Pay No Attention To The Regulation Behind The Curtain: The Implications Of The Return To Title Iv (R2t4) Federal Aid Policy On Time To Degree, Apri Medina

Department of Educational Administration: Dissertations, Theses, and Student Research

Federal aid programs and their effect on student persistence, stopout, and completion have long been studied, but current literature does not fully capture the temporal nature of these programs due to insufficient methods, imprecise data, or both. Using event history methodologies, I leverage a unique level of access to data at a public four-year, research intensive university to explore how the Return to Title IV federal aid withdrawal policy, one of the most prominent yet understudied aspects of federal financial aid policies, influences time to degree. The treatment of this policy is associated with a 58.6% reduced risk (reduced conditional …

Do Lactic Acid Bacteria In Fermented Foods Persist In The Gastrointestinal Tract: An In Vitro Investigation, Chloe M. Christensen, Car Reen Kok, Robert W. Hutkins

UCARE: Research Products

It is now well established that the composition of the gastrointestinal (GI) microbiota has a profound influence on intestinal and overall health. Many contemporary diseases such as type two diabetes, IBS and obesity are associated with a dysbiotic microbiota. Therefore, researchers are interested in how diet and specific dietary components can modulate the gastrointestinal microbiota and possibly repair a dysbiotic state. One approach to improve gastrointestinal health is by consuming probiotics and prebiotics or a diet rich in fermented foods containing live microbes. Fermented foods are popular because of their enhanced preservation, safety, organoleptic, functionality, and nutritional properties. These fermented …

Zika Virus Induces Apoptosis In Retinal Pigmented Epithelial Cells, Madison Kraus, Philma Glora Muthuraj, Sathish Kumar Natarajan

UCARE: Research Products

Zika virus is a single-stranded positive sense RNA virus that attracted international attention in 2015 when an epidemic broke out in Brazil. This vector borne virus is transmitted by mosquitos and is especially concerning for pregnant women because the virus can affect the development of the fetus. One known complication is the destruction of infected retinal epithelial cells. The transduction of light is interrupted due to cell death and results in blind spots. Previous research has demonstrated that palmitoleate can reduce the observed levels of apoptosis in ZIKAV infected JEG-3 cells. We have also seen the protection of retinal pigmented …

Economic Impacts Of Food Fraud, Syed Imran Ali Meerza, Konstantinos Giannakas, Amalia Yiannaka

Cornhusker Economics

When you order seafood at a restaurant, do you get what you pay for? Scientists from Oceana, a non-profit marine conservation organization, conducted one of the largest seafood fraud investigations on 1,215 samples from 674 outlets in 21 states of the United States (U.S.) over the period 2010 to 2012.They found that there is, on average, a 33% probability that you do not actually get what you pay for. The share of mislabeled food was (a quite remarkable) 74% for seafood sold in sushi restaurants and 18% of seafood sold in gro-cery stores with mislabeled “red snapper” and “tuna” accounting …

Comparative Effects Of Technical-Grade And Formulated Chlorantraniliprole To The Survivorship And Locomotor Activity Of The Honey Bee, Apis Mellifera (L.), Jennifer R. Williams, Daniel R. Swale, Troy D. Anderson

Department of Entomology: Faculty Publications

Background: The loss of honey bee colonies is a nationally recognized problem that demands attention from both the scientific community and the beekeeping industry. One outstanding threat is the unintended exposure of these pollinators to agricultural pesticides. Anthranilic diamides, such as chlorantraniliprole, are registered for use in stone and pome fruits, vegetables, turf, and grains. There are few publicly available studies that provide an analysis of chlorantraniliprole effects on the survivorship and locomotion activity of beneficial, pollinating insects such as honey bees. The data gathered in this study provide the acute toxicity, 30-day survivorship, and locomotor activity of honey …

Research Tools And Their Uses For Determining The Thermal Inactivation Kinetics Of Salmonella In Low-Moisture Foods, Soon Kiat Lau

Department of Food Science and Technology: Dissertations, Theses, and Student Research

The reputation of low-moisture foods as safe foods has been crumbling over the past decade due to repeated involvement in foodborne illness outbreaks. Although various pasteurization technologies exist, a majority are thermal processes and have not been well-characterized for pasteurizing low-moisture foods. In addition, the nature of a low-moisture food matrix introduces various experimental complications that are not encountered in high-moisture foods. In this dissertation, the development, building instructions, and characterization of various open source tools for studying the inactivation kinetics of microorganisms in low-moisture foods are described. The first tool is the TDT Sandwich, a dry heating device for …

Effects Of Pesticide Residue Accumulation On Honey Bee (Apis Mellifera L.) Development And Implications For Hive Management, Jennifer Weisbrod

Department of Entomology: Dissertations, Theses, and Student Research

Honey bee (Apis mellifera L.) colonies face annual declines of 40% in the United States. Pesticides play a role in these losses. Pesticide residues picked up from the environment as well as beekeeper-applied compounds may accumulate within the hive. The effects of pesticide accumulation in comb on bee health mitigation of accumulation have not been well-studied. To examine pesticide exposure on health and development, chlorothalonil and the amitraz metabolite N-(2,4-dimethylphenyl) formamidine (DMPF), two pesticides found in hive products, were applied to brood comb. Queen bees were caged onto treated and control comb and egg laying and development were assessed. …

The Effects Of Temperature On Evacuation Rates And Absorption Efficiency Of Flathead Catfish, Zach Horstman Apr 2020

