The Future Of U.S. Agricultural Biotechnology And Trade: Summary Of A Roundtable Discussion Oct 2023

The Future Of U.S. Agricultural Biotechnology And Trade: Summary Of A Roundtable Discussion

Yeutter Institute Publications

The United States is one of the largest and highest-yield agricultural producers and exporters in the world. But the world’s population is growing, and growing conditions themselves are changing. Farms face shifting weather patterns, more frequent and severe storms, floods, and drought, and changing pest and disease pressures. What farmers plant and how they plant it are evolving to keep pace with these changes. Future global food security, sustainability, and U.S. leadership in agriculture will depend on the domestic and international policy and regulatory landscapes for agricultural biotechnology innovation. The Clayton Yeutter Institute of International Trade and Finance at the …

Optimizing Crop Productivity And Fertility Practices In Intermediate Wheatgrass, Roberta Bianchin Rebesquini Oct 2023

Optimizing Crop Productivity And Fertility Practices In Intermediate Wheatgrass, Roberta Bianchin Rebesquini

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

The intensive management associated with many annual crops often includes recurring tillage, fertilization, and pesticide applications, which contribute to environmental concerns such as water pollution and soil erosion. Intermediate wheatgrass (Thinopyrum intermedium), recognized under the trade name Kernza®, is a perennial grass that can be managed to produce grain and biomass while providing desired environmental benefits such as soil conservation and nutrient cycling. There has been limited research on best management practices and crop productivity for this alternative dual-use crop in Nebraska. A field experiment was conducted beginning in 2021 to assess nitrogen (N), phosphorus (P), and potassium …

Waterfowl infected with avian influenza A viruses (IAVs) shed infectious virus into aquatic environments, providing a mechanism for transmission among waterfowl, while also exposing the entire aquatic ecosystem to the virus. Aquatic invertebrates such as freshwater snails are likely exposed to IAVs in the water column and sediment. Freshwater snails comprise a significant portion of some waterfowl species’ diets, so this trophic interaction may serve as a novel route of IAV transmission. In these experiments, tadpole snails (Physa spp.) were exposed to a low-pathogenicity IAV (H3N8) to determine whether snails can accumulate the virus and, if so, how long virus …

Wetlands are responsible for 20%–31% of global methane (CH4) emissions and account for a large source of uncertainty in the global CH4 budget. Data-driven upscaling of CH4 fluxes from eddy covariance measurements can provide new and independent bottom-up estimates of wetland CH4 emissions. Here, we develop a six-predictor random forest upscaling model (UpCH4), trained on 119 site-years of eddy covariance CH4 flux data from 43 freshwater wetland sites in the FLUXNET-CH4 Community Product. Network patterns in site-level annual means and mean seasonal cycles of CH4 fluxes were reproduced accurately in tundra, boreal, and temperate regions (Nash-Sutcliffe Efficiency ∼0.52–0.63 and 0.53). …

Highly pathogenic avian influenza viruses (HPAIVs) of the A/goose/Guangdong/1/1996 lineage H5 clade 2.3.4.4b continue to have a devastating effect on domestic and wild birds. Full genome sequence analyses using 1369 H5N1 HPAIVs detected in the United States (U.S.) in wild birds, commercial poultry, and backyard flocks from December 2021 to April 2022, showed three phylogenetically distinct H5N1 virus introductions in the U.S. by wild birds. Unreassorted Eurasian genotypes A1 and A2 entered the Northeast Atlantic states, whereas a genetically distinct A3 genotype was detected in Alaska. The A1 genotype spread westward via wild bird migration and reassorted with North American …

Beneath The Surface: An Investigation Into The Relation Between Power, Dehumanization, And Objectification In And Initial Social Interaction, Lillian Hefner Oct 2023

Beneath The Surface: An Investigation Into The Relation Between Power, Dehumanization, And Objectification In And Initial Social Interaction, Lillian Hefner

Honors Program: Senior Projects (Public)

Objectification theory suggests that women are disproportionately affected by objectification leading them to experience more negative health outcomes such as depression and eating disorders. Further research on objectification and synthesis of leading theories in the area suggest that power may be one factor likely to predict the objectification and dehumanization of women. One important dimension of this objectification and dehumanization is the environment in which it occurs. Few studies examine a social/dating context as the current study does. We expected the men in the study who felt a stronger sense of power during the interaction would exhibit more objectification of …

Existing Evidence For Influenza B Virus Adaptations To Drive Replication In Humans As The Primary Host, Matthew J. Pekarek, Eric A. Weaver Sep 2023

Existing Evidence For Influenza B Virus Adaptations To Drive Replication In Humans As The Primary Host, Matthew J. Pekarek, Eric A. Weaver

