Adult Phyllostomid (Bat) Enamel By Scanning Electron Microscopy - With A Note On Dermopteran Enamel, K. S. Lester, S. J. Hand, F. Vincent Sep 1987

Adult Phyllostomid (Bat) Enamel By Scanning Electron Microscopy - With A Note On Dermopteran Enamel, K. S. Lester, S. J. Hand, F. Vincent

Scanning Microscopy

This study assesses the enamel of five phyllostomids of differing feeding habits; only one example of the microchiropteran super-family Phyllostomoidae having previously been studied by SEM. A dermopteran was also examined to ascertain whether the enamel might reveal insectivore, chiropteran or primate characteristics.

The five phyllostomids were found to display the additional crystallite discontinuity feature (minor boundary plane or seam) which is a major characteristic of all the bats we have so far examined with the exception of two megachiropterans. The enamel of the four fruit and nectar feeders (Phyllostomus, Carollia, Glossophaga and Artibeus) is essentially similar and different to …

An Overview Of Platelet Structural Physiology, J. G. White Sep 1987

An Overview Of Platelet Structural Physiology, J. G. White

Scanning Microscopy

Marion Barnhart and her colleagues used light, phase contrast and scanning electron microscopy to provide a clear picture of platelet surface changes developing in response to aggregating agents. This review, in honor of Marion and her work, has sought to expand the horizon provided through study of surface alterations by peeling back the membrane of the platelet to reveal the dynamic world within. A cytoskeleton consisting of a circumferential microtubule and submembrane actin filaments supports the discoid shape of the resting cell. Following exposure to aggregating agents in suspension, to foreign surfaces or denuded blood vessels and to fibrin strands …

Scanning Electron Microscopy Of Oral Mucosa In Vivo And In Vitro: A Review, Kristina Arvidson, Roland C. Grafström, Anders Pemer Sep 1987

Scanning Electron Microscopy Of Oral Mucosa In Vivo And In Vitro: A Review, Kristina Arvidson, Roland C. Grafström, Anders Pemer

Scanning Microscopy

The oral mucosa is classified by function into lining, masticatory and specialized oral mucosa, with regional structural adaptation. In this review, the surface structures of the human oral mucosa have been studied in the scanning electron microscope (SEM). Regional variations in regard to keratinization, cell arrangements and microplications with related specific structures observed in SEM are described and correlated with the appearance of similar areas observed in the light microscope. Furthermore, human oral tissue and cell cultures have also been studied. These systems offer usable and complementary models for performing similar studies in vitro under controlled experimental conditions. We now …

Scanning Microscopic Observations On Dental Caries, Sheila J. Jones, Alan Boyde Aug 1987

Scanning Microscopic Observations On Dental Caries, Sheila J. Jones, Alan Boyde

Scanning Microscopy

This paper presents findings made using special techniques of imaging and/or of specimen preparation to investigate the changes in tooth structure which occur in caries. We have studied both coronal and root caries in enamel, dentine and cementum using scanning electron and confocal scanning optical microscopy.

In preparation for backscattered electron (BSE) imaging in the SEM, teeth were stored in 70% ethanol until further dehydration in ethanol and embedding in polymethylmethacrylate (PMMA). Longitudinally cut surfaces were diamond polished and coated with carbon or silver before BSE imaging. Important changes in the distribution of densities in both enamel and dentine occurred …

Storage Of Sperm In The Reptilian Oviduct, Daniel H. Gist, Jeffrey M. Jones Aug 1987

Storage Of Sperm In The Reptilian Oviduct, Daniel H. Gist, Jeffrey M. Jones

Scanning Microscopy

Female reptiles have the ability to store sperm within their reproductive tracts for extended periods of time. Sites of sperm storage in lizards and snakes include both the anterior vagina and the infundibulum. Vaginal receptacles are found in between longitudinal folds (snakes) and in tubules formed by invaginations of the epithelium (lizards). Infundibular receptacles are alveolar or tubular in structure and are formed from invaginations into the lamina propria of the oviduct wall. In turtles, sperm are stored in the posterior portion of the tuba, in tubular albumen-secreting glands. Sperm may be embedded within the cells of the receptacles, but …

