The association of promazine-HCl with bovine serum albumin, ammonium detergent micelles, and hemoglobin-free-rabbit-erythrocyte membranes were measured using the changes in the absorption and the emission of the interactants plus the fluorescent probe 1-anilino-8-naphthalene sulfonate. The binding of a promazine to serum protein and membrane did not alter the emission of the drug. However, the binding did produce a measurable quenching of the membrane and protein fluorescence. The quenching reaction was pH sensitive for both the protein and the membrane. Promazine binding to serum protein produced a hyperchromic change in the drugs absorption spectrum which paralleled the quenching reaction. The binding …

Described are the results of harvesting the above-ground parts of an 18-year-old aspen stand and the observations made on the initial suckering of the new stand. The yields for the stand of 17.7 cunits per acre were greater than predicted yields of comparable material for the site at age 35. Suckering on the area the first year after cutting averaged 37,000 aspen stands per acre and 2.6 feet in height.

A Simple Microradiometric Technique For The Rapid Measurement Of Monoamine Oxidase Activity In Rat Testicular Minced And Teased-Tubular Preparations, Ronald L. Urry

Two radiometric techniques are described for the measurement of monoamine oxidase activity in testicular tissue preparations. These techniques utilize 5-HT-2-14C (serotonin) as the substrate and measure its conversion into 5-HIAA-14C (hydroxy-indole acetic acid). The first technique described uses a substrate concentration of 0.9 µM (0.05 µCi) in an incubation volume of 3.1 ml and 120 mg of teased-tubules. Samples are incubated 1 hour. This assay was found suitable for use with teased-tubule preparations but not for preparations containing interstitual cells. The in vitro development of Eimeria magna was studied using various cell types and compared with corresponding development observed in the intestinal epithelial cells of rabbits at 3, 4 and 5 days after inoculation. Leighton tubes were inoculated with sporozoites and the cultures examined at various intervals after incubation with phase-contrast or interference-contrast microscopy. Mucosal scrapings from infected rabbits were also examined. Mature first- and second-generation schizonts were seen in Madin-Darby bovine kidney (MDBK) cells and epithelioid embryonic bovine liver cells, whereas no development beyond the sporozoite occurred in fibroblastic, embryonic, bovine liver cells, lamb trachea cells and aggregates of …

The Regulation Of Purine Biosynthesis In A Phosphoribosylpyrophosphate Aminotransferase-Deficient Mutant Of Salmonella Typhimurium, Wen-Cherng Tsai

The regulation of the alternate first enzyme of purine de novo synthesis (ribose-5'-phosphate aminotransferase) has been examined in pur-F 341, a purine-requiring mutant of Salmonella typhimurium deficient in the primary first enzyme (PP-ribose-P amidotransferase, EC 2.4.2.14). The ribose-5'- phosphate aminotransferase is feed-back inhibited by AMP, ADP, GDP and adenosine but not by ATP, GTP, GMP and free adenine. Pyrimidines (except CDP and UDP) and thiamine compounds show no significant inhibitory effect on ribose-5'-phosphate aminotransferase activity. The alternate enzyme seems to be also controlled at the gene-level by one or more intra-cellular purines (repression-derepression). The role of the aminotransferase in …

The purpose of this paper is to advocate the use of computer modeling as a tool in the study of homeotherm energetics. The relationships between measurable environmental variables and components of productivity were reviewed and discussed as a basis for data input in the models. Photoperiod, temperature, rainfall, and wind were suggested as measurable environmental parameters. The facets of productivity which can be predicted from these parameters are the timing, the energetic cost, and the energetic capacity for productivity.

Amino Acid And Protein Metabolism During Development Of The Alfalfa Weevil Hypera Postica (Gyllenhal), Darrel J. Gunderson

Amino acid and protein metabolism were studied during growth and development of the alfalfa weevil. The amino acids were separated and identified by two-dimensional thin layer chromatography on cellulose plates and quantitative determinations were made with a ninhydrin spectrophotometric method. The radioactive isotope glucose-U-C14 was used to determine amino acid requirements and their rates of synthesis. A total of 23 amino acids were identified in the fourth instar larvae. Of these, 21 were free amino acids and 17 were protein amino acids. Limited concentrations of methionine, Y-aminobutyric acid and tryptophan in the insect were directly correlated with their dietary …

