Studies On The Effect Of Glycolysis And Pi(3,5)P2 On V-Atpase Activity, Dennis Ralph Dominguez

We studied two signals that regulate the yeast vacuolar proton pump, V-ATPase, glucose and PI(3,5)P2.  V-ATPase regulation by glucose is linked to the glycolytic enzyme PFK-1. Chapter one investigates the relationship between V-ATPase and glycolysis in cells overexpressing the PFK-1 subunit Pfk2p.  We measured: (i) PFK-1 activity to determine the effect on PFK-1 function; and (ii) NADH autofluorescence to determine the effect on glycolysis. PFK2 overexpression decreased PFK-1 catalytic activity. However, the NADH synthesis rate increased by 1.4 – 2.6-fold, suggesting that excess Pfk2p enhanced the apparent glycolytic capacity of the cells. In the face of increasing population, development pressures, and climate change, many regions around the world face freshwater shortages. Planned potable water reuse can improve sustainability and reliability of water supplies by providing drinking water from wastewater. Most potable reuse research has focused on large coastal communities with relatively high mean household incomes. However, the US Department of Interior predicts that “hot spots” of conflict over water in the arid West are “highly likely” in numerous small-to-medium-sized inland communities with low-to-moderate household income levels. Search In T Cell And Robot Swarms: Balancing Extent And Intensity, George M. Fricke

This work investigates effective search and resource collection algorithms for swarms. Deterministic spiral algorithms and L ́evy search processes have been shown to be optimal for single searchers. We extend these strategies to swarms of robots and populations of T cells and measure performance under a variety of conditions.

Search extent and intensity lie on a continuum: more intensive patterns search thoroughly in the local area, while extensive patterns cover more area but may miss targets nearby. The phosphodiester backbone of DNA is maintained by DNA ligases. In human cells, there are three genes that encode DNA ligase polypeptides with distinct but overlapping functions. A series of small molecule inhibitors of human DNA ligases were previously identified using a rational structure-based approach. Three of these inhibitors, L82, a DNA ligase I selective inhibitor, and L67, an inhibitor of DNA ligases I and III, and L189, an inhibitor of all three human DNA ligases, have related structures. Alone In A Crowd: Perceived Social Relationships And Health Outcomes For Clergy, Christopher G. Ford

The purpose of this study was to understand more about the relative contributions of different measures of social relationships to mental and physical health. This was done through secondary analysis of a clergy health intervention (n = 616), a population with unique professional and personal relationship characteristics. Hierarchal multiple regression was used in three steps to control for demographics, measures of perceived social relationships (e.g., social support, social engagement, social isolation, and relationship satisfaction), and whether or not clergy worked alone or with clergy colleagues. A Nation at Risk (ANAR) represented a paradigm shift in national education policy and public education discourse. Through their ability to self-renew and differentiate, hematopoietic stem/progenitor cells (HSPCs) maintain the adult blood and immune systems. The microenvironment, or niche, in which HSPCs reside, serves as a critical regulator of HSPC functions. As previous work has identified the tetraspanin CD82 as a mediator of HSPC-niche interactions, we aimed to determine the mechanism by which this occurs. Our data demonstrate that CD82 expression and scaffolding regulate HSPC interactions with niche components by organizing the α4 integrin subunit into tightly packed nanoclusters. In this dissertation, I explore agricultural practices as a window into ecocultural communication. Using agricultural practices of villagers in Village G, Oman, as a case study, I explore the ways in which villagers and government officials conceptualize humanature relations and the forces that enhance and/or impede these relations.
