Agrobacterium Mediated Transformation Of Anti-Stress Genes Into Cauliflower (Brassica Oleracea Var. Botrytis L.). 2. Transformation And Confirmation Of Stress Tolerance., Emr Metwali, Mp Fuller, Aj Jellings
School of Biological and Marine Sciences
Integration of APX and SOD anti-stress genes into cauliflower (Brassica oleracea var. botrytis L.) plants was achieved by using Agrobacterium tumefaciens - mediated transformation method. Cauliflower explants (hypocotyls and cotyledons) and Agrobacterium tumefaciens strains (APX, SA, TA) were used in this experiment. The procedure utilizes polymerase chain reaction (PCR) amplification of insert DNA directly after isolation of individual colonies without the necessity of separate procedures for DNA isolation and purification. Integration of the introduced stress gene (APX and SOD) in the plants was confirmed by using β-glucuronidase gene (GUS) and leaf disc assays as a gene fusion and diagnostic marker, …

Energy Productivity And Efficiency Of The ‘Gher’ (Prawn-Fish-Rice) Farming System In Bangladesh, S Rahman, Bk Barmon
School of Geography, Earth and Environmental Sciences
Gher' farming is a unique system that incorporates the joint operation of three enterprises: freshwater prawn, fish and HYV rice, and is expanding rapidly in the coastal regions of Bangladesh because of its proven high income earning potential. In this paper, the sustainability of this system is evaluated by analysing its performance in terms of energy use by applying a stochastic distance function approach which revealed interesting and unexpected results. The prawn enterprise which is the key income earning component is found to be technically inefficient while the rice enterprise is found to be efficient. The net energy balance and …

In the world of smartphone devices, iPads and digital billboards, our visual landscape is changing fast - from static display to moving, time-based displays. Yet traditional illustration often manipulates time with more grace than many of the slightly awkward digital examples, such as those clamouring from screens on cashpoint displays and digital posters on bus stops and London’s rail and tube stations. It must be acknowledged that the occasions where such displays seemingly move for the sake of being able to do so (where the elements of their design move over time but do not always develop the content over …

A New Taxonomy For Describing And Defining Adherence To Medications, B Vrijens, S De Geest, Da Hughes, K Przemyslaw, Et Al
Peninsula Medical School
Interest in patient adherence has increased in recent years, with a growing literature that shows the pervasiveness of poor adherence to appropriately prescribed medications. However, four decades of adherence research has not resulted in uniformity in the terminology used to describe deviations from prescribed therapies. The aim of this review was to propose a new taxonomy, in which adherence to medications is conceptualized, based on behavioural and pharmacological science, and which will support quantifiable parameters. A systematic literature review was performed using MEDLINE, EMBASE, CINAHL, the Cochrane Library and PsycINFO from database inception to 1 April 2009. The objective was …

Investigating Interaction Effects In An Array Of Multi-Mode Wave Energy Converters, Ej Ransley, Dm Greaves
School of Engineering, Computing and Mathematics
A number of wave energy device developers have now successfully tank-tested scale-model prototypes and several are attempting full-scale deployment at sea [1]. Many believe the superior survivability of simple, buoy-like designs make them the most economically viable solutions [2, 3]. Developers of so called ‘point-absorbers’ hope to install multiple devices in arrays, offering considerable savings in terms of moorings, grid connections and maintenance. It is recognised that the additional hydrodynamic interactions between devices, from scattered and radiated waves within the array, can significantly alter the surface elevation and enhance the interaction factor, q, defined as the ratio of power from …

Predicting Ecosystem Shifts Requires New Approaches That Integrate The Effects Of Climate Change Across Entire Systems., Bayden D. Russell, Christopher D.G. Harley, Thomas Wernberg, Nova Mieszkowska, Stephen Widdicombe, Jason M. Hall-Spencer, Sean D. Connell
School of Biological and Marine Sciences
Most studies that forecast the ecological consequences of climate change target a single species and a single life stage. Depending on climatic impacts on other life stages and on interacting species, however, the results from simple experiments may not translate into accurate predictions of future ecological change. Research needs to move beyond simple experimental studies and environmental envelope projections for single species towards identifying where ecosystem change is likely to occur and the drivers for this change. For this to happen, we advocate research directions that (i) identify the critical species within the target ecosystem, and the life stage(s) most …

BACKGROUND: Realist synthesis is an increasingly popular approach to the review and synthesis of evidence, which focuses on understanding the mechanisms by which an intervention works (or not). There are few published examples of realist synthesis. This paper therefore fills a gap by describing, in detail, the process used for a realist review and synthesis to answer the question 'what interventions and strategies are effective in enabling evidence-informed healthcare?' The strengths and challenges of conducting realist review are also considered. METHODS: The realist approach involves identifying underlying causal mechanisms and exploring how they work under what conditions. The stages of …

