Origin And Epigenetic Regulation Of Cutaneous T Cell Lymphoma, Carly M. Harro

The pathogenesis of Cutaneous T cell lymphoma (CTCL) remains unclear. Using conditional knockout mice, we found that ablation of the genomic organizer Special AT-rich sequence-binding protein-1 (Satb1) caused malignant transformation of mature, skin-homing, Notch-activated CD4+ and CD8+ T cells into progressively fatal lymphoma. Mechanistically, Satb1 restrained Stat5 phosphorylation and the expression of skin-homing chemokine receptors in mature T cells. H3K27 and H3K9 trimethylation occluded the SATB1 promoter in Sézary cells, while inhibition of SUV39H1/2 methyltransferases (unlike EZH2 inhibition), restored protective SATB1 expression and selectively abrogated the growth of primary Sézary cells more effectively than romidepsin. Algae, photosynthetic aquatic organisms, have recently caught attention due to their high growth rate and the high CO2 capture ability. Biofuels and bioproducts from algae are considered promising alternatives because of their lower carbon footprint. Higher education administrators are tasked with supporting and retaining students with increasing needs. These needs often include emotional and mental health issues but can worsen to include suicidality and violence toward others. Traditional campus approaches for supporting students and intervening for violence, such as counseling and campus safety, have been reactionary rather than proactive. Behavioral Intervention Teams (BITs) have emerged as a mechanism for heading off violence before it occurs while also supporting students who may never engage in violence but need support. Female sexual desire disorder is often discussed in the literature, but there is a paucity of data on how sex therapists conceptualize and treat this disorder. To address a gap in the current literature, this study collected the perspectives and attitudes of clinicians working with patients presenting with low sexual desire. This qualitative study aimed to understand how sex therapists’ conceptualization of female sexual desire disorder is used to make decisions about diagnosis, treatment, and assessing patient progress. Approximately 33 million American adults had a movement disorder associated with medication use, ear infections, injury, or neurological disorders in 2008, with over 18 million people affected by neurological disorders worldwide. Physical therapists assist people with movement disorders by providing interventions to reduce pain, improve mobility, avoid surgeries, and prevent falls and secondary complications of neurodegenerative disorders. Current gait assessments used by physical therapists, such as the Multiple Sclerosis Walking Scale, provide only semi-quantitative data, and cannot assess walking quality in detail or describe how one’s walking quality changes over time. Although many behavior analysts recognize the value of conducting a functional analysis (FA), some report being hesitant to conduct the assessment (Oliver et al., 2015). Two potential barriers to conducting FAs include setting limitations (Roscoe et al., 2015) and lack of trained staff (Deochand et al., 2020). Researchers have addressed these limitations by developing variations of the traditional FA and demonstrating that those procedures could be taught across various populations. The cumbersome and time-consuming process of generating new mouse strains and multiallelic experimental animals often hinders the use of genetically engineered mouse models (GEMM) in cancer research. In chapter 2, we describe the development and validation of an embryonic stem cell (ESC)-GEMM platform for rapid modeling of melanoma in mice. The platform incorporates 12 clinically relevant genotypes composed of combinations of four driver alleles (LSL-BrafV600E, LSL-NrasQ61R, PtenFlox, and Cdkn2aFlox) and regulatory alleles to spatiotemporally control the perturbation of genes of interest. Capillary formation and maturation can only occur through a series of highly orchestrated events that requires the coordinated efforts of both endothelial cells (ECs) and pericytes. While ECs are perfectly capable of creating tube networks on their own, the maturation and stabilization of these tube networks require the presence of a second cell type, pericytes, to create stable capillary networks. α-Helix accounts for the largest class of protein secondary structures and plays a key role in mediating protein-protein interactions (PPIs). Small-molecule α-helix mimetics mimic the structural features of α-helical hot spots and offer a powerful tool to unravel complex signaling networks and mitigate aberrant PPIs. Myelodysplastic Syndromes (MDS) are heterogeneous bone marrow (BM) failure malignancies characterized by constitutive innate immune activation, NLRP3 inflammasome (IFM) driven pyroptotic cell death, and the induction of interferon-stimulated genes (ISG). MDS often diagnostically resembles other hematologic disorders, as such, additional tools are needed for the discrimination from overlapping disorders. We observed that oxidized mitochondrial DNA (ox-mtDNA) is released upon cytolysis. We hypothesize that levels of ox-mtDNA are high in MDS, allowing us to use it as a diagnostic biomarker; and that cell free ox-mtDNA is contributing to feedforward BM failure.

Infants diagnosed with neonatal opioid withdrawal syndrome (NOWS) exhibit neurobehavioral dysregulation that may make them challenging to feed, potentially impeding positive early maternal-infant interactions. The goal of this study was to investigate dyadic behavior during the maternal-infant feeding interaction, specifically infant dysregulation and maternal sensitivity, between dyads with opioid exposure in the neonatal intensive care unit (NICU). This dissertation is an ethnographic case study that analyzes the gendered socio-cultural and socio-economic implications for a community that was displaced in order to develop a geothermal energy system in Kenya. The implications are rendered particularly interesting because the displaced community is also one of the communities served by the geothermal power grid. The analysis employs a composite framework of Feminist Political Ecology and the Political Ecology of Climate Change Mitigation. This dissertation furthers our understanding of how we (humans) sense and interact withforces, humans, and robots in three distinct cases, which affect robotics and rehabilitation. Chapter 2 examines physical therapists’ (PTs’) consistency when applying assistance forces during rehabilitation. Chapter 3 explores how control performance of an unstable system is affected by the system’s degrees of freedom (DoFs). Chapter 4 investigates the effects of the task speed and direction of perturbing forces on a dynamic task where position is controlled through self-generated forces.
