Protease Activity In The Plasma Of American Oysters, Crassostrea Virginica, Experimentally Infected With The Protozoan Parasite Perkinsus Marinus, P. Muñoz, K. Vance, M. Gomez-Chiarri

Perkinsus marinus is responsible for disease and mortality of the American oyster, Crassostrea virginica. To inves-tigate the interactions between P. marinus and oyster hemocytes, protease activity was measured in plasma of oysters collected 4 hr, 24 hr, 4 days, and 2 mo after experimental infection with P. marinus. A significant increase in protease activity was observed in oyster plasma 4 hr after injection with P. marinus, followed by a sharp decrease within 24 hr. Gelatin-impregnated gel electrophoresis showed the presence of 2 major bands (60 and 112 kDa) and 3 less prevalent bands (35, 92, and 200 kDa) with metalloproteinaselike …

Functions Of The Activation Loop In Csk Protein-Tyrosine Kinase, Xiaofeng Lin, Sungsoo Lee, Gongqin Sun

Autophosphorylation in the activation loop is a common mechanism regulating the activities of protein-tyrosine kinases (PTKs). PTKs in the Csk family, Csk and Chk, are rare exceptions for lacking Tyr residues in this loop. We probed the function of this loop in Csk by extensive site-specific mutagenesis and kinetic studies using physiological and artificial substrates. These studies led to several surprising conclusions. First, specific residues in Csk activation loop had little discernable functions in phosphorylation of its physiological substrate Src, as Ala scanning and loop replacement mutations decreased Csk activity toward Src less than 40%. Marine Science Journal Prices : A Case Study, Judith B. Barnett

Approximately 350 English-language journals in the interdisciplinary field of marine science are published. Two groups, one of fity titles, and one of fourteen are analyzed for price increases between 1997 and 2003. Within the larger context of escalating journal prices in all scientific fields, the marine science journals have increased in price approximately 71% during this period, while general science titles have increased 34%.

Taming The Rugged Landscape: Techniques For The Production, Reordering, And Stabilization Of Selected Cluster Inherent Structures, Dubravko Sabo, J. D. Doll, David L. Freeman

We report our studies of the potential energy surface (PES)of selected binary Lennard-Jones clusters. The effect of adding selected impurity atoms to a homogeneous cluster is explored. Inherent structures and transition states are found by combination of conjugate gradient and eigenvector-following methods while the topography of the PES is mapped with the help of a disconnectivity analysis. We show that we can controllably induce new structures as well as reorder and stabilize existing structures that are characteristic of higher-lying minima.

Impact Of Ectoparasites On The Demography Of A Small Reef Fish, Rachel J. Finley, Graham E. Forrester

In marine systems, empirical studies demonstrating effects of macroparasites on host population dynamics are relatively few. We tested the effects of a copepod macroparasite infecting the gills of a small coral reef fish, the bridled goby Coryphopterus glaucofraenum. Fish that were naturally infected and uninfected were tagged as individuals and tracked in the field for 5 mo. Parasitism was associated with an increase in gill ventilation rate, and a reduction in feeding. More importantly, parasitized fish showed significantly reduced growth (by 66%) and gonad mass (by 68%) compared to uninfected fish, and parasitism increased instantaneous mortality by a factor …

In experiments comparing biodegradable, plastic and wooden imidacloprid-treated spheres for control of Rhagoletis mendax Curran, the mean number of flies caught on plexiglas panes below each sphere type was not significantly different for the entire season. However, the mean time spent by R. mendax flies alighting on biodegradable imidacloprid-treated spheres was significantly greater (2.6×) than plastic imidacloprid-treated spheres. During 2001, significantly fewer larvae were found in blueberries harvested from bushes that had wooden imidacloprid-treated spheres hung within the canopy compared with bushes where biodegradable and plastic imidacloprid-treated spheres were deployed. The full-length sequence of tick salivary gland cDNA coding for a protein similar to metalloproteases (MP) of the reprolysin family is reported. The Ixodes scapularis MP is a 488 amino acid (aa) protein containing pre- and pro-enzyme domains, the zinc-binding motif HExxHxxGxxH common to metalloproteases, and a cysteine-rich region. In addition, the predicted amino-terminal sequences of I. scapularis MPs were found by Edman degradation of PVDF-transferred SDS/PAGE-separated tick saliva proteins, indicating that these putative enzymes are secreted. Furthermore, saliva has a metal-dependent proteolytic activity towards gelatin, fibrin(ogen), and fibronectin, but not collagen or laminin. Accordingly, I. scapularis saliva has a …

Acid in the stomach is thought to be a barrier to bacterial colonization of the intestine. Escherichia coli, however, has three systems for acid resistance, which overcome this barrier. The most effective of these systems is dependent on transport and decarboxylation of glutamate. GadX regulates two genes that encode isoforms of glutamate decarboxylase critical to this system, but additional genes associated with the glutamate-dependent acid resistance system remained to be identified. The gadX gene and a second downstream araC-like transcription factor gene, gadW, were mutated separately and in combination, and the gene expression profiles of the mutants …

Science progresses whenever new ways of explaining natural phenomena are revealed. New ideas can only be put to use after the results and significance of research have been brought to the attention of both the scientific and lay communities. The Milwaukee Public Museum Dig-A-Dinosaur Program is a model for scientists who want to conduct research as well as engage and educate the public in their field of study. An array of rewards is derived from including amateurs as members of a research team. Such collaborations strongly increase interest in and comprehension of science as a process. The ability of E. coli strains to colonize the mouse large intestine has been correlated with their ability to grow in cecal and colonic mucus. In the present study, an E. coli MG1655 strain was mutagenized with a mini-Tn5 Km (kanamycin) transposon, and mutants were tested for the ability to grow on agar plates with mouse cecal mucus as the sole source of carbon and nitrogen. One mutant, designated MD42 (for mucus defective), grew poorly on cecal-mucus agar plates but grew well on Luria agar plates and on glucose minimal-agar plates. Human DEC (differentially expressed in chondrocytes), mouse STRA (stimulated with retinoic acid), and rat SHARP (split and hairy related protein) proteins constitute a new and structurally distinct class of the basic helix-loop-helix proteins. In each species, two members are identified with a sequence identity of >90% in the basic helix-loop-helix region and ∼40% in the total proteins, respectively. Recently, we have reported that DEC1 is abundantly expressed in colon carcinomas but not in the adjacent normal tissues.
