SUMMARIZING ACCOMPLISHMENTS OF THE LTER NETWORK OFFICE DURING THE FOURTH YEAR OF COOPERATIVE AGREEMENT DEB—9634135 - The Network Office played key roles in three major LTER activities during 2000-2001. Preparations for a mid-term site review in May provided an opportunity to review and revise NET goals and led to an intensive effort to prioritize activities in support of these goals. Within grasslands, precipitation, fire, nitrogen (N) addition, and extreme temperatures influence community composition and ecosystem function. The differential influences of these abiotic factors on Chihuahuan Desert grassland communities was examined within the Sevilleta National Wildlife Refuge, located in central New Mexico, U.S.A. Although fire is a natural disturbance in many grasslands, fire during drought temporarily increased forb cover and decreased grass cover for several years. The seasonal timing of fire was not important, rather community recovery was more influenced by the persistent drought conditions. Analysis Of Genes Required For Quiescent Cell Formation In Stationary Phase Cultures Of Saccharomyces Cerevisiae, Melissa R. Wilson

Yeast cells in stationary phase cultures, after several days growth in rich, glucose-based medium (YPD), are separable by density-gradient centrifugation into two fractions. The heavier, quiescent cells are mostly virgin daughters whereas the less-dense, non-quiescent cells, are a typical mixture of daughters to aged cells. Quiescent cells can also be separated based on expression of specific GFP-tagged proteins, including many that are localized to the mitochodria. Proliferation and migration are critical steps within normal mammary development and breast cancer progression. While 17β-estradiol (E2) stimulates proliferation in normal and breast cancer cells through estrogen receptor-α (ERα) and G Protein-Coupled Estrogen Receptor-1 (GPER) [1-3], GPER regulation of E2-dependent migration has not been fully examined. GPER upregulates pathways necessary for increased migration, including vimentin, matrix metalloproteinase-9, and mitogen activated protein kinase (MAPK) signaling [1, 4-6]. GPER may also increase phospho-focal adhesion kinase (p-FAK) through Src activation [7, 8]. As a tumor develops, it produces transforming growth factor-β (TGF-β) to activate normal fibroblasts in the stroma, transforming them into cancer-associated …

Developmental events in the temporal bones shift the pattern of a given speech sounds acoustic profile through the time children are mapping linguistic sound systems. Before age 5 years, frequency information in vowels is differentially accessible through the years children are acquiring the sound systems of their native language(s). To model the acoustic effects caused by developing temporal bones, data collected to elicit steady-state vowels from adult native speakers of English and Diné were modified to reflect the form of children's hearing sensitivities at different ages based on patterns established in the psychoacoustic literature. Skin cancer is the one of the most diagnosed cancers in the United States with increasing incidence over the past two decades. There are three major forms of skin cancer but melanoma is the deadliest. It is estimated that 76,690 new diagnoses of melanoma and 9,480 deaths will occur in 2013. Melanoma accounts for approximately 1.6% of all cancer related deaths and is the 5th leading diagnosed cancer in the United States. Proteins without signal peptides can still be released into extracellular space via ER/Golgi-independent unconventional secretory pathway(s) that remain to be revealed. A class of leaderless proteins including the pro-inflammatory cytokine interleukin-β (IL-β) is processed in the cytosol by proteolytic machineries such as the inflammasome prior to secretion. Studies in yeast uncovered a new unconventional secretory pathway that is dependent on GRASP (Golgi Reassembly and Stacking Protein) and autophagy proteins. Virus-like particles (VLPs) comprised of viral structural proteins that self-assemble into particles resembling the native virion represent a relatively novel vaccine development strategy. Both safe and immunogenic, VLPs can be used as vaccines against the virus from which they are derived, but can also be used to present heterologous epitopes from other pathogens to the immune system. Both techniques result in high-titer antibody responses against the target epitope. Indeed, vaccines of VLPs are already available, including the two vaccines targeting Human Papillomavirus (HPV). The most common cause of cancer deaths among women is breast cancer. Current breast cancer chemotherapies rely on passive diffusion of the drug into the tumor interior, which leads to a sub-optimal drug concentration in some tumor regions. One important goal for cancer pharmaceutics is optimizing drug targeting to tumors. We propose to develop active drug delivery, mediated by the patient's immune cells. We hypothesize that circulating monocytes, known to infiltrate tumors, are effective biologically-active carriers of multifunctional nanoparticles. Peripheral blood mononuclear cells (MS) were isolated from the spleens of transgenic mice that are immunocompetent and spontaneously develop mammary tumors. …

My thesis examines the discourses present in digital video projects created by queer youth, ages 13-20, who participated in The Reel Queer Youth video mentorship program between 2009 and 2012. I used textual analysis, specifically discourse and critical technocultural discourse analysis to explore constructions of identity in youth and experiences present in digital video projects and the contexts these discourses were produced within. I identified three overarching discursive themes: (a) call for more complex understandings of queer youth identities and experiences; (b) concern regarding gender binaries; and (c) change and self-responsibility. The Department of Family and Community Medicine (DFCM) at the University of New Mexico is initiating phase III of a three-phase study examining the gastrointestinal health effects in residents who use a private well in an area with on-site wastewater treatment and disposal system (septic tanks). These residents show an increased exposure to detectable Cryptosporidium relative to a control (unexposed) population of residents who use municipal water as their source of water. This exposure was found in three different communities with significantly different hydrogeologic conditions. One community was located in the inner valley of the Rio Grande where depth to …
