Phlip Peptide Targets Nanogold Particles To Tumors, Lan Yao, Jennifer Daniels, Anna Moshnikova, Sergey Kuznetsov, Aftab Ahmed, Donald M. Engelman, Yana Reshetnyak, Oleg A. Andreev

Physics Faculty Publications

Progress in nanomedicine depends on the development of nanomaterials and targeted delivery methods. In this work, we describe a method for the preferential targeting of gold nanoparticles to a tumor in a mouse model. The method is based on the use of the pH Low Insertion Peptide (pHLIP), which targets various imaging agents to acidic tumors. We compare tumor targeting by nonfunctionalized nanogold particles with nanogold–pHLIP conjugates, where nanogold is covalently attached to the N terminus of pHLIP. Our most important finding is that both intratumoral and i.v. administration demonstrated a significant enhancement of tumor uptake of gold nanoparticles conjugated …

Enhanced Taupathy And Ad-Like Pathology In Aged Primate Brains Decades After Infantile Exposure To Lead (Pb), Syed Waseem Bihaqi, Nasser H. Zawia

Biomedical and Pharmaceutical Sciences Faculty Publications

Late Onset Alzheimer Disease (LOAD) constitutes the majority of AD cases (∼90%). Amyloidosis and tau pathology, which are present in AD brains, appear to be sporadic in nature. We have previously shown that infantile lead (Pb) exposure is associated with a change in the expression and regulation of the amyloid precursor protein (APP) and its beta amyloid (Aβ) products in old age. Here we report that infantile Pb exposure elevated the mRNA and protein levels of tau as well as its transcriptional regulators namely specificity protein 1 and 3 (Sp1 and Sp3) in aged primates. These changes were also accompanied …

Was That Ethical? Feminist Critics’ Response To The “Queerness” Of Modernist Women’S Writing, Meridith M. Kruse Jan 2013

Was That Ethical? Feminist Critics’ Response To The “Queerness” Of Modernist Women’S Writing, Meridith M. Kruse

Journal of Feminist Scholarship

This article employs insights from contemporary theories of ethical reading to conduct a case study of feminist critics’ reaction to the queerness of modernist women’s writing. My aim is to develop a set of practices and principles for ethically responding to queerness in literary texts and everyday life, as well as contribute feminist acumen to the current claim that the humanities are the best site to train students how to do justice to texts. The introduction utilizes theories of ethical reading set forth by Jane Gallop and Eve Kosofsky Sedgwick to develop a preliminary framework of ethical response. The subsequent …

Regulation Of Hemolysin Gene Expression And The Effects Of Metabolism Mutations On The Virulence Of Vibrio Anguillarum, Xiangyu Mou Jan 2013

Regulation Of Hemolysin Gene Expression And The Effects Of Metabolism Mutations On The Virulence Of Vibrio Anguillarum, Xiangyu Mou

Open Access Dissertations

Vibrio anguillarum is the causative agent of vibriosis, a fatal hemorrhagic septicemic disease. V. anguillarum infects more than 50 fresh and salt-water fish species including various species of economic importance to the larviculture and aquaculture industry. In vibriosis, V. anguillarum invades its host fish through the intestine and skin. Infected fish usually die with systemic infection of V. anguillarum. Bacterial hemolysins are exotoxins that cause lysis of erythrocytes in the host and thus the release of the intracellular heme, and are therefore identified as important virulence factors. Moreover, hemolysins are known to also cause lysis in other cell types, …

Effects Of Ciona Intestinalis Distal-Less-B Nonexpression And Overexpression On Phenotype And Downstream Targets, Matthew David Blanchette Jan 2013

Effects Of Ciona Intestinalis Distal-Less-B Nonexpression And Overexpression On Phenotype And Downstream Targets, Matthew David Blanchette

Open Access Dissertations

Ciona intestinalis can serve as a useful model for developmental studies in the chordate lineage due to its basal position in the chordate phylogeny. It shows a simplified chordate body plan during its development, during which important genetic pathways are conserved with vertebrates, and its developmental gene regulation can be manipulated through the insertion of transgenic DNA by electroporation. The Distalless (Dll) genes are a family of homeobox genes in chordates that are homologous to the single Dll gene of other metazoan animal groups and code for developmental factors that play a role in determining multiple developmental cell fates. These …