Modelling Of Bacterial Growth With Shifts In Temperature Using Automated Methods With Listeria Monocytogenes And Pseudomonas Aeruginosa As Examples, M Salih, I Mytilinaios, Hk Schofield, Rjw Lambert
School of Geography, Earth and Environmental Sciences
Time to detection (TTD) measurements using turbidometry allow a straightforward method for the measurement of bacterial growth rates under isothermal conditions. Growth rate measurements were carried out for Listeria monocytogenes at 25, 30 and 37°C and for Pseudomonas aeruginosa over the temperature range 25 to 45°C. The classical three-parameter logistic model was rearranged to provide the theoretical foundation for the observed TTD. A model was subsequently developed for the analysis of TTD data from non-isothermal studies based on the Malthusian approximation of the logistic model. The model was able to predict the TTD for cultures of L. monocytogenes or P. …

The Environmental Behaviour Of Beryllium-7 And Implications For Its Use As A Sediment Tracer, Alex Taylor
The Environmental Behaviour Of Beryllium-7 And Implications For Its Use As A Sediment Tracer, Alex Taylor
School of Geography, Earth and Environmental Sciences Theses
The use of cosmogenic beryllium-7 (7Be) as a soil and sediment tracer relies upon a number of important assumptions which to date have not been fully underpinned by supporting data. As a catchment management tool 7Be offers unique potential to assess the effects of recent land use or climate change but further research is required to provide confidence in key data and elucidate sources of uncertainty. Through a range of laboratory and field studies, this thesis aims to explore knowledge gaps relating to i) the temporal and spatial dynamics of 7Be activity in rainfall which has importance in the context …

Teachers As Mediators: An Exploration Of Situated English Teaching, Peter Kelly, Ulrike Hohmann, Nick Pratt, Hans Dorf
Teachers As Mediators: An Exploration Of Situated English Teaching, Peter Kelly, Ulrike Hohmann, Nick Pratt, Hans Dorf
Plymouth Institute of Education
Within the context of lower secondary English teaching in South West England, this study identifies in broad terms the competing goals between which English teachers mediate and the explicit and hidden tensions that result. To understand the interactions of competing goals, teachers’ goal-oriented behaviours are referenced to a set of idealised ‘role types’ based on the dimensions of goals, norms, discourses and practices. It is asserted that competing goals, significant to particular educational circumstances, emanate from various sometimes contradictory local, national and perhaps broader social and cultural influences on practice. Yet the teachers observed moved smoothly between goal-oriented behaviours in …

Rips, Currents And Snags: Investigating The Complexities Of Teaching Swimming And Water Safety Within Primary Schools., Tj Lynch
Plymouth Institute of Education
It is possible for swimming and water safety to be skilfully connected to any of the eight key learning curriculum areas (discipline subjects) by primary school teachers, and can be argued that teachers will have increased opportunity and autonomy to do this with the development of a National Curriculum Framework. While this proposal seems simple on the surface, there are underlying complexities that may act as possible barriers.

Growth Curve Prediction From Optical Density Data, I Mytilinaios, M Salih, Hk Schofield, Rjw Lambert
School of Geography, Earth and Environmental Sciences
A fundamental aspect of predictive microbiology is the shape of the microbial growth curve and many models are used to fit microbial count data, the modified Gompertz and Baranyi equation being two of the most widely used. Rapid, automated methods such as turbidimetry have been widely used to obtain growth parameters, but do not directly give the microbial growth curve. Optical density (OD) data can be used to obtain the specific growth rate and if used in conjunction with the known initial inocula, the maximum population data and knowledge of the microbial number at a predefined OD at a known …

Effect Of Ph And Temperature On The Morphology And Phases Of Co-Precipitated Hydroxyapatite, H. R. Le, K. Y. Chen, C. A. Wang
School of Biological and Marine Sciences
This paper reports a high-yield process to fabricate biomimetic hydroxyapatite nano-particles or nano-plates. Hydroxyapatite is obtained by simultaneous dripping of calcium chloride and ammonium hydrogen phosphate solutions into a reaction vessel. Reactions were carried out under various pH and temperature conditions. The morphology and phase composition of the precipitates were investigated using scanning electron microscope and X-ray diffraction. The analyses showed that large plates of calcium hydrophosphate are formed at neutral or acidic pH condition. Nanoparticles of hydroxyapatite were obtained in precipitates prepared at pH 9–11. Hydroxyapatite plates akin to seashell nacre were obtained at 40 °C and pH 9. …