Levels of potassium in nectar of onion, Allium cepa L., varied from 3600 to 13,000 parts per million, about ten times higher than that of nectar of competing flora tested. An open-pollinated amphidiploid derived from the interspecific cross, A. cepa x A. fistulosum L., was the only onion with nectar potassium level as low as that of competing flora, and it was more attractive to honey bees, Apis mellifera L., than was the common onion. Addition of potassium salts to sugar solutions at the rate of 1500, 3000, 4500, 6000, or 7500 parts per million potassium clearly reduced uptake of …

Purification And Properties Of L-Asparaginase (L-Asparagine Amidohydrolase, Ec 3.5.1.1) From Bacillus Kaustophilus, Amin Abdul-Jabbar Al-Selami

L-asparaginase was isolated and purified 60 fold from Bacillus kaustophilus by heat treatment, ammonium sulfate fractionation, adsorption chromatography on glass beads and preparative electrophoresis. The purified sample from ammonium sulfate precipitation and adsorption chromatography on glass beads could be stored at 4 C for several weeks and could withstand freezing and thawing without loss of activity. Kinetic studies showed that this L-asparaginase has a temperature optimum of 74 C, an activation energy of 11,400 calories, a pH optimum of 9.2 in sodium carbonate-bicarbonate buffer and Michaelis constant for asparagine of 3.7 x 10-3 Mat 74 C and pH 9.2. …

Cryptogamic soil crusts within the Colorado Plateau were studied to determine their effect on infiltration rates, potential sediment production, permeability, and several chemical properties of the soil. Six different crust stages were identified. Undisturbed soil cores were used to determine intrinsic permeability under three treatments and disturbed soil samples were analyzed for pH, percent organic matter, soil texture, Ca+Mg content and total conductivity.

It was found that the cryptogamic crust had little effect on soil chemical properties. Analysis of undisturbed soil core data indicates that high cryptogamic cover tends to decrease intrinsic permeability; this effect was reinforced when cores were …

A mathematical model was developed to predict water content profiles, evapotranspiration, water flow from or to the water table, root extraction and root water potential at the surface as functions of time under unsteady state conditions.

The model was tested in the field at The Hullinger Farm near Vernal, Utah, in 1970 and 1971. Comparison of water content-depth profiles show excellent agreement at the end of a 9- day run in 1970 on oats seeded to alfalfa. In 1971 with alfalfa as the crop, the data show best agreement, between predicted and computed water content-depth profiles, 48 hours after any …

Exponential rates of digestion are described for brown trout and whitefish for July, October, December (1969), and April (1970). The slope of the line fitted to the digestion data from each month was defined as the instantaneous rate of digestion and applied to an exponential growth model to determine the instantaneous consumption rate.

The digestion and consumption rates were applied to field measurements of percent fullness to determine the amount of food material ingested during a 24 hour period. Brown trout consumed 127, 24, 19, and 84 mean percent of their stomach capacity in the July, October, December, and April …

This experiment was designed to measure in situ concentrations of NH4+ and NO3--N in a soil of the arctic coastal tundra plain, to determine if nitrification was taking place in this soil and to determine if the vascular plants growing in this soil could assimilate NH4+-N.

A small tobacco mosaic virus nucleoprotein unit, thought to be the basic infectious unit of TMV, was isolated and purified from systemically-infected Turkish tobacco leaves. This small TMV particle was characterized by a doughnut shaped form as shown in the electron micrographs and by the induction of gray lesions when inoculated on Scotia bean leaves. Four carbodiimides, organic chemical condensing reagents, were compared for effectiveness in inducing polymerization of this starting material into longer rod-shaped viral particles. Dicyclohexyl carbodiimide, a wax-like solid, was found to be the most suitable reagent when used with an organic solvent such as ethanol in …

Six herbicides (diuron, karbutilate, propham, chloropropham, atrazine and simazine) were tested for their effects on Chlorocuccum hypnosporum, Chlorella pyrenoidosa and Anabaena sp. Experiments were conducted to determine minimum lethal dosages, algicidal versus algistatic concentrations and the uptake of 14CO2 fixation by diuron containing cultures, as well as the uptake of radioactive diuron.

All three investigated species have shown a high degree of sensitivity to diuron at or below 1 ppm. Karbutilate was found to inhibit the growth of Chlorella pyrenoidosa and Anabaena sp. at lower concentrations than were required in the case of Chlorococcum hypnosporum.