Assessment Of Mammal Biodiversity In Coffee-Dominated Landscapes Of India And Costa Rica, Stephanie Amanda Caudill Jan 2013

Assessment Of Mammal Biodiversity In Coffee-Dominated Landscapes Of India And Costa Rica, Stephanie Amanda Caudill

Open Access Dissertations

The fate of many tropical wildlife species depends on the way in which human-dominated landscapes are managed. Conservation strategies that incorporate agricultural landscapes are imperative for the survival of native fauna. Coffee is a major cash crop worldwide and is grown in tropical regions of the world that are often designated as biodiversity hotspots. Shade coffee, an agroforestry system in which coffee is grown under the shade of intentionally managed and/or native tree cover, has shown promise to provide habitat and support the diversity of bird, bat, and insect communities, but few studies have focused on non-volant mammals living in …

A Comparison Of Tripolar Concentric Ring Electrodes To Disc Electrodes And An Eeg Real-Time Seizure Detector Design, Xiang Liu Jan 2013

A Comparison Of Tripolar Concentric Ring Electrodes To Disc Electrodes And An Eeg Real-Time Seizure Detector Design, Xiang Liu

Open Access Dissertations

The electroencephalogram (EEG) is broadly used for research of brain activities and diagnosis of brain diseases and disorders. Although the EEG provides good temporal resolution, millisecond or less, it does not provide very good spatial resolution. There are two main reasons for the poor spatial resolution, (1) the blurring effects of the head volume conductor, and (2) poor signal to noise ratio. The surface Laplacian of the potential distribution was found to increase the spatial resolution. Several potential interpolation based methods were previously developed to estimate the surface Laplacian. However, these methods are generally complicated in terms of computation, which …

Study Of Glycation And Advanced Glycation On A Humanized Monoclonal Antibody, Hong Liu

Open Access Dissertations

In the presented work, we reviewed the protein glycation effect on recombinant monoclonal antibodies (MAbs), and demonstrated an in vitro glycation model on rhuMAb A was a successful tool to evaluate site-specific glycation. We also implemented this model to study MAb glycation in three aspects: the formation of glycation, the change of glycation adducts under thermal stress, and the control of glycation adducts stability by formulation compositions.

A protein characterization study demonstrated that the in vitro forced glycation model generated seven glycated amino acid sites on rhuMAb A. The lysine residue K49 was the preferably site, while others sites were …

Precautionary Protocol For Educational Professionals In Caring For A Newly Diagnosed Student With Type I Diabetes, Sandra Berard

Senior Honors Projects

Type I Diabetes in children has become a major critical disease in today’s elementary schools. The statistics reveal that “there are 25.8 million people in the United States, or 8.3% of the population, who have diabetes” (American Diabetes Association, 1). It is estimated that of those with Type I “only 5% of people with diabetes have this form of the disease” (ADA, 1). The problem is increasing among students at younger ages each and every year. Currently, students are being diagnosed with this autoimmune disease as early as four years of age. The American Diabetes Association states that “about 1 …

Fisheries, Animal and Veterinary Sciences Faculty Publications

The activin type IIB receptor (Acvr2b) is the cell surface receptor for multiple transforming growth factor β (TGF-β) superfamily ligands, several of which regulate muscle growth in mammals. To investigate the role of the Acvr2b signaling pathway in the growth and development of skeletal muscle in teleost fish, transgenic rainbow trout (RBT; Oncorhynchus mykiss) expressing a truncated form of the acvr2b-2a (acvr2b▵) in muscle tissue were produced. High levels of acvr2b▵ expression were detected in the majority of P1 transgenic fish. Transgenic P1 trout developed enhanced, localized musculature in both the epaxial and hypaxial regions …

