Virtually all fields of biology have benefited from the advancements in comparative genomics technologies, specifically in the study of evolution. In this dissertation I develop and use comparative genomic technologies to investigate the novel SARS-CoV-2 virus, assembly the first genome of the black lace domestic angelfish and identify germline genetic variants associated with altered breast cancer-specific survival. Our genome tiling array for the novel coronavirus presents a rapid and cost-effective method to sequence the entire viral genome and can be used to track the rapid evolution of viral variants in the population. Hydrogen sulfide (H2S) is a gasotransmitter critical in maintaining microcirculation homeostasis. Impaired microcirculation occurs in multiple disease states such as peripheral vascular disease, diabetes mellitus (DM), and hypertension. Early detection and identification of patients with DM who are at risk for heart attack, stroke and amputation due to microvascular disease is crucial. Human skin is an accessible vascular bed that provides an opportunity to non-invasively measure H2S, which could be used as a biomarker to evaluate microvascular health.

A Thin Line Of Social Justice: Challenges And Changes In Navigating Relations Of Power In Graduate Schools Of Education, Heather Catherine Sands

This dissertation brings together three qualitative research articles to interrogate a disjuncture between curriculum development in graduate schools of education and the relations of power it fosters in teacher education and counselor education. In applying biopower and intersectional analyses throughout each article, this dissertation contends that the recognition of power relations within these fields helps to highlight hegemonic patterns of teaching and counseling that occur in settings of classrooms and sessions. Three themes emerged within this dissertation: resistance, community, and subjectivity. Article One demonstrates a gap between teachers and school counselors with cyberbullying directed toward LGBTQ+ students. The Article leverages …

Epidermal growth factor receptor (EGFR) signal transduction is initiated via ligand (EGF) binding, followed by dimerization, autophosphorylation of the C-terminal tails, and recruitment of proteins that form a larger signaling complex to propagate the signal. We explored the relationship between receptor mobility and signaling using single-particle tracking (SPT) by examining the diffusional dynamics of EGFR and two truncation mutants to understand whether mobility changes are correlated with signaling. Results revealed that phosphorylation of the C-terminal tail of EGFR is required for maximal reduction in mobility that occurs with EGF stimulation, indicating that receptor mobility is a read-out for receptor signaling. …

Species ranges are shifting due to anthropogenic climate change. However, it is unclear how climate variability affects species recruitment, an essential step in range expansion. Recruitment is challenging in drylands, where soil moisture is highly unpredictable. In Southwestern North America, creosotebush (Larrea tridentata) has expanded into Chihuahuan Desert grassland (CDG) dominated by black grama (Bouteloua eriopoda), and black grama is expanding into Great Plains grassland (GPG) dominated by blue grama (Bouteloua gracilis). Recent breakthroughs in nanophotonics have brought new opportunities to control and manipulate light at the nanoscale. The optical properties of metallic nanostructures have attracted particular interest because of their plasmon resonances, which couple strongly with visible light, and generate large near-field enhancements in their vicinity. In the first part of this thesis, we investigate the fundamental limits of the local density of photonic states near nanostructures by analyzing a sum rule relating its spectral integral to the field induced by a static dipole. The home range (HR) is a fundamental manifestation of an animal’s spatial behavior, but its study has been limited by unclear, unreliable, and unstandardized methodological approaches for measurement. The lack of a comprehensive evaluation of the influence of components of the HR estimation process has hampered consensus over best practices for collecting and analysing movement data. This leads to wide variation in practice, thwarting direct comparability between HR estimates and curtailing the cohesive power of cross-study findings necessary for testing hypotheses and developing appropriate conservation and management strategies. Natural killer (NK) cells are lymphocytes of the native immune system that play a pivotal role in host defense and immune surveillance. While the conceptual view of NK-neoplasms is evolving, little is known about the rare NK lymphoblastic leukemia (NK-LL), which remains as a provisional entity in the 2016 WHO Classification. The goal of this study is to characterize NK-LL cases and compare with other CD56 co-expressing acute leukemias. We identified 105 cases, diagnosed as NK-LL (6), CD56+ acute undifferentiated leukemia (AUL) (6), CD56+ T-lymphoblastic leukemia (T-LL) (51), and CD56+ acute myeloid leukemia (AML) (42). Heart Rate Variability (HRV) is a useful tool for clinical practice and substance use research, as it may provide insight into liability for relapse or development of addiction, as well as provide indicators of recovery outcomes and response to treatment. Psychotherapy would benefit from an integrated understanding of physiological regulation systems such as what are measured by HRV, as HRV is linked to psychopathology and substance use disorders.
HRV was measured in non-treatment-seeking adult drinkers as they underwent a motivational interviewing (MI) session. Subjects’ HRV from the interview was then regressed on therapist MI-consistent behaviors. The subjects’ drinking at both …

