Glioblastoma is the most common malignant brain tumor in adults. In contrast to several cancers, where the tumor suppressor gene TP53 is altered, most glioblastoma patients retain a non-mutant TP53 gene. However, the survival of these patients is similar to that of mutant patients, suggesting that p53 is nonfunctional. To test this hypothesis, we analyzed data from The Cancer Genome Atlas, identified potential regulators of p53, and we will test their function in patient-derived cells.

Non-Expert Summary:

The neuron-specific gene family (NSG1-3) plays crucial roles in regulating the movement and processing of key proteins within neurons, particularly those involved in Alzheimer's disease and synaptic strength regulation. While NSG1 is known for its importance in proper synaptic function, NSG2's role is less understood. Surprisingly, deleting NSG2 selectively enhanced learning and memory, contrary to predictions. Electrophysiology experiments revealed NSG1 and NSG2 have opposite effects on synaptic plasticity, shedding light on their distinct molecular mechanisms.

Background:

The HEALthy Brain and Child Development (HBCD) is the largest multi-site longitudinal prospective study in the US. The national cohort is composed of local pregnant persons and children who continue with HBCD for 5+ years. HBCD seeks to examine brain and child development amongst substance and environmental exposures, social, cultural, and genetic influences.

Methods:

Repeated electroencephalography (EEG) assessments are performed throughout HBCD, starting at Visit 3 (3-9 months). EEG signals and corresponding stimuli are analyzed to measure event-related potentials (ERPs). EEG acquisition includes 4 age-adjusted tasks recorded via MagstimEGI 128 channel net, EKG lead, and Net Station Acquisition. Video …

Spreading depolarizations (SDs) have been strongly linked to advancing secondary injury in ischemic stroke but may also provide beneficial effects to brain tissue that is not metabolically depleted. In the current study, we sought to identify whether SDs initiated remotely can alter secondary injury expansion. Infarct size was significantly reduced in mice with transient MCAo when SDs were induced in brain regions located outside of the vulnerable stroke penumbra suggesting a neuroprotective effect of SD.

This thesis foregrounds traditional Irish music and the role it plays in building community. I demonstrate how the genre is defined and transmitted plays an important role in how inclusive the genre can be and where changes need to be made to be more welcoming to new individuals. Playing or singing in musical groups has been shown to combat loneliness but there has been little focus on specific music genres, especially those that are instrumental in nature. Through the development of microreactors, a free piston Stirling engine (FPSE) is an excellent candidate to support the development of the microreactors applications. This study proposes a 20 kWe design of the FPSE for microreactor applications. The model has been developed and analyzed through a MATLAB code that was verified and benchmarked with both RE-1000 engine’s experiment data and Sage software. The MATLAB code has been developed from scratch using the first and second order approximation to simulate the engine’s steady state operation. Neuroplasticity In The Dentate Gyrus: A Multimethod Investigation On Hippocampal Ltp Following Prenatal Alcohol Exposure, Monica Goncalves-Garcia

Studies have demonstrated alcohol-related deficits in LTP associated with histaminergic and glutamatergic impairments. The histaminergic H3R inverse agonist ABT-239 has shown promise in reversing these deficits. The present study focuses on another H3R inverse agonist, SAR-152594, and its impact on LTP deficits following moderate PAE. The findings reveal that systemic administration of 1 mg/kg of SAR-152594 reverses deficits in potentiation fEPSPs in adult male rats exposed to moderate PAE. Time-frequency analyses of evoked responses indicate PAE-related reductions in power during the fEPSP and increased power during later components of evoked responses, both reversed by SAR-152594. Protein Interacting with C Kinase-1 (PICK1) is a scaffolding protein that offers a promising solution to support individuals suffering from substance use disorders. While many efforts have been put towards identifying small-molecule drugs with the potential to target PICK1 and ultimately support individuals suffering from the effects of substance use disorders, all efforts have so far fallen short. This thesis explores the monolithic integration of antimonide based narrow bandgap semiconductors on commercially available substrates for mid-wave infrared (MWIR) devices. The research focuses on the integration and growth of antimonide based alloys using the interfacial misfit dislocation (IMF) arrays to enhance the cost efficiency, manufacturability and performance. The study particularly investigates the relaxation mechanisms of antimonide alloys on InAs and InP substrates, assessing the impact of mismatch strain on the quality of epilayers. The IMF arrays are examined using high-resolution transmission electron microscopy (HR-TEM) and x-ray diffraction (XRD), revealing information about the effective strain relief mechanisms behind IMF arrays …

This study proposed a neutrosophic set framework with TreeSoft Set for sustainable waste valorization selection. The neutrosophic set is used to overcome uncertainty and vague information in the evaluation process. The neutrosophic set has three membership degrees: truth, indeterminacy, and falsity. The multi criteria decision-making (MCDM) methodology deals with various criteria to evaluate waste valorization. The VIKOR method is an MCDM method used to rank the alternatives. In this paper we present an open source framework developed in Python and consisting of three distinct classes designed to manipulate in a simple and intuitive way both symbolic representations of neu trosophic sets over universes of various types as well as mappings between them. The primary objective of this article is to develop a mathematical model and determine the optimal policies of an inventory system involving power demand and controlled deterioration through preservation technology. This model comes in handy in a power demand-oriented inventory system with demand high at the end of the period. The model incorporates backlogged shortages and linear holding cost. The triangular neutrosophic numbers (TNN’s) are used for a nuanced representation of uncertain and imprecise inventory-related expenses. Medical image processing has become a critical research area due to the vast amounts of digital image data available. However, medical images often suffer from poor illumination and low visibility of significant structures, requiring image enhancement to improve image quality before processing. In this paper, we propose a technique for enhancing medical images by removing noise and improving contrast based on three different enhancing transforms. The proposed technique embeds the image into a neutrosophic fuzzy domain, where it is mapped into three different levels of trueness, falseness, and indeterminacy, and each level is processed individually using the enhancement transforms. The Shortest path problem is highly relevant in our daily lives, addressing uncertainties like traffic conditions and weather variations. To handle such uncertainties, we utilize Fuzzy Numbers. This paper focuses on Bipolar Neutrosophic Fuzzy Numbers, which have dual positive and negative aspects. They provide a robust framework for representing arc (node/edge) weights, signifying uncertain travel times between nodes. Importantly, these weights can change over time in bipolar neutrosophic fuzzy graphs.
