The aim of this thesis is to investigate the role that shame plays as a subversive tool in the works of Québécoise author Nelly Arcan. Dealing with themes such as sexuality, gender, suicide, and resentment, Arcan’s works deal with that which can be deemed “shameful,” and she deliberately speaks the unspeakable. First in constructing an understanding in how a shame-based vocabulary is employed throughout her works, we then study how this shame subverts expectations and adds weight to political questions relevant to the widespread use of electronic media. Gen Z is defined as including persons born after 1996 and, in 2018, the first Gen Z would have been twenty-two years old, the historically traditional age that many complete undergraduate studies and enter law school. With Gen Z entering law schools, the legal academy has been wholeheartedly preparing for the arrival of the first truly digital native generation in a myriad of ways. The piezoelectric response has been a measure of interest in density functional theory (DFT) for micro-electromechanical systems (MEMS) since the inception of MEMS technology. Piezoelectric-based MEMS devices find wide applications in automobiles, mobile phones, healthcare devices, and silicon chips for computers, to name a few. Piezoelectric properties of doped aluminum nitride (AlN) have been under investigation in materials science for piezoelectric thin films because of its wide range of device applicability. In this research using rigorous DFT calculations, high throughput ab-initio simulations for 23 AlN alloys are generated.

In this dissertation, I defend the view that, contrary to popular opinion, vulnerability is not merely susceptibility to harm but also openness to unanticipated change and transformation. Drawing on the work of Simone de Beauvoir, Judith Butler, Erinn Gilson, Gilles Deleuze, and Benedict Spinoza, I also aim to show that vulnerability is not a static property of some individuals but rather a relational process that is both universal and differently distributed. For over forty years, Brazil, its subnational governments, Indigenous communities, other nations, non-governmental organizations, corporations, and individuals have worked to conserve the Amazon rainforest through a staggering number of diverse international initiatives. While some initiatives have supported Brazil in decreasing the rate of deforestation over the past fifteen years, the 2019 fires demonstrated that destruction continues. Left unchecked, this irreversible destruction promises to amplify. Fortunately, the long history of global involvement in Amazon conservation provides ample lessons for effective, place-based deforestation prevention. Thoughtful and coordinated international action can address the current lethal combination of destructive factors: Brazil’s environmentally hostile federal …

Phosphorylation is a necessary posttranslational modification that regulates protein function and directs cell signaling outcomes. Current methods to measure protein phosphorylation cannot preserve the heterogeneity in phosphorylation across individual proteins. The single-molecule pull-down (SiMPull) assay was developed to investigate the composition of macromolecular complexes via immunoprecipitation of proteins on a glass coverslip followed by single-molecule imaging. The current technique is an adaptation of SiMPull that provides robust quantification of the phosphorylation state of full-length membrane receptors at the single-molecule level. Imaging thousands of individual receptors in this way allows for quantifying protein phosphorylation patterns. The present protocol details the optimized …

Manganese-enhanced magnetic resonance imaging (MEMRI) holds exceptional promise for preclinical studies of brain-wide physiology in awake-behaving animals. The objectives of this review are to update the current information regarding MEMRI and to inform new investigators as to its potential. Mn(II) is a powerful contrast agent for two main reasons: (1) high signal intensity at low doses; and (2) biological interactions, such as projection tracing and neural activity mapping via entry into electrically active neurons in the living brain. The Department of Energy/National Nuclear Security Administration (DOE/NNSA) and Sandia National Laboratories /New Mexico (SNL/NM) personnel submitted the Mixed Waste Landfill (MWL) Annual Long-Term Monitoring and Maintenance Report for the April 2021 through March 2022 reporting period to the New Mexico Environment Department (NMED) in accordance with the requirements of Section 4.8.1 of the MWL Long-Term Monitoring and Maintenance Plan (LTMMP). The LTMMP became effective on January 8, 2014.

