Tritrichomonas foetus (TF) is a significant reproductive pathogen of cattle, and sample collection, handling, transport, and testing are significant hurdles to surveillance programs. Recent methods have been developed that allow for the direct detection of TF using a reverse transcription real-time PCR (direct RT-qPCR) approach. To evaluate these methods, a comparative analysis was conducted to assess the technical performance of this assay with a commercially available real- time PCR (qPCR) assay. The serine palmitoyltransferase (SPT) complex catalyzes the first and rate-limiting step in sphingolipid biosynthesis in all eukaryotes. ORM/ORMDL proteins are negative regulators of SPT that respond to cellular sphingolipid levels. However, the molecular basis underlying ORM/ORMDL-dependent homeostatic regulation of SPT is not well understood.We determined the cryo–electron microscopy structure of Arabidopsis SPT-ORM1 complex, composed of LCB1, LCB2a, SPTssa, and ORM1, in an inhibited state. A ceramide molecule is sandwiched between ORM1 and LCB2a in the cytosolic membrane leaflet. Ceramide binding is critical for the ORM1-dependent SPT repression, and dihydroceramides and phytoceramides differentially affect this repression. A hybrid β sheet, formed …

Chlamydia trachomatis (CT) causes the most prevalent sexually transmitted bacterial disease in the United States. The lack of drug selectivity is one of the main challenges of the current antichlamydial pharmacotherapy. The metabolic needs of CT are controlled, among others, by cylindrical proteases and their chaperones (e.g., ClpX). It has been shown that dihydrothiazepines can disrupt CT-ClpXP. Based on this precedent, we synthesized a dihydrothiazepine library and characterized its antichlamydial activity using a modified semi-high-throughput screening assay. Invasive species are suspected to be major contributors to biodiversity declines worldwide. Counterintuitively, however, invasive species effects are likely scale dependent and are hypothesized to be positively related to biodiversity at large spatial scales. Past studies investigating the effect of invasion on biodiversity have been mostly conducted at small scales (<100 m2) that cannot represent large dynamic landscapes by design. Therefore, replicated experimental evidence supporting a negative effect of invasive plants on biodiversity is lacking across many landscape types, including large grasslands.

The (simw501);Ore genotype is a strain of Drosophila melanogaster that has impaired mitochondrial function as a result of a disrupted interaction between a mitochondrially-encoded tRNATyr and the nuclear-encoded synthetase that aminoacylates mt-tRNATyr. We hypothesized that disruption of mitochondrial protein synthesis and oxidative phosphorylation (OXPHOS) in this genotype decreases mitochondrial translation and results in the sterility of (simw501);OreR males at 28oC. The results of Mitotracker staining showcased the reduced Mean Fluorescence Intensity (MFI), which confirms that sterility has an energetic basis, yet also that mature sperm function is compromised. Attention deficit hyperactivity disorder (ADHD) can provide many challenges for college students due to difficulties focusing, multi-tasking, and staying organized. Colleges are required to offer academic accommodations to students with disabilities in an attempt to provide students with an equitable learning environment. However, accommodations may be ineffective if they are highly unattainable, unused by the student once granted, or hindered by university faculty.

BACKGROUND: Stable flies [Stomoxys calcitrans (L.)] are economically important pests of cattle and other livestock. As an alternative to conventional insecticides, we tested a push–pull management strategy using a coconut oil fatty acid repellent formulation and an attractant-added stable fly trap.
RESULTS: In our field trials we found that weekly applications of a push–pull strategy can reduce stable fly populations on cattle as well as a standard insecticide (permethrin). We also found that the efficacy periods of the push–pull and permethrin treatments following on-animal application were equivalent. Traps with an attractant lure used as the pull component of the …

Climatic data from different years and experiments conducted in Nebraska were used to estimate four comfort thermal indices and to predict the risk of heat stress and its relationship with pen surface temperature (PST). These included the temperature–humidity index (THI), the adjusted THI (THIadj), the heat load index (HLI), and THIPST using pen surface temperature instead of air temperature. Respiration rates (RR), tympanic temperatures (TT), and panting scores (PS) were also collected in each year and from each location. NUTR/GLST 498b: Global Research Experiences in Nutrition and Health
Introduction: Zambia is a multilingual country that uses 8 different languages for instruction including English and 7 other indigenous languages.
Methods: Survey research conducted between May-June 2022 on 6-7th graders within 9 Zambian schools. Classroom observations made and teachers interviewed.
Results: In Eastern and Southern Provinces, Zambian teachers speak an average of 5 languages while students speak an average of 2. Both teachers and students say English remains the most important language followed by first languages.
Conclusion: Continued research on language-in-education policies and impacts on student performance must be conducted if “One Zambia, One Nation” is meant to promote all …

