, Cultural Semiosis And The Africana Intellectual Heritage, Samuel T. Livingston Jan 2014

Speech Is My Hammer, It's Time To Build: Hip Hop, Cultural Semiosis And The Africana Intellectual Heritage, Samuel T. Livingston

Journal of Hip Hop Studies

The article examines Hip Hop music's relationship with African cultural symbolism and the discipline of Africana Studies. The author maintains that Africana Studies must reclaim the study of cultural semiosis, which may be used to contextualize Hip Hop praxis. Examining semeiotic traces within African and Afrodiasporic primary sources, including Hip Hop lyrics, the article posits that Hip Hop is the latest development in a long tradition of Afro-Kemetic oral artistry, semeiotic systems and the uses of these dual literacies as modes of resistance and affirmations of Black historical and cultural agency. The article adapts Harryette Mullen's literary model of African …

Listen To The Story: Banksy, Tyler The Creator, And The Growing Nihilistic Mindset, Duri Long

Journal of Hip Hop Studies

Art, as an expression of feelings, worldviews, and personal beliefs, is a reflection of our environment and how we interact with it. In this way, urban art such as rap music and graffiti can serve as a lens through which we are able to examine the state of the urban environment. Building on community literature that addresses the presence of nihilism in rap music, this work will establish that nihilism is a prevalent theme in the work of two artists: Tyler the Creator's rap music and BANKSY's graffiti art. By examining the growing subculture and appeal of urban art in …

Side A, Side B, Hong Zhang Jan 2014

Side A, Side B, Hong Zhang

Theses and Dissertations

I believe that aural and visual language can contribute and inspire one another. Sound evokes the emotions and the senses. It is a catalyst for our imagination. It can be combined with imagery for the purpose of communication, harmoniously or contrastively. Both have corresponding characteristics—color/timbre, brightness/pitch, length/duration, scale/volume, composition/rhythm—and thus provide ways of transforming each other back and forth as input and output. Computer technology has increased the ease of transforming and manipulating the two. In my thesis project, I explore sound visualization, sound/image transformation and combination.

Investigating The Effects Of Applied Electric Fields On Microglial Cell Behaviour, Eman Bani Jan 2014

Investigating The Effects Of Applied Electric Fields On Microglial Cell Behaviour, Eman Bani

Theses and Dissertations

As surveyors of the central nervous system (CNS), microglial cells play an integral part in the inflammatory response following traumatic injuries. Thus, they have been implicated in the limited capability of neurons to regenerate in the CNS. Additionally, the roles of endogenous electric fields in the regenerative process of neurons in the mammalian peripheral nervous system (PNS) or amphibian CNS have long been studied. Further, previous studies in our lab have shown that physiological electric fields are capable of directing behaviours in astrocytes and schwann cells. Therefore in this study, a BV-2 microglia cell line was utilized to investigate whether …

Identification Of Peptidases In Highly-Pathogenic Versus Weakly-Pathogenic Naegleria Fowleri Amebae, Ishan Vyas Jan 2014

Identification Of Peptidases In Highly-Pathogenic Versus Weakly-Pathogenic Naegleria Fowleri Amebae, Ishan Vyas

Theses and Dissertations

Naegleria fowleri, a free-living ameba, is the causative agent of Primary Amebic Meningoencephalitis. Highly-pathogenic mouse-passaged amebae (Mp) and weakly-pathogenic axenically-grown (Ax) N. fowleri were examined for peptidase activity. Zymography and azocasein peptidase activity assays demonstrated that Mp and Ax N. fowleri exhibited a similar peptidase pattern. Prominent for whole cell lysates, membranes and conditioned medium from Mp and Ax amebae were the presence of an activity band of approximately 58kDa and 100 kDa bands susceptible to the action of cysteine and metallopeptidase inhibitors, respectively. Further roles of the peptidases during the invasion process were examined by in vitro invasion assays …

Using Structural Analysis To Investigate The Function Of Suppressor Of Ikk-Epsilon (Sike), Sean W. Mckinley Jan 2014