Carbonate precipitation in perennially ice-covered Lake Fryxell, McMurdo Dry Valleys, Antarctica produces a modern analog for Antarctic paleolake carbonates. Lake Fryxell contains a steep oxycline in the photic zone, where dissolved oxygen falls from supersaturation to zero. In the lake’s benthic microbial mats, oxygen concentrations are higher than in the water column due to microbial photosynthesis. These mats contain carbonate cements that precipitated in a discrete episode from mat pore waters. Precipitation continued through seasonal fluctuations in oxygenation, demonstrated by variable concentrations of oxidized and reduced manganese and iron within carbonates. This dissertation presents the first Systemic Functional Linguistics-based analysis of the teaching/learning of computational thinking through computer programming and comprehensive analysis of discourse of a whole computer programming course at any educational level. The current educational research raises questions about the nature of authentic computational

Persistently low population sizes, when coupled with reduced interpopulation connectivity, can impede the long-term viability of species in fragmented landscapes. Riparian-associated species in the arid American Southwest now face a series of threats due to fragmented populations and changing environmental conditions. During the last century, riparian habitats have deteriorated due to the synergistic effects of livestock grazing, increasing incidence of fire, and other anthropogenic impacts potentially have made local populations smaller, less demographically stable, and susceptible to the negative impacts of genetic drift and stochastic events. Climate change related drought is projected to harm maize production. Water use strategies can help mitigate the impact of drought on crop yield. However, little is known about maize metabolic response to drought at different developmental stages. To shed light on this, drought conditions were applied to maize at the six-leaf stage (V6), twelve leaf stage (V12), and tassel stage (VT). V6 and VT took eight days to achieve a low stomatal conductance threshold, but V12 took 16 days. Differential gene expression analysis of the transcriptome indicates that V6 showed the most response with 53 impacted metabolic pathways, many of …

Adverse childhood experiences (ACEs) are common, potentially traumatic events that impact one's health and well-being. These harmful experiences increase the risk of chronic comorbidities, mental health problems, and substance use disorders; this risk exponentially increases in children and adolescents with four or more ACEs. They are associated with socially disadvantaged populations and structural violence in communities. ACEs are categorized into abuse, neglect, and household challenges, which are proven to contribute to the longevity of health, behavioral, social problems, and even premature death through an accretion of studies. Fire-exclusion has acted as a major perturbation on dry conifer forests, increasing tree density and, in mixed-conifer forests, the dominance of shade-tolerant species. Restoration efforts aim to reverse these effects by reducing stand density, restoring relative proportions of tree species, and reintroducing recurrent fire, but the long-term effects of repeated burning on tree regeneration have not been quantified. We analyzed two decades of seedling and overstory data from the Teakettle Experimental Forest in the southern Sierra Nevada to determine how thinning and repeated burning affect seedling establishment and overstory recruitment. Gastrointestinal complaints are often reported during high altitude ascent (>2500m), though their etiology is unknown. One explanation is hypoxia-mediated intestinal barrier dysfunction. High altitude exposures can result in splanchnic hypoperfusion and hypoxemia causing hypoxic and oxidative stress. Exertion may worsen hypoxia-induced intestinal injury via greater splanchnic hypoperfusion and hypoxemia. We propose that these stressors injure the intestinal barrier leading to increased permeability, bacterial translocation, and local/systemic inflammation which may contribute to gastrointestinal complications or Acute Mountain Sickness (AMS). The purpose of this study was to capture the student voice regarding the value, importance, and relevance of visual and performing arts education in public schools. The host institution for the study was an urban school district in the southern central United States serving 75,000 students, with 55% students identified as Hispanic, and 52% of students experiencing poverty. Lizards rely heavily on environmental temperatures to thermoregulate and maintain a body temperature (Tb) that optimizes physiological function and maximizes surface activity time. While early research noted the propensity for lizards to thermoregulate by shuttling between different thermal environments, it was long assumed that evaporative cooling via panting was an ineffective means of thermoregulation. However, evaporative cooling can potentially lower a lizard’s Tb significantly below air temperature, and thus allow lizards to extend activity periods during prolonged heat exposure. Plate reader-based methods for high-throughput measurement of growth rate, cellular survival, and chronological lifespan are a compelling addition to the already powerful toolbox of budding yeast Saccharomyces cerevisiae genetics. These methods have overcome many of the limits of traditional yeast biology techniques, but also present a new bottleneck at the point of data-analysis. Herein, we describe SPOCK (Survival Percentage and Outgrowth Collection Kit), an R-based package for the analysis of data created by high-throughput plate reader based methods. Sunlight contains about 9% UV radiation with roughly one-third of it penetrating the atmosphere and reaching the Earth’s surface. UV radiation from the sun is classified into three different types: UVA (315- 499 nm), UVB (280- 314nm), and UVC (100- 279 nm). Prolonged exposure to artificial UV radiation or direct sunlight can still induce many adverse effects such as sunburn, weakening of the immune system, and skin cancer. Terrestrial ecosystems are critical to human and ecological processes but many gaps in our knowledge remain regarding how terrestrial plant communities assemble and respond to global change. I used field experiments distributed around the world, including long-term experiments from the Sevilleta National Wildlife Refuge (SNWR) in New Mexico and deserts of the southwestern U.S., to evaluate the consequences of drought and other abiotic stressors on plant communities. Dominant grasses were particularly important for the productivity and structure of grasslands at SNWR. The Cyphochilus white beetle has received significant attention recently for its exceptional visible-light-scattering properties, owing to the beetle scale's anisotropic nano-fibrillar network structure. Inspiration from the remarkable optical properties of these biological species has directed research efforts to fabricating biomimetic nano-fibrillar structures with polymer materials. In this work, electrospun silk fibroin polymer films reproduce the white beetle scale's anisotropic nature to draw on its optical scattering properties and further improve the radiative-cooling capabilities of silk fabrics. An anisotropically restructured silk film with a mean fiber diameter of a few hundred nanometers substantially increases optical scattering strength in the visible spectrum …