Maternal Hydration And Infant Weight: A Case-Control Study In Breastfeeding Infants Delivered By Cesarean Section, Diane J. Thulier Jan 2013

Maternal Hydration And Infant Weight: A Case-Control Study In Breastfeeding Infants Delivered By Cesarean Section, Diane J. Thulier

Open Access Dissertations

Growing evidence suggests that infants may experience increased weight loss due to maternal medical interventions during labor such as intravenous fluid hydration. When breast fed infants lose ≥ 7% of their birth weight, formula supplementation is often initiated. Formula supplementation is a variable unequivocally associated with shorter breastfeeding duration. Supplementation should not be initiated if the cause of infant weight loss is unrelated to breastfeeding adequacy. The purpose of this study is to examine the impact of intrapartum maternal hydration and regional anesthesia on infant weight.

A retrospective, case-control research study was designed. Data was collected from maternal and infant …

Seasonal Particle And Carbon Dynamics In The Eastern Bering Sea, Matthew S. Baumann Jan 2013

Seasonal Particle And Carbon Dynamics In The Eastern Bering Sea, Matthew S. Baumann

Open Access Dissertations

The ocean margins of high-latitude seas, such as the eastern Bering Sea, are recognized as important areas for the enhanced scavenging removal of particle reactive chemicals and for the potential to sequester atmospheric carbon dioxide via photosynthetic conversion to biogenic particles and subsequent downward particle transport to deeper waters. This region is expected to warm in the future, and with the warming will come a reduction in the extent and duration of seasonal sea-ice. This physical process exerts an important control on the timing, location, and magnitude of the spring primary production bloom. Within the context that 2008-2010 are characterized …

Sequence Effects On The Repair And Replication Of Arylamine-Dna Adducts, Vipin Jain Jan 2013

Cancer is a collection of diseases defined by uncontrolled cell growth and dissemination of abnormal cells. It is the second most deadly disease in the United States. Exposure to chemicals, both endogeneous and exogeneous is one of the major causes of cancer initiation. The role of chemicals in carcinogenesis was observed back in the eighteenth century when surgeon Pott observed a large number of scrotal cancer cases in chimney sweeps and associated it with to the soot exposure. However, research in the field of chemical carcinogenesis was renewed when Yamagawa and Ichikawa reported the development of malignant tumor on the …

Application Of Silver Nanoparticles In Drinking Water Purification, Hongyin Zhang Jan 2013

Application Of Silver Nanoparticles In Drinking Water Purification, Hongyin Zhang

Open Access Dissertations

Nanotechnology is an emerging and fast-growing technology. Currently, there are more than 1,317 nanotechnology-based products on the market. Silver nanoparticles account for more than 23% of all nano-products. The extensive application of the silver nanoparticle (AgNP) results in their inevitable release into the environment. Silver nanoparticles are known as excellent antimicrobial agents, and therefore they could be used as alternative disinfectant agents. On the other hand, released silver nanoparticles could pose a threat to naturally occurring microorganisms.

In Chapter 1, we introduce the background information on the environmental fate, toxicological effects, and application of AgNP and review the current knowledge …

Probiotic Strains For Shellfish Aquaculture: Protection Of Eastern Oyster, Crassostrea Virginica, Larvae And Juveniles Againsl Bacterial Challenge, Murni Karim, Wenjing Zhao, David C. Rowley, David R. Nelson, Marta Gomez-Chiarri

Fisheries, Animal and Veterinary Sciences Faculty Publications

Bacterial pathogens, including several Vibrio spp. and Roseovarius crassostreae, cause severe mortality of larval and juvenile eastern oysters. The introduction of beneficial bacterial isolates in oyster hatcheries and nurseries for the biocontrol of bacterial diseases is a good alternative to the use of antibiotics. The goal of this study was to screen and characterize marine bacterial isolates as potential agents to prevent larval and juvenile mortality by the oyster pathogens Vibrio tubiashii and R. crassostreae. Screening of bacterial isolates from Rhode Island marine organisms and environment using agar-based assay methods for detection of antimicrobial activity against oyster pathogens led …

