Modulation Of Fast And Slow Inactivation In Two Cardiac Nav Channel Isoforms By Sdz 211-939, Tyce Jeffrey Kearl

Here we report a hitherto unknown effect of a synthetic inactivation inhibitor on inactivation in cardiac sodium channels (Nav1.5) from two different species: human and bovine. SDZ 211-93 9 stabilized the slow inactivated-state in both channels as seen by an increased steady-state probability of slow inactivation. SDZ also destabilized the fast-inactivated state and increased the amplitude of persistent currents. SDZ modulated conductance parameters, open-state fast inactivation time constants, and activation kinetics of hNav1.5, but not bNav1.5. These findings will aid future studies designed to elucidate the binding site and molecular mechanisms of inactivation inhibitors …

Ferritin is the iron storage protein found in humans, animals, plants, fungi and bacteria. We are interested in how iron is loaded and stored in mammalian ferritin. Ferrous iron must be oxidized to ferric iron in order to be stored in ferritin. It is generally believed that ferritin does the loading itself, dependant upon a "ferroxidase activity." Oxidation of iron can result in the production of the hydroxyl radical which can cause oxidative damage to surrounding proteins and other biomolecules. Increasing concern about the environmental impacts of greenhouse gases and PM 2.5 particulates has prompted many researchers to examine the processes of gaseous loss of nitrogen (N) from agricultural land. As agricultural production becomes more competitive and producers strive to become more efficient by reducing input costs, they will increasingly employ practices such as the rotational stocking, also called Management Intensive Grazing (MIG). R-type channels are voltage dependent calcium channel subtypes. To date, the only variant of the R-type calcium channel that has been isolated and cloned is the Cav2.3 variant, encoded by the gene Cchral (Yamazaki et al., 1998). The channel was named "R-type" because it was initially characterized by its resistance to nifedipine (a blocker of L-type calcium channels), Ω-conotoxin GVIA (an N-type calcium channel blocker) and Ω -Aga IVA (a blocker of P/Q-type calcium channels) (Hille 2001). A strain of Escherichia coli was genetically modified to co-express human heme oxygenase-1 and ferritin. The E. coli were then grown with varying amounts of hemin to see if the iron released upon degradation of the hemin by heme oxygenase-1 is loaded into ferritin. Following incubation, the ferritin was purified and the amount of iron loaded into ferritin determined. It was found that ferritin purifed from E. coli expressing human heme oxygenase-1 contained more iron than E. coli that did not contain human heme oxygenase-1. Matrix metalloproteases (MMPs) are proteolytic enzymes secreted by cardiac fibroblasts. Altered expression of MMPs plays an important role in congestive heart failure and other cardiovascular diseases1,2. Emerging evidence has led us to consider the role of phosphatidylinositol 4-kinase (PtdIns 4-kinase) in regulating MMP secretion in cardiac fibroblast cells3-7. PtdIns 4-kinase is a lipid kinase involved in various cellular processes including protein secretion. This project was designed to test the hypothesis that PtdIns 4-kinase controls MMP secretion. The purpose of this grounded theory research was to determine what differentiates people with disabilities who leave Social Security Disability Insurance (SSDI) benefits due to increased income from those who cannot. Fifteen individuals, former and current participants in a Medicaid Buy-In program, were interviewed. Within the context of health and disability, four theoretical propositions were identified: education, opportunity to work, interpersonal support, and secure housing. Higher benefit levels were also found to be an effective barrier for many.

