This thesis explores the story of per- and polyfluoroalkyl substances (PFAS) in New England wastewater systems. PFAS compounds, known for their environmental persistence, toxic nature, and heavy integration into our daily lives, has recently become an infamous topic in environmental engineering due to the discovery of contaminated agricultural lands, water resources, and food systems. In response to these daunting discoveries, regulators have begun looking to reduce point source pollution of PFAS, including that of wastewater systems and facilities. Phytoplankton population dynamics are significant indicators of ecosystem health and marine food web structure. Phytoplankton population dynamics are also highly variable, especially within complex coastal ocean systems. To capture temporal phytoplankton population dynamics, a weekly time series measuring temperature, salinity, tide height, chlorophyll a concentration, nutrients, and phytoplankton biodiversity was performed at two sites in New Hampshire over an annual cycle. Despite being geographically close in proximity, the coastal and estuarine locations were very different in terms of blooming periods, community composition, and ecological factors. Over the years bacteria have developed resistance against antibiotics. Overuse and misuse of antibiotics is one of the main reasons of this increased bacterial resistance. A recently discovered peptide antibiotic known as teixobactin has shown potent activity against Gram-positive bacteria including methicillin-resistant Staphylococcus. aureus (MRSA), Mycobacterium tuberculosis, and vancomycin-resistant enterococci, (VRE). The structure of teixobactin consists of several uncommon amino acids including D-amino-acids and L-allo-enduracididine. L-allo-Enduracididine is a unique amino acid residue consisting of a 5-membered guanidine ring, which offers a great challenge for its synthesis. Forest-dwelling fungi are critical to ecosystem function in their roles as decomposers and nutrient cyclers, food sources, pathogens, and mycorrhizal symbionts. In northern New England, a region with a substantial timber industry, the degree to which fungal communities are resilient to timber harvest, as well as their ability to recolonize post-disturbance, may impact ecosystem function and forest regeneration. In this thesis, we investigate the impacts of (1) timber harvest on fungal fruiting, an important element of fungal reproduction, and (2) the dispersal mechanisms by which fungi may re-colonize harvested areas. GLI3 is an important protein traditionally found in the Sonic Hedgehog pathway. It is involved in numerous events that take place during embryogenesis such as the differentiation and patterning of cells in the body. Monocytes are important innate immune cells which infiltrate the site of inflammation when a pathogen invades our body. Gli3 has also been shown to be upregulated in monocytes following pathogen stimulation, suggesting Gli3 could have an important role in mediating the release of inflammatory signals known as cytokines. Salmonids play a major role in aquaculture globally and their production is expected to increase as the human population rises and wild fish stocks decline. Sea lice infestation is the largest economic burdens to salmonid net pen aquaculture with hundreds-of-millions of dollars lost due to the treatment, labor, and loss of biomass associated with these parasitic copepods. Chemotherapeutics were originally used to treat these infestations, but their effects on nontarget species resulted in them being banned in many countries. Many salmonid farmers now utilize cleanerfish technology to resolve this financial stressor. Lumpfish (Cyclopterus lumpus) are efficient delousers in salmonid aquaculture …

Most nutrients and contaminants within municipal wastewaters are removed through physical, chemical, and biological treatment methods. Per- and polyfluoroalkyl substances (PFAS) are a class of contaminants of emerging concern (CECs) that are concentrated in wastewater treatment facilities (WWTFs) from municipal, industrial, and commercial sources in communities. PFAS are poorly removed in conventional municipal wastewater treatment because these facilities are generally not designed for treatment of low-level CECs. Some PFAS leave WWTFs through aqueous effluent, while others partition to solids and are handled in sludges due to properties based on the degree of fluorination, chain length, and headgroup. Eastern oyster, Crassostrea virginica, populations have been declining steadily over the past several decades across the North American east coast. Great Bay Estuary (GBE), located in New Hampshire, is experiencing this loss and restoration efforts have been put into effect. This work addresses restoration needs by characterizing abundances of settled spat and two early stages of C. virginica, D-hinge and veliger, in GBE from June 2020 through November 2020. Abundances are compared based on the collection site and the physicochemical data recorded on each sampling date. Tomatoes are a high value crop grown worldwide. Indeterminate varieties are commonly grown in high tunnel structures throughout New England for the fresh market. Indeterminate tomato plants often suffer from a phenomenon called ‘June drop’ in which the plant’s first four to five cluster of fruit set perfectly but the subsequent two to three clusters have poor set and plant productivity drops suddenly. While cluster thinning (e.g., reducing the number of fruit allowed to mature per cluster) has been successfully shown to increase fruit size, it has generally not increased marketable yield. Biogenic volatile organic compounds (VOCs) play crucial roles in ecosystems at multiple scales, ranging from mediating soil microbial interactions to contributing to atmospheric chemistry. However, soil VOCs and how they respond to environmental change remains understudied. We aimed to assess how 2 abiotic global change drivers, soil warming and simulated nitrogen (N) deposition, impact soil VOC emissions over time in a temperate forest. We characterized the effect of warming, N deposition, and their interaction on the composition and emissions of soil VOCs during the growing season of 2 consecutive years. Controls On Buffering And Coastal Acidification In A Temperate Estuary, Christopher W. Hunt, Joseph Salisbury, Douglas Vandemark

Estuaries may be uniquely susceptible to the combined acidification pressures of atmospherically driven ocean acidification (OA), biologically driven CO2 inputs from the estuary itself, and terrestrially derived freshwater inputs. This study utilized continuous measurements of total alkalinity (TA) and the partial pressure of carbon dioxide (pCO2) from the mouth of Great Bay, a temperate northeastern U.S. estuary, to examine the potential influences of endmember mixing and biogeochemical transformation upon estuary buffering capacity (β–H). Observations were collected hourly over 28 months representing all seasons between May 2016 and December 2019. Results indicated that endmember mixing explained most of the observed variability …

Neuroscience is the field that deals with studying the biological and chemical processes underlying our brain’s operations. In this commentary, I give an overview of my experience learning the most commonly used research technique in neuroscience: electrophysiological recording. In 2021, I participated in the Research Experience and Apprenticeship Program (REAP) under the guidance of Dr. Paula Mouser of UNH’s Department of Civil and Environmental Engineering. I researched the fate and transport of several estrogenic compounds in a Connecticut wastewater treatment facility. The popularity of oral contraceptives has led to an increase in hormonal contaminants in surface water bodies. Estradiols and ethinyl estradiols, the two estrogenic compounds that I intended to study, can negatively impact the development of reproductive organs in male fish, such as reducing testes size as well as lowering sperm count. Behind The Scenes: Turtle Trapping And Connectivity Mapping For Endangered Blanding’S Turtle Conservation In Southern New Hampshire, Maeve Kelley

Freshwater turtles are experiencing dramatic population declines due to human-caused stressors. Roads, in particular, have significantly increased turtle mortality rates through vehicle collisions. The Blanding’s turtle (Emydoidea blandingii) is state endangered in New Hampshire and is listed as endangered on the International Union for Conservation of Nature (IUCN) Red List. Blanding’s turtles are especially vulnerable to population declines because they reproduce late in life and their young have low survival rates. Adult Blanding’s turtles migrate from ponds to upland habitats to nest and hatchlings must later travel across the landscape to reach ponds. These overland movements increase the risk of …