The field of Spanish historiography has overwhelmingly been shaped by theories of Convivencia or anti-Convivencia, of total harmony or complete violence. The interpersonal connections made between individuals of different faiths—Islam, Judaism, and Christianity—often contravene institutional regulation that prohibited sexual and familial connections and dissuaded casual camaraderie, complicating and disagreeing with histiorgraphic (anti-)Convivencia traditions. In place of an (anti-)Convivencia framework, modern theories of sexuality, as first championed by Michele Foucault, can explain discrepancies between individual action and institutional regulation through a matrix of power, identity, and interaction. The purpose of this mixed methods study was to examine graduate student preferences for the types of learning environments that support ethical development in research ethics education. The initial quantitative phase of this study used the Learning Environment Preferences (LEP) survey to provide a baseline measure of intellectual development up to position 5 of the Perry scheme (Moore, 2000; Perry, 1970). Climate change in the American Southwest is altering the composition of species assemblages. However, the resulting patterns in mean trait values and functional diversity are poorly understood. Bees assemblages in Southwestern drylands are exceptionally diverse, and vary greatly in their morphologic traits. In this study we focused on two questions: Have community-weighted mean trait values shifted over time and/or with aridity, consistent with the hypothesis that aridification is driving bee assemblage change? The objective of this thesis project was to investigate the effects of tungsten on breast cancer metastasis to the bone niche, using the 4T1 orthotopic breast cancer model. Oral tungsten (15 ppm) exposure did not affect primary tumor growth. However, following tungsten exposure there were marked changes in the bone niche, including increased metastasis of 4T1 tumor cells and increased osteolysis. Further analysis of the bone niche indicates that enhanced metastasis is associated with a pro-tumorigenic immune suppressive environment, including increased number of granulocytic myeloid derived suppressor cells and increased gene expression of the pro-inflammatory cytokine IL-1β and the activated …