The purpose of this study was to evaluate observed parenting practices among first-generation Spanish-speaking Latinos living in Utah. Participants included 50 families with a child between the ages of 4 and 9. Parents and their child engaged in a behavioral observational task that was coded for parenting dimensions and styles to determine if they predict child outcomes among Latino families. Parents completed the Child Behavior Checklist to assess for behavioral problems. Observations were coded using the Parenting Scale, developed specifically for this study. In general, parents received high ratings on warmth and demandingness, and lower ratings on autonomy granting. Parenting …

The increasing bacterial resistance to available antibiotics requires the search for new antibacterial compounds to be broadened. This study investigated the antimicrobial properties of two secondary metabolites from fluorescent pseudomonads -- syringopeptin 25A, a lipodepsipeptide produced by Pseudomonas syringae pv. syringae, and a rhamnolipid mixture produced by Pseudomonas aeruginosa. The rate of antimicrobial action was determined by monitoring the rate of uptake of propidium iodide during exposure to the compounds. Inhibition was also confirmed by the microbroth dilution method to determine the MI Cs. The research in this dissertation focused on determining antioxidant effects of various natural antioxidants in cooked meat systems. Milk mineral (MM), spices, and raisin paste were used in cooked meat systems to verify their potential antioxidant properties.

The MM study determined the antioxidant activity of 1.5% MM added to uncured cooked beef meatballs, and possible additive effects of MM in combination with 20-ppm or 40-ppm sodium nitrate in cooked beef sausages. There was no additive inhibition of lipid oxidation in samples containing 20-ppm or 40-ppm sodium nitrite plus 1.5% MM. Cooked meat yield was not different between control meatballs and …

Use of an induced blanket reactor (IBR) to break down organic matter into methane is a financially attractive method to reduce the environmental impact of animal or industrial waste. Different fiber types were incorporated in an extruded expanded high-protein snack food. Three dietary fibers (powdered cellulose, wheat fiber, and oat fiber) were selected based on ease of extrusion, percent total dietary fiber, visible expansion, and commercial availability. A high-fiber, high-protein snack food containing whey protein, normal cornstarch, and pregelatinized waxy cornstarch was extruded using the three selected fibers. The fibers replaced the normal cornstarch at 30, 60, and 80% yielding extrudates with three fiber levels (18, 36, and 48%). Cigarette smoke contains sufficient carbon monoxide to induce maternal and fetal hypoxia. Hypoxia is a known teratogen, and consequently maternal smoking has been the focus of many studies on adverse birth outcomes, including cleft lip and palate. Current literature of epidemiological studies on smoking and clefts suggests a modest but statistically significant increase in risk of clefting associated with maternal smoking. This study used family systems theory as a lens to examine fathers' relationships with young adolescent children as it relates to the areas of time spent together, communication between generations, and paternal satisfaction in raising an early adolescent child. Similarities and differences of scores on the Parent Success Indicator were examined for 191 Caucasian American (n=110) and African American (n=81) fathers of 10- to 14-year-old-children. Results showed that increased amounts of time that fathers report spending with children was significantly associated with paternal satisfaction, communication between generations, fathers' use of time, parental frustration, and parental teaching. …

There is very little research to date on infant development in developing countries, including Paraguay. The present study aimed to determine the effects of Pastoral del Nino, an ECD program, in rural Paraguay by assessing two groups of infants. Group 1 (n = 50) was receiving no intervention and group 2 (n =58) was participating in Pastoral del Nino. Both groups were matched for SES and age of infant. Results indicate that group 2 performed better on the Bayley Scales of Infant Development Mental Development Index. Group 1 had a mean MDI score of 77.96 and group 2 …

