The dynamics of N and its interactions with labile dissolved organic C (DOC), bacteria, and phytoplankton were studied to determine potential effects of dissolved organic matter (DOM) and light on N dynamics in surface waters of the Mississippi River (USA) plume in the Gulf of Mexico. Bacterial uptake of added labeled N compounds ( 15NH4+ or 15N-labeled dissolved free amino acids. DFAA) was stimulated more by high-molecular-weight (HMW, >l kDa) DOM than by low-molecular-weight (LMW, < l kDa) DOM. An index that inversely indicated the presence of labile DOC was defined as the fraction of assimilated Amino acid-15N that was Recovered as 15N -Ammonium (ANRA), following the additions of high-levels (4 µM) of 15N -DFAA. …

The histones H2B, H3 and H4, the transition protein TP1 and protamine were localised using ultrastructural immunocytochemistry in nuclei of rabbit spermatids and spermatozoa. Histones are present in round spermatid nuclei and are lost during the elongation of nuclei. TP1 and protamine appear simultaneously in all nuclei during this period. TP1 is located at the periphery of chromatin cords, while protamine seems to be located at random in the same cords. TP1 is lost in most elongated sprematids during step 13 of spermiogenesis, and the protamine stays in all sperm nuclei. TP1 remains persent in some old spermatids and ejaculated …

Community respiration, net nutrient fluxes and heterotrophic bacterial production were investigated in the Mississippi River (USA) plume during May 1992 using dark bottle incubations of unfiltered water. Highest rates of community O2 consumption and dissolved inorganic carbon regeneration were observed at intermediate (10 to 27%0) plume salinities. Plume surface 02 consumption rates were 2- to 4-fold greater than rates reported previously during the summer and winter. Heterotrophic bacterial production ([3H]-leucine incorporation) was also highest at intermediate salinities and 2- to 4-fold greater than rates reported from other seasons. Net regeneration of NH4+ …

Habitual exercise provides protection against fatal coronary heart disease, extends longevity, and enhances quality of life. National surveys show less physical activity in older men and women compared with middle-aged and younger persons; older women are particularly sedentary. Although there are still few longitudinal studies on exercise and physical activity in older individuals, the data support a positive relationship between physical activity and health function in older individuals. The data further show that with regular physical activity, health and fitness are maintained or even increased over time in older individuals. This study examined differences between primary and intermediate teachers concerning teacher behaviors, teacher communications, grouping, control, and materials. 6 primary classrooms (grades 1 and 2) and 5 intermediate classrooms ( grades 4 and 5) were each observed for 4 45-minute periods. In addition, observers, teachers, and 5 students from each classroom responded to 2 vignettes depicting classroom situations and 1 vignette asking respondents to describe a lesson on nutrition. Responses were coded for teacher behaviors, goals, and instructional methods. Analyses of observational data showed that in comparison with teachers in intermediate grades, primary teachers used significantly more sanctions, procedural communications …

Glycation, oxidation, and browning of proteins have all been implicated in the development of diabetic complications. We measured the initial Amadori adduct, fructoselysine (FL); two Maillard products, N epsilon-(carboxymethyl) lysine (CML) and pentosidine; and fluorescence (excitation = 328 nm, emission = 378 nm) in skin collagen from 39 type 1 diabetic patients (aged 41.5 +/- 15.3 [17-73] yr; duration of diabetes 17.9 +/- 11.5 [0-46] yr, [mean +/- SD, range]). Sediment cores were collected inside and outside of a bed of a bromophenol-producing marine polychaete, Notomastus lobatus, and examined for impact of the bromophenols on sediment microflora and meiofauna around N. lobatus burrows. No significant differences were found between microbial parameters measured inside and outside of the N. lobatus bed. Integrated 6 cm cores taken adjacent to N. lobatus burrows contained 1.2 x 109 bacteria ml-1. Cell numbers were similar at control sites within the bed, but away from burrows, and not significantly different from cell numbers (1.0 X 109 cells ml-1) at a …

The RAPD (or AP-PCR) DNA fingerprinting method was used to distinguish among clinical isolates of Helicobacter pylori, a bacterium whose long term carriage is associated with gastritis, peptic ulcers and gastric carcinomas. This method uses arbitrarily chosen oligonucleotides to prime DNA sythesis from genomic sites to which they are fortuitously matched, or almost matched. Most 10-nt primers with > 60% G + C yielded strain-specific arrays of up to 15 prominent fragments, as did most longer ( > 17-nt) primers, whereas most 10-nt primers with 50% G +C did not. Time courses for ingestion, retention and release via feces of microbial food was investigated using 2 bivalves with different feeding strategies, Potamocorbula amurensis, and Macoma balthica. The results showed 2 pathways for the uptake of food material in these clams. The first is represented by an initial label pulse in the feces. The second pathway operates over longer time periods. Inert 51Cr-labeled beads were used to determine time frames for these pathways. The first pathway, involving extracellular digestion and intestinal uptake, is relatively inefficient in the digestion of bacterial cells by P. amurensis but more efficient in …

Formation Of Pentosidine During Nonenzymatic Browning Of Proteins By Glucose, Daniel G. Dyer, James A. Blackledge, Suzanne R. Thorpe, John W. Baynes

A fluorescent compound has been detected in proteins browned during Maillard reactions with glucose in vitro and shown to be identical to pentosidine, a pentose- derived fluorescent cross-link formed between arginine and lysine residues in collagen (Sell, D. R., and Monnier, V. M. (1989) J. Biol. Chem. 264, 21597- 2 1602). Pentosidine was the major fluorophore formed during nonenzymatic browning of ribonuclease and lysozyme by glucose, but accounted for <1% of nondisulfide cross-links in protein dimers formed during the reaction. Pentosidine was formed in greatest yields in reactions of pentoses with lysine and arginine in model systems but was also formed from glucose, fructose, ascorbate, Amadori compounds, 3-deoxyglucosone, and other sugars. Pentosidine was not formed from peroxidized polyunsaturated fatty acids or malondialdehyde. Its formation from carbohydrates was inhibited under nitrogen or anaerobic conditions and by aminoguanidine, an inhibitor of advanced glycation and browning reactions. Pentosidine was detected in human lens proteins, where its concentration increased gradually with age, but it did not exceed trace concentrations (55 Fmol/mol lysine), even in the 80-year-old lens. Although its precise carbohydrate source in vivo is uncertain and it is present in only trace concentrations in tissue proteins, pentosidine appears to be a useful biomarker for assessing cumulative damage to proteins by nonenzymatic browning reactions with carbohydrates.

Glycation, oxidation, and nonenzymatic browning of protein have all been implicated in the development of diabetic complications. The initial product of glycation of protein, fructoselysine (FL), undergoes further reactions, yielding a complex mixture of browning products, including the fluorescent lysine-arginine cross-link, pentosidine. Alternatively, FL may be cleaved oxidatively to form N(epsilon)-(carboxymethyl)lysine (CML), while glycated hydroxylysine, an amino-acid unique to collagen, may yield N(epsilon)-(carboxymethyl)hydroxylysine (CMhL).