Nebraska Center for Virology: Faculty Publications

Influenza B virus (IBV) is one of the two major types of influenza viruses that circulate each year. Unlike influenza A viruses, IBV does not harbor pandemic potential due to its lack of historical circulation in non-human hosts. Many studies and reviews have highlighted important factors for host determination of influenza A viruses. However, much less is known about the factors driving IBV replication in humans. We hypothesize that similar factors influence the host restriction of IBV. Here, we compile and review the current understanding of host factors crucial for the various stages of the IBV viral replication cycle. While …

Did Cover Crop Or Animal Manure Ameliorate Corn Residue Removal Effects On Soil Mechanical Properties After 10 Years?, Hans W. Klopp, Humberto Blanco-Canqui, Michael Sindelar, Virginia L. Jin, Marty R. Schmer, Richard B. Ferguson

Department of Agronomy and Horticulture: Faculty Publications

Crop residue removal may negatively affect soil mechanical properties, which are key components of soil quality. To evaluate potential long-term effects, we assessed the 10-yr impact of corn (Zea mays L.) residue removal (59 % of non-grain biomass annually) on surface soil mechanical properties (0–20 cm). We also evaluated whether adding carbon (C) amendments, such as using a winter rye (Secale cereale L.) cover crop or surface-applying cattle manure (24 Mg ha− 1 biannually) can ameliorate the effects of crop residue removal. This long-term study was under irrigated no-till continuous corn on a silt loam soil in south-central …

Although several reports have hypothesized that exercise may increase skeletal muscle protein lactylation, empirical evidence in humans is lacking. Thus, we adopted a multifaceted approach to examine if acute and subchronic resistance training (RT) altered skeletal muscle protein lactylation levels. In mice, we also sought to examine if surgical ablation-induced plantaris hypertrophy coincided with increases in muscle protein lactylation. To examine acute responses, participants’ blood lactate concentrations were assessed before, during, and after eight sets of an exhaustive lower body RT bout (n = 10 trained college-aged men). Vastus lateralis biopsies were also taken before, 3-h post, and 6-h post-exercise …

Background Macrophages are highly plastic innate immune cells that play key roles in host defense, tissue repair, and homeostasis maintenance. In response to divergent stimuli, macrophages rapidly alter their functions and manifest a wide polarization spectrum with two extremes: M1 or classical activation and M2 or alternative activation. Extracellular vesicles (EVs) secreted from differentially activated macrophages have been shown to have diverse functions, which are primarily attributed to their microRNA cargos. The role of protein cargos in these EVs remains largely unexplored. Therefore, in this study, we focused on the protein cargos in macrophage-derived EVs.

Myocarditis is a predominant cause of congestive heart failure and sudden death in children and young adolescents that can lead to dilated cardiomyopathy. Lymphocytic myocarditis mediated by T cells can result from the recognition of cardiac antigens that may involve CD4 or CD8 T cells or both. In this report, we describe the generation of T cell receptor (TCR) transgenic mice on a C57BL/6 genetic background specific to cardiac myosin heavy chain (Myhc)-α 334–352 and make the following observations: First, we verified that Myhc-α 334–352 was immunogenic in wild-type C57BL/6 mice and induced antigen-specific CD4 T cell responses despite being …

A reliable and simple Agrobacterium-mediated transformation system for the unicellular green algae model organism Chlamydomonas reinhardtii has been developed. The protocol has been successfully employed with both neomycin phosphotransferase II (nptII) and the phleomycin resistance (bleI) genes coupled with the selective agents paromomycin and zeocin, respectively. A set of binary vectors were assembled that carry the selectable marker cassettes under control either of the Rbcs2 alone or fused to the HSP270A leader sequence, PsaD, or ß-tubulin2 promoters. The corresponding T-DNA elements also harbored a cassette with a codon-optimized version of yellow fluorescence protein (YFP) under …

Centromeres are epigenetically specified by the histone H3 variant CENP-A. Although mammalian centromeres are typically associated with satellite DNA, we previously demonstrated that the centromere of horse chromosome 11 (ECA11) is completely devoid of satellite DNA. We also showed that the localization of its CENP-A binding domain is not fixed but slides within an about 500 kb region in different individuals, giving rise to positional alleles. These epialleles are inherited as Mendelian traits but their position can move in one generation. It is still unknown whether centromere sliding occurs during meiosis or during development. Here, we first improve the sequence …

Single activation of peroxymonosulfate (PMS) in a homogeneous system is sometimes insufficient for producing reactive oxygen species (ROS) for water treatment applications. In this work, manganese spinel ferrite and graphitic carbon nitride (MnFe2O4/g-C3N4; MnF) were successfully used as an activator for PMS under visible light irradiation to remove the four-mostdetected- hormone-contaminated water under different environmental conditions. The incorporation of g-C3N4 in the nanocomposites led to material enhancements, including increased crystallinity, reduced particle agglomeration, amplified magnetism, improved recyclability, and increased active surface area, thereby facilitating the PMS activation and electron …

