Phosphorylation is a necessary posttranslational modification that regulates protein function and directs cell signaling outcomes. Current methods to measure protein phosphorylation cannot preserve the heterogeneity in phosphorylation across individual proteins. The single-molecule pull-down (SiMPull) assay was developed to investigate the composition of macromolecular complexes via immunoprecipitation of proteins on a glass coverslip followed by single-molecule imaging. The current technique is an adaptation of SiMPull that provides robust quantification of the phosphorylation state of full-length membrane receptors at the single-molecule level. Imaging thousands of individual receptors in this way allows for quantifying protein phosphorylation patterns. The present protocol details the optimized …

Spirituality is generally found to have a significant but small positive association with well-being; however, the associations between spirituality and well-being vary greatly. One organizing framework for understanding the varying associations may be the degree to which spirituality and well-being measures capture connection, the pinnacle of which may be conceptualized as Oneness. The purpose of this paper is twofold: to begin exploring this hypothesis by examining the associations between beliefs in Oneness and dimensions of subjective well-being and to test possible conflating and contributing factors in the associations between beliefs in Oneness and subjective well-being. Technologies Of Territoriality: Indigeneity, Surveillance, And The State, Elspeth Iralu

American Studies ETDs

This dissertation examines the global spatial surveillance of Indigenous peoples, nations, and territories in the twenty-first century through a multi-site relational analysis of colonial surveillance and Indigenous cartography in the United States, India, and Palestine. Analyzing Indigenous graphic novels, video games, virtual reality, performance protests, and visual art, I demonstrate how air and the aerial perspective actively shape what happens on and below the ground. I argue that Indigenous experiences of and responses to colonial and counterinsurgent surveillance are not limited by the geographic and legal bounds of nation-states but are rather linked through global histories of militarization and colonialism. …

Within recent decades, spectral methods have become an important technique in numerical computing for solving partial differential equations. This is due to their superior accuracy when compared to finite difference and finite element methods. For such spectral approximations, the convergence rate is solely dependent on the smoothness of the solution yielding the potential to achieve spectral accuracy. We present an iterative approach for solving the two-dimensional Helmholtz problem posed on a rectangular domain subject to Dirichlet boundary conditions that is well-conditioned, low in memory, and of sub-quadratic complexity. In 2014 and 2019 the New Mexico Acequia Association attended events organized by the Water Tribunal of Valencia in Spain commemorating the fifth and tenth anniversaries when the acequias of Valencia and Murcia were listed by UNESCO as examples of the Intangible Cultural Heritage of Humanity. The documentary report provides the context for and significance of the two visits along with a series of photographs that tell the story for the benefit of the citizens of New Mexico.

BACKGROUND: Estimates of the humoral immune response to incident human papillomavirus (HPV) infections are limited.

METHODS: In this post hoc analysis of 3875 women aged 16-23 years from a 4-valent HPV vaccine trial (NCT00092482), HPV seroprevalence on day 1 was measured with a 9-valent HPV (HPV 6/11/16/18/31/33/45/52/58) competitive Luminex immunoassay and compared with cervical/external genital HPV detection by polymerase chain reaction. In the control group, among women who were HPV DNA‒negative on day 1, seroconversion following initial HPV detection was estimated using Kaplan-Meier methods.

RESULTS: Type-specific HPV seropositivity among women with no day 1 cervical/external genital HPV detection was 0.6%-3.6%. …

Manganese-enhanced magnetic resonance imaging (MEMRI) holds exceptional promise for preclinical studies of brain-wide physiology in awake-behaving animals. The objectives of this review are to update the current information regarding MEMRI and to inform new investigators as to its potential. Mn(II) is a powerful contrast agent for two main reasons: (1) high signal intensity at low doses; and (2) biological interactions, such as projection tracing and neural activity mapping via entry into electrically active neurons in the living brain. Purpose: To accommodate the unprecedented number of critically ill patients with pneumonia caused by coronavirus disease 2019 (COVID-19) expansion of the capacity of intensive care unit (ICU) to clinical areas not previously used for critical care was necessary. We describe the global burden of COVID-19 admissions and the clinical and organizational characteristics associated with outcomes in critically ill COVID-19 patients.

Methods: Multicenter, international, point prevalence study, including adult patients with SARS-CoV-2 infection confirmed by polymerase chain reaction (PCR) and a diagnosis of COVID-19 admitted to ICU between February 15th and May 15th, 2020.