Tooth Enamel Structure In The Koala, Phascolarctos Cinereus: - Some Functional Interpretations, William G. Young, Maria Mcgowan, Terrence J. Daley Aug 1987

Tooth Enamel Structure In The Koala, Phascolarctos Cinereus: - Some Functional Interpretations, William G. Young, Maria Mcgowan, Terrence J. Daley

Scanning Microscopy

The purpose of this study is to determine whether the structural organization of Pattern 2 marsupial enamel in the Koala is disposed to resist wear on the sectorial crests of the molar teeth. The orientation of wear on the crests is uniformly delineated by parallel scratches on their polished surfaces. Twin blades, a leading and a trailing edge of enamel are formed on each crest by wear into dentine on which the differential wear at enamel to dentine interfaces indicates that the direction of wear is labial to lingual.

96 leading and trailing edges from 12 koala molars were examined …

Somites And Axon Guidance, Kathryn W. Tosney Aug 1987

Somites And Axon Guidance, Kathryn W. Tosney

Scanning Microscopy

The somites are arrayed in a repeating pattern along the longitudinal axis of the embryo, as are the developing sensory and sympathetic ganglia and the spinal nerves. This pattern is not a coincidence: the somite imposes a segmental pattern on the cells and axons that invade it. Both neural crest cells and axons prefer the anterior portion of the sclerotome (the ventral part of the somite) for outgrowth. What differences in anterior and posterior sclerotome are responsible? I used scanning electron microscopy to ask whether these populations differed on the tissue level in chick embryos. This study shows that differences …

Ultrastructural Analysis Of Enamel Formation During In Vitro Development Using Chemically-Defined Medium, P. Bringas Jr., M. Nakamura, E. Nakamura, J. Evans, H. C. Slavkin Jul 1987

Ultrastructural Analysis Of Enamel Formation During In Vitro Development Using Chemically-Defined Medium, P. Bringas Jr., M. Nakamura, E. Nakamura, J. Evans, H. C. Slavkin

Scanning Microscopy

To test the hypothesis that enamel biomineralization is regulated by sequential expression of an intrinsic genetic program, we designed experiments to determine if a serumless, chemically-defined medium is permissive for position-dependent ameloblast differentiation and subsequent enamel tissue-specific biomineralization in vitro. In the absence of serum or other exogenous growth factors, Swiss Webster strain mouse embryonic (15-and 16-days gestation) mandibular first molar tooth organs (cap stage) developed within 21 days in vitro into well-defined molar tooth organs expressing dentine and enamel biomineralization. Analysis of data obtained from von Kossa histochemistry for calcium salt formation, as well as ultrastructural information obtained …

Characteristics Of An Electric/Magnetic Quadrupole Detector For Low Voltage Scanning Electron Microscopy, M. Brunner, R. Schmid Jul 1987

Characteristics Of An Electric/Magnetic Quadrupole Detector For Low Voltage Scanning Electron Microscopy, M. Brunner, R. Schmid

Scanning Microscopy

The electric/magnetic quadrupole detector allow's efficient detection of secondary electrons in low voltage scanning electron microscopy without introducing disturbing aberrations. The original detector of this type was built in 1986; it has now been equipped with scintillator-photomultiplier assemblies on both positive electrodes. Their signals, A and B, can be combined to A+B or A-B, thus enhancing or suppressing different types of contrast. The aberration disc produced by the present design of detector was estimated to have a diameter of 10 nm. Experimentally, no deterioration of image resolution was observed. The collection efficiency was predicted to be 26% and can be …

Ultrastructure Of Spermatozoa From Domesticated Birds: Comparative Study Of Turkey, Chicken And Guinea Fowl, R. J. Thurston, R. A. Hess Jul 1987

Ultrastructure Of Spermatozoa From Domesticated Birds: Comparative Study Of Turkey, Chicken And Guinea Fowl, R. J. Thurston, R. A. Hess

Scanning Microscopy

Typical for non-passerine birds, the spermatozoa of the turkey, chicken and guinea fowl were vermiform with a maximum width of 0.5-0. 7μm and lengths of 90μm for chicken sperm, and 75-80μm for those of the turkey and guinea. An acrosome, nucleus, midpiece and flagellum could be distinguished by SEM.