Migration And Stopover Ecology Of Songbirds And Bats Along A Major Ecological Barrier, Adam D. Smith Jan 2013

Migration And Stopover Ecology Of Songbirds And Bats Along A Major Ecological Barrier, Adam D. Smith

Open Access Dissertations

Migration is a physiologically and energetically demanding period in the life cycle of a migratory animal. Most migrant songbirds alternate between periods of nocturnal flight, when energy is used, and stopover, when energy is accumulated for the next flight period; indeed, songbirds spend most of their time and energy on stopover. Conversely, the habits of migratory bats remain largely enigmatic, but there is reason to expect that models of songbird migration can inform bat migration research. I investigated, at multiple scales, the migration ecology of songbirds and bats and the stopover ecology of songbirds along the Atlantic Coast of southern …

Identification Of Salivary Antigens From The Deer Tick Ixodes Scapularis As Novel Vaccine Candidates, Martin Becker Jan 2013

Identification Of Salivary Antigens From The Deer Tick Ixodes Scapularis As Novel Vaccine Candidates, Martin Becker

Open Access Master's Theses

Ticks act as vectors for a number of different pathogens such as Borrelia burgdorferi the causative agent of Lyme disease. The most prominent tick vector in the United States is the deer tick Ixodes scapularis. Tick bites are of special public health concern since there are no vaccines available against most tick transmitted pathogens. Based on the observation that host animals such as guinea pigs or humans can develop adaptive immune responses to tick bites, anti-tick vaccination is a potential approach to mitigate health risks associated with tick bites.

The study presented here aimed at identifying immunogenic salivary proteins …

Soil Quality In Rhode Island Pastures Grazed By Different Types Of Livestock, Alissa H. Becker Jan 2013

Soil Quality In Rhode Island Pastures Grazed By Different Types Of Livestock, Alissa H. Becker

Open Access Master's Theses

Historically, much of the New England landscape was converted to pasture for grazing animals and harvesting hay. Today, consumer demand for local, sustainably produced food, the number of livestock farms and the land area used for farming are increasing in Rhode Island. In order to make sound management decisions, it is important to characterize the effects of livestock on the quality of pasture soils. Although pastures remain a fairly common part of the New England landscape, little research has been published on the effects of grazing animals on pasture soil quality. To assess this, I examined soil quality in farms …

Comparative Phylogeography And A Mitochondrial Dna Barcode For Identifying Three Sympatric Lagomorphs In The Northeastern United States, Mary E. Sullivan

Open Access Master's Theses

The New England cottontail (Sylvilagus transitionalis), New England’s endemic cottontail, has been experiencing dramatic population declines and is estimated to exist in only 14% of its historical range. The New England cottontail is currently a candidate for endangered species listing under the Endangered Species Act of 1973. New England cottontails may be distinguished accurately from eastern cottontails (S. floridanus) with the use of non-invasive genetic techniques (e.g., fecal sample collection) that allow sampling of large geographical areas with minimal cost. A restriction enzyme technique has been published based on NlaIII (New England Biolabs Inc, Massachusetts) cut …

The Distribution And Ontogeny Of Neuromast Receptor Organs And A Comparison Of Methods For Chemical Albation Of The Lateral Line System In Two Cichlid Fishes, Emily Anne Becker Jan 2013

The Distribution And Ontogeny Of Neuromast Receptor Organs And A Comparison Of Methods For Chemical Albation Of The Lateral Line System In Two Cichlid Fishes, Emily Anne Becker

Open Access Master's Theses

The lateral line system is composed of a series of mechanoreceptors called neuromasts, which are found on the head and trunk in specific patterns. On the head, larger canal neuromasts (CNs) are enclosed in cranial canals and smaller superficial neuromasts (SNs) are embedded in the skin in lines or clusters. Among species the lateral line canals can be narrow, widened, branched, or reduced. Through the use of fluorescent stains, Scanning Electron Microscopy, and cleared and stained specimens the distribution and ontogenetic appearance of SNs and CNs were mapped in two species of Lake Malawi cichlids with divergent adult lateral line …
