Transesterification Of Waste Vegetable Oil By Thermomyces Lanuginosus Lipase Immobilized On Zeolite Zsm-5, Ye Deng

In this thesis, the effect of organic solvent-cosolvent system, temperature, methanol concentration, water content and used enzyme washing method on transesterification of waste vegetable oil catalyzed by Thermomyces lanuginosus lipase immobilized on zeolite ZSM-5 was investigated. Four organic solvents, one co-solvent, three reaction temperatures, three methanol addition methods, four different water content and two used-enzyme washing methods were assessed. Optimal conditions were obtained with n-hexane as solvent, 5 vol.% tert-butanol as co-solvent, 25°C reaction temperature, batch addition of 3 equivalent molar methanol, water content equivalent to 7.5% by weight, and used-enzyme washing method B. Activity Rhythms Expressed By Juvenile American Horseshoe Crabs, Limulus Polyphemus, Elizabeth Anne Dubofsky

The goals of this study were to determine if juvenile horseshoe crabs, Limulus polyphemus, express daily or tidal patterns of activity and how light and tidal cycles influence these patterns. When exposed to a light:dark cycle (n=24), 63% of juveniles exhibit daily patterns of locomotion and 25% of juveniles express circatidal patterns. When subsequently exposed to constant darkness, 17% express circadian rhythms of activity, 25% express a combination of circadian and circatidal patterns, and 46% express circatidal patterns of activity. An Investigation Of Two Methods For Assessing The Vertical Structure Of Forest Stands, Daniel S. Maynard

In this study we investigate the limitations of two methods for assessing forest structure: vertical point sampling with a camera and laser point quadrat sampling. Vertical point sampling with a camera is a method by which the height squared per unit area of a forest can be quickly estimated. First, we derive the bias incurred for failing to adjust for slope when implementing this sampling method, and we show that slope can generally be ignored as long as the majority of sample points occur on slopes less than 35 degrees. This feasibility study investigated the removal of an emerging organic contaminant, 1,4 dioxane, from groundwater using point-of-entry (POE) treatment techniques in response to its discovery in some small New Hampshire groundwater-based private drinking water systems. The New Hampshire Department of Environmental Services (NHDES) is evaluating future treatment options for dioxane contamination of these small, groundwater-based private systems. Treatment technologies assessed for dioxane removal included: air stripping, carbon adsorption, direct UV photolysis, and UV-peroxide (H2O2) oxidation. Small streams often present the first opportunity for dissolved greenhouse gases to exchange with the atmosphere and can be potential hot spots for evasion. In this study three streams in southeastern New Hampshire representing differing landuse were monitored for emissions of nitrous oxide, methane, and carbon dioxide. Average emission rates of N2O varied from -84 mug N m-2 day-1 to 2,561 mug N m-2 day-1 and correlated strongly with NO 3- concentration. One stream, Rum Brook, was found to be a net sink for N2O from the atmosphere. Methane emissions varied from 1.1 mg C m-2 day-1 to 21 mg …

Under this work an automated 5 inch single-cell PEM fuel cell was retrofitted with a computer controlled syringe pump and operated as a DAFC. Catalyst synthesis procedures were developed in-house for DAFC catalysts preparations. Six catalysts, PtSn/C, Pt2Sn/C, Pt 2SnRu/C, PtSnRu0.5/C, PtSn0.5Ru/C and Pd/C were prepared, and their mass composition estimated through mass balance calculations. A series of experiments for VI performance data and power output were performed in the DAFC at different temperatures (75°C, 80°C, 85°C) and 1M ethanol flow rates (0.32ml/min, 1ml/min, 2ml/min) with a fixed catalyst loading of 1mg/cm2 platinum. River temperature is an influential variable for biologic activity in arctic rivers. Regulated discharge regimes can have significant impacts on water temperature on both short and long time scales as water discharged through subterranean power stations is often a significantly different temperature than water flowing in the bypass reach. As a result, river temperatures downstream of a hydropower station's discharge point are influenced by the power station's electric production schedule and the relative temperatrures of bypass flows. This study documents soil fungal communities at the Harvard Forest Chronic Nitrogen Addition experiment, which was established in 1989 to test the effects of long-term nitrogen fertilization on ecosystem processes. Researchers at this site have observed an accumulation of soil carbon in the nitrogen fertilized plots and a decrease in fungal biomass, ligninolytic enzyme activity, and rates of litter decay. We hypothesized that decreased decomposition rates in nitrogen-fertilized plots were due to changes in the structure of the fungal community, especially Basidiomycetes, the primary decomposers of lignin in this ecosystem. Degradation of dissolved organic carbon by microbial and photolytic processes was examined along the main stem of the Lamprey River Watershed located in southeastern New Hampshire. Eight sites were chosen and sampled biweekly throughout the seasonal hydrograph. Lab incubations were employed to assess microbial degradation of dissolved organic carbon (DOC) where one set of samples was exposed to natural sunlight for a day to assess photolytic degradation. Mean biodegradable dissolved organic carbon (BDOC) throughout the study period was 5.8% with no significant variation observed between sites. Endothelial inflammation plays major roles in all phases of atherosclerotic process and both innate immunity and endothelial adhesion molecules contribute to endothelial inflammation. In this thesis, there are three main parts regarding the endothelial cell inflammation and multiple protein detection. In the first part, we applied multiple antibodies (Abs) to measure changes in expression levels of six proteins in response to inflammatory stimulation, and we observed different dynamic behaviors in them. Interdisciplinary research and education are a growing emphasis in United States institutions of higher education but relatively little is known about the doctoral students engaged in these atypical programs. The purpose of this study was to understand the socialization process of 18 students involved in a large-scale, federally funded, interdisciplinary research project focused on sustainability at one university. Using Weidman, Twale, and Stein’s framework of graduate student socialization, themes emerged related to (a) their distinctive characteristics and cultures, (b) the learning process, (c) balance, (d) uncertainty, and (d) support. Recommendations for policy and practice are included.

Harvard Forest LTER (HFR) is a two decade-strong, integrated research and educational program investigating responses of forest dynamics to natural and human disturbances and environmental changes over broad spatial and temporal scales. HFR engages >30 researchers, >200 graduate and undergraduate students, and dozens of institutions in research into fundamental and applied ecological questions of national and international relevance. Through LTER I–IV, HFR has added historical perspectives, expanded its scope to the New England region, integrated social, biological, and physical sciences, and developed education and outreach programs for K-12, undergraduate, and graduate students, along with managers, decision-makers, and media professionals.