Using Structural Analysis To Investigate The Function Of Suppressor Of Ikk-Epsilon (Sike), Sean W. Mckinley

Theses and Dissertations

The innate immune system provides the body’s first line of defense against pathogenic challenge through pathogen recognition and initiation of the immune response. Among the various cellular mechanisms of pathogen recognition in mammals, Toll-like receptor 3 (TLR3) recognizes viral dsRNA. Stimulation of TLR3 signaling pathway leads to transcription of pro-inflammatory cytokines and type-1 Interferons. Suppressor of IKKε (SIKE) interacts with two kinases in the signaling pathway, IKKε and TANK binding kinase 1 (TBK1), inhibiting the transcription of type I interferons. Recently, the Bell Laboratory discovered that SIKE blocks TBK1-mediated activation of type I interferons by acting as a high affinity, …

Group Via Calcium-Independent Phospholipase A2 Regulates Bcl-Xl Protein Levels In Mice Lung, Sang-Jin Nam Jan 2014

Group Via Calcium-Independent Phospholipase A2 Regulates Bcl-Xl Protein Levels In Mice Lung, Sang-Jin Nam

Theses and Dissertations

With previous indication of the Group VIA phospholipase A2 (iPLA2β) enzyme regulating ER-stress induced apoptosis in β-cells by regulating the anti-apoptotic protein Bcl-xL via alternative splicing, our lab postulated iPLA2β to be utilizing a similar mechanism to regulate apoptosis in mice lung. Our previous lab work has shown implications of lung function compromise in iPLA2β-/- mice, and we speculated the cause to be due altered lung architecture stemming from the attenuation of apoptosis. The western blot analysis in this study suggested that iPLA2β is involved in the regulation of Bcl-xL, but the mRNA ratios of the splice variants suggested that …

Transformational Directing: An Analysis On How Leadership Affects The Creative Process, James Clayton Winters Jan 2014

Transformational Directing: An Analysis On How Leadership Affects The Creative Process, James Clayton Winters

Theses and Dissertations

Leadership is often ascribed as a quality one either possess or doesn’t. We often hear phrases like “She is born leader” or “All eyes are on you”, these are thrown around without discretion in the classroom, conference rooms, social gatherings, and business meetings. This work contends that leadership and in particular leadership that grows and transforms its participants is less an inherent character trait, than an outlook and set of core principles that lead to transformation. Through the application of current and developing business leadership theory and its effect on creativity and a cross analysis of “Best in Class” theatre …

Investigating The Role Of The Synaptic Transcriptome In Ethanol-Responsive Behaviors, Megan A. O'Brien Jan 2014

Investigating The Role Of The Synaptic Transcriptome In Ethanol-Responsive Behaviors, Megan A. O'Brien

Theses and Dissertations

Alcoholism is a complex neurological disorder characterized by loss of control in limiting intake, compulsion to seek and imbibe ethanol, and chronic craving and relapse. It is suggested that the characteristic behaviors associated with the escalation of drug use are caused by long-term molecular adaptations precipitated by the drug’s continual administration. These lasting activity-dependent changes that underlie addiction-associated behavior are thought, in part, to depend on new protein synthesis and remodeling at the synapses. It is well established that mRNA can be transported to neuronal distal processes, where it can undergo localized translation that is regulated in a spatially restricted …

Evaluation Of The Allometric Exponents In Prediction Of Human Drug Clearance, Da Zhang Jan 2014

Evaluation Of The Allometric Exponents In Prediction Of Human Drug Clearance, Da Zhang

Theses and Dissertations

Background. Allometric scaling (AS) is widely used in predicting human clearance (CL) based on animal data. Substantial prediction errors have been commonly observed and various modifications to AS have not provided a broad reliable improvement. In this study, an extensive data set was assembled including animal and human systemic CL and physiochemical properties. The allometric exponents were calculated based on multiple species AS and single-species AS methods. The correlations between the allometic exponents and physiochemical properties were evaluated in an attempt to find covariates that may explain the inter-compound variability in the allometric exponents. Lastly, the statistical approaches in analyzing …