The acrosomal cap projected over a perforatorium surrounded by granular material. At its base, the cap encircled apical projections of the nucleus. The nucleus consisted of dense chromatin, and formed a concave implantation fossa where it joined the midpiece of the tail. For turkey and chicken spermatozoa, the neck region of …

External Shape Of Enamel Crystals, H. Warshawsky Jun 1987

External Shape Of Enamel Crystals, H. Warshawsky

Scanning Microscopy

Biological hydroxyapatite crystals are either small, as in bone and dentin, or large as in enamel. Enamel crystallites are unique since each is initiated and grows in length, thickness and width until the entire layer of enamel is secreted. In maturation, these extremely long crystallites grow only in thickness and width. Crystal growth in vitro follows physico-chemical principles, but lacks biological intelligence; in vivo this intelligence is contributed by protein templates. The location of the organic template in enamel is congruent with the crystallite, constituting the crystal ghost. Since crystals cannot accommodate proteins, the explanation is logically inconsistent. In sections, …

Scanning Electron Microscopy Of Vascular Smooth Muscle Cells From Rat Muscular Arteries, William J. Krizmanich, Robert M. K. W. Lee Jun 1987

Scanning Electron Microscopy Of Vascular Smooth Muscle Cells From Rat Muscular Arteries, William J. Krizmanich, Robert M. K. W. Lee

Scanning Microscopy

The use of scanning electron microscopy (SEM) for the examination of medial smooth muscle cells (SMC) is limited by the presence of connective tissue mass in the adventitia obscuring the detailed structure of the SMC. This study deals with the description of a method which selectively removes the connective tissue, allowing three dimensional observation of SMC using SEM. A number of methods were tried. One method involving post fixation with 2% Os04 used in conjunction with HCl proved most successful in removing the extracellular matrix of large mesenteric arteries from spontaneously hypertensive rats (SHR) and age-matched Wistar-Kyoto normotensive rats …

Analysis Of Clean Transition Metal Surfaces By Core Level Spectroscopy, M. C. Desjonquères, D. Sébilleau, G. Tréglia, D. Spanjaard, C. Guillot, D. Chauveau, J. Lecante Jun 1987

Analysis Of Clean Transition Metal Surfaces By Core Level Spectroscopy, M. C. Desjonquères, D. Sébilleau, G. Tréglia, D. Spanjaard, C. Guillot, D. Chauveau, J. Lecante

Scanning Microscopy

The shifts in the binding energy of core electrons detected by high resolution X-ray photoelectron spectroscopy are a very sensitive probe of the chemical bonding of the excited atom. Since the surface atoms have their geometrical environment perturbed, their core levels are shifted from their bulk positions. A very large number of experiments have been performed on the 4f core level positions of tantalum and tungsten for various orientations of the surface plane. Systematic trends have been put forward and explained by theoretical models. Furthermore, the analysis of the angular variation of the core level line intensities gives structural information …

The Fine Structure Of The Human Placental Villus As Revealed By Scanning Electron Microscopy, G. J. Burton Jun 1987

The Fine Structure Of The Human Placental Villus As Revealed By Scanning Electron Microscopy, G. J. Burton

Scanning Microscopy

Scanning electron microscopy of the placenta has a history of only twenty years. During that time, however, there have been dramatic advances in instrument technology coupled with the refinement of preparative techniques designed to reduce fixation artifacts to a minimum. As a result many of the early claims must be amended or suitably qualified, and this is one aim of the present review.

Much new data on the internal structure of the placental villus is also presented. By means of the partial digestion technique it is now possible to describe the three dimensional configuration of the various components of the …

The Use Of Scanning Electron Microscopy In Studying Enamel Caries, R. P. Shellis, A. S. Hallsworth May 1987

The Use Of Scanning Electron Microscopy In Studying Enamel Caries, R. P. Shellis, A. S. Hallsworth

Scanning Microscopy

SEM studies related to carious change in dental enamel are reviewed, and their contribution to understanding the mechanism of formation of the early enamel lesion and of its repair evaluated. SEM has contributed significantly to understanding the mechanism of enamel dissolution at the level of the single crystal. Etching studies have yielded useful information on the effect of enamel structure on the pattern of acid dissolution at the microscopic level and have highlighted the importance of re- precipitation phenomena in modifying the pattern of mineral loss. High-resolution studies have provided interesting quantitative data on changes in crystal size, and also …

