Ecophysiology Of Metal-Reducing Aeromonas And Shewanella Strains From Great Bay Estuary, Victoria Knight

The seasonal abundance and distribution of mesophilic aeromonads at seven sites in Great Bay Estuary, NH were monitored over a 24 month period. Total heterotrophic bacteria, total and fecal coliforms, salinity, temperature, pH, total suspended solids, and chlorophyll were also measured in the samples. Aeromonads were isolated at each site throughout the year with the highest incidence and population density in the late summer to early fall. Aeromonad abundance correlated positively with the incidence of coliforms, temperature, and salinity.

Slow sand filters are an established treatment method for water with low turbidity. They usually are effective for the removal of turbidity, microorganisms (including cysts of Giardia and Cryptosporidium), and particles, but they require significant periods of time for cleaning. In the 1950's, West Hartford, CT developed a harrowing process to reduce the time and labor required for cleaning at that plant. A 1988 study observed those filters had higher removal rates for non-particulate dissolved organic carbon and UV absorbing materials, surrogates for trihalomethane formation, than did filters at two other plants cleaned by the conventional scraping method.

Nucleotide composition of a DNA molecule is a product of base substitution. Variation in nucleotide composition indicates a change in the pattern of substitution at either the level of the underlying mutational spectrum or the constraints imposed by natural selection. This work explores patterns of nucleotide usage within and between animal mitochondrial genomes and the evolutionary mechanisms that have shaped these patterns. Fourfold degenerate sites are expected to reflect the underlying mutational spectrum. Three simple measures of compositional bias, taking into account the strand-specific nature of nucleotide distribution in mtDNA, reveal considerable variation among fourfold degenerate sites of metazoan mitochondrial …

The analytical electrophoresis apparatus (AEA) is capable of generating and monitoring the electrophoretic migration of macroions. The oligonucleotide pd(A)$\sb{20}\cdot$pd(T)$\sb{20}$ was use as a model to compound to evaluate the range and validity of AEA measurements under a variety of electric fields and solvent conditions. A broad range of electric fields yield consistent, reproducible values. The charge determination from different procedures, steady state electrophoresis (SSE) and electrophoretic mobility, have not been consolidated into a consistent theory but advancements in the scope and understanding of the AEA's potential have been made. This dissertation integrates the concepts of circular causality, emergence, and hierarchical organization through an investigation of the evolutionary dimension within economic thought. The different sections of this dissertation represent complementary perspectives on this theme.

The first chapter introduces the concept of circular causality and the technique of causal diagramming. These tools are used in the second and third chapters to highlight the evolutionary dimension within the history of economic thought. Molecular Identification, Coastal Distribution And Recruitment Of Cucmaria Frondosa Larvae In The Western Gulf Of Maine, Dorothy E. Medeiros-Bergen

Understanding of the physical transport of larvae requires information on adult source populations, larval distributions, larval behavior and physical oceanography. Identification of three species (Cucumaria frondosa, Psolus fabricii and Chiridota laevis) of holothurian larvae in the western Gulf of Maine was done with a simple detection protocol, using oligonucleotide probes, for the 16S rRNA portion of the mitochondrial DNA genome. More than 2000 larvae and new recruits were analyzed. This technique revealed that of the three species present in the plankton, Cucumaria larvae dominated the samples comprising $>$90% of the larvae and 95% of new recruits during 1993. Cucumaria …

Aluminum (Al) has been suggested to be an important stress factor in forest decline due to its mobilization in soil following atmospheric deposition of acidic pollutants. A major goal of our research is to develop physiological and biochemical markers of stress in trees using cell cultures and whole plants. Needles of red spruce (Picea rubens) collected from several sites in the northeastern United States and red spruce cells grown in suspension cultures were examined for polyamine and inorganic-ion content. The calA, calC, and calD mutants of Escherichia coli are known to be sensitive to Ca2+ (R. N. Brey and B. P. Rosen, J. Bacteriol. 139:824-834, 1979). In the absence of any added stimuli for chemotaxis, both the calC and the calD mutants swam with a tumbly bias. Both the calC and the calD mutants were defective in chemotaxis as measured by computer analysis, use of swarm plates, and capillary assays. The calA mutant was only slightly defective in motility and only slightly impaired in chemotaxis. Chemotactically wild-type cells had an intra-cellular free-Ca2+ level of about 105 nM. Outdoor mesocosm experiments were used to examine the response of eelgrass communities to excess nutrient loading and reduced light that simulated coastal eutrophication. A series of replicated manipulations conducted between 1988 and 1990 demonstrated the effects of reduced available light and increased loading of nitrogen plus phosphorus on habitats dominated by eelgrass Zostera marina L. Shade and nutrients each significantly affected eelgrass growth, morphology, density, and biomass. We found no significant interactions between the effects of shade and the effects of nutrients on any plant characteristics except leaf length.
