Review Of Erosion And Sedimentation Control Programs In The Piscataqua Region, Piscataqua Region Estuaries Partnership, Ken Hickey, Cayce Dalton, Jeff Clifford
PREP Reports & Publications
The Piscataqua Region Estuaries Partnership (PREP) seeks to minimize adverse impacts to water resources associated with construction site development activities. In order to achieve this goal, PREP must understand the strengths and weaknesses of existing erosion and sedimentation control (E&SC) programs in the 52 municipalities of the PREP water shed (Figure 1-1). A detailed understanding of the existing E&SC programs will enable PREP and other stakeholders to identify and implement actions to improve E&SC programs and minimize adverse impacts. This report provides a review and assessment of existing erosion and sedimentation control programs and a set of recommendations for improving …

Studies have already detected intensification of precipitation events consistent with climate change projections. Communities may have a window of opportunity to prepare, but information sufficiently quantified and localized to support adaptation programs is sparse: published literature is typically characterized by general resilience building or regional vulnerability studies. The Fourth Assessment Report of the IPCC observed that adaptation can no longer be postponed pending the effective elimination of uncertainty. Methods must be developed that manage residual uncertainty, providing community leaders with decision-support information sufficient for implementing infrastructure adaptation programs. This study developed a local-scale and actionable protocol for maintaining historical risk …

An Enhanced Method For Sequence Walking And Paralog Mining: Topo® Vector-Ligation Pcr, Benjamin B. Orcheski, Thomas M. Davis
Biological Sciences
Background
Although technological advances allow for the economical acquisition of whole genome sequences, many organisms' genomes remain unsequenced, and fully sequenced genomes may contain gaps. Researchers reliant upon partial genomic or heterologous sequence information require methods for obtaining unknown sequences from loci of interest. Various PCR based techniques are available for sequence walking - i.e., the acquisition of unknown DNA sequence adjacent to known sequence. Many such methods require rigid, elaborate protocols and/or impose narrowly confined options in the choice of restriction enzymes for necessary genomic digests. We describe a new method, TOPO® Vector-Ligation PCR (or TVL-PCR) that innovatively …

Intellectual Growth For Undergraduate Students: Evaluation Results From An Undergraduate Research Conference, Sharyn J. Potter, Eleanor Abrams, Lisa Townson, Cameron P. Wake, Julie E. Williams
Sociology
We describe the development and evaluation of the university-wide, weeklong undergraduate research conference at the University of New Hampshire. Despite increases nationally in the number of undergraduate research conferences (URC), there has been little research examining the social and educational impact of these events on student presenters. We describe the development and evaluation of the university-wide, weeklong URC at the University of New Hampshire. A survey administered to URC participants over a four year period revealed that research culminating in a presentation at the URC was one of the more influential events students experienced during their undergraduate years and students …