Mechanism Of Action Of Syringomycin On The Red Beet Plasma Membrane, Ashok P. Bidwai May 1987

Mechanism Of Action Of Syringomycin On The Red Beet Plasma Membrane, Ashok P. Bidwai

Biology

Syringomycin is a peptide toxin produced by the phytopathogen Pseudomonas syringae pv syringae. It is a major virulence factor produced by this organism and is involved in the development of a number of destructive plant diseases. Syringomycin preferentially stimulated the vanadate sensitive ATPase associated with the plasma membrane of red beet. The stimulation of ATPase activity was not due to a collapse of the ion gradients or to a detergent like breakdown of the membranes. Ionophores did not affect the stimulation of ATPase activity. Deoxycholate eliminated the ability of the toxin to stimulate ATPase activity, and the Zwittergent 3-14 …

The Effects Of Temperature Tempering And Transport Stress On Rainbow Trout (Salmo Gairdneri), Kevin Dee Christopherson May 1987

The Effects Of Temperature Tempering And Transport Stress On Rainbow Trout (Salmo Gairdneri), Kevin Dee Christopherson

Biology

The effects of capture, handling, and the high fish density of transport were compared with the added stressor of a temperature change or a brief hypoxic period. Temperatures from 5°C to 19°C were tested on fish acclimated to 14°C. The effects of a 30 minute and 60 minute temper were compared with a sudden temperature change. Plasma osmolality, plasma chloride, plasma glucose and hematocrit, all responded to the handling and simulated transport. Numbers of circulating lymphocytes were greatly reduced and numbers of erythocytes increased. The mean corpuscular volume of the erythocytes also declined over a 24 hour period. Leucocrit, thrombocyte …

Localization Of Dictyostelium Plasmids By Chromatin Analysis, Hassan Ashktorab May 1987

Localization Of Dictyostelium Plasmids By Chromatin Analysis, Hassan Ashktorab

Biology

Nuclear localization of endogenous plasmids in the cellular slime molds Dictyostelium discoideum and Dictyostelium mucoroides was investigated. Micrococcal nuclease resistant DNA fragments fractionated by gel electrophoresis were present in a banded pattern corresponding to lengths of multiples of approximately 160 bp that have been associated with nucleosome structure in D. discoideum. The same pattern was observed for the plasmids Ddp1 and Ddp2 in D. discoideum as for chromosomal DNA (discoidin genes) and extrachromosomal rDNA (17 S rRNA genes). The presence of the D. discoideum plasmids in chromatin establishes that they are located in the nucleus. Both transcribed and nontranscribed …

An Analysis Of Dna Polymorphisms In Hereditary Multiple Exostoses, Brian K. Moloney May 1987

An Analysis Of Dna Polymorphisms In Hereditary Multiple Exostoses, Brian K. Moloney

Biology

Hereditary multiple exostoses (HME) is a bone disease characterized by multiple benign bone tumors which develop only during active skeletal growth. Previous studies have demonstrated an autosornal dominant pattern of inheritance. In this study, several cellular oncogenes and human chromosomal fragments were used as probes to identify any restriction fragment length polymorphisms in the genomes of HME affected horses and humans. Sequences homologous to the human oncogenes c-fes, c-myb, c-myc, c-raf-1 and c-sis were identified in the horse genome. Polymorphisms were identified using c-raf-1 hybridized to Bam HI and Pst I digested horse DNA, but neither appeared linked to HME. …

The Multidimensional Concept Of Religiosity And Its Application To The Construction Of Mormon Religiosity Scales, Jeff Black May 1987

The Multidimensional Concept Of Religiosity And Its Application To The Construction Of Mormon Religiosity Scales, Jeff Black

Undergraduate Honors Capstone Projects

Until the 1960's, the concept of religiosity in sociology was very underdeveloped. Although many earlier social scientists were interested in religion, the concept of religiosity was conceptually and theoretically unsophisticated. Religiosity or religiousness was most often determined by church attendance or sometimes simply church membership. In the last two decades, much has been done to rectify this condition.