The Middle Rio Grande riparian zone, named the Bosque, provides cultural, aesthetic, environmental, recreational, and historical value to the residents of the Middle Rio Grande valley. Most of the Bosque has not experienced successful native cottonwood (Populus deltoides ssp. wislizenii) recruitment since the completion of the Cochiti reservoir and most of the Bosque cottonwood forest is senescing. This decline of the native species in the Bosque can be attributed to highly managed hydrology of the riparian zone. Excessive Tibial Overgrowth Associated Ankle Valgus Deformity After Pediatric Open Mid-Shaft Tibial Fracture: A Case Report, Ahmed M. Thabet, Rami Khalifa, Rasheed Alnajjar, Amr Abdelgawad

Tibial overgrowth after pediatric tibial fractures has been reported in the literature. Multiple factors can affect the amount of overgrowth, including age and gender of patients, type of fracture (open vs closed), degree of communion, reduction techniques, and fixation method. We describe a 10-year-old boy with a 2-cm overgrowth after sustaining an isolated open tibia fracture with an intact fibula, which is more than what has been previously described in the literature for tibia fractures. This overgrowth was associated with asymmetrical growth of the distal tibial physis, asymmetrical growth between tibia and fibula, and ankle valgus deformity.

The ongoing coronavirus disease 2019 (COVID-19) pandemic is caused by infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Human natural defense mechanisms against SARS-CoV-2 are largely unknown. Serine proteases (SPs) including furin and TMPRSS2 cleave SARS-CoV-2 spike protein, facilitating viral entry. Here, we show that FXa, a SP for blood coagulation, is upregulated in COVID-19 patients compared to non-COVID-19 donors and exerts anti-viral activity. Mechanistically, FXa cleaves the SARS-CoV-2 spike protein, which prevents its binding to ACE2, and thus blocks viral entry. Furthermore, the variant B.1.1.7 with several mutations is dramatically resistant to the anti-viral effect of FXa compared …

The Department of Energy/National Nuclear Security Administration (DOE/NNSA) and Sandia National Laboratories /New Mexico (SNL/NM) personnel submitted the Mixed Waste Landfill (MWL) Annual Long-Term Monitoring and Maintenance Report for the April 2020 through March 2021 reporting period to the New Mexico Environment Department (NMED) in accordance with the requirements of Section 4.8.1 of the MWL Long-Term Monitoring and Maintenance Plan (LTMMP). The LTMMP became effective on January 8, 2014.

This Article analyzes some of the recent rules, guidance documents, policy-based administrative decisions, and rulemaking procedures used by the United States Patent and Trademark Office (USPTO), and concludes that the USPTO is improperly promulgating substantive rules sub rosa via, inter alia, updates to the Trial Practice Guide (TPG), an ostensibly nonbinding document that controls many broad substantive and procedural aspects of the Patent Trial and Appeal Board (PTAB), and in doing so, avoids appellate, Congressional, and stakeholder review of such decisions.