Allostatic Load Index Effectively Measures Chronic Stress Status In Zoo-Housed Giraffes, Haley N. Beer, Lisa K. Karr, Trenton C. Schrader, Dustin T. Yates

Department of Animal Science: Faculty Publications

For giraffes, few standardized methods exist for quantifying chronic stress. Allostatic load index is quantified from a panel of multi-system stress biomarkers to estimate cumulative stress. Our objective was to determine whether a panel of biomarkers selected for their role in allostatic load would reflect the number of documented stress events experienced by giraffes. Cortisol, DHEA-S, cholesterol, non-esterified fatty acids (NEFA), and fructosamine were determined in serum samples from zoo-housed giraffes (n = 18). These were correlated with the overall number and frequency of ZIMS-documented stress events experienced prior to blood collection. We also compared giraffes grouped by high vs. …

Zea E-Books Collection

For 40 years Wayne Mollhoff conducted a personal bird census every January. He explains: "After having run several Breeding Bird Survey routes, and participated in several Christmas Bird Counts, I became curious to see what might be found on a winter count under the more tightly controlled parameters of a census, as contrasted with Christmas counts done with variable numbers of observers."

The count was set up similarly to the USGS Breeding Bird Survey routes with 50 stops, one-half mile (800 meters) apart, all birds counted for 3 minutes, with birds counted at one stop not counted again at following …

Broken Tails In Holstein Dairy Cattle: A Cross-Sectional Study, Hannah E. Olsen, Karly N. Anderson, Katherine C. Creutzinger, Kurt D. Vogel

Department of Animal Science: Faculty Publications

Dairy cows are regularly handled when moved to the milking parlor and during other routine procedures. Low-stress handling methods are important in avoiding negative welfare states for dairy cattle. Tail twisting is used by some handlers to prompt cattle movement. However, when used inappropriately with excessive force, tail twisting can lead to a broken tail. The aim of this cross-sectional study was to determine cow-level factors that may be associated with the prevalence of broken tails in dairy cattle. A subset of 229 Holstein dairy cows (68 primiparous and 161 multiparous) at a single dairy were assessed for broken tails …

Preclinical studies have shown that chronic alcohol abuse leads to alterations in the gastrointestinal microbiota that are associated with behavior changes, physiological alterations, and immunological effects. However, such studies have been limited in their ability to evaluate the direct effects of alcohol-associated dysbiosis. To address this, we developed a humanized alcoholmicrobiota mouse model to systematically evaluate the immunological effects of chronic alcohol abuse mediated by intestinal dysbiosis. Germ-free mice were colonized with human fecal microbiota from individuals with high and low Alcohol Use Disorders Identification Test (AUDIT) scores and bred to produce human alcohol-associated microbiota or human control-microbiota F1 progenies. …

Emerging infectious disease (EID) represents an existential threat to humanity. EIDs are increasing in frequency and impact because of climate change and other human activities. We are losing the battle against EIDs because of improper assessment of the risk of EID. This stems from adherence to a failed paradigm of pathogen-host associations that suggests EIDs ought to be both unpredictable and rare. That, in turn, leads to policies suggesting that crisis response is the best we can do. Real-time and phylogenetic assessments show EIDs to be neither rare nor unpredictable—this is the parasite paradox that shows the failures of the …

Background: Intrauterine growth restriction (IUGR) is associated with reduced β2 adrenergic sensitivity, which contributes to poor postnatal muscle growth. The objective of this study was to determine if stimulating β2 adrenergic activity postnatal would rescue deficits in muscle growth, body composition, and indicators of metabolic homeostasis in IUGR offspring.

Methods: Time-mated ewes were housed at 40°C from day 40 to 95 of gestation to produce IUGR lambs. From birth, IUGR lambs received daily IM injections of 0.8 μg/kg clenbuterol HCl (IUGR+CLEN; n = 11) or saline placebo (IUGR; n = …

Species distribution models often suggest strong links between climate and species' distribution boundaries and project large distribution shifts in response to climate change. However, attributing distribution shifts to climate change requires more than correlative models. One idea is to examine correlates of the processes that cause distribution shifts, namely colonization and local extinction, by using dynamic occupancy models. The Cape Rock-jumper (Chaetops frenatus) has disappeared over most of its distribution where temperatures are the highest. We used dynamic occupancy models to analyse Cape Rock-jumper distribution with respect to climate (mean temperature and precipitation over the warmest annual quarter), …