Sequencing-based genetic association analysis is typically performed by first generating genotype calls from sequence data and then performing association tests on the called genotypes. Standard approaches require accurate genotype calling (GC), which can be achieved either with high sequencing depth (typically available in a small number of individuals) or via computationally intensive multi-sample linkage disequilibrium (LD)-aware methods. This study aimed to evaluate the use of peroxyacetic acid (PAA), buffered vinegar (BV), and cultured dextrose fermentate (CDF) to reduce Salmonella on artificially inoculated raw chicken livers, one of the most consumed offal around the world. Samples were inoculated with a 5-strain cocktail of poultry-borne Salmonella to obtain 106 CFU/g and immersed for 90 s with agitation in one of the following treatments: distilled water (control), 450 ppm PAA, 2.0% (w/v) BV, or 1.5% (w/v) CDF, prior to storing at 4oC. The first data update of the atlas of ticks in Germany published in 2021 is presented here. This atlas provides maps based on georeferenced tick locations of 21 species endemic in Germany as well as three tick species that are regularly imported to Germany. Water quality governance encompasses multiple “wicked” interacting problems that manifest within social-ecological systems. Concerned governments, institutions, and actors concerned with addressing these issues must wrestle with complex systems that span time, space, and scale. This complexity of connected systems requires the participation of multiple actors across political boundaries, problem areas, and hydrologic domains. In Lake Champlain (US), frequent cyanobacteria blooms negatively affect property values, recreational activities, and public infrastructure, in addition to their impacts on the aquatic ecosystem. Higher education culture is steeped in institutional identity and ties to history. But what happens when that history is challenged, and an institution must change its name? While a merger was not initially intended for Kearney State College, merging into the University of Nebraska system was the only way to reflect the change and growth that emulated the type of institution it had become. Despite a growing focus on access to and use of emergent literacy assessment in early childhood, little is known about early childhood teachers’ data practices and their associations with children’s emergent literacy skills. A questionnaire was used to confirm and elaborate findings from prior qualitative work (Schachter & Piasta, 2022) investigating U.S. teachers’ emergent literacy data practices. We focused on how teachers gathered data (data gathering), what they learned from those data (data knowledge), and how they used those data in their practice (data use) along with associations between the practices and children’s emergent literacy skills. The market value of agricultural land in Nebraska increased by 14% over the prior year to an average of US $3,835 per acre, according to the 2023 Nebraska Farm Real Estate Market Survey. This marks the second-largest increase in the market value of agricultural land in Nebraska since 2014 and the highest non-inflation-adjusted state-wide land value in the 45-year history of the survey.

The consumer demand for fresh produce (vegetables and fruits) has considerably increased since the 1980s for more nutritious foods and healthier life practices, particularly in developed countries. Currently, several foodborne outbreaks have been linked to fresh produce. The global rise in fresh produce associated with human infections may be due to the use of wastewater or any contaminated water for the cultivation of fruits and vegetables, the firm attachment of the foodborne pathogens on the plant surface, and the internalization of these agents deep inside the tissue of the plant, poor disinfection practices and human consumption of raw fresh produce. …

Some Pseudomonas species are common meat spoilage bacteria that are often associated with the spoilage of fresh meat. The recently reported ability of these bacteria to also spoil cooked and vacuum packaged meat products has created the need to investigate all potential routes of spoilage they may be able to utilize. The objective of this experiment was to determine if spoilage Pseudomonas spp. survive thermal processing and grow during refrigerated storage under vacuum. Heavy metals are commonplace in nature, and are typically found in scarce quantities in all drinking sources. However, in high enough concentrations, they can have a myriad of negative health effects. This study seeks to find the concentrations of such metals in the Lincoln water supply utilizing the resources of the Water Center at the University of Nebraska at Lincoln. To test for discrete concentrations of trace elements, a method of using Inductively Coupled Plasma Mass Spectrometry (ICP-MS) was implemented. Within exercise physiology, the study of factors potentially mediating interorgan crosstalk during and after exercise is a fascinating field of research. As exercise activates a plethora of metabolic pathways in several tissues, organs and systems, examining the underlying biological mechanisms contributing to exercise related metabolic benefits is imperative. Since two decades, the skeletal muscle is known to secrete humoral factors into the circulation in response to exercise, originally described as “myokines” by Pedersen et al. (2003). Climate forecasts suggest the Great Plains of North America have increased risk of droughts during global warming. Environmental factors have potential to influence turtle populations in aquatic habitats through temperature-dependent sex determination and influences on food availability. Long-term studies are critical to evaluate the influence of climatic variation on turtles. We used a 12-year set of mark-recapture data collected from painted turtles (Chrysemys picta, n = 162) in a pond in Keith County, Nebraska during 2005–2016 to assess variation in sex ratio and growth dynamics. Forward masking is generally greater for Gaussian noise (GN) than for low-fluctuation noise maskers, i.e., GN disruption. Because the minimal hearing loss that is associated with older age may affect GN disruption differently than more significant hearing loss, the current study explored the contribution of minimal hearing loss associated with older age to GN disruption. GN disruption was measured using three masker-signal delays (25, 75, and 150 ms) for three adult groups: younger participants with normal hearing (NH), older participants with minimal hearing loss, and older participants with sensorineural hearing loss. Immigrants receive mental health services less than their U.S.-born counterparts, even after accounting for the lower rate of mental health difficulties often found among these populations. Stigma is one such barrier that is significant in reducing mental health service utilization among immigrant populations. This study explores how immigrants perceive stigma as a barrier to seeking help for trauma-related mental health concerns. Using qualitative data from a study on an adaptation of Written Exposure Therapy in Latinx immigrants (Andrews et. al, 2022), I examined the role of stigma in participants’ perceived barriers to care and their proposed strategies for reducing them. …

Introduction: Intellectual disability (ID) is a significant limitation in both intellectual ability and adaptive functioning, but many studies of participants with ID only include a measure of overall intellectual functioning when describing their samples. The purpose of this perspective article was to provide a starting point for future research regarding the utility of including measures of both intellectual and adaptive functioning in research focused on ID. In this article, we discuss the differences and similarities between the constructs of intellectual and adaptive functioning, how they are measured, and the benefits of using both measures to describe participant abilities.