The purpose of this study was to examine how welfare status, gender, and the interaction between welfare status and gender relate to the utilization of counseling services related to marriage (i.e., "marital help-seeking"). This study was a secondary analysis of the Utah Marriage Survey data set from 2003. Two samples were utilized in this study, and generated a total sample of I ,316 participants. All species of Tapiridae and Rhinocerotidae are threatened or endangered in the wild. Captive populations have been established for most of these species, but successful management has proved challenging. Effective ex situ conservation strategies, however, rely on the ability of zoological institutions to maintain and breed these endangered species. In this study, I examined the captive environment to identify the factors associated with reproduction, mortality, and health of rhinos and tapirs. The distribution of the cliff chipmunk in Idaho was previously considered to include only the Raft River Valley and the Goose Creek Basin. A pilot study was conducted in 2003 and 2004. Thirty-five cliff chipmunk presence locations and 124 absence locations were recorded. Habitat variables of elevation, slope, deviation from south, distance to water, and vegetation type were extracted for all of the absence and presence points by means of GIS analysis. My work focused on the study of road effects and mitigation of negative impacts of roads on wildlife. Two different studies were conducted on Interstate 15, in southern Utah. My first study reported on road effects on small mammal communities. The results suggested that overall, there was no clear effect on small mammal populations relative to distance from the road. Most small mammal species did not appear to be negatively affected by the presence of the road. Instead, the road seemed to have either a neutral or a positive effect. Ethylene is an endogenously synthesized plant hormone that dissipates quickly in field conditions and seldom exceeds five nmol mol-1. Ethylene can accumulate to 1000 times this level in closed environments. The best-known effects of ethylene are its impacts on fruit ripening and senescence, yet ethylene influences growth and development throughout the plant life cycle. At low, continuous concentrations (20 to 50 nmol mol-1), ethylene reduces yield of many plants. The distribution of species with respect to physiognomic type, patch size, shape, and environmental heterogeneity was assessed for a mosaic landscape comprised of conifer forest, deciduous forest, and shrub-steppe physiognomic types in the southwest comer of Grand Teton National Park, Wyoming. Youth with inflammatory bowel disease (IBD) have higher rates of depression than healthy youth. A cognitive-behavioral treatment, primary and secondary control enhancement training-physical illness (PASCET-PI), for depressed adolescents with IBD was associated with reductions in depressive symptoms. The purpose of this study was to examine the salience of nonspecific processes (factors inherent in any human relationship) and specific processes (factors related to therapy content) during PASCET-PI sessions and their association with improvements in psychological functioning.

This study investigated the utility of applying the social constructionist perspective to adolescent sexual minority identity development, disclosure, and identity explorations. Differences between middle and late adolescents and male and females were examined. No differences were found between middle and late adolescents on measures of identity development and identity exploration; however, differences in identity disclosure were found regarding history of accidental discovery of sexual orientation. Biological sex differences were found for identity development, disclosure, and exploration. Relationships between same- and opposite-sex attractions, behaviors, romantic experiences, and self-labels are presented. Many desert perennials are long-lived, but there are few data on ages or population structures of desert plants. We used 2 methods to estimate ages of a population of Yucca brevifolia (Joshua tree) in southwestern Utah from a 14-year census of plant sizes. Plant height at the 1st census ranged from 0.08 m to 6.0 m, and trees grew in height at a mean rate of 3.75 cm ⋅ yr–1. Plants also increased slowly in basal diameter (0.142 cm ⋅ yr–1) and branch length (0.024 m ⋅ yr–1), but basal diameter varied greatly from …

The water vapor permeability of fat barrier films has been associated with structural characteristics such as polymorphism, crystal size, and chemical composition, among others. However, no mechanistic models have been proposed to describe this relationship. In this study, we have determined the effects of processing conditions on the structure and physicochemical characteristics of four fats and their relationship to water vapor permeability. Results suggest that the solids' volume fraction and the domain size of the fat crystals seem to be the most important factors controlling water vapor migration. Solenopsis invicta Buren populations were confirmed in southern New Mexico, resulting in a quarantine of Doña Ana County in 1998; however, Solenopsis xyloni McCook remains the dominant fire ant. This study compared the queen weights and egg production of polygyne colonies of both species. There was no difference in live weights of the major worker caste; however, queens were very different in weight. In this study, S. invicta queens weighed almost twice as much as S. xyloni (S. invicta = 11.32 ± 0.25mg vs. S. xyloni = 6.40 ± 0.30mg) and produced over 50% as many eggs as S. xyloni. …