A transgender person faces obstacles trying to negotiate a gender-binary world. Going through a TSA checkpoint is no different. A substantial number of transgender persons have reported that they were detained and examined because they were transgender.1 Why this situation persists and what policy reforms should be implemented to alleviate it are the subjects of this Essay. In my dissertation, I used morphological, sanger sequencing and next-generation sequencing data sets to explore the evolutionary relationships and study cryptic diversity of a extant Neotropical myotine radiation. Using a integrative approach, I described a new species of Myotis from the montane and premontane forest of Panama, Costa Rica and Eastern Ecuador, refining our undestanding of the taxonomy and systematics of the Myotis pilosatibialis complex of bats. Patients with core-binding factor (CBF) acute myeloid leukemia (AML), caused by either t(8;21)(q22;q22) or inv(16)(p13q22)/t(16;16)(p13;q22), have higher complete remission rates and longer survival than patients with other subtypes of AML. However, ∼40% of patients relapse, and the literature suggests that patients with inv(16) fare differently from those with t(8;21). We retrospectively analyzed 537 patients with CBF-AML, focusing on additional cytogenetic aberrations to examine their impact on clinical outcomes. Trisomies of chromosomes 8, 21, or 22 were significantly more common in patients with inv(16)/t(16;16): 16% vs 7%, 6% vs 0%, and 17% vs 0%, respectively. In contrast, del(9q) and loss of …

Prior research suggests that a scarcity of dementia specialists could hamper access to disease-modifying Alzheimer’s treatments. We describe alternative approaches on how to leverage specialist time for memory care in this narrative synthesis based on 17 semi-structured interviews and a targeted literature review on memory care approaches that leverage specialist time. We identified four types of approaches: community primary care practices empowered with better tools and training; primary care memory clinics; specialty memory clinics and; specialty memory centers. Several approaches to use specialist time efficiently have been implemented and some but not all evaluated. The optimal approach may depend on …

Clinical trial data and real-world evidence suggest that the AS04-adjuvanted vaccine targeting human papillomavirus types 16 and 18 (AS04-HPV-16/18) vaccine provides nearly 90% protection against cervical intraepithelial neoplasia grade 3 or higher irrespective of type, among women vaccinated before sexual debut. This high efficacy is not fully explained by cross-protection. Although AS04-HPV-16/18 vaccination does not affect clearance of prevalent infections, it may accelerate clearance of newly acquired infections. We pooled data from 2 large-scale randomized controlled trials to evaluate efficacy of the AS04-HPV-16/18 vaccine against clearance of nontargeted incident infections. Managing diabetes requires a detailed self-care regimen of physical activity, eating well, testing glucose, healthy weight maintenance, and positive emotional outlook in order to maintain health outcomes. Education, clear communication, and sufficient support are needed to address challenging aspects of daily self-care at the onset of diagnosis and throughout the management of the illness. However, many people diagnosed with diabetes are unable to successfully manage their condition. Native Americans are twice as likely to have diabetes than Whites. The aim of this study was to help men convicted of child sexual offenses learn to recognize and engage their character strengths over 12 months. Participants were six men convicted of contact and noncontact (internet) child sexual offenses. All participants were members of a community-based reintegration group for registered citizens. In the first weeks of the study, participants received the Values in Action Inventory of Strengths survey. The survey, containing 240 questions—10 items for each of the 24 character strengths outlined—helped participants identify their top character strengths. This dissertation investigates the effects of disturbance (a catastrophic forest fire and decadal-level multi-day flood) on populations of submerged aquatic macrophytes (SAMs) in the streams and rivers of the Jemez Mountains of northern New Mexico, USA. Using phenomenology as a lens of analysis, I investigate how immersive theatre engages audiences in a more direct and sensory way than traditional theatre. In a proscenium theatre the action is seen from the same angle. The theatre itself becomes a phenomenon in audience’s minds, each performance subtly influencing how the audience perceives the next. I investigate how relationships between audience and performers are altered in immersive experiences with no delineation between the space audience and actors occupy. The human brain consists of 86 billion neurons compared to rodent brain, which consists of 75 million neurons, and the underlying mechanisms for the increased complexity of the primate cortex are not fully elucidated. However, an increase in specific cell populations, including calretinin-expressing (CALB2/CR) interneurons (INs), has been identified. In my thesis work, I demonstrate that human pluripotent stem cells (hPSCs) differentiated to neurons by default patterning give rise to this CR population in vitro, providing a method to study their development.

The purpose of this qualitative Self-Study of Teacher Education Practices was to explore and tell a personalized account of myself as an instructional coach striving for culturally responsive pedagogical practices through multicultural literature in pre-kindergarten through sixth grade classrooms.