Parental preference of girls and boys was studied in this research. Situations of recreation, personal and moral needs, places and things, and social events constituted the range of choices in the children's selection for one parent or the other as a companion for the activity.

Ninety children (30 preschool, 30 third graders, and 30 sixth graders) were tested individually by an interview schedule. The Parent-Affinity Perception Scale developed for this research contained 30 items. Children were asked to choose "Mother" or "Father" as a response for each item.

The Swan Peak Formation in north-central Utah thickens westward, from zero feet near Logan to 687 feet in the Promontory Range. The unit is subdivided into three distinct members: 1) A lower member of interbedded shales, limestones, and quartzites; 2) A middle member of interbedded shales and brown quartzites; and 3) An upper member of white quartzites. The Swan Peak thins southward toward the east-west-trending Tooele Arch in the area of study; this thinning probably reflects both lesser deposition and greater subsequent erosion there than elsewhere. Twenty four cattle, six each of healthy cows and calves, and cows and calves with brisket disease, were obtained, examined and slaughtered. The concentrations of calcium, chloride, cobalt, copper, iron, magnesium, molybdenum, phosphorus, potassium, sodium and zinc; and percent absolute dry matter and percent ash were determined in tissues selected from the following: cardiac, hepatic, renal, osseous, whole blood and blood serum. In addition, certain physical and biological parameters were recorded for each animal. The results were analyzed as a 2 x 2 factorial, segregating the effects of age and brisket disease, and the age-disease interaction.

Investigations of limited and selected areas along the western shore of Bear Lake were carried out to determine the relative algal productivity with relation to nutrient concentrations and incident sunlight. The primary objective of the study was to determine the effects of nutrient changes in the littoral zone on the western shore on the algal productivity and to establish a baseline of data.

Attempts were made to adapt the microtiter hemagglutination inhibition assay technique for the assay of enterotoxin A. The presence of a potent hemagglutinin in crude and partially purified preparations and the instability of sensitized erythrocytes prevented its use for routine analysis of enterotoxin from culture media and foods.

A capillary tube immunological assay was developed in which 1 μ g of enterotoxin/ml was detected in less than 1 hr . Interfacial reaction of antisera and enterotoxin solutions in a 1 mm internal diameter capillary tube allowed rapid detection and serological typing of enterotoxins.

The effects of various land uses on water quality in Summit Creek were evaluated during the period beginning March 13, 1971 and ending October 27, 1971. Potential sources of pollution investigated were: (1) septic tank use, (2) feedlot runoff, (3) urban runoff, (4) rural runoff.

Samples were collected from five sampling stations on 16 separate days during the sampling period. Analyses were performed to determine the following constituents: ammonia, nitrite, nitrate, total phosphorus, orthophosphate, coliform bacteria, chloride, suspended solids, volatile suspended solids, total carbon, organic carbon, temperature and pH.

An intensive creel census and marking program was conducted in 1969 and 1970 to make possible population estimates and estimates of harvest of cutthroat trout in the Snake River in Teton County, Wyoming. Stock density determinations made in one of the five study areas provided an estimate of 400 and 992 cutthroat trout, eight inches or more in length, per mile of stream in 1969 and 1970 respectively.

Harvest data provide an estimate of 5,207 and 5,903 cutthroat trout harvested in 1969 and 1970 respectively. The harvest estimates obtained in this study are not comparable to those made in 1967 …

Female American cockroaches showed definite age-dependent changes in levels of activity of the microsomal mixed function oxidase chain. Cytochrome P-450 levels, EPN-detoxication, and p-nitroanisole O-demethylation activities were very low in young adult insects but increased steadily reaching a peak at about 100 days in fat body and at about 90 days in midgut and hindgut. The activities then declined rapidly reaching levels of young insects at about 130 to 140 days of age. NADPH-NT-reductase activity was high in young insects and declined during the first few weeks of adult life. This activity too showed a peak at about …

The purpose of this thesis is to evaluate the potential effectiveness of the Rich County 701 Plan as a planning and management aid in controlling the development around Bear Lake. The success of the plan in integrating cultural data and natural resource data is judged on the basis of professionally accepted techniques of resource analysis and on the availability of pertinent planning data.

Guidelines for evaluating the land use controls for Bear Lake that will result from the Plan are derived from the shoreland ordinances of Wisconsin and Minnesota and the lakeshore experiences of New Hampshire and Vermont.