Optical Brightener Study Of Spruce Creek, Town Of Kittery, Me, Fb Environmental Associates, Inc.
PREP Reports & Publications
Elevated bacteria levels in the Spruce Creek Watershed have led to shellfish bed closures in the Spruce Creek Estuary and impaired waters listings by the State of Maine. Since 2005, the Town of Kittery, Spruce Creek Association and numerous partners have assessed the local waters to attempt to determine the sources of bacteria impairment. Stormwater outfalls and residential septic systems have been identified as potential sources of impairment. To help address these concerns, the Town of Kittery received a grant in 2009 through the Piscataqua Region Estuaries Partnership to further investigate potential bacteria hotspots. Project staff worked with Federal, State …
Stewardship Plan Isinglass River Conservation Reserve, Strafford, Nh, Ellen Snyder, Dan Sperduto
Stewardship Plan Isinglass River Conservation Reserve, Strafford, Nh, Ellen Snyder, Dan Sperduto
PREP Reports & Publications
In addition to the significant frontage on the Isinglass River, the Hanson Lot supports myriad wetlands – vernal pools, beaver flowages, forested swamps, meadow marshes, and scrub-shrub wetlands. These are embedded within a hemlock – oak – beech - pine forest that was heavily logged in 1998. A plan to build a large 58- to 70-lot residential subdivision on the property was averted by the acquisition of the Hanson Lot as permanent conservation land. The Bedford property is also mostly wooded; the forest was carefully managed by previous owners and therefore retains a mix of tree species and ages. Together …
Evaluation Of Post Restoration Conditions At Little River Marsh In North Hampton, New Hampshire, David M. Burdick, Chris Peter, Rober Vincent
Evaluation Of Post Restoration Conditions At Little River Marsh In North Hampton, New Hampshire, David M. Burdick, Chris Peter, Rober Vincent
Natural Resources & the Environment
The goals of the restoration project at Little River were two-fold: 1) restore ecological function of the marsh by reestablishing regular tidal flooding; and 2) reduce the frequency and severity of local flooding to surrounding landowners. Our report documents substantial progress toward the first goal using regional monitoring protocols to assess four functional areas: hydrology, soils, vegetation and nekton. Installation of two large box culverts (each 1.8 by 3.6 m in cross-section) greatly enhanced tidal flow and freshwater drainage. The tide range increased from 0.5 to 1.4 m during spring tides and now even neap high tides flood most of …

Diarylamidines: High Potency Inhibitors Of Acid-Sensing Ion Channels, Xuanmao Chen, Liyan Qiu, Minghua Li, Stefan Grunder, Beverley A. Orser, Zhi-Gang Xiong, John F. Macdonald
Molecular, Cellular & Biomedical Sciences
Acid-sensing ion channels (ASICs) are proton-gated cation channels that are predominantly expressed in the nervous system. ASICs are involved in a number of neurological diseases such as pain, ischemic stroke and multiple sclerosis but limited tools are available to target these channels and provide probes for their physiological functions. Here we report that the anti-protozoal diarylamidines, 4′,6-diamidino-2-phenylindole (DAPI), diminazene, hydroxystilbamidine (HSB) and pentamidine potently inhibit ASIC currents in primary cultured hippocampal neurons with apparent affinities of 2.8 μM, 0.3 μM, 1.5 μM and 38 μM, respectively. These four compounds (100 μM) failed to block ENaC channels expressed in oocytes. Sub-maximal …

Testing Of Great Bay Oysters For Two Protozoan Pathogens, Douglas E. Grout
PREP Reports & Publications
Two protozoan pathogens, Haplosporidium nelsoni (MSX) and Perkinsus marinus (Dermo) are known to be present in Great Bay oysters. With funds provided by the Piscataqua Region Estuaries Partnership (PREP), the Marine Fisheries Division of New Hampshire Fish and Game Department, (NHF&G) continues to assess the presence and intensity of both disease conditions in oysters from the major beds within the Great Bay estuarine system. Histological examination of Great Bay oysters has also revealed other endoparasites.

Vegetation albedo is a critical component of the Earth's climate system, yet efforts to evaluate and improve albedo parameterizations in climate models have lagged relative to other aspects of model development. Here, we calculated growing season albedos for deciduous and evergreen forests, crops, and grasslands based on over 40 site‐years of data from the AmeriFlux network and compared them with estimates presently used in the land surface formulations of a variety of climate models. Generally, the albedo estimates used in land surface models agreed well with this data compilation. However, a variety of models using fixed seasonal estimates of albedo …

[1] We are preparing to return humans to the Moon and setting the stage for exploration to Mars and beyond. However, it is unclear if long missions outside of low-Earth orbit can be accomplished with acceptable risk. The central objective of a new modeling project, the Earth-Moon-Mars Radiation Exposure Module (EMMREM), is to develop and validate a numerical module for characterizing time-dependent radiation exposure in the Earth-Moon-Mars and interplanetary space environments. EMMREM is being designed for broad use by researchers to predict radiation exposure by integrating over almost any incident particle distribution from interplanetary space. We detail here the overall …