The Effects Of Temperature On Evacuation Rates And Absorption Efficiency Of Flathead Catfish, Zach Horstman

School of Natural Resources: Dissertations, Theses, and Student Research

Knowledge of fish gastric evacuation rates are a necessary component for both field and laboratory studies when trying to understand feeding rates, modeling energy budgets, and understanding trophic dynamics of aquatic ecosystems. Many freshwater fish encounter a broad range of environmental temperatures across life history stages, but the mechanistic link between temperature and physiological processes often remains poorly understood. We designed three recirculating aquatic systems capable of rearing Flathead Catfish Plyodictis olivaris in order to quantify gastric evacuation rates and gross energy absorption at three temperatures (17º C, 22º C and 25º C). We examined the relationship between temperature and …

Diverse Aging And Health Inequality By Race And Ethnicity, Kenneth F. Ferraro, Blakelee R. Kemp, Monica M. Williams Apr 2020

Diverse Aging And Health Inequality By Race And Ethnicity, Kenneth F. Ferraro, Blakelee R. Kemp, Monica M. Williams

Department of Sociology: Faculty Publications

Although gerontologists have long embraced the concept of heterogeneity in theories and models of aging, recent research reveals the importance of racial and ethnic diversity on life course processes leading to health inequality. This article examines research on health inequality by race and ethnicity and identifies theoretical and methodological innovations that are transforming the study of health disparities. Drawing from cumulative inequality theory, we propose greater use of life course analysis, more attention to variability within racial and ethnic groups, and better integration of environmental context into the study of accumulation processes leading to health disparities.

Development Of A Sandwich Elisa Targeting Cashew Ana O 2 And Ana O 3, Morganne Schmidt Apr 2020

Development Of A Sandwich Elisa Targeting Cashew Ana O 2 And Ana O 3, Morganne Schmidt

Department of Food Science and Technology: Dissertations, Theses, and Student Research

Cashew nut is the second leading tree nut allergy in the US. Cross-contact of cashew nut poses potential food safety risks for individuals with cashew allergies. Highly-processed foods, such as HTST/UHT cashew milks may lead to problems in cashew protein detection by current allergen detection methods. Therefore, the aim of this study was to develop a robust sandwich ELISA for detection of highly processed cashew proteins. Commercial cashew ELISAs were evaluated for their robustness and sensitivity in detecting cashew milk cashew protein. After unreliable results were determined, cashew Ana o 2 (11S) and Ana o 3 (2S) were semi-purified using …

Dowdy, Shannon Keating, Alan R. Lemmon, Emily M. Lemmon, William E. Conner, Clare H. Scott Chialvo, Susan Weller, Rebecca B. Simmons, Melissa S. Sisson, Jennifer M. Zaspel

Department of Entomology: Faculty Publications

Anchored hybrid enrichment (AHE) has emerged as a powerful tool for uncovering the evolutionary relationships within many taxonomic groups. AHE probe sets have been developed for a variety of insect groups, though none have yet been shown to be capable of simultaneously resolving deep and very shallow (e.g., intraspecific) divergences. In this study, we present NOC1, a new AHE probe set (730 loci) for Lepidoptera specialized for tiger moths and assess its ability to deliver phylogenetic utility at all taxonomic levels. We test the NOC1 probe set with 142 individuals from 116 species sampled from all the major lineages of …

The Role Of Conformational Dynamics In Isocyanide Hydratase Catalysis, Medhanjali Dasgupta Apr 2020

The Role Of Conformational Dynamics In Isocyanide Hydratase Catalysis, Medhanjali Dasgupta

Department of Biochemistry: Dissertations, Theses, and Student Research

Post-translational modification of cysteine residues can regulate protein function and is essential for catalysis by cysteine-dependent enzymes. Covalent modifications neutralize charge on the reactive cysteine thiolate anion and thus alter the active site electrostatic environment. Although a vast number of enzymes rely on cysteine modification for function, precisely how altered structural and electrostatic states of cysteine affect protein dynamics, which in turn, affects catalysis, remains poorly understood.

Here we use X-ray crystallography, computer simulations, site directed mutagenesis and enzyme kinetics to characterize how covalent modification of the active site cysteine residue in the enzyme, isocyanide hydratase (ICH), affects the protein …

School of Veterinary and Biomedical Sciences: Faculty Publications

For more than 30 years, lipid droplets (LDs) were considered as an inert bag of lipid for storage of energy-rich fat molecules. Following a paradigm shift almost a decade ago, LDs are presently considered an active subcellular organelle especially designed for assembling, storing and subsequently supplying lipids for generating energy and membrane synthesis (and in the case of hepatocytes for VLDL secretion). LDs also play a central role in many other cellular functions such as viral assembly and protein degradation. Here, we have explored the structural and functional changes that occur in hepatic and adipose tissue LDs following chronic ethanol …

Honeybee Microbiome Is Stabilized In The Presence Of Propolis, Perot Saelao, Renata S. Borba, Vincent Ricigliano, Marla Spivak, Michael Simone-Finstrom

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Honeybees have developed many unique mechanisms to help ensure the proper maintenance of homeostasis within the hive. One method includes the collection of chemically complex plant resins combined with wax to form propolis, which is deposited throughout the hive. Propolis is believed to play a significant role in reducing disease load in the colony due to its antimicrobial and antiseptic properties. However, little is known about how propolis may interact with bee-associated microbial symbionts, and if propolis alters microbial community structure. In this study, we found that propolis appears to maintain a stable microbial community composition and reduce the overall …
